A proposal of gene transfer scheme from diploid to hexaploid in oats was described. The main idea of this scheme are (1) use Avena magna which has two genomes partially in common with two genomes of the hexaploid Avena sativa or a common genome and the rest genomes partially commonn, and which lead to more regular pairing between them rather than AABB genome type to get 6x-amphiploid as a bridge between ploidy level. Cross between Avena strigosa and Avena magna is compatible and further give 42% seed set, (2) extract tetraploid derivatives which have in corporated desired genes from Avena strigasa to Avena magna, (3) Synthetic petaploid provide 2n=21 chromosome number in female gametes, which lead to complete pairing or nearly so in progenies with Avena sativa, (4) eventually homozygous lines will be produced by selfing the heterozygous (regarding to$A^{As}$ genome) at final step.
The multiple ear and tiller line (MET), which was selected from a local maize collection and has been selected for earliness by selfing for five generations, was compared its plant characteristics with four U.S. leading hybrids. 1. The general plant growth was a little retarded by poor soil fertility and structure. 2. The fresh weight of the MET line was about 97.8% of the U.S. check varieties. 3. The dry weight of the MET line was also about 94.5% of the check U.S. hybrids (1,695 kg / 10a.). 4. The tillering and eating habits of the MET line had contributed toward increasing fresh and dry weight of the MET line. 5. The average plant height of the MET line was much less than those of the check varieties, due to the selfing depression manifested by the MET line, (234.7cm. of the MET vs 250.7cm. of the check) 6. Apparent differences of disease resistance was observed among varieties studied. The degree of plants infected with the black stripe mosaic virus was over 20% for the check U.S. hybrids, while less than 5% of the MET line was infected with the disease. 7. The grain yield per 10 a of the MET line was far below the check hybrids due to the inbreeding depression of the MET line. However, the kernel number per unit area of the MET line with small size kernel was much greater than that of the check hybrids. The 100 kernel weight of the MET line was around 10 grams. 8. The average fresh and dry matter weight of the three ($sorghum{\times}sudan$ grass) varieties was very much alike with those of the MET line. 9. In conclusion, the use of the MET line for silage production may have dual adventages because of its low seed price and of its small size kernels, which may reduce the amount of seeds required for planting in an unit area.
Two strains which have good traits in quality and speedy growing were selected based on previous report(Ryu, 2007) to breed a new strain carrying the traits in it. KNR2322 carrying high quality and KNR2503 carrying a speedy growing trait were cultivated to harvest monokaryons. The monokaryotic mycelia were characterized in growth rate, color, and stability. Selfing of monokaryons from KNR2322 and crossing between KNR2322- and KNR2503-derived monokaryons were performed. Two parental lines, $KNR2322-4{\times}37-6{\times}12(B)$ showing thremmatological values in weight and quality, 68.5g and 6.5 and $KNR2322-30{\times}KNR2503-60(A)$ was harvested in 13.5 days after scraping old medium were selected. Crossing between A and B resulted in two high quality and speedy growing strain, A8B8 and A8B10. Their days for harvest after scraping, weight, and quality were 14.0 days, 88.8g and 92.7g, 7.0 and 7.1. Originality tests were conducted with genomic DNAs of A8B8 and Keuneutari 3ho(commercial strain) by DNA fingerprinting a confrontation culture. The results showed polymorphism and a inhibition line between them. It suggested that the new strain have originality from the commercial strain. A8B8 have been registered in Korea Seed and Variety Service with as "Saesongi 1ho".
This study was carried out to investigate the tissue-specific expression of ${\beta}$-glucuronidase (gus) gene driven by endosperm-specific promoter (Z4 promoter) in the transgenic tobacco and to find out inheritance pattern of transgene to the next generation. Tobacco (Nicotiana tabaccum cv. Havana SR1) was transformed with Agrobacterium tumerfaciens LBA4404 harboring BV3 construct containing gus gene driven by Z4 promoter and a kanamycin resistant gene. Seven hundred bp PCR products, indicating the presence of npt II gene, were found in the all eight transformants by PCR analysis using nptII primers. To study the expression pattern of the two different kind of promoters, leaf disks of the Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. As a result, leaf disks of Z4pro-gus-transformed plants showed very weak and partial positive gus activity. In contrast, leaf disks of 35Spro-gus-transformed plants showed relatively strong positive gus activity. To investigate the expressed position of Z4 promoter, seeds from Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. Z4pro-gus-transformed seeds showed positive gus activity restricted to the endosperm. However, the blue-colored product in 35Spro-gus-transformed seeds was observed in all the area including endosperm. Kanamycin resistance assay showed that transgenes were stably inherited to next generation in all lines.
This study was conducted to obtain basic information on cross breeding for developing bolting resistant variety of Angelica gigas NAKAI which has been subjected to severe yield reduction resulted from flower-stalk initiation during the growth. Compatibility of selfing and/or crossing pollination among 5 local cultivars were tested. Results obtained are summarized as follows. In case of three year old plant, all cultivars emerged March 14 and bolted from May 12 to May 13. Ponghwa cultivar was flowered 3 to 5 days earlier than other cultivars. Flower duration continued 41 to 43 days. Interval of flowering date from main stem to first primary branch, from first to second or second to third primary branch was 9 to 13 days. Emergency of stamen proceeded 4~8 days of pistil initiation demonstrating that Angelica gigas NAKAI is protandrous plant. Furthermore initiation of first pistil was 1~4days later than formation of last stamen in the same flower cluster. Percent pollination of Angelica gigas NAKAI was 52% under natural condition, so called open-pollination, but bagging by enveloping flowers was as low as 0.4~2.4%. In the result of compatibility test, all local cultivars tend to show both of self-and cross-compatibility. Inpollination, ‘Muju’$\times$‘Muju’combination showed highest fertility up to 40.8% whereases cross pollination, ‘Pyeongchang’$\times$‘Inje’combination, showed 67.0% fertility.
Fourteen Acer pseudosieboldianum populations in South Korea were used to estimate genetic diversity, genetic differentiation and genetic relationships using seven AFLP primer combinations. The average of effective alleles ($A_e$), the proportion of polymorphic loci (%P) and Shannon's diversity index (I) was 1.4, 82.2% and 0.358, respectively. The expected heterozygosity ($H_e$) under Hardy-Weinberg equilibrium was 0.231 and the expected heterozygosity (Hj) from Bayesian inference was 0.253. The level of genetic diversity was moderate compared to those of Genus Acer and lower than those of other species having similar ecological niche and life history. The inbreeding coefficient within populations ($F_{IS}$) from Bayesian method was 0.712 and it could be influenced by selfing or biparental inbreeding to induce homozygote excess. The level of genetic differentiation was 0.107 from AMOVA (${\Phi}_{ST}$) and 0.110 from Bayesian method (${\Phi}^{II}$). The genetic differentiation was lower than those of other species having similar ecological niche and life history. Ulleungdo population had the lowest level of genetic diversity and was genetically the most distinct population from others in the study. We consider that founder effect and genetic drift might be occurred to reduce genetic diversity and then the geographical isolation might interrupt gene flow to aggravate it.

'Baekjung' adaptable to high temperature was made by crossing between monokaryon derived from selfing of brown strain and monokaryon derived from Korea white strain. In the condition that temperature is maintained at $10^{\circ}C$ without low temperature of $4^{\circ}C$ suppressing treatment and wrapping during cultivation period, it showed good productivity than Uri1ho(control). The optimum temperature of mycelial growth was $30^{\circ}C$ and that of fruiting body initiation and development were $14^{\circ}C$ and $7^{\circ}C$, respectively. The days for the fruiting and yield were 7 days and $277{\pm}11.2$ g per 1,100 ml bottle, respectively. This variety needed high concentration of carbon dioxide up to 4,000 ppm for the good quality.

A total of eleven Chinese cabbage accessions were used for microspore culture and were grown to take basal data. Based on the collected data, breeding materials were chosen to develop new improved Chinese cabbage cultivars. The range of microspore-derived embryoid taken from flower buds was 1.6 to 35.4 embryoids. The embryoids from IT26110 and IT26153 among the Chinese cabbages were more than 34 per flower bud. The viability rate after cold treatment was low from 0.2 to 11.7%. The range of fertility rate was 7.7 to 58.8% in general but the IT26118, IT26122, IT26128, IT26130, and IT26164 were more than 50%. The result of their seed production ability by selfing was 11.9 seeds per siliqua in IT26128 while the others were less than 10 seeds. In the microspore culture using parents of different hereditary, the number of embryoids, the number of plants, the rate of fertility and their pure seed production ability appeared to be very different in doubled haploid lines obtained from fertile plants of Chinese cabbage.
This work was undertaken to study the effect of stigmatic and cut-style pollination on self seed set in Lilium longiflorum and L. ${\times}$ formolongi, and their crosses as the female parent with other cultivars/genotypes. Stigmatic pollination of L. longiflorum cv. Gelria and Lorina resulted in cent per cent fruit set with mean number of seeds/fruit of 189 and 70, respectively. However, cut-style pollination resulted in 87% and 40% fruit set in Gelria and Lorina, respectively. The corresponding mean number of seeds/fruit was 53 and 20. In L. ${\times}$ formolongi, stigmatic pollination set 80% fruits with 736 seeds/fruit. On the other hand, cut-style pollination resulted in 47% fruit set with 81 seeds/fruit. The intraspecific stigmatic pollination of L. longiflorum cv. Gelria and Lorina with other cultivars formed 60% fruits with a mean number of 18 seeds/fruit. However, only 20% fruit set with mean number of seeds/fruit of 7 was recorded when cut-style pollination of L. longiflorum cultivar were pollinated with other cultivars/genotypes. In the intraspecific pollination of L. ${\times}$ formolongi cv. Raizan with Novia, fruit set resulting from stigmatic and cut-style pollination was 75% and 50%, respectively with the corresponding mean number of seeds/fruit of 579 and 98. It was concluded that self as well as intraspecific seed set in the two species of Lilium gets considerably reduced as a result of cut-style pollination.

This experiment was conducted to investigate the propagation system by seed to check growth, flowering, and making seed of three-year-old cactus of potted Mammillaria goldii. M. theresae, and M. pseudopeclinata. Plant height and width of them was 2.5~2.8 cm, and 2.8~3.1 cm, and seed formation of Mammillaria goldii and M. theresae had internal body taking130 to 135 days to ripening seeds, however, M. pseudopectinata had externals taking just 26 days to ripening seeds. The mean seeded pod from April to June of M. goldii was 50.0% and the number of seeds in a pod was 25.3. In M. theresae, the mean seeded pod from April to June was 57.3% and the number of seeds in a pod was 35.0. However, the highest rate of seeded pod on May of M. pseudopectinata was 75.0% and the number of seed in a pod was 66.0. The sib crossing was much more ripening seed fertilization than that of self cross ing in the Mammillaria. The self crossing was formed from 12.0% to 20.0% of seeded pod, from 12.0 to 16.0 number of seed in a pod. However, sib crossing obtained 58.0% of seeded pod, 30 seeds in a pod of M. goldii, 65.0% seeded pods and 40 seeds in a pod of M. theresae, and 75.0% seeded pod, 68 seeds in a pod of M. pseudopectinala as the highest of them.
