Studies on Quality Evaluation of Current Sesame Oils Sold in Markets (시중 유통참기름의 품질평가에 관한 연구)

  • In the present study, an attempt was made to use FV (Fatty acid ratio & Villavecchia reaction) value determination as a reliable method for the detection and analysis of the adulteration of sesame oils. FV value was defined as fatty acid ratio, C18 : I + C18 : 2/C16 : ${\times}C18$ : 3, times modified Villavecchia-Suarez test value. Seventy-four sesame oils collected from markets were evaluated using this method. Only II among 74 collected sesame oils were found to be pure sesame oil by FV value determination. In 63 adulterated sesame oils, it was revealed 23 samples were adulterated soybean oil, to with rice bran oil, 10 with sesame dregs extract oil, 8 with perilla seed oil, 7 with corn oil, 3 with cotton seed oil, and 2 with rape seed oil.

Detection of Collapse Buildings Using UAV and Bitemporal Satellite Imagery (UAV와 다시기 위성영상을 이용한 붕괴건물 탐지)

  • In this study, collapsed building detection using UAV (Unmanned Aerial Vehicle) and PlanetScope satellite images was carried out, suggesting the possibility of utilization of heterogeneous sensors in object detection located on the surface. To this end, the area where about 20 buildings collapsed due to forest fire damage was selected as study site. First of all, the feature information of objects such as ExG (Excess Green), GLCM (Gray-Level Co-Occurrence Matrix), and DSM (Digital Surface Model) were generated using high-resolution UAV images performed object-based segmentation to detect collapsed buildings. The features were then used to detect candidates for collapsed buildings. In this process, a result of the change detection using PlanetScope were used together to improve detection accuracy. More specifically, the changed pixels acquired by the bitemporal PlanetScope images were used as seed pixels to correct the misdetected and overdetected areas in the candidate group of collapsed buildings. The accuracy of the detection results of collapse buildings using only UAV image and the accuracy of collapse building detection result when UAV and PlanetScope images were used together were analyzed through the manually dizitized reference image. As a result, the results using only UAV image had 0.4867 F1-score, and the results using UAV and PlanetScope images together showed that the value improved to 0.8064 F1-score. Moreover, the Kappa coefficiant value was also dramatically improved from 0.3674 to 0.8225.

Development of PCR Diagnosis System for Plant Quarantine Seed-borne Wheat Streak Mosaic Virus (식물검역 종자전염 Wheat Streak Mosaic Virus의 PCR 검사시스템 개발)

  • Wheat streak mosaic virus (WSMV), a member of the genus Tritimovirus in Potyviridae, severely impacts wheat and corn seed worldwide, but has yet to be detected in Korea, and hence, every effort should be made to prevent its introduction. To prevent WSMV from entering the country, it is necessary to prepare a specific, sensitive, simple, and fast detection method for routine application to plant quarantine procedures. For this reason, a two-step diagnosis system consisting of RT-PCR and nested PCR is being used for WSMV detection. In addition, a novel positive control was developed for use with the system. WSMV has been detected in seed sweet corn from Japan and seed wheat from USA by a two-step diagnosis system, the details of which are described in this study. After sequence analysis, similarities of 80.6 and 100.0% with other isolates were determined by BLAST. They showed the same topology, which was classified as 4 genotypes by various phylogenetic trees, using a poly protein encoding sequence amplification. In this analysis, WSMV-JSweet-corn2868 (JX845574) is classified as clade B, while WSMV-Uwheat1944-1 (KC754959) and WSMV-Uwheat1944-2 (KC754960) belong to clade D.

High-sensitivity ZnO gas Sensor with a Sol-gel-processed SnO2 Seed Layer (Sol-Gel 방법으로 제작된 SnO2 seed layer를 적용한 고반응성 ZnO 가스 센서)

  • A metal oxide semiconductor gas sensor is operated by measuring the changes in resistance that occur on the surface of nanostructures for gas detection. ZnO, which is an n-type metal oxide semiconductor, is widely used as a gas sensor material owing to its high sensitivity. Various ZnO nanostructures in gas sensors have been studied with the aim of improving surface reactions. In the present study, the sol-gel and vapor phase growth techniques were used to fabricate nanostructures to improve the sensitivity, response, and recovery rate for gas sensing. The sol-gel method was used to synthesize SnO2 nanoparticles, which were used as the seed layer. The nanoparticles size was controlled by regulating the process parameters of the solution, such as the pH of the solution, the type and amount of solvent. As a result, the SnO2 seed layer suppressed the aggregation of the nanostructures, thereby interrupting gas diffusion. The ZnO nanostructures with a sol-gel processed SnO2 seed layer had larger specific surface area and high sensitivity. The gas response and recovery rate were 1-7 min faster than the gas sensor without the sol-gel process. The gas response increased 4-24 times compared to that of the gas sensor without the sol-gel method.

Sequential Defect Region Segmentation according to Defect Possibility in TFT-LCD Image (TFT-LCD영상에서 결함 가능성에 따른 순차적 결함영역 분할)

  • Defect region segmentation of TFT-LCD images is performed by combining defect pixels detected by a defect detection method into defect region, or by using morphological operations to segment defect region. Therefore, the result of segmentation of the defect region is highly dependent on the defect detection result. In this paper, we propose a method which segments defect regions sequentially according to the possibility of being included in defect regions in TFT-LCD images. The proposed method repeats the process of detecting a seed using the median value and the median absolute deviation of the image, and segments the defect region using the seeded region growing method. We confirmed the superiority of the proposed method to segment defect regions using pseudo-images and real TFT-LCD images.

  • White mold (or Sclerotinia stem rot), caused by Sclerotinia species, is a major air, soil, or seed-transmitted disease affecting numerous crops and wild plants. Microscopic or culture-based methods currently available for their detection and identification are time-consuming, laborious, and often erroneous. Therefore, we developed a multiplex quantitative PCR (qPCR) assay for the discrimination, detection, and quantification of DNA collected from each of the three economically relevant Sclerotinia species, namely, S. sclerotiorum, S. minor, and S. nivalis. TaqMan primer/probe combinations specific for each Sclerotinia species were designed based on the gene sequences encoding aspartyl protease. High specificity and sensitivity of each probe were confirmed for sclerotium and soil samples, as well as pure cultures, using simplex and multiplex qPCRs. This multiplex assay could be helpful in detecting and quantifying specific species of Sclerotinia, and therefore, may be valuable for disease diagnosis, forecasting, and management.

Characterization of Fennel Flavors by Solid Phase Trapping-Solvent Extraction and Gas Chromatography-Mass Spectrometry

  • Headspace solid phase trapping solvent extraction (HS-SPTE) and GC-MS was applied for the characterization of volatile flavors from fennel, anise seed, star-anise, dill seed, fennel bean, and Ricard aperitif liquor. Tenax was used for HS-SPTE adsorption material. Recoveries, precision, linear dynamic ranges, and the limit of detection in the analytical method were validated. There were some similarities and distinct differences between fennel-like samples. The Korean and the Chinese fennels contained trans-anethole, (+)-limonene, anisealdehyde, methyl chavicol as major components. The volatile aroma components from star anise were characterised by rich trans-anethole, (+)-limonene, methyl chavicol, and anisaldehyde. Additionally, principal component analysis (PCA) has been used for characterizing or classifying eight different fennel-like samples according to origin or other features. A quite different pattern of dill seed was found due to the presence of apiol (dill).

Identification and Pathogenicity of Microorganisms Associated with Seed-Rhizome Rot of Gingers in Underground Storage Caves (토굴저장 생강의 부패에 관여하는 미생물의 동정 및 병원성)

  • Microorganisms associated with seed-rhizome rot of gingers preserved in three underground storage caves were identified with respect to rot types. Rot patterns were grouped into 4 different types : yellow soft rot, brown rot, localized ring rot, and water-soaked rot. Water-soaked rot was highest in frequency with 40% and ring rot the least with 14%. Causal pathogens differed with rot type, yellow soft rot by Erwinia carotovora and Pseudomonas aeruginosa, brown rot by Fusarium solani and Pseudomonas aeruginosa, localized ring rot by F. solani, and water-soaked rot by Pythium spinosum and P. ultimum. Pythium myriotylum, the causal pathogen of ginger rhizome rot which occurs severely in fields was rarely detected from storage seed-rhizomes suggesting its minor involvement with storage rot. Pathogenic Pythium isolates were frequently obtained from both rhizome surface and inner tissues of rotten rhizomes. Detection frequency of Pythium isolates in inner tissues decreased as increasing distance from rhizome surface. In wound-inoculation tests, above pathogens caused a varying degree of rot on healthy rhizomes at 15$^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$ with increasing severity at higher temperatures.

  • 본 논문에서는 렌즈의 흠집을 추출할 수 있는 퍼지 기법을 이용한 렌즈 흠집 검출 방법을 제안한다. 제안된 방법은 렌즈 영상을 그레이 영상으로 변환한 후, 캐니 마스크를 이용하여 렌즈의 경계선을 추출한다. 추출된 렌즈의 경계선에 대해 평균 이진화와 모폴로지를 이용하여 렌즈 경계선을 보정한다. 렌즈 경계선이 보정된 영상에서 Seed Fill 알고리즘을 적용하여 렌즈의 내부 영역만을 추출한다. 추출된 렌즈의 내부 영역에 해당하는 원 영상에서 소벨 마스크를 적용하여 렌즈 내부 영역의 에지를 추출한다. 렌즈 내부 영역에서 추출된 에지 객체들의 정보를 이용하여 흠집과 비흠집을 분류하는 퍼지 기법을 적용하여 흠집 영역을 추출한다. 본 논문에서 제안된 렌즈의 흠집 검출 방법의 성능을 평가하기 위해 CHEMI, MID, HL, HM 시력 보정용 렌즈를 대상으로 실험한 결과, 제안된 방법이 흠집을 효과적으로 검출하는 것을 확인하였다.

Detection of Radiation Induced Markers in Oranges Imported from the United States of America (미국산 오렌지의 Radiation Induced Marker 검색)

  • Radiation induced markers were investigated for the detection of irradiated oranges imported from America. In the DNA comet assay, the non-irradiated and irradiated samples showed the comets with long tails in both seed and flesh. Though this tendency was maintained for 6 weeks, identification of non-irradiated or irradiated samples was impossible. In the thermoluminescence (TL) measurement, the non-irradiated samples revealed a glow curve with low intensity at about 28$0^{\circ}C$, while the irradiated samples showed with higher intensity at around 18$0^{\circ}C$. There were no remarkable changes in detection properties for 6 weeks after irradiation. The TL ratio of area for TL$_1$ glow curve to TL$_2$ was below 0.1 for the non-irradiated samples and 0.5 or more for the irradiated ones during storage. In the electron spin resonance (RSR) measurement, irradiated oranges showed an unspecific central signal in all parts (seed, flesh and peel), so the detection for radiation treatment of oranges was impossible. Based on the results, DNA comet assay and ESR were not useful for the detection, but TL was appropriate to search radiation induced markers of oranges during storage period. The detectable period during storage is confirmed by sensory evaluation.
