Comparison of Physicochemical Properties and Antioxidative Activities of Sunflower Sprout According to Germination Day (발아일수에 따른 해바라기 싹의 이화학적 특성 및 항산화성 비교)

  • It is well known that sunflower (SF) sprout has more beneficial effects than SF seed due to increased levels of phytochemical components such as vitamins, total phenolics, and isoflavones during germination. This study investigated the physicochemical properties and antioxidative activities of SF seed during both germination and cultivation. In a proximate analysis, the water content of SF groat was 9.17% and then increased to 15.32% on the 11th day after seeding. On a dry weight basis, crude fat content decreased while the contents of carbohydrates, crude protein, and crude ash increased. As cultivation proceeded, the contents of minerals were in decreasing order of K, Mg, Ca, Na, Zn, Fe, Mn, and Cu, in which SF sprout cultivated for 9 or 11 days contained the highest mineral contents. Though vitamin C was not detected on SF groat, the content of vitamin C continuously increased up to the 5th day of cultivation and then decreased gradually. Vitamin E content in SF groat was higher than that in SF sprout. It was also found that the vitamin E content in SF sprout was the highest on the 5th day of cultivation. Daidzin was not detected in SF groat, but its concentration reached a maximum on the 5th day of cultivation in SF sprouts. Furthermore, higher amounts of daidzein were observed on the 3rd, 5th, and 9th days of cultivation. The highest total isoflavone content was observed on either the 3rd or 5th day of cultivation. The highest content of total phenolics was observed on the 5th of cultivation. When DPPH radical and peroxyl radical scavenging activities of SF sprout were measured in order to measure antioxidant efficacy, it was found that 5 day-cultivated SF sprout had the highest scavenging activities. In conclusion, SF sprout cultivated for 9 or 11 days was found to be a good source of minerals. Furthermore, the fifth-day after seeding was the optimal time for the production of SF sprout with effective natural antioxidant activity and high amounts of functional components such as vitamins, total phenolics, and isoflavones.

Analysis of Functional Compounds on the Seeds of Sorghum (Sorghum bicolor Moench) and Their Sprouts (수수의 종자와 새싹채소의 기능성분 분석)

  • This study was done to examine antioxidant compounds, antioxidative activity and chemical properties in organically cultivated sorghum seed and their sprout, and to examine as a basic data for various application methods as an organic farming product of sorghum. The crude protein and mineral contents in the sprout had 2.5 and 1.6~9.87 folds more than sorghum seed, respectively. Total polyphenol content as antioxidant was more in the sprout showing $4,135mg\;GAE\;g^{-1}$ than the seed, whereas total flavonoid content was relatively higher in the seed showing $3.86mg\;GAE\;g^{-1}$. ABTS radical scavenging activity in the seed was observed by higher level as $15.78mg\;TE\;g^{-1}$. However, The sprouts with and without natural light were no significant difference. DPPH radical scavenging activity in the seed exhibited activity of 11 folds or more compared to that of the sprout. The highest activity was detected from the seeds and the sprouts in Donganmae. Although sorghum was no significant correlations among the varieties, it showed correlation with between the light-treated condition and antioxidation.

Effects of Sprout Length, $Ca0_2$ Coating and Seeding Depth on Seedling Stand and Early Growth in Puddled-soil Drill Seeding of Rice (벼 무논골뿌림재배시 최아장, 과산화석회 처리 및 파종심도가 입모수와 초기생육에 미치는 영향)

  • This study was carried out to clarify the effect of sprout length, burried depth of seed and CaO$_2$ seed coating on emergence, seedling stand and early growth of rice in puddled-soil drill seeding at Junbuk in 1995. The cultivar tested was Dongjinbyeo(Japonica type). Although the longer sprouted seeds were caused the better seedling stand, 4mm sprout length of seeds appeared to be appropriate for good seedling stand and seeding uniformity, while 6mm sprouted seed resulted in ununiform seedling establishment. Rapid emergence and increased seedling stand were obtained by CaO$_2$ seed coating, which had effect more on soaked seed than on sprouted seed, but the seedling stand of sprouted seeds significantly reduced at deeper than 1cm seeding depth and also emergence rate of sprouted seeds significantly reduced at deeper than 2cm seeding depth.

Chemical Compositions and DPPH Radical Scavenger Activity in Different Sections of Safflower (홍화의 부위별 화학성분과 DPPH Radical 소거 활성)

  • Chemical compositions and DPPH radical scavenger activity in different sections of safflower (Carthamus tinctorious L.) were investigated. Protein contained 28.39% in sprout and fat contained 20.47% in seed, respectively. Linoleic acid as predominant unsaturated fatty acid of safflower contained 80.01% in sprout and 78.27% in seed. Glucose contained 1253.6mg% in sprout and fructose contained 970.7mg% in sprout. Sucrose contained 912.0mg% in flower bud. Succinic acid was included 2795.3 mg% in flower, malic acid was included 2054.8mg% in leaf. K as minerals contained 2826.8mg% in leaf and 2613,6 mg% in sprout, Ca contained 1999.8mg% in leaf and 1160.9mg% in sprout. Total phenolics contained 5.8%, 4.7%, 4.4% in flower, sprout and leaf, and total flavonoid contained 6.5%, 2.5%, 2.0% in flower, sprout and leaf, respectively Serotonin-I (Ν- [2- (5-hydroxy - l Η- indol -3- yl)ethyl] ferulamide) as serotonin compounds was determined 147.7mg% in seed, serotonin - II (Ν-[2-(5-hydroxy-lΗ-indo-1-3yl )ethyl]-p-coumaramide) was determined 155.4 mg% in seed. Acacetin as flavonoid compounds was contained 116.5mg% in seed. Luteolin as flavonoid compounds was identified 388.3mg% in sprout, luteolin 7-glucoside was determined 692.3mg% in leaf, respectively. DPPH radical scavenger activity was measured by DPPH method, it was shown higher 114.2% in ethanol extract of flower and 113.6% in ethanol extract of leaf than 88.05% of 100 ppm BHA as chemical antioxidant.

Germination Ability of Pregerminated and Dried Rice Seed (벼 최아후 건조종자의 발아능력)

  • In order to investigate vigor of rice seed pregerminated, some organic and inorganic component content, response of ecotypes, and effect of gibberellin treatment, under different levels of hastening of germination, were examined. Protein content was high in dried seed, the larger size in length of pregerminated seed, the less protein content, but lipid content was a reverse tendency to protein one. Potassium content was low in dried seed, the larger size in length of pregerminated seed, the more potassium content, but increase dramatically at above 10 mm in length of sprout. Magnesium content showed a same tendency to potassium one. The ability of regermination was high in early maturing cultivar, but almost lost in the pregerminated seeds with above 5 mm in length of sprout. The regermination percentage of dried seed was lower than that of pregerminated seed with 0.1∼0.5 mm in length, but was similar to that of pregerminated one with 1∼3 mm in length of sprout. The effect of gibberellin on regermination percentage was highest at 10 ppm and showed high with small sized one of pregerminated seed. The effect of gibberellin on plant height, fresh weight, and dry weight showed a same tendency to regermination percentage.

Correlations between the Growth Period and Fresh Weight of Seed Sprouts and Pixel Counts of Leaf Area

  • Purpose: This study was carried out to predict the growth period and fresh weight of sprouts grown in a cultivator designed to grow sprouts under optimal conditions. Methods: The temperature, light intensity, and amount of irrigation were controlled, and images of seed sprouts were acquired to predict the days of growth and weight from pixel counts of leaf area. Broccoli, clover, and radish sprouts were selected, and each sprout was cultivated in a 90-mm-diameter Petri dish under the same cultivating conditions. An image of each sprout was taken every 24 hours from the 4th day, and the whole cultivating period was 6 days, including 3 days in the dark. Images were processed by histogram inspection, binary images, image erosion, image dilation, and the overlay image process. The RGB range and ratio of leaves were adjusted to calculate the pixel counts for leaf area. Results: The correlation coefficients between the pixel count of leaf area and the growth period of sprouts were 0.91, 0.98, and 0.97 for broccoli, clover, and radish, respectively. Further, the correlation coefficients between the pixel count of leaf area and fresh weight were 0.90 for broccoli, 0.87 for clover, and 0.95 for radish. Conclusions: On the basis of these results, we suggest that the simple image acquisition system and processing algorithm can feasibly estimate the growth period and fresh weight of seed sprouts.

Establishment of Seed Treatment for Healthy Production of Peanut Sprout (청정 땅콩나물 생산을 위한 종자처리기술 확립)

  • The present study was conducted to develop seed treatment for the production of healthy and clean peanut sprout. Dry heat treatment of peanut seeds reduced the incidence of the rot. The seed treatment condition at $52^{\circ}C$ for 10 h. was the most efficient without inhibiting seed viability significantly. Seeds were dark cultured at $27^{\circ}C$ for up to 9 days. The treatment of Indole-B and gibberellic acid influenced germination, T50, fresh, dry weight, hypocotyl length, hypocotyl length diameter, root length, number of lateral root and epicotyl of peanut sprout. There were no differences in the germinability of peanut seeds between gibberellic acid treatment methods but higher fresh weight was observed in the GA3 solution spray after 2 hour water soaking. The general growth and lateral root development of peanut sprouts were suppressed by Indole-B which is used for inhibiting root formation and promoting hypocotyls. The treatment of gibberellic acid promoted hypocotyl elongation, but it did not influence on the growth of hypocotyls and root system.

Contents of Minerals and Vitamines in Soybean Sprouts (콩나물의 무기이온 및 비타민 함량)

  • Soy sprouts cultured in a lab with five soybean cultivars and three commercial soy sprouts purchased in a market were investigated to compare the mineral and vitamine contents. In case of minerals, K, Ca, Mg, and Na were found as major minerals and a small quantity of Fe, Zn, Mn, and Cu was also analyzed in soy sprout. The all minerals were distributed in the cotyledon and the hypocotyl but their concentration orders were different between in cotyledon and in hypocotyl. After boiling, much of minerals eluted in water, the pattern of release of the minerals from the soy sprout to water was similar in all soy sprouts. The vitamin C was concentrated a great amount in hypocotyl than in cotyledon whereas vitamin B was found in the cotyledon generally. And their contents were reduced after boiling owing to heat-mediated degrading. The vitamin C and B contents were different according to cultivars, which shows chemical composition of soybean seed was important to soy sprout characters. The present results provide a basis for attempt to improve seed quality in soybean breeding program for soy sprout.

Effects of Chitosan on Production and Rot Control of Soybean Sprouts

  • The practicality of utilizing chitosan as a natural antimicrobial compound to reduce soybean sprout rot was tested. Domestic and imported soybean seeds were soaked for 6 hours in solutions containing different levels of chitosan and acetic acid (glacial), and cultured at $25^{\circ}C$ for 6 days. In case of domestic seeds, soaking with 1,000ppm chitosan increased germination percentage, hypocotyl thickness, total length, and fresh weight of sprouts by 4%, 5%, 2%, and 1%, respectively. The total sprout yield was increased by chitosan in a concentration-dependent manner in that 1,000ppm chitosan resulted in 8% increment of total yield (7.47kg sprouts/kg seed). Chitosan significantly reduced sprout rot percentage to 7.0% compared to control (13.8%), and consequently enhanced marketable sprout yield by 39%. Compared to domestic seeds, the imported soybean seeds exhibited very low germination percentage regardless of chitosan treatments. Chitosan, nevertheless, consistently induced yield increment and rot decrement in imported soybean sprouts. Although 100ppm acetic acid was effective in reducing sprout rot percentage down to 11.8%, its yield-increasing effects were not as prominent as chitosan. In conclusion, soaking soybean seeds with chitosan seems to be a practical method to enhance the efficiency of soybean sprout production.

Effects of Environmental Conditions on Incidence of Bacterial Soft Rot in Soybean Sprout (환경요인이 콩나물 무름병 발생에 미치는 영향)

  • Incidence of soybean sprout rot by Erwinia carotovora subsp. carotovora was examined under several artificial conditions. Under higher temperatures over 3$0^{\circ}C$, disease incidence was increased and the rate of soft rot incidence was 22% at 35$^{\circ}C$. Artificial injuries of inner cotyledon and seed coat induced the disease above 70% and inhibited the soybean sprout growth. Relative humidity above 90% increased the soft rot to 33% and inhibited soybean sprout growth. When the leaked water collected from soybean sprout was reused for irrigation, the disease incidence was increased.
