Quality Characteristics of Bread Added on Chia seed Powder (치아시드 분말을 첨가한 식빵의 품질특성)

  • To examine the quality characteristics of bread made of different amounts of chia seed powder, a functional food ingredient, this study made bread by adding chia seed powder by 1%, 3%, and 5% of the flour weight. When bread dough properties were determined using mixograph, all doughs were appropriate for bread-making. In the evaluation of fermentation rate, the dough with 3% of chia seed powder CS 3 showed the most favorable fermentation rate during the first 60-min fermentation of the bread. According to CrumbScan, the fineness of bread pores were higher in control group than bread loaves containing chia seed powder. The volume of bread in control group was significantly lower than those of other experimental groups. The volume and specific volume were highest when chia seed powder was added by 3%. According to preference analysis, appearance and taste was best in CS 3, showing the highest overall score in preference. Therefore, the optimum amount of chia seed powder is identified to be 3% in bread-making.

Quality Evaluation for Vegetable Use in Local Soybean Cultivars with Various Seed Coat Color

  • From the evaluation of physical properties such as springiness, gumminess, adhesiveness, chewiness and hardness by the texture analyzer, vegetable soybean lines with green seed-coat were best as compared with those with black, brown, mixed, and yellow seed-coats. A panel test evaluated on the basis of taste, sweetness, chewiness, and total scores also indicated that soybean lines with green seed-coat were the best. The total scores of panel test was decreased in the order of green > yellow> black> brown seed-coat colored soybean. The mean value of sucrose content obtained by HPLC analysis was highest in black seed-coat colored soybean, and followed by green, yellow, and brown soybeans. The highest sucrose content (8.22%) was observed in 180362, a soybean line with green seed-coat. The full-season type soybeans showed much higher sucrose content than summer types which are mainly cultivated on farmer's fields for vegetable purposes. The final 13 lines selected from 300 colored soybeans showed nearly the same panel scores as Miwongreen. However, these lines had a great deal of variation in sucrose content, and much higher readings in texture analysis than Miwongreen, especially in chewiness and hardness which were the most important properties in vegetable soybeans.

Agronomic Characters and Their Correlation Coefficient on Black Seeded Soybeans Collected in Chonnam Province

  • In order to obtain the genetic information on the quantitative characters of black seeded soybeans, which would be needed to improve selection efficiency for breeding high yielding genotype, 45 varieties of black seeded soybeans collected in Chonnam, Korea were grown and variations of several important characters were observed. Heritability of each observed character, phenotypic and genotypic correlations among the characters and contribution of each yield component on grain yield through path coefficient analysis were estimated. Both number of pods per plant and 100-seed weight showed not only high heritability but also highly significant phenotypic and genotypic correlation with seed yield, and hence it was desirable to select plants with more number of pods per plant and higher 100-seed weight than raise seed yield of black seeded soybeans collected in Chonnam. In addition, number of pods per and 100-seed weight were proved to be the most influential variables on the viability of seed yield by path coefficient analysis. Since these showed the high heritability of number of pods per plant, selection of plants with higher 100-seed weight would be more efficient for breeding high yielding genotype.

The comparative gene expression concern to the seed pigmentation in maize (Zea mays L.)

  • Maize seed pigmentation is one of the important issue to develop maize seed breeding. The differently gene expression was characterized and compared for three inbred lines, such as the pigment accumulated seed (CM22) and non-pigmented seed (CM5 and CM19) at 10 days after pollination. We obtained a total of 63,870, 82,496, and 54,555 contigs by de novo assembly to identify gene expression in the CM22, CM5, and CM19, respectably. In differentially expressed gene analysis, it was revealed that 7,044 genes were differentially expressed by at least two-fold, with 4,067 upregulated in colored maize inbred lines and 2,977 upregulated in colorless maize inbred lines. Of them,18 genes were included to the anthocyanin biosynthesis pathways, while 15 genes were upregulated in both CM22/5 and CM22/19. Additionally, 37 genes were detected in the metabolic pathway concern to the seed pigmentation by BINs analysis using MAPMAN software. Finally, these differently expressed genes may aid in the research on seed pigmentation in maize breeding programs.

Development of On-line Sorting System for Detection of Infected Seed Potatoes Using Visible Near-Infrared Transmittance Spectral Technique (가시광 및 근적외선 투과분광법을 이용한 감염 씨감자 온라인 선별시스템 개발)

  • In this study, an online seed potato sorting system using a visible and near infrared (40 1100 nm) transmittance spectral technique and statistical model was evaluated for the nondestructive determination of infected and sound seed potatoes. Seed potatoes that had been artificially infected with Pectobacterium atrosepticum, which is known to cause a soil borne disease infection, were prepared for the experiments. After acquiring transmittance spectra from sound and infected seed potatoes, a determination algorithm for detecting infected seed potatoes was developed using the partial least square discriminant analysis method. The coefficient of determination($R^2_p$) of the prediction model was 0.943, and the classification accuracy was above 99% (n = 80) for discriminating diseased seed potatoes from sound ones. This online sorting system has good potential for developing a technique to detect agricultural products that are infected and contaminated by pathogens.

Chemical Components of Seed Oil of Sapium japonicum Pax. et Hoffm. (사람주나무 종실유의 화학적 조성)

  • Physico-chemical characteristics and chemical composition of seed oil of Sapium japonicum were determined by various analysis methods. Physio-chemical characteristics of the extracted oil from fresh seed were specific gravity (0.928), refraction index (1.477), acid value (2.30), saponification number (190.0), and iodine value (126.0). Neutral lipid (93%) in seed oil was identified as major components, followed by glycolipid (4.9%) and phospholipid (1.3%). In GC analysis, nine fatty acids were presented in the seed oil. Among fatty acids, predominant fatty acids were oleic acid (45.8%) and linoleic acid (35.6%). The contents of fatty acids of seed oil somewhat varied with their storage period. Composition of fatty acids from lipid fractions which were isolated on silica open column was investigated. Major fatty acids in three lipid fractions were linoleic acid and linolenic acid. The fatty acid contents of fresh seed oil derived from each lipid fraction were generally high comapred to storage seed oil. From the above results, seed oil of Sapium Japonicum could be useful in cosmetics, detergents and a few pharmaceuticals.

Genetic Variation of Flower Production in Breeding Seedling Seed Orchards of Quercus acuta and Q. glauca

  • This study was conducted to test the significant difference of fertility variation among families and to select superior families for acorn production in the breeding seedling seed orchards (BSSOs) of Quercus acuta and Quercus glauca. The seed orchards were located in Jeju island and established by seedlings raised from selected parents for genetic testing in 2006. In the spring of 2021, the numbers of female and male flower were counted from 5 to 10 individuals per family in the BSSOs. To test statistical significance of which parameter is not satisfied through the normality test, we used a nonparametric analysis. Correlation analysis was performed to quantify the association between female and male flower production. As the results, the significant difference of flower production among families was found in both seed orchards. The averages of female flower production were 65.3 and 181.9 in Q. acuta and Q. glauca. The positive Spearman's rank correlation was existed between male and female flower production. Broad-sense heritability on female and male flower production were 0.191 and 0.147 in Q. acuta, and 0.285 and 0.068 in Q. glauca, respectively. Sexual asymmetry (e.g., maleness index) between female and male, and contribution variation among families (e.g., parental balance) were analyzed to find reasonable alternatives in the management of seed orchards. Effective population size of seed crops was predicted as a concept of status number. Loss of gene diversity (accumulation of group coancestry) would not be alarming in the BSSOs. Our results would be helpful to select breeding materials for establishing new seed orchards and to supply genetically improved seeds of evergreen oaks, which is one of the backbones of the strategy of carbon sink in the 2050 Carbon Neutrality of Korea Forest Service.

Brassica rapa Sec14-like protein gene BrPATL4 determines the genetic architecture of seed size and shape

  • Seed size traits are controlled by multiple genes in crops and determine grain yield, quality and appearance. However, the molecular mechanisms controlling the size of plant seeds remain unclear. We performed functional analysis of BrPATL4 encoding Sec14-like protein to determine the genetic architecture of seed size, shape and their association analyses. We used 60 $T_3$ transgenic rice lines to evaluate seed length, seed width and seed height as seed size traits, and the ratios of these values as seed shape traits. Pleiotropic effects on general architecture included small seed size, erect panicles, decreased grain weight, reduced plant height and increased sterility, which are common to other mutants deficient in gibberellic acid (GA) biosynthesis. To test whether BrPATL4 overexpression is deleterious for GA signal transduction, we compared the relative expression of GA related gene and the growth rate of second leaf sheath supplied with exogenous $GA_3$. Overexpression of BrPATL4 did not affect GA biosynthesis or signaling pathway, with the same response shown under GA treatment compared to the wild type. However, the causal genes for the small seed phenotype (D1, SRS1, and SRS5) and the erection of panicles showed significantly decreased levels in mRNA accumulation compared to the wild type. These results suggest that the overexpression of BrPATL4 can control seed size through the suppression of those genes related to seed size regulation. Although the molecular function of BrPATL4 is not clear for small seed and erect panicles of BrPALT4 overexpression line, this study provides some clues about the genetic engineering of rice seed architecture.

Seed Characteristics and Germination Properties according to Change of Cone Production in Pinus densiflora Stands (소나무 우량 임분의 구과 생산량에 따른 종자 및 발아 특성)

  • This study was carried out to obtain the several data for improving the seed supply in Pinus densiflora. Seed characteristics and germination properties according to change of cone production from 2003 to 2006 were analyzed in three plus stands of P. densiflora. P. densiflora showed good production at interval of two years in the periodicity of seed production. Seed potential and percent of filled seed (PFS) among the major seed characteristics showed positive relations to the change of cone production, whereas percent of developed seed (PDS) and seed efficiency showed opposite trend. As a result of regression analysis between seed characteristics and cone production, seed potential ($R^2=0.682$, p<0.01) and PFS ($R^2=0.161$, non-significant) presented positive correlation with cone production while PDS ($R^2=0.418$, p$R^2=0.308$, non-significant) presented negative correlation. In germination properties percent of germination (PG) showed similar tendency to cone production, while mean germination time (MGT) showed opposite tendency. As a result of regression analysis between germination characteristics and cone production, PG showed positive correlation, whereas MGT showed negative correlation. Therefore, we could expect high germination percentage and short period for germination when we harvested cones at good cone production time.

Studies on the Protein of Peach seed flour (복숭아 종자의 단백질에 관한연구)

  • The proteins of peach seed four was examined to utilize protein source. The peach seed flour contained 20.38% of crude proteins. The extractability of salt souble proteins of seed were 70% and recovery rate of main protein fractions separated by sephadex G-200 were about 51%. The electrophoretic analysis showed 11 bands and molecular weight showed 14,000-110,000 in seed proteins. The Amino acid of peach seed flour and isolated were mainly composed of arginine, asparic acid. glutamic acid, glycine and leucine. The solubility of isolated peach seed protein was lowest at pH 5.5.
