Porcine LMNA Is a Positional Candidate Gene Associated with Growth and Fat Deposition

  • Crosses between Korean and Landrace pigs have revealed a large quantitative trait loci (QTL) region for fat deposition in a region (89 cM) of porcine chromosome 4 (SSC4). To more finely map this QTL region and identify candidate genes for this trait, comparative mapping of pig and human chromosomes was performed in the present study. A region in the human genome that corresponds to the porcine QTL region was identified in HSA1q21. Furthermore, the LMNA gene, which is tightly associated with fat augmentation in humans, was localized to this region. Radiation hybrid (RH) mapping using a Sus scrofa RH panel localized LMNA to a region of 90.3 cM in the porcine genome, distinct from microsatellite marker S0214 (87.3 cM). Two-point analysis showed that LMNA was linked to S0214, SW1996, and S0073 on SSC4 with logarithm (base 10) of odds scores of 20.98, 17.78, and 16.73, respectively. To clone the porcine LMNA gene and to delineate the genomic structure and sequences, including the 3'untranslated region (UTR), rapid amplification of cDNA ends was performed. The coding sequence of porcine LMNA consisted of 1,719 bp, flanked by a 5'UTR and a 3'UTR. Two synonymous single nucleotide polymorphisms (SNPs) were identified in exons 3 and 7. Association tests showed that the SNP located in exon 3 (A193A) was significantly associated with weight at 30 wks (p<0.01) and crude fat content (p<0.05). This association suggests that SNPs located in LMNA could be used for marker-assisted selection in pigs.

  • Salmonella typhimurium can encounter a wide variety of environments during its life cycle. In nature, S. typhimurium can experience and survive dramatic acid stresses that occur in diverse ecological niches ranging from pond water to phagolysosomes. These survival mechanism is aquired by the Acid Tolerance Response(ATR) in Salmonella. The ATR of S. typhimurium is a complex inducible phenomenon in which exposures to slight or moderate low pH will produce a stress response capable of protecting the organism against more severe acid challenges. ATR in Salmonella has two different systems that are called RpoS dependent and independent. We found that ATR in anaerobic was showed RpoS independent because rpoS$\Omega$AP had ATR as S. typhimurium UK1. Using the P22 MudJ(Km, lacZ) operon fusion technique and a lethal selection procedure combining low pH(pH4.5) and sodium acetate(10mM, pH4.5), we isolated LF487 aatA::MudJ which showed acid sensitive in anaerobic condition. aatA locus was determined at 12 min on Salmonella Genetic Map. The survival rate of aatA mutant was showed significantly diminished at pH4.3 than virulent wild type Salmonella in anaerobic condition(5% $CO_2$, 5% H$_2$, 90% $N_2$). Therefore isolated gene was confirmed important gene for anaerobic ATR system.

Automatic Classification Algorithm for Raw Materials using Mean Shift Clustering and Stepwise Region Merging in Color (컬러 영상에서 평균 이동 클러스터링과 단계별 영역 병합을 이용한 자동 원료 분류 알고리즘)

  • In this paper, we propose a classification model by analyzing raw material images recorded using a color CCD camera to automatically classify good and defective agricultural products such as rice, coffee, and green tea, and raw materials. The current classifying agricultural products mainly depends on visual selection by skilled laborers. However, classification ability may drop owing to repeated labor for a long period of time. To resolve the problems of existing human dependant commercial products, we propose a vision based automatic raw material classification combining mean shift clustering and stepwise region merging algorithm. In this paper, the image is divided into N cluster regions by applying the mean-shift clustering algorithm to the foreground map image. Second, the representative regions among the N cluster regions are selected and stepwise region-merging method is applied to integrate similar cluster regions by comparing both color and positional proximity to neighboring regions. The merged raw material objects thereby are expressed in a 2D color distribution of RG, GB, and BR. Third, a threshold is used to detect good and defective products based on color distribution ellipse for merged material objects. From the results of carrying out an experiment with diverse raw material images using the proposed method, less artificial manipulation by the user is required compared to existing clustering and commercial methods, and classification accuracy on raw materials is improved.

High Utilization of Photovoltaic Power System in Rural Green Village Location Analysis and Evaluation using GIS - With Chubumyeon, Keumsan, Chungnam province - (GIS를 이용한 태양광 발전시스템의 활용도 높은 농촌 그린빌리지 적정입지 평가 - 충청남도 금산군 추부면을 중심으로 -)

  • The composition of rural Green Village requires higher utilization of renewable energy in those selected rural villages. The purpose of this study is to select the best results of rural green villages when using photovoltaic power system(PV system). 10 different rural villages in Chubumyeon, Keumsan, Chungnam province, were selected as study villages. This study shows measured solar radiation data, a 20-year time series data, and GIS spatial analysis; and whose were used to predict the photovoltaic power generation. PV system is used as a form with capacity of 3kWp to use for personal and public houses. Generation data was calculated by the town, where the economics of the Green Village location analysis was performed; and the solar radiation's correction factor was calculated by the 20-year time series data and measured data by study villages. By applying to the data of DEM, slope and aspect of the study villages were found, therefore performed. Spatial analysis tools were performed by using solar radiation map's tools. Those data found were used to calculate the average needed energy every months. When used the properly calculated data, towns performed economical energy consumption in rural Green Village. Every study villages have showed very high potential for PV system. Sungdangri ranked at the first (7,401kWp/year), Jangdaeri follows behind to the second (7,203kWp/year) and Yogwangri at third (7,89kWp/year) which shows higher developed energy than other study villages. The areas covered of these three towns are as follows: Sungdangri at $33,300m^2$, Jangdaeri covers $18,000m^2$ and Yogwangri shows $46,800m^2$. With these results, analyzing the potentials using GIS spatial analysis before installation of PV system was possible. Also different villages and topography in study villages have showed various results by the area. For convenience and to shorten research time, it is possible and enough to use solar radiation tools when studying spatial analysis of solar radiation.

Comparative Genomic and Genetic Functional Analysis of Industrial L-Leucine- and L-Valine-Producing Corynebacterium glutamicum Strains

  • Corynebacterium glutamicum is an excellent platform for the production of amino acids, and is widely used in the fermentation industry. Most industrial strains are traditionally obtained by repeated processes of random mutation and selection, but the genotype of these strains is often unclear owing to the absence of genomic information. As such, it is difficult to improve the growth and amino acid production of these strains via metabolic engineering. In this study, we generated a complete genome map of an industrial L-valine-producing strain, C. glutamicum XV. In order to establish the relationship between genotypes and physiological characteristics, a comparative genomic analysis was performed to explore the core genome, structural variations, and gene mutations referring to an industrial L-leucine-producing strain, C. glutamicum CP, and the widely used C. glutamicum ATCC 13032. The results indicate that a 36,349 bp repeat sequence in the CP genome contained an additional copy each of lrp and brnFE genes, which benefited the export of L-leucine. However, in XV, the kgd and panB genes were disrupted by nucleotide insertion, which increase the availability of precursors to synthesize L-valine. Moreover, the specific amino acid substitutions in key enzymes increased their activities. Additionally, a novel strategy is proposed to remodel central carbon metabolism and reduce pyruvate consumption without having a negative impact on cell growth by introducing the CP-derived mutant $H^+$/citrate symporter. These results further our understanding regarding the metabolic networks in these strains and help to elucidate the influence of different genotypes on these processes.

A Study of Planning for Gumswae-dong Garden Heritage Maintenance (고산 윤선도 금쇄동 정원유적 정비에 관한 연구)

  • Gosan Yoon, Sundo's Gumswae-dong Garden Heritage has been designated as a National Historical Site 432 including Hyunsan old fortress wall. The site requires maintenance planning. For the plan, it also requires the consideration of Gosan's written documents describing the garden heritage and the site survey with on-site inspections for authentic maintenance approaches. It should be based on the thorough comprehension of historical remains. The site is a traditional ancient garden, so its approaches should be different with a historic structure and building. For the planning, the study conducts the interview of residents and experts, literature review, the investigation of historical materials, site survey, and the analysis of aerial photography. The results are following:5) Firstly, the paper suggests three types of an excavation area selection: core, recommend and investigation. Secondly, of 22 landscapes named by Gosan, it has the plan of guidance facilities, vegetation maintenance, safety facilities, landscape maintenance as view points, pathway maintenance, deck, and halting place. Thirdly, it also suggests pathway plan for authentic garden promenade according to the literature of Gumswaedong-Gi, an old map and aerial photography(1967, 1976, 1990), and interviews with residents. Fourthly, it suggests vegetation refurbishment at the site to check erection time and to require a preservation plan. In a case of no historic remains part, it also has a plan of amenities for visitors and wetlands for biodiversity of ecology and landscape. Finally, although it requires excavation and more historical evidences for the Hyunsan fortress wall, it suggests a maintenance plan of Pyeonchuksseong and Hyeopchuksseong partially.

An Effective Extraction Algorithm of Pulmonary Regions Using Intensity-level Maps in Chest X-ray Images (흉부 X-ray 영상에서의 명암 레벨지도를 이용한 효과적인 폐 영역 추출 알고리즘)

  • In the medical image application the difference of intensity is widely used for the image segmentation and feature extraction, and a well known method is the threshold technique that determines a threshold value and generates a binary image based on the threshold. A frequently-used threshold technique is the Otsu algorithm that provides efficient processing and effective selection criterion for choosing the threshold value. However, we cannot get good segmentation results by applying the Otsu algorithm to chest X-ray images. It is because there are various organic structures around lung regions such as ribs and blood vessels, causing unclear distribution of intensity levels. To overcome the ambiguity, we propose in this paper an effective algorithm to extract pulmonary regions that utilizes the Otsu algorithm after removing the background of an X-ray image, constructs intensity-level maps, and uses them for segmenting the X-ray image. To verify the effectiveness of our method, we compared it with the existing 1-dimensional and 2-dimensional Otsu algorithms, and also the results by expert's naked eyes. The experimental result showed that our method achieved the more accurate extraction of pulmonary regions compared to the Otsu methods and showed the similar result as the naked eye's one.

A Positioning Study of National Food: In Perspective of Korean, American, Chinese Food Tourists (세계음식 브랜드 포지셔닝에 대한 연구: 한국, 미국, 중국 음식관광객을 대상으로)

  • This study was conducted to derive a positioning map using multidimensional scaling method to understand how the brand image of national foods including Korean food, Chinese food, Japanese food, Thai food, and Vietnamese food is perceived by domestic and foreign tourists. In order to achieve the research purpose, this study collected 250 data through online and offline surveys for potential food tourists who are interested in visiting overseas. Except the unfaithful responses or missing values, 202 data were analyzed. As a result, first, 8 factors which are considered to be important by food tourists were extracted. Second, the result of similarity analysis using ALSCAL and PROXSCAL did not show that the foods of the five countries were very similar, but all countries seemed to be more likely to compete with each other. Third, attribute selection also indicates that mean value of food taste (3.88), national image (3.82), and sufficient food quantity (3.65) had high level of importance, respectively. These results may provide practical implications for development of branding strategy in food tourism.

An Effective Method for Dimensionality Reduction in High-Dimensional Space (고차원 공간에서 효과적인 차원 축소 기법)

  • In multimedia information retrieval, multimedia data are represented as vectors in high dimensional space. To search these vectors effectively, a variety of indexing methods have been proposed. However, the performance of these indexing methods degrades dramatically with increasing dimensionality, which is known as the dimensionality curse. To resolve the dimensionality curse, dimensionality reduction methods have been proposed. They map feature vectors in high dimensional space into the ones in low dimensional space before indexing the data. This paper proposes a method for dimensionality reduction based on a function approximating the Euclidean distance, which makes use of the norm and angle components of a vector. First, we identify the causes of the errors in angle estimation for approximating the Euclidean distance, and discuss basic directions to reduce those errors. Then, we propose a novel method for dimensionality reduction that composes a set of subvectors from a feature vector and maintains only the norm and the estimated angle for every subvector. The selection of a good reference vector is important for accurate estimation of the angle component. We present criteria for being a good reference vector, and propose a method that chooses a good reference vector by using Levenberg-Marquardt algorithm. Also, we define a novel distance function, and formally prove that the distance function lower-bounds the Euclidean distance. This implies that our approach does not incur any false dismissals in reducing the dimensionality effectively. Finally, we verify the superiority of the proposed method via performance evaluation with extensive experiments.

Visualized Determination for Installation Location of Monitoring Devices using CPTED (CPTED기법을 통한 모니터링 시스템 설치위치 시각화 결정법)

  • Needs about safety of residents are important in urbanized society, elderly and small-size family. People are looking for safety information system and device of CPTED. That is, Needs and Installations of CCTV increased steadily. But, scientific analysis about validity, systematic plan and location of security CCTV is nonexistent. It is simply put these devised in more demanded areas. It has limits to look for safety of residents by increasing density of CCTVs. One of the characteristics of crime is clustering and stong interconnectivity. So, exploratory spatial data of crime is geo-coded using 2 years data and carried out cluster analysis and space statistical analysis through GIS space analysis by dividing 18 variables into social economy, urban space, crime prevention facility and crime occurrence index. The result of analysis shows cluster of 5 major crimes, theft, violence and sexual violence by Nearest Neighbor distance analysis and Ripley's K function. It also shows strong crime interconnectivity through criminal correlation analysis. In case of finding criminal cluster, you can find criminal hotspot. So, in this study I found concept of hotspot and considered technique about selection of hotspot. And then, selected hotspot about 5 major crimes, theft, violence and sexual violence through Nearest Neighbor Hierarchical Spatial Clustering.
