Screening of Tomato Cultivars Resistant to Bacterial Canker by Seedling Test (유묘검정법을 이용한 궤양병 저항성 토마토품종 선발)

  • Bacterial canker, caused by Clavibacter michiganensis subsp. michiganensis, is a very damaging disease to tomato (Lycopersicon esculentum) farm in Korea. It infects tomato, spreads through the xylem and causes bacterial wilt and canker. Selection of resistant cultivar is the best way to prevent or reduce the occurrence of the disease. Thirty-nine tomato cultivars, twenty-one cherry tomato cultivars and thirteen rootstock tomato cultivars were inoculated with Clavibacter michiganensis subsp. michiganensis, to evaluate tomato cultivarspecific resistance against bacterial canker. In the evaluation of 73 major commercial cultivars, 'Sunmyung', 'Sweet', 'Akiko', 'Dadaki', 'Match', 'Magnet', 'Friend', and 'Greenpower' were found to have a high level of resistance to bacterial canker of tomatoes.

  • In an experiment to evaluate the various aspects of symptom of witches' broom of jujube tree with two different kinds of scions, taken from diseased plant and stem- grafted upon sound stocks-one cut before wintering(stored in cellar), the other after wintering, it was observed that the disease rate of those cut before wintering(Nov.) was $\98\%$ whereas those cut after wintering(March) showed much lower rate, $3\%$ initially and gradually increased up to $39\%$. In another experiment of diseased bud grafted into healthy seedling, the finding made in the following year was that 14 stocks(only one of which salt union by callusing) were infected out of the given 23- in the initial stage only 4, yet gradually increased to the number of 14. The shoots from the ground portion of the diseased stock were in general more quickly subjected to the disease than the others. Under natural condition, the diseased trees develop at first seemingly the same leaves as healthy ones; it is not until the branches and loaves grow to a considerable degree that the symptom appears. Once appearing, tile disease grows and the symptom continues to appear as late as in Sept. and early Oct., causing tile plant to develop the extraordinary branches and leaves of extremely reduced size, the typical symptom of witches' broom. Such phenomenon can be observed in the experiments of the foregoing paragraphs. And the suckers and roots of diseased plant are bound to be infected, it was found out with no exception. Viewing from the results of the above experiments and observation, it is believed that the low temperature during winter causes the virus in the above-ground portion of diseased plant to diminish or inactivated, and subsequently the virus in the roots moves up or multiply in the sprouting season.

Occurrence of Leaf Spot on Eriobotrya japonica Caused by Entomosporium mespili in Korea (Entomosporium mespili에 의한 비파나무 점무늬병 발생)

  • Since 2008, a new leaf spot disease has been found in loquat tree (Eriobotrya japonica) at a nursery station in Goheung, Korea. Above 50% of the seedlings developed the spot symptoms on the leaves. The infected plants were generally reduced in the seedling vigour and early defoliation was occurred. The infected leaves developed grayish lesion with dark reddish brown margin. The pathogen developed round to ellipsoidal acervuli on the lesion. Based on morphological characteristics of the pathogen, the fungus was identified as Entomosporium mespili. The cultured pathogen successfully reproduced the same disease symptom on the leaves of loquat tree and found to be E. mespili. A monoconidial culture was deposited in Korean Agricultural Culture Collection (KACC 44727). This is the first report of E. mespili causing leaf spot of loquat tree in Korea.

Detection of "Candidatus Phytoplasma Asteris" Associated with Black Locust Witches' Broom in Korea ("Candidatus phytoplasma asteris" Group에 속하는 아까시나무 빗자루병 검출)

  • Typical phytoplasma witches' broom symptoms were observed in black locust (Robinia pseudoacacia L.) in Korea. The symptoms of the disease were showing abnormally small leaves, shortened intemodes and proliferation of shoots. The phytoplasmas were detected consistently in all the symptomatic samples by the amplification with phytoplasma universal primer pairs P1/P7 and R16F2n/R2, and the expected size was 1.8 kb and 1.2 kb. However, the phytoplasma DNA was not detected in healthy seedling. Based on sequence analysis of amplified region, this phytoplasma has close homologies with aster yellow, mulberry dwarf, maize bushy stunt, ash witches' broom and sumac witches' broom phytoplasmas, more than 99.2% but showed homologies with black locust witches' broom (GeneBank Accession No. AF 244363), and jujube witches' broom, 88.6% and 87.7%, respectively. This phylogetic analysis indicates that the black locust witches' broom phytoplasma founded in korea should be classified in the Candidatus phytoplasma asteris (16Sr I) group and clearly distinct from the black locust witches' broom group 16Sr III (peach X-disease phytoplasma group).

Evaluation of Resistance to Ralstonia solanacearum in Tomato Genetic Resources at Seedling Stage

  • Bacterial wilt of tomatoes caused by Ralstonia solanacearum is a devastating disease that limits the production of tomato in Korea. The best way to control this disease is using genetically resistant tomato plant. The resistance degree to R. solanacearum was evaluated for 285 tomato accessions conserved in the National Agrobiodiversity Center of Rural Development Administration. These accessions of tomato were originated from 23 countries. Disease severity of tomato accessions was investigated from 7 days to 14 days at an interval of 7 days after inoculation of R. solanacearum under greenhouse conditions. A total of 279 accessions of tomato germplasm were susceptible to R. solanacearum, resulting in wilt and death in 70 to 90% of these plants. Two tomato accessions were moderately resistant to R. solanacearum. Only four accessions showed high resistance against R. solanacearum. No distinct symptom of bacterial wilt appeared on the resistant tomato germplasms for up to 14 days after inoculation of R. solanacearum. Microscopy of resistant tomato stems infected with R. solanacearum revealed limited bacterial spread with thickening of pit membrane and gum production. Therefore, these four resistant tomato germplasms could be used in tomato breeding program against bacterial wilt.

Impact of Rhizosphere Competence of Biocontrol Agents upon Diseases Suppression and Plant Growth Promotion

  • Root colonization of biocontrol agents via seed treatment was investigated and a compatible combination, Gliocladium virens G872B and Pseudomonas putida Pf3, in colonizing cucumber rhizosphere was confirmed through the study. Much higher number of fungal and bacterial propagules were detected when two isolates were inoculated together. The presence of Pf3 in root system was greatly helpful to G872B to colonize at root tip. The mechanism of this phenomenon is partially elucidated through the results of in vitro experiments and the observations of scanning electron and fluorescence microscope. Addition of Pf3 cells resulted earlier germination of G872B conidia and increased mycelial growth. And the more number of germinated conidia on seed coat, the more vigorous hypal streching and sporulation on the root surface were observed in coinoculated treatment. The propagules of G872B on the cucumber root when they were challenged against the pathogenic Fusarium oxysporum, were even higher than that of G872B treated alone, and the magnitude of such a difference was getting grater toward the root ip and the population of F. oxysporum on the root was reduced by seed inoculation of G872B. The rhizosphere competence was obviously reflected to disease suppression and plant growth promotion that induced by the given isolate. Green house experiments revealed that the combined treatment provided long-term disease suppression with greater rate and the larger amount of fruit yield than single treatments. Through this study the low temperature growing Pseudomonas fluorescens M45 and MC07 were evaluated to apply them to the winter crops in field or plastic film house. In vitro tests reveal that M45 and MC07 inhibited the mycelial growth of Pythium ultimum, Rhizoctona solani and Phytophthora capsici and enhanced growth of cucumber cotyledon in MS agar. This effect was more pronounced when the bacteria were incubated at 14$^{\circ}C$ than at 27$^{\circ}C$. And disease suppression and plant growth promotion in green house were also superior at low temperature condition. Seed treatment of M45 or soil treatment of MC07 brought successful control of damping-off and enhanced seedling growth of cucumber. The combined treatment of two isolates was more effective than any single treatment.

Analysis of Control Efficacy of Bacterial Fruit Blotch Caused by Acidovorax avenae subsp. citrulli in Recent Issues (최근 문제시 되는 수박 과일썩음병에 대한 방제효과 분석)

  • Bacterial fruit blotch (BFB) caused by Acidovorax avenae subsp. citrulli is defective disease to watermelon cultivated areas. To control of BFB, we investigated control efficiency to use commercial antibacterial pesticides. Growth inhibition zone on medium were formed as oxolinic acid WP and oxytetracycline WP. Control efficacy of four anti-bacterial pesticides on seed and seedling stage were performed. As a results, oxytetracyclin WP is shown over 90% control efficiency on seed and acibenzolar-S-methyl + mancozeb WP shown over 90% control efficiency on seedling stage Hot-water treatment method could be possible to reduced infection rate on seed. The conditions of hot-water treatments are $50{\sim}55^{\circ}C$ on 20~30 minutes. These results suggested that the methods were helpful watermelon seedling nursery to control of the bacterial fruit blotch by A. avenae subsp. citrulli.

Enhancement of Biomass Production in Chinese Milk Vetch (Astragalus sinicus L.) by Controlling Alopecurus aequalis with Sethoxydim under Poor CMV Seedling Stand (자운영 입모부족시 Sethoxydim 처리가 둑새풀 방제 및 자운영 녹비량 증가에 미치는 영향)

  • Technology development for sufficient dry matter production of Chinese milk vetch (CMV) is most important in CMV-rice cultivation system in order to provide sufficient nutrients to rice plants. However, when the CMV plants are dominated by the weed, especially Alopecurus aequalis, the CMV growth could be reduced due to light and nutrient competition. In addition, A. aequalis is potential host of the rice dwarf virus disease. Therefore, control of A. aequalis is necessary to enhance the biomass production of CMV plants when CMV stands are insufficient. The use of chemical like sethoxydim (20%, ai) showed the highest control rate of 84% at early stage and was reduced as application was delayed. A. aequalis control did not change the CMV seedling stand before and after herbicide treatment and the reseeding stand in fall was rather increased 2.2 to 2.6 times. On the other hand, in untreated control, the CMV stand at May 15 and reseeding stand in fall was significantly reduced as compared with the before herbicide treatment. Control of A. aequalis increased the CMV dry matter production by 164% for 50% CMV coverage rate and 63% for 25% CMV coverage rate. This is equivalent to $12.3{\sim}16.4\;kgN$/10a which is greater than the recommended nitrogen rate of 9kg/10a. The result indicates that the control of A. aequalis is an efficient way to enhance dry matter production in CMV-rice cultivation system especially when CMV stand is poor.

Optimization of In vitro Cultures for Production of Seedling and Rootstock of Rehmannia glutinosa(Gaertn.) DC. (지황 배양묘 및 종근 생산을 위한 기원검증 및 최적기내배양조건 확립)

  • Rehmannia glutinosa(Gaertn.) DC. is a herbaceous perennial plant and belonging to the Scrophulariaceae and used as roots for medicinal part and purpose. R. glutinosa is and usually used for fresh rehmannia root or prepared rehmannia root. However, it is very difficult to propagate using the seeds because of lack germination so it is propagated using the vegetative method as the rootstock. Currently, propagation and harvesting using the rootstock of R. glutinosa has difficulties about production of the high quality and quantity in R. glutinosa because of root rot disease. To optimize in vitro cultures and to improve the rootstock and seedling of R. glutinosa after morphological and genetical determination, 5 plant culture media (MS, DJ, LS, QL, and WPM) were used in this study then WPM was selected for better growth, for multiplication condition(WPM + IAA 1.0 mg/L + IBA 0.5 mg/L), and for root enlargement condition(WPM + NAA 0.1 mg/L) of R. glutinosa. Based on these results, in vitro seedlings of R. glutinosa were transferred to soil for acclimation with environment adaptation and shown the positive effects about root enlargement and root formation. Therefore, it can be used for high quality of R. glutinosa production and production of the rootstock based on propagation using in vitro seedlings of R. glutinosa.

Control of Bakanae Disease of Rice by Seed Soaking into the Mixed Solution of Procholraz and Fludioxnil (Prochlornz와 fludioxonil 혼용침지소독에 의한 벼 키다리병 방제)

  • These experiments were conducted to improve the effect of seed disinfection on rice seed severely infected Bakanae disease by seed soaking into mixed solution of prochloraz EC and fludioxonil FS. We investigated the effects of various concentrations of two fungicides mixed solution on spore germination and mycelial growth of Fusarium fujikuroi. Mycelial growth was inhibited 100% at $10{\mu}g$/ml of prochloraz and 33.3% at $80{\mu}g$/ml of fludioxonil. Spore germination was inhibited 81.4% at $40{\mu}g$/ml of prochloraz. Interestingly, mixed solution of $5{\mu}g$/ml or $10{\mu}g$/ml of each fungicide inhibitied 100% of mycelial growoth and 99.2% of spore germination, respectively. Severely infected rice seeds soaked into mixed solution composed of $125{\mu}l$/ml of prochloraz and $50{\mu}l$/ml of fludioxonil showed 2.1% of disease symptoms compared to 20.4% of prochloraz $125{\mu}l$/ml, but higher concentrations of prochloraz decreased the seedling stand rate. When the seed soaking time was longer and temperature was higher, control effect on Bakanae disease was improved, but seedling stand was lower about 80% over $35^{\circ}C$.
