땃두릅나무 재분화 유식물체의 순화 체계 및 생육 특성 (Establishment of Acclimatization System and Growth Characteristics for Regenerated Plants of Oplopanax elatus Nakai)

  • Background: Oplopanax elatus is widely distributed at high altitudes (about 1,100 m) in China, Russia and Korea. It is hard to propagate, breed, and difficult to grow. Hence, it has been designated as a rare and endangered medicinal plant. A study was conducted to establish a system for large scale seedling production of Oplopanax elatus in vitro and to find the ideal environment for its seedling growth. Methods and Results: In this study, the explants produced under in vitro conditions during our previous study were grouped into three categories (under 10 mm, 10 mm - 30 mm and above 30 mm) based on plant height and were transferred to the growth-chamber and greenhouse for two weeks in each setting for acclimatization. The plantlet category of above 30 mm showed good performance, and was further evaluated under three acclimatization methods as follows: three different growth media (commercial soil, commercial soil + perlite, commercial soil + sand), four shading levels (0%, 50%, 70%, 90%) and four altitude levels (157 m, 218 m, 601 m, 870 m) in Gangwon province of South Korea. As results, O. elatus seedlings showed better growth characteristics at 870 m of altitude, 70% shading level and in the commercial soil compared to other treatments. Conclusions: The regenerated seedlings of Oplopanax elatus obtained through plant tissue culture would be advantageous for use in large scale seedling production systems paired with a good acclimation method. For obtaining optimal results, it is recommended that seedling be acclimatized in a high altitude environment.

적외선등을 이용한 실외 실험적 온난화 처리에 따른 소나무와 전나무의 종자 발아 및 유묘 생존율 (Seed Germination and Seedling Survival Rate of Pinus densiflora and Abies holophylla in Open-field Experimental Warming Using the Infrared Lamp)

  • 본 연구는 지구 온난화와 관련한 대응 대책 마련을 위해서 대기 온도 상승에 따른 주요 침엽수의 종자 발아 및 발아 후 유묘의 생존율 변화를 알아보고자 수행하였다. 소나무와 전나무를 대상으로 적외선등을 이용하여 대조구 보다 온난화 처리구의 대기 온도를 $3^{\circ}C$ 높게 유지하는 실외 실험적 온난화 처리를 실시하였다. 온난화 처리에 따른 대기 온도 상승으로 두 수종 모두 발아율이 높아지는 경향을 보였지만, 전나무에서만 유의적 차이가 나타났다. 소나무와 전나무 두 수종 모두 온난화 처리구가 대조구보다 평균발아일수는 유의적으로 감소하였으며, 온난화 처리구에서 높은 발아속도와 발아세를 보였다. 온도 상승과 토양 수분 감소에 따라 전나무 유묘의 고사율은 증가하였지만, 소나무는 온난화 처리에 따른 차이를 보이지 않았다. 향후, 지구 온난화에 따른 대기와 토양 온도 상승 및 토양 수분감소로 양묘과정에서 종자 발아 및 유묘 생존율의 변화가 예상되기 때문에 지구 온난화에 대응할 수 있는 양묘기술 개선이 필요할 것으로 판단된다.

파종전 무기유황 관주처리가 묘삼의 생육 및 뿌리썩음병 발생에 미치는 영향 (Effect of Irrigation of Sulfur Solution before Sowing on Growth and Root Rot Disease of Seedling in Ginseng Nursery)

  • To control the disease of root rot in ginseng nursery, inorganic sulfur solution of 0.1%, 1.0%, and 2.0% were irrigated by amount of $10{\ell}$ per $3.3m^2$ before sowing. On the last ten days of July, Fusarium solani and F. oxysporum were similarly detected by 44.8% and 43.8%, respectively, while Cylindrocarpon destructans was low detected by 4.4% in the diseased seedling. The more sulfur's concentration was increased, the more soil pH was decreased. Soil pH was decreased from 5.87 to 4.59 by the irrigation of sulfur solution of 1.0%. The more sulfur's concentration was increased, the more electrical conductivity (EC) of soil was increased. EC was increased from 0.27 dS/m to 1.28 dS/m by the irrigation of sulfur solution of 1.0%. Irrigation of sulfur solution was effective on the inhibition of damping-off caused by Rhizoctonia solani in ginseng seedling. Control value for damping-off by the irrigation of sulfur solution of 1.0% and 2.0% were 75.7%, and 78.5%, respectively. Growth of leaf was inhibited by the irrigation of sulfur solution of 2.0%. Root weight per $3.3m^2$ showed the peak in sulfur solution of 1.0%, while survived-root ratio and root weight per plant were decreased in the level of 2.0%. Survived-root ratio of seedling in sulfur solution of 1.0% was distinctly increased by 4.7 times compare to the control, but control value for root rot was relatively low as 49.2%. Mycelium growth of C. destructans, F. solani, and R. solani were distinctly inhibited by the increase of sulfur's concentration in vitro culture using PDA medium.

돈분 처리에 의한 빈도리나무 유묘생육 및 무기양분 흡수에 관한 연구 (A Study on Seedling Growth and Absorption of Inorganic Nutrients of Deutzia Crenata Seedlings Treated with Swine Manure)

  • 본 연구는 빈도리나무를 대상으로 고형 돈분의 시비 농도에 따른 빈도리나무 종자의 발아율, 묘목의 생장량, 건중량, 체내 무기양분 흡수량 및 토양의 화학적 변화 등을 조사하여 빈도리나무 유묘의 생장에 고형 돈분의 적정 시용량에 대한 자료를 얻고자 실시하였다. 1. 고형 돈분 처리시 빈도리나무의 종자발아율은 대조구가 가장 높았으며, 고농도 돈분 처리시 발아율은 감소하는 경향을 나타냈다. 2. 묘의 생장량은 돈분 처리구가 대조구 보다 모두 높은 생장량을 보였고, 돈분 1.0% 처리구에서 묘고 생장이 가장 컸다. 3. 건중량은 1.0% 처리구에서 가장 높게 나타났고, 체내 무기양분 흡수량은 대부분 1.0% 처리구에서 높았으며, 2.0%에서 급격히 흡수량이 떨어졌다. 4. 빈도리나무 식재 토양의 경우 돈분을 고농도로 처리할수록 pH는 낮아지고, 토양 중 잔류된 전질소, 유효인산, 치환성 K, Na 및 Mg 함량은 높아졌다.

투광율과 토양수분함량이 묘삼(苗蔘) 생육에 미치는 영향 (Effect of Light Transmission Ratio and Soil Moisture Content on Growth Characteristics of Seedling in Panax ginseng C. A. Meyer)

  • 비닐하우스 내에서 일평균 투광율을 21.2%, 24.8%, 30.3%와 같이 3수준으로, 토양수분을 11.0%, 12.5%, 15.3%, 18.9%와 같이 4수준으로 조절하여 묘삼의 생육 및 수량을 조사한 결과는 다음과 같다. 엽록소 함량은 투광율이 낮은 조건에서 토양수분함량의 감소에 따라 완만히 감소되었으나 투광량이 높은 조건에서 토양수분함량이 낮아지면 엽록소 함량은 현저히 감소되었다.고온장해율은 투광율이 낮은 조건에서 토양수분함량이 11.0%처럼 매우 낮을 때에만 현저히 증가되었지만 투광량이 높은 조건에서는 토양수분함량의 감소에 따라 고온장해율은 급격히 증가되었다. 3.3$m^2$(칸)당 생근중, 주당 생근중 및 사용가능묘삼수는 투광율이 증가되고 토양수분함량이 감소될수록 뚜렷이 감소되었는데, 토양수분함량이 적절한 조건(18.9%)에서는 투광율이 증가되어도 묘삼수량의 감소는 적었으나 토양수분함량이 부족한 조건에서는 투광율이 증가될수록 묘삼수량은 현저히 감소되었다. 적변율은 투광율이 높고 토양수분함량이 많을 때 현저히 증가되었는데, 투광율이 높은 조건에서 토양수분함량이 감소되면 적변율도 차차 감소되었으며, 투광율이 낮은 조건에서 적변율은 토양수분함량에 따라 큰 변화를 보이지 않았다.

벼 육모일수에 따른 양분흡수와 배유양분 소모 (Nutrient Absorption and Endosperm Consumption in Rice Seedling)

  • 벼 육묘 일수에 따른 양분 흡수 양상을 밝혀 어린모 기계이앙 상자육묘시 시비의 기초자료로 활용하고자 1990년 모래를 HCI로 세척후 시비량을 달리하고 동진벼를 상자당 200g을 파종하여 시험한 결과를 요약하면 다음과 같다. 1. 상토의 질소 함유율은 모래(N-lg), 인공상토 모두 파종 후 3일부터 감소하였으며, 파종 후 11일부터는 모래(N-lg)에서 질소가 검출되지 않았다. 2. 식물체 질소 함유율은 파종 후 9일까지는 모래 N-lg, 인공상토, 모래 N-0g의 순으로 높았으나 파종후 11일부터는 인공상토>모래 N-g>모래 N-0g의 순으로 높았다. 3. 질소 흡수량은 출아 직후인 파종 후 3일부터 인공상토>모래 N-lg>모래 N-0g의 순으로 많았으며, 파종 후 9일까지는 인공상토와 모래 N-1g간에 별 차이가 없었으나 파종 후 11일부터는 흡수량 차이가 컸다. 4. 인산의 흡수량은 파종 후 7일부터 인공상토>모래 N-lg>모래 N-0g의 순으로 많았고 칼리 흡수량은 모래의 시비량간에는 차이가 없었다. 5. 모의 초장과 건물중은 파종 후 3일부터, 엽수는 파종 후 5일부터 인공상토>모래 N-lg>모래 N-0g의 순으로 많았다. 6. 배유 소모율은 인공상토>모래 N-lg>모래 N-0g의 순으로 많았으나 배유 의존도(배유소모량/지상부 건물중)는 모래 N-0g>모래 N-lg>인공상토의 순으로 많았다.

연초용(煙草用) 상토재료(床土材料)로서의 왕겨숯(燻炭)의 질산중화효과(窒酸中和效果) (Neutralization of Rice Hull Charcoal with Nitric Acid Solution and its Neutritional Effect on Tobacco Seedling)

  • Rice hull was reduced to ash by carbonization grades to illcuidate alkalinity increase and extract of inorganic nutrients in the rice hull charcoal. Alkaline reaction of water extraction was neutral at less carbonized charcoal, but much carbonized ash from 65% weight loss reached over 10 of pH value, also origin shape of rice hull was maintained until near 65% carbonized grade. Therefore, physical properties sustained good condition for seedling bed. The more charcoal carbonized to ash, the pH value and concentration of inorganic nutrient in their extracts were increased gradually. Nitric acid concentrations for neutralizing extract from charcoal were stronger in proportion to the carbonized grade but 0.1 N nitric acid solution was very reasonable to neutralize the 65% carbonized charcoal for mixing with heavy texture acidy soil(pH 5.3) of uncultivated deep horizon to transplant the tobacco seedlings. Volume ratio mixing for seedling bed is adequate at five of ash to one of acid solution. Neutralization with nitric acid solution also accelerated extraction of the inorganic nutrient in rice hull ash. Tobacco seedlings grown on bed mixed with neutralized rice hull charcoal and soil had shown better results on the agronomic measurement than alkaline ash bed, and phosphorus and cations were uptaken more amounts.

배추 뿌리혹병(Plasmodiophora brassicae) 방제를 위한 육묘용 상토와 농약처리 효과 (Effects of Seeding Bed Media and Fungicide on Control of Clubroot Disease of Chinese cabbage Caused by Plasmodiophora brassicae)

  • 배추 뿌리혹병을 방제하기 위하여 상토의 종류와 농약 병용처리에 대한 효과를 조사한 결과 온실시험에서 피트 모스, 바로커 상토에 육묘한 배추에서의 발병도가 48.3, 54.2%에 비해 황토, 마사토 상토에 육묘한 배추에서는 발병도가 10.8, 18.7%이었고 포장시험에서는 피트모스 상토 75.7%, 바로커 36.6$, 마사토21.7%, 황토 14.1% 순이었다. 후루설파마이드 분제를 토양에 혼화처리하였을때는 봄재배에서 피트모스 상토에서는 발병도 4.5%에 비해 황토 상토에서는 0.4%이었고 가을재배에서는 10.1, 5.7%이었다. 이상의 결과에서 피트모스, 바로커 상토보다 황토, 마사토 상토를 사용하고 후루설파마이드 분제로 약제처리하면 뿌리혹병 방제에 효과적일 것으로 생각된다.

Effect of Early Seeding on Seedling Establishment and Yield in Direct Dry Seeding Rice at Honam Plain Area of Korea

  • To examine the seedling stand and growth as affected by early seeding dates of dry direct seeded rice in the Honam plain area of Korea, Dongjinbyeo was seeded at six seeding dates from early March to late April in rice fields of silty loam soil(Jeonbuk series) at the National Honam Agricultural Experiment Station (NHAES) for two years, 1996 and 1998. Seedling stand decreased slightly. with an early seeding date, but it produced more than the optimum seedling number except for the seeding of 25 March in 1996. Days to emergence was significantly longer, as seeding date was earlier, and days to emergence by early seeding was shortened only by 8 days because the mean air temperature was lower in 1996 than average, while in 1998, the reduction effect was nine to twenty five days because the mean air temperature was higher than average. In early seeding, various weeds occurred at the emergence date of rice and dominant weeds were Alopecurus aequailis, Ludwigia prostata and Rorippa islandica. NH$_4$$^{+}$ -N content in the soil at the 5th leaf stage and maximum tillering stage were lower, as the seeding date was earlier when nitrogen was split applied as basal and top dressed in 1996, while it was not significantly different among seeding dates when nitrogen was intensively applied as a top dressing in 1998. Tiller number at the maximum tillering stage and panicle number/m$^2$ were more, as seeding date was earlier in 1996, while it was not different in 1998. Filled grain rate and 1,000 grain weight was not different among the seeding dates. Milled rice yield was significantly decreased in the seeding before the middle of March, but in the seeding after late March, it was not varied when compared with the normal seeding date in 1996, while in 1998, there was no difference among seeding dates. From the above results, in consideration of seedling stand, weed occurance, rice growth and milled rice yield, the critical optimum early seeding time in the southern plain area may lie in early April. But it was suggested that when soil moisture is proper for seeding practices, seeding amount is increased and nitrogen is applied after plumule emergence of rice, milled rice yield may not be reduced in the seeding of middle or late March, compared with the seeding in April.l.
