Label-free and sensitive detection of purine catabolites in complex solutions by surface-enhanced raman spectroscopy

  • Purine catabolite screening enables reliable diagnosis of certain diseases. In this regard, the development of a facile detection strategy with high sensitivity and selectivity is demanded for point-of-care applications. In this work, the simultaneous detection of uric acid (UA), xanthine (XA), and hypoxanthine (HX) was carried out as model purine catabolites by surface-enhanced Raman Spectroscopy (SERS). The detection assay was conducted by employing high-aspect ratio Au nanopillar substrates coupled with in-situ Au electrodeposition on the substrates. The additional modification of the Au nanopillar substrates via electrodeposition was found to be an effective method to encapsulate molecules in solution into nanogaps of growing Au films that increase metal-molecule contact and improve substrate's sensitivity and selectivity. In complex solutions, the approach facilitated ternary identification of UA, XA, and HX down to concentration limits of 4.33 𝜇M, 0.71 𝜇M, and 0.22 𝜇M, respectively, which are comparable to their existing levels in normal human physiology. These results demonstrate that the proposed platform is reliable for practical point-of-care analysis of biofluids where solution matrix effects greatly reduce selectivity and sensitivity for rapid on-site disease diagnosis.

  • 본 연구는 불소계 폴리실록산이미드 공중합체 막을 이용하여 휘발성 유기 화합물(Volatile Organic Compounds; VOCs)의 일종인 dichloromethane(DCM)을 물로부터 투과증발 분리하는 것을 목적으로 한다. 유연한 사슬을 갖는 diamine-terminated polydimethylsiloxane(SIDA)과 경직된 구조를 갖는 불소계 방향족 디아민 단량체(2-(perfluorohexyl)ethyl-3,5-diaminobenzene PFDAB)과 4,4'-(hexafluoroisopropylidene)diphthalic anhydride(6FDA)를 이용하여 일련의 소수성 불소계 폴리실록산이미드 공중합체 막을 제조하여 다이아민의 구성성분에 따른 분리능의 변화를 상온에서 관찰하였다. 공중합체 막 내의 다이아민 성분 중 SIDA의 함량은 0, 25, 50, 75, 100 mol%로 조절되었으며, 원액의 조성은 DCM 0.05 wt%를 사용하였다. 소수성이 높고 자유부피가 큰 SIDA의 함량이 증가함에 따라 소수성 용매인 DCM의 수착도 및 수착선택도가 증가하는 경향을 나타내었으며, 확산계수 및 확산선택도 또한 증가하여 궁극적으로는 투과플럭스와 투과선택도가 증가하는 것을 확인할 수 있었다.

Chitosan/Pebax 복합 막에 의한 기체투과 특성 (Gas Permeation Characteristics by Chitosan/Pebax Composite Membranes)

  • Chitosan과 Pebax[poly(ether-block-amide)의 mass ratio를 1 : 1, 1 : 2, 1 : 10, 1 : 30으로 하여 Chitosan/Pebax 복합 막을 제조하였다. 기체투과 실험은 압력 $6kgf/cm^2$ 하에서 35, 45, $65^{\circ}C$로 온도를 달리하여 진행되었고, Chitosan/Pebax 복합 막에 대한 $N_2$$CO_2$의 기체투과도와 선택도($CO_2/N_2$) 등을 조사하였다. $N_2$$CO_2$의 투과기체에 대해 Chitosan에 가해지는 Pebax의 함량이 증가할수록 $N_2$$CO_2$의 기체투과도는 모두 증가하였다. 선택도($CO_2/N_2$)는 Chitosan/Pebax 복합 막에서 6.9~44.3의 값을 가지며, Pebax의 함량이 증가할수록 그리고 온도가 낮을수록 큰 값을 보였다.

막반응기에서의 수성가스전이반응의 성능 분석 (Performance Analysis of Water Gas Shift Reaction in a Membrane Reactor)

  • 본 연구는 1차원 반응기 모델을 이용한 수치 시뮬레이션을 통해 수소투과량, 수소선택도, 사용된 촉매의 양, 급송흐름에서의 $H_2O/CO$ 조성비 및 Ar sweep gas가 막반응기(membrane reactor)에서의 수성가스전이반응의 성능에 미치는 영향을 분석하였다. 막반응기에서 평형상태보다 향상된 수소수율을 얻기 위해선 적어도 100 이상의 수소선택도를 가져야 함이 관찰되었으며, 수소투과량이 계속 증가될 경우에는 수소수율의 증가폭이 점차 감소됨이 보였다. 낮은 수소투과량의 경우에는 촉매량이 증가할수록 초기엔 증가된 CO 전환율을 보이다가 점차 그 증가폭이 감소되었으며, 높은 수소투과량의 경우에는 촉매의 양과 무관하게 높은 CO 전환율이 관찰되었다. 급송흐름에서의 $H_2O/CO$ 조성비가 1.5 이상인 경우엔 수소투과량이 막반응기에서의 CO 전환율에 미치는 영향이 미미하였고, 막반응기에서 평형상태보다 향상된 CO 전환율을 얻기 위해선 적어도 $6.7{\times}10^{-6}mol\;s^{-1}$ 의 Ar 몰유속이 필요함이 밝혀졌다.

  • 친환경 냉매인 R-1234yf를 제조하기 위하여 오불화프로펜인 1,2,3,3,3-pentafluoropropene의 수소화반응을 수행하였다. 수소화반응 촉매로 팔라듐이 담지된 탄소입자를 제조하여 실험에 사용하였다. 반응조건에 따른 수소화반응 활성을 규명하기 위하여 WHSV, 반응온도 및 반응물의 조성에 따른 반응성능에 대하여 조사하였다. 일정한 실험 조건에서 WHSV는 주 생성물의 선택도에 영향이 없으며, 반응온도의 경우 높은 온도에서 우수한 생성물 선택도를 보였다. 수소와 반응물의 비의 경우 1.5 미만에서 우수한 생성물 선택도를 보였다. 또한 반응조건에 따른 생성물의 수율을 예측할 수 있는 모델식을 통계학적 접근을 통해 완성하였다.

Characterization of Via Etching in $CHF_3/CF_4$ Magnetically Enhanced Reactive Ion Etching Using Neural Networks

  • This study characterizes an oxide etching process in a magnetically enhanced reactive ion etching (MERIE) reactor with a $CHF_3/CF_4$ gas chemistry. We use a statistical $2^{4-1}$ experimental design plus one center point to characterize the relationships between the process factors and etch responses. The factors that we varied in the design include RF power, pressure, and gas composition, and the modeled etch responses were the etch rate, etch selectivity to TiN, and uniformity. The developed models produced 3D response plots. Etching of $SiO_2$ mainly depends on F density and ion bombardment. $SiO_2$ etch selectivity to TiN sensitively depends on the F density in the plasma and the effects of ion bombardment. The process conditions for a high etch selectivity are a 0.3 to 0.5 $CF_4$ flow ratio and a -600 V to -650 V DC bias voltage according to the process pressure in our experiment. Etching uniformity was improved with an increase in the $CF_4$ flow ratio in the gas mixture, an increase in the source power, and a higher pressure. Our characterization of via etching in a $CHF_3/CF_4$ MERIE using neural networks was successful, economical, and effective. The results provide highly valuable information about etching mechanisms and optimum etching conditions.

Highly sensitive xylene sensors using Fe2O3-ZnFe2O4 composite spheres

  • Pure ZnFe2O4 and Fe2O3-ZnFe2O4 hetero-composite spheres were prepared by ultrasonic spray pyrolysis of a solution containing Zn- and Fe-nitrates. Additionally, the sensing characteristics of these spheres in the presence of 5 ppm ethanol, benzene, p-xylene, toluene, and CO (within the temperature range of 275-350 ℃) were investigated. The Fe2O3-ZnFe2O4 hetero-composite sensor with a cation ratio of [Zn]:[Fe]=1:3 exhibited a high response (resistance ratio = 140.2) and selectivity (response to p-xylene/response to ethanol = 3.4) to 5 ppm p-xylene at 300 ℃, whereas the pure ZnFe2O4 sensor showed a comparatively lower gas response and selectivity. The reasons for the superior response and selectivity to p-xylene in Fe2O3-ZnFe2O4 hetero-composite sensor were discussed in relation to the electronic sensitization due to charge transfer at Fe2O3-ZnFe2O4 interface and Fe2O3-induced catalytic promotion of gas sensing reaction. The sensor can be used to monitor harmful volatile organic compounds and indoor air pollutants.

Selective Cytotoxic Effects of Doenjang (Korean Soybean Paste) Fermented with Bacillus Strains on Human Liver Cell Lines

  • This report compares the selective cytotoxic effects of Doenjang fermented by various Bacillus strains (Bacillus sp. SS9, SSA3, and PM3) on human liver cell lines with that of conventional Doenjang (DTY, DTG, and DTK) and commercial Doenjang (DCM, DCD, and DCS). To investigate selective cytotoxic effects of Doenjang extracts, the cell density of HepG2 (Hepatocellular carcinoma) and CCL-13 (cells derived from human normal liver) was estimated after addition of the extracts by using a viable cell counting method. The maximum selectivity ratio ($IC_{50}$value against CCL-13/$IC_{50}$ value aganist HepG2) was observed by PM3 (extracts of Doenjang fermented with Bacillus sp. PM3). As for morphological changes shown by the addition of PM3 into HepG2 and CCL-13 cultures, HepG2 was significantly disrupted, however, CCL-13 was not affected. Also, the growth rate of HepG2 was decreased significantly by the addition of PM3. Consequently, PM3 showed a more detrimental effect on HepG2 than that on CCL-13.

고밀도 플라즈마에 의한 $Y_2O_3$ 박막의 식각 메커니즘 연구 (Etch Mechanism of $Y_2O_3$ Thin Films in High Density Plasma)

  • In this study, $Y_2O_3$ thin films were etched with inductively coupled plasma (ICP). The etch rate of $Y_2O_3$ , and the selectivity of $Y_2O_3$ to YMnO$_3$were investigated by varying $Cl_2$/($Cl_2$+Ar) gas mixing ratio. The maximum etch rate of $Y_2O_3$ , and the selectivity of $Y_2O_3$ to YMnO$_3$ were 302/min, and 2.4 at $Cl_2$/($Cl_2$+Ar) gas mixing ratio of 0.2 repetitively. In x-ray photoelectron spectroscopy (XPS) analysis, $Y_2O_3$ thin film was dominantly etched by Ar ion bombardment, and was assisted by chemical reaction of Cl radical. These results were confirmed by secondary ion mass spectroscopy(SIMS) analysis. YCl, and $YC_3$ existed at 126.03 a.m.u, and 192.3 a.m.u, respectively.

  • To increase the sensitivity and the selectivity of the sensors to TMA gas, the composition ratio and the growth conditions of the ZnO films are studied. Annealing of the ZnO films in the various time ranges and temperatures in the oxygen is carried out to enhance the stbility of the electrical resistance. Pt/Ti heater deposited on backside of the substrates in used to control the operating temperature of th esensor. The ZnO thin film sensors doped to 4.0 wt% $Al_{2}$O$_{3}$ 1.0wt.% TiO$_{2}$ and 0.2wt.% V$_{2}$O$_{5}$ exhibited a high sensitivity and an excellent selectivity for TMA gas. The sensors made with the thin films annealed at 700$^{\circ}$C for 60 minutes in the oxygen atmosphere had a good stability and linearity. The heater deposited in the ratio of 1 to 1 (Pt:Ti) had a good heating properties. The sensors fabricated using above conditions showed a good response to the actual gases of a mackerel at a step of deterioration after death.
