Investigating the Subsea Sandwich Pipeline Integrity under Complex Loadings (선형 매칭 기법을 활용한 해저 샌드위치 파이프의 복합하중 영향도 분석)

  • Subsea pipelines are widely used to transport hydrocarbons from ultra-deep seawater to facilities on the coast. A sandwich pipe is a pipe-in-pipe system in which the annulus between the two concentric steel pipes is filled with polymer cores and fillers for insulation and structural reinforcement. Sandwich pipeline is always exposed to complex loading such as bending moment, bulking, internal and external pressures caused by installation, operation and environmental factors. This research provides insights into the structural integrity of sandwich pipeline exposed to complex loading conditions using a linear matching method (LMM). The finite element model of the sandwich pipeline has been generated from previous research, and the model validation is performed by comparing the results of the linear analysis between the two models. The temperature dependent material properties are used to simulate the behavior of real pipeline, and the elastic-perfectly plastic (EPP) model has been taken into account for the material non-linearity. Numerical results provide comprehensive insights into the structural response of the sandwich pipeline under monotonic and cyclic loading and provide notable points about the evaluation of the plastic collapse limit and the elastic shakedown limit of the sandwich pipeline.

Cryopreservation of Filefish (Thamnaconus septentrionalis) Sperm

  • The present study examined the possibility of long term storage, by cryopreservation in liquid nitrogen, of the sperm of Filefish (Thamnaconus septentrionalis), and the changes in motility, survival rate and ultrastructure of the sperm after freezing and thawing. The sperm was collected by stripping and stored on ice until experiments. For selection of the immobilizing solution, diluted artificial seawater (ASW) of 20, 30 and 40% were tested. The sperm motility was significantly inhibited in 30% ASW, and restored entirely after 100% ASW was added again. Two cryoprotectants, dimethyl sulfoxide ($Me_2$SO) and glycerol, were added to 30% ASW to formulate the extenders at the concentrations between 5 to 20% by volume for freezing. The sperm was diluted at the ratio of 1 :6 with the extenders, inserted into 0.5ml plastic straws and frozen at a freezing rate of $50^{\circ}C$/min to $-100^{\circ}C$ after equilibration for 10 min at room temperature, followed by plunging into liquid nitrogen. The straws were thawed in a $30^{\circ}C$ water bath for 15 sec. The highest post-thawed sperm motility and survival rate were obtained with 5% glycerol Afterward, the effect of different freezing rates was examined using 5% glycerol as a cryoprotectant, and the rate of $20^{circ}C/min to $-80^{\circ}C$ showed the best result Some ultrastructural changes of sperm, such as the detachment of plasmatic and nuclear membranes, destruction of mitochondria, were observed after cryopreservation. Morphological normality of the sperm in 5% glycerol frozen at the ratio of 1$0^{\circ}C$/min to $-80^{\circ}C$ was better than that of others.

Effects of Storage Temperature on the Survival of Vibrio mimicus K-1 in Seawater and Arkshell (해수와 피조개에서 Vibrio mimicus K-1의 생존에 대한 보관온도의 영향)

  • The cell density changes of Vibrio mimicus K-1 in sea water and arkshell feeding it were examined at various temperature. The strain was suspended in sterilized sea water and storaged at experimental temperature $(5,\;10,\;15,\;20,\;and\;28^{\circ}C)$). At intervals of up to 10 days, aliquots of each suspension were plated onto BHI agar. At 5 and $10^{\circ}C$, the plate counts of V. mimicus K-1 showed a rapid decline, which 3s known to be a reault of this bacterium's entering into the viable but non culturable state. At 20 and $28^{\circ}C$, however, V. mimicus K-1 are stable over the 10 days experimental periods. V. mimicus K-1 was fed to arkshell, which was subsequently stored at temperatures ranging from 5 to $20^{\circ}C$ for 10 days. The samples of arkshell were homogenized and plated at intervals to determine the cell density of V. mimicus K-1 and total aerobic population of bacteria present. At 5 and $10^{\circ}C$, the numbers of V. mimicus K-1 in sea water rapid decreased over the 10 days experimental periods. However, little change of V. mimicus K-1 density was observed in shellstock arkshell at 5 and $10^{\circ}C$. While, V. mimicus K-1 density was decreased more rapidly to level below limit of dectection in shucked arkshell at same temperature. Incubation at the higher temperature $(20^{\circ}C)$ resulted in large increase in total aerobic bacterial number of shellstock arkshell. These results suggest that even with proper storage, indigenous levels of V. mimicus may remain sufficiently high in shellstock arkshell to produce infection in compromise hosts.

Effects of Water Temperature and Salinity on the Oxygen Consumption Rate of Juvenile Spotted Sea Bass, Lateolabrax maculatus (점농어, Lateolabrax maculatus 치어의 산소 소비율에 미치는 수온과 염분의 영향)

  • An experiment was conducted to investigate the effects of three water temperatures (15, 20 and $25^{\circ}C$) in combination with three salinities (0, 15 and 30 psu) on the oxygen consumption rate of juvenile spotted sea bass, Lateolabrax maculatus (mean body weight $5.5{\pm}0.3g$). The oxygen consumption rates of L. maculatus were measured in triplicate for 24 hours using a continuous flow-through respirometer. Water temperature resulted in significant differences in the mean oxygen consumption rate of L. maculatus (p<0.001), but salinity and combinations of salinity and water temperature did not have (p>0.05). The oxygen consumption increased with increasing water temperatures in all experimental salinity regimes (p<001). Mean oxygen consumption rates at 15, 20 and $25^{\circ}C$ ranged 328.8~342.3, 433.9~441.0 and 651.5~659.9 mg $O_2\;kg^{-1}\;h^{-1}$, respectively. $Q_{10}$ values did not vary with salinity, bud varied with water temperature. $Q_{10}$ values ranged 1.63~1.75 between 15 and $20^{\circ}C$, 2.24~2.26 between 20 and $25^{\circ}C$, and 1.92~1.98 over the full temperature range. The energy loss by metabolic cost increased with increasing water temperatures in all experimental salinity regimes (p<0.001) Mean energy loss rates at 15, 20 and $25^{\circ}C$ ranged 224.6~233.8, 296.3~301.2 and $444.9{\sim}450.7kJ\;kg^{-1}\;d^{-1}$, respectively. These data suggest that the culture of juvenile spotted sea bass is possible without energy loss by salinity difference in freshwater as well as seawater after salinity acclimation. Thus, this result has an application for culture management and bioenergetic model for growth of this species.

Coastal Eutrophication caused by Effluent from Aquaculture Ponds in Jeju (제주연안 육상양식장 밀집지역 주변해역의 영양염 과잉 요인)

  • This study investigated the temporal-spatial distribution and variations in water quality parameters (temperature, salinity, pH, DO, COD, SPM, DIN, DIP, silicate, TN, TP, and chlorophyll-a) in the coastal area of Jeju, Korea, adjacent to aquaculture ponds (Aewol-ri, Haengwon-ri, Pyosun-ri, and Ilkwa-ri). Data were collected bimonthly from February 2010 to December 2011. A principal component analysis (PCA) identified three major factors controlling variations in water quality during the sampling period. Aquaculture effluent water led to large changes in nutrient levels. The highest nutrient values were observed during the investigation period. The relatively large increase in organic matter at the sampling stations coupled with sea area runoff events during the summer rainy period. Variation in chlorophyll-a concentration was mainly driven by meteorological factors such as air temperature and rainfall in the coastal areas of Aewol and Haengwon. In the coastal areas of Pyosun and Ilkwa, pollution was caused by anthropogenic factors such as discharge of aquaculture effluent water. High nutrient concentrations at the majority of the coastal stations indicate eutrophication of coastal waters, especially within a distance of 300 m and depth of 10m from drainage channels. Coastal eutrophication driven by aquaculture effluent may be harmful inshore. Events such as eutrophication may potentially influence water pollution in aquaculture ponds when seawater intake is detected because of aquaculture effluent water.

Environmental Factors and Variation Characteristics of Zooplankton Communities in Gamak Bay (가막만 동물플랑크톤 군집의 변동특성과 환경요인)

  • Spatio-temporal patterns in the zooplankton communities, physicochemical environmental factors, and dominant species were examined in Camak Bay from April, July, September, and November, 2003. A total of 40 taxa were sampled with a mean abundance of total zooplankton ranging from $270{\sim}1,803inds/m^3$. The most abundant species was Acartia erythraea, followed by Eurytemora pacifica, Podon polyphemoides, A. omorii, and Paracalanus parvus s. 1 during the study period. Multivariate analysis revealed significant differences in community structure among three regions: the middle part of the sampling area (B), sites 1, 2 (A) and other sites (C). The number of species and diversity (H') varied significantly among the three regions (p<0.05), but no differences in the mean abundances were observed (p>0.05). In order to investigate factors affecting the correlations between the abundance of dominant species, all zooplankton and environmental factors ware calculated using a correlation analysis. Of these, the abundance of dominant taxon showed positive correlation with seawater temperature, salinity and dissolved oxygen (p<0.05). In statistical analysis showed that the environmental factors influencing the variation of the communities ere predominantly temperature, salinity, and dissolved oxygen.

Geochemical Characteristics of Devonian Bitumen Carbonates in Alberta, Canada (캐나다 데본기 비투멘 탄산염암의 지화학적 특성 연구)

  • The objective of this study is to investigate inorganic characteristics of Devonian bitumen carbonates in Alberta using two drilling cores, Saleski 03-34-88-20w4 and Saleski 08-01-88-20w4, taken from the Core Research Center (CRC) of Canada. The bitumen carbonates are mainly composed of less than 0.2 mm dolomites and some carbonate includes small amount of quartz and calcite. The bitumen carbonates from two cores are interpreted to have formed in similar sedimentary environments and dolomitization processes. Carbonates from Saleski 03-34-88-20w4 core were formed under higher inflow of clastic sediment than those from Saleski 08-01-88-20w4 core. Range of crystallization temperature of dolomites in the both bitumen carbonate cores is about 40~$55^{\circ}C$. Dolomitizing fluid of the bitumen carbonates would be Devonian seawater. Bitumen carbonates from Cairn Formation, compared with the CRC cores, have experienced a similar crystallization temperature, but dolmititizing fluid of the bitumen carbonates from Cairn Formation have been modified from the isotopic exchange with continental crust.

Spawning Induction and Egg Development of Surf Clam, Spisula sachalinensis (북방대합, Spisula sachalinensis의 산란유발 및 난발생)

  • In order to obtain the basic information for seedling production of surf clam, Spisula sachalinensis, spawning induction and egg development were investigated. $NH_4OH$ addition and serotonin injection could induce the spawning in surf clam. Water temperature rising, sperm suspension immersion, UV-ray irradiated seawater and $H_2O_2$ addition less affected on induction of spawning than $NH_4OH$ and serotonin did. On the other hand, males were more sensitive to the treatments than females. The response time to initial spawning in the case of $NH_4OH$ addition was $3\~4$ hours. However in the case of serotonin injection, it was within 5 minutes. The number of eggs released by $NH_4OH$ addition were significantly more than those released by serotonin injection. The serotonin injection induced higher rates of germinal vesicle breakdown than the $NH_4OH$ addition. Fertilizing and hatching rates of the eggs also were the similar results. Eggs of surf clam were demersal isolated eggs and averaging $65.2{\pm}±1.8\;{\mu}m$ in diameter after spawning. Optimum range of water temperature for the development of egg was $15\~20^{\circ}C$, The required time for development of D-shaped larvae was 42 hours at $15^{\circ}C$ and 27 hours at $20^{\circ}C$, respectively.

Seasonal Photosynthetic Performance of Laminaria japonica sporophytes on the Southeastern Coast of Korea (한국 동남해안에서 다시마(Laminaria japonica) 포자체의 광합성의 계절변화)

  • Rates of photosynthesis and respiration of Laminaria japonica sporophytes cultivated on the southeastern coast of Korea were monthly measured in situ and under constant temperature and nitrogen concentration in laboratory from February to July 1996 in order to understand the seasonal photosynthetic performance of this alga. P-I (the relationship between photosynthsis and irradiance) parameters measured in situ varied seasonally. Photosynthetic capacity ($P_{max}$) reached its maximum in March (6.64 mg $O_2{\cdot}gdw^{-1}{\cdot}h^{-1}$) and gradually decreased thereafter. Photosynthetic efficiency (${\alpha}$), which ranged from 0.026 to 0.106, generally showed a similar pattern with the $P_{max}$, curve. Correlation between respiration and $P_{max}$, was not significant (Spearman's rank correlation, p>0.05); respiration rate, which varied between 0.25 and 0.83 mg $O_2{\cdot}gdw^{-1}{\cdot}h^{-1}$, showed no gradual decline from March. $P_{max}$ in situ significantly correlated with the relative growth rate of frond weight (Spearman's rank correlation, p<0.01) and this result means that the amount of accumulated materials in body increased with the increment of $P_{max}$. Compared $P_{max}$, in situ with that in the laboratory. The lower $P_{max}$ in February was probably caused by the effect of lower seawater temperature at this time. The decrease in $P_{max}$ after March, however, was primarily attributable to the thickness of blade as the plants got old. Comparison of P-I parameters measured for different size groups in April, explained the negative relationship between $P_{max}$ and frond thickness.

Development of Ecologically Suitable Habitat Model for the Sustainable Sea Cucumber Aquafarm (지속가능한 해삼 양식장 조성을 위한 생태적합 서식처 모형 개발)

  • We investigated the tidal current, hydrographic data, and benthic environment of major sea cucumber (Holothuroidea, de Blainville, 1834) habitats in Baengnyeongdo, Jindo and Uljin to understand the optimal environmental or ecological habitat for sea cucumbers. The three study areas were characterized by a cold-water mass of temperatures ranging $12{\sim}18^{\circ}C$, with an active circulation between the surface and deep waters. According to an analysis of the tidal current map, a strong flow velocity of $100{\sim}120cm\;s^{-1}$ appeared in Baengnyeongdo and Jindo. The three sea cucumber habitats showed the common characters of a bottom sediment composed of sand-silt, a diverse seaweed colony and benthic organisms, and boulders and rocks which provide a hideout for the organisms. We aimed to draw the optimal habitat condition for sea cucumbers in Korea, and the result showed that the low water temperature, rapid water flow, active vertical mixing between surface and deep waters, bottom composed by sand-silt, large rocks, and diverse seaweed colony and benthic organism were important factors. The optimal habitat for Juvenile sea cucumbers was the intertidal areas characterized by a muddy bottom, reef, and seaweed. The optimal habitat for adult sea cucumbers was characterized by a place where sand and mud are mixed, and the body size of the sea cucumber was proportional to water depth, and the relatively large boulders and rocks compared to the intertidal area.
