Study on the Salt Tolerance of Rice and Other Crops in Reclaimed Soil Areas 12. On the Salt Tolerance of the Rice Seeldings Grown under the Land and Water Conditions in the Reclaimed Salty Areas (간척지에서 수도 및 기타작물의 내염성에 관한 연구 12. 육묘와 수묘의 염분간척지에서의 내염성에 관하여)

  • The rice variety, Kwanok, was reared in the water and land beds and transplanted to the reclaimed soil area, having an average salt concentration of 0.39%. Two levels of the moderate and late season cultures with 4 treatments were used. The K and Si contents of the stem part of land bed seedlings were somewhat smaller, but total carbohydrate remarkably larger, the C/N ratio was accordingly greater than water bed seedlings. The rooting ability of land bed seedlings was vigorous markedly in culture solutions, to which added various concentrations of NaCl, The rooting ability of each seedling water not much declined in theculturing solution of up to 9.4mmhos/cm, (0.6%) of salt concentration, but it was drastically declined in the salt concentration over that. It seemed that the critical salt concentration for the rooting rice plant. The land bed seedlings in each salty condition markedly decreased compared with the water bed seeldings in transpiration rate and it showed a stronger drought resistance and contained a large amount of chlorophyll at transplanting stage, and also showed higher stability of chlorophyll at rooting stage in the salt treatment. The number of panicles, panicle weight, number of grains per panicle and ratio of matured grains of the rice plant grown by the land bed seedlings were much greater and 1,000 grain weight was less than from water bed seedlings. The cultural practices with the land bed seedlings increased the rough rice yields by 15% and 11%, respectively, compared with the yields of the moderate and late season cultures by water bed seedlings.

Effects of Nursery Condition on the Establishment and the Growth of Sesame Seedling (참깨묘 양성조건이 활착 및 생육에 미치는 영향)

  • An experiment was conducted to establish the appropriate tranaplanting method of sesame seedlings. Covering of polyethylene film on nursery bed resulted in more adventitious roots and better growth after transplanting. Nursery soil showed the best results when it was mixed with the equal amounts of soil, sand and compost. The most healthy seedlings were raised by spot seeding of 5${\times}$5 cm distance and 4 cm bed-depth. Compared to direct seeding on June 15, sesame seeded on June 10 followed by transplanting on June 30 flowered 4 days earlier and produced more capsules per plant and 18% more seed yields.

The Effects of Phosphate Starvation on the Activities of Acid and Alkaline Phosphatase, Fructose-1,6-bisphosphatase, Sucrose-phosphate Synthase and Nitrate Reductase in Melon (Cucumis melo L.) Seedlings

  • Plants response to phosphate starvation include the changes of activity of some enzymes, such as phosphatases, fructose-1,6-bisphosphatase, sucrose-phosphate synthase and nitrate reductase. In this study, to determine the effects of phosphate starvation on the change of activities of acid and alkaline phosphatase, fructose-1,6-bisphosphatase, sucrose-phosphate synthase, and nitrate reductase were studied in melon seedlings (Cucumis melo L.). The content of the protein and chlorophyll tended to relatively reduced in melon seedlings subjected to phosphate starvation. Acid phosphatase activity in first and second leaves of melon seedlings was relatively higher than that of third and fourth leaves of seedlings in 14 days after phosphate starvation treatment, respectively. Active native-PAGE band patterns of acid phosphatase in melon leaves showed similar to activities of acid phosphatase, whereas alkaline phosphatase activity was different from the change in the activity of acid phosphatase. Inorganic phosphate content in melon seedlings leaves was constant. The changes of Fructose-1,6-bisphosphatase and sucrose phosphate synthase activities showed similar patterns in melon seedlings leaves, and between these enzymes activities and phosphate nutrition negatively related. Fructose-1,6- bisphosphatase and sucrose phosphate synthase activities showed significant difference in second and fourth leaves, but nitrate reductase showed significant difference in first and second leaves in 14days after phosphate starvation treatment. We concluded that phosphate nutrition could affect the distribution of phosphate, carbon and nitrogen in melon seedlings.

Effect of Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 Inoculation on Seed Germination and Early Growth of Maize and Sorghum-sudangrass hybrid Seedling under Different Salinity Levels

  • Salinity is one of the most relevant abiotic factor limiting crop yield and its net primary productivity. In addition, salinity induces an increased stress ethylene synthesis in plants which, in turn, exacerbate the responses to the stressor. Bacterial single or co-inoculation effect was tested using previously characterized plant growth promoting (PGP) bacteria Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 on maize and sorghum-sudan grass hybrid under different concentrations of NaCl. Non-inoculated maize and sorghum-sudangrass hybrid showed 33.4% and 20.0% reduction in seed germination under highest NaCl (150 mM) level tested. However, under the same NaCl concentration, co-inoculation with B. iodinum RS16 and M. oryzae CBMB20 PGP strains increased the seed germination in maize (16.7%) and sorghum-sudangrass hybrid (4.4%). In Gnotobiotic growth pouch experiments conducted for maize and sorghum-sudangrass hybrid, co-inoculation of PGP B. iodinum RS16 and M. oryzae CBMB20 mitigated the salinity stress and promoted root length by 22.9% and 29.7%, respectively. Thus the results of this study could help in development of potential bioinoculants that may be suitable for crop production under saline conditions.

The applicable evaluation of biodegradable polymer coated-mulching paper for afforestation seedlings (생분해성 고분자 코팅 조림묘목용 mulching mat 원지의 적용성 평가)

  • Recently, as the function of largest supplier of biomass for "low carbon green growth", the necessity for systematic management of afforestation areas is emphasizing. The forestation of seedling, besides the afforestation cost itself, is required some additional follow-up management costs, like mowing and fertilizing of forestation area, and removal of bindweed. The mulching mat for afforestation seedlings is available for rooting of little seedlings as well as initial forestation expenses. Mulching technique is also used to control soil temperature and moisture by covering the surface of ground. In this study, the paper based-mulching film coated with biodegradable polymer and functional additive was specially produced using laboratory bar coater, and analyzed for its degradable behavior. Coating colors were prepared by dissolving PE (polyester) 80 % and PLA(polylactic acid) 20 % in chloroform and finally applied to handsheet prepared by preceding study conditions. Base paper and polymer-coated paper were artificially aged by 2 kinds of degradation methods, which are soil degradation by microorganism and light degradation by 257 nm UV wavelengths. Strength property, oxidation index and morphological property were evaluated by reduction rates of tensile strength, FTIR spectra ratio of carboxyl and carbonyl group and SEM micrograph. As these results, polymer coated-paper was superior to base paper in degradation behaviors, having results with lower reduction rate of strength properties.

Effect of UV-B Radiation on the Leaf Growth of Rice Seedling (벼 유묘의 엽생장에 대한 자외선의 효과)

  • Rice seedlings were used to examine the effect of UV-B radiation on leaf elongation and development. Leaf elongation in both rice seedlings showed differently depending on each leaf age. UV-B radiation strongly reduced leaf elongation, 58-66% compared to without UV-B radiation, of two rice seedlings, therefore, those seedlings could not grow further. Both control and plants grown under UV-B regime showed a diurnial fluctuation in growth rate, showing maximum growth during the light period and minimum during the dark period. Leaf growth at the third leaf stage by UV-B treatment was considerably reduced by 1.7-fold than the control whereas at the fifth leaf stage was not changed. Hydrogen peroxide was considerably increased in the second phase of UV-B-induced response as catalase and peroxidase are deactivated with an increase of UV-B radiation.

Studies on Short Term Hardening Method of Tree Seedlings for Afforestation of Cut-Rock Slope (암반절개사면 녹화용 강건묘목의 속성육묘법에 관한 연구)

  • This study was carried out to develop a short term hardening method of tree seedlings of Rhus chinensis Mill., Evodia daniellii Hemsley and Parthenocissus tricuspidata(Sieb. et Zucc.) Planck for afforestation on a concave and a crack of cut-rock slope. The seedlings were grown in a cylinder shaped pot made of polyvinyl net with the soil media of peatmoss, vermiculite, clay, compost, fertilizer, and absorbant(40:25:19:15:1:0.1, v:v). They were cultivated in a greenhouse for four months and in field condition for two months. During the last three months of the growing period the seedlings were hardened by periodic desiccation and irrigation in 4 to 10 days interval. The hardened seedlings showed lower leaf water potential, higher leaf osmotic pressure, and lower T/R ratio than those before the hardening. The hardened seedlings survived well on the soil medium in the concave of cut-rock slope.

Isolation of Antifungal Compound and Biocontrol Potential of Lysobacter antibioticus HS124 against Fusarium Crown Rot of Wheat

  • Fusarium graminearum is the main cause of substantial economic loss in wheat production. The aim of this study is to investigate biocontrol potential of Lysobacter antibioticus HS124 against F. graminearum and to purify an antifungal compound. In preliminary study, n-butanol crude extract revealed destructive alterations in the hyphal morphology of F. graminearum and almost degraded with $1,000{\mu}g\;mL^{-1}$ concentration. For further study, the antifungal compound extracted from the n-butanol crude extract of L. antibioticus HS124 was identified as N-Butyl-tetrahydro-5-oxofuran-2-carboxamide ($C_9H_{16}NO_3$) using NMR ($^1H-NMR$, $^{13}C-NMR$, $^1H-^1H\;COSY$, HMBC, and HMQC), and HR-ESI-MS analysis. To our knowledge, N-Butyl-tetrahydro-5-oxofuran-2-carboxamide may be a novel compound with molecular weight of 186.1130. The minimum inhibitory concentration value of antifungal compound was $62.5{\mu}g\;mL^{-1}$ against F. graminearum. In an in vivo pot experiment, crown rot disease from F. graminearum was inhibited when wheat seeds were treated with both HS124 culture and F. graminearum. Growth of wheat seedling was enhanced by treatment of HS124 compared to control. Our results suggest that L. antibioticus HS124 characterized in this study could be successfully used to control F. graminearum and could be used as an alternative to chemical fungicides in modern agriculture.

Effect of Water Condition and Air Circulation on Fruit Fermentation of Oriental Melon (Cucumis melo L. var. makuwa Makino) (수분조건 및 송풍처리가 참외의 발효과 발생에 미치는 영향)

  • This study was conducted to assess effects of soil moisture condition and ventilation on development of fermented fruit of oriental melon. In higher soil moisture condition (-10 kPa), roots absorbed more water and transpiration was decreased under low temperature and high humidity conditions. Development of fermented fruit may be come from absorbed water concentrated placenta with higher soluble solid. Fermented fruit was developed with higher level at Shintozoa and Elite, was developed with lower level at Hongtozoa, and was not developed at self-rooting of oriental melon. Ventilation on fruit did not affect development of fermented fruit at Shintozoa, Elite, Hongtozoa and self-rooting seedling. Ca contents of flesh and placenta of fruit under ventilation treatment were higher than control but contents of Acetaldehyde, Ethanol and Ethylacetate were lower.

The Comparison of Resistance of Sweet Potato Cultivars to Sclerotium Rot Caused by Sclerotium rolfsii (품종 간 고구마 흰비단병(Sclerotium rolfsii) 발생 정도 비교)

  • This study was carried out to assay the disease incidence degree of Sclerotium rot caused by Sclerotium rolfsii in sixteen most popular commercial cultivars of sweet potato (Ipomoea batatas) in Republic of Korea. The degree of disease incidence was evaluated on pot experiments. In pot experiments using artificial inoculation, the Sclerotium rolfsii caused a stem rot on seedling of sweet potato plants and causes a crown rot on lower stems near or at the soil line at favorable environmental conditions. White mycelial mats and sclerotia were formed at the infection sites. Plants severely infected were fell over or died because lower stems near soil surface were rotten. The degree of disease incidence was varied according to cultivars. Two cultivars, Bio-mi and Deayumi, were very resistant, while five cultivars, Shinjami, Shingeonmi, Hongmi, Yeonjami, and Shinhung-3 were highly susceptible.
