Accumulation and Depuration of Fluoranthene, a Polycyclic Aromatic Hydrocarbon, in Rockfish Sebastes schlegeli (조피볼락 (Sebastes schiegeii)에서 다환성방향족탄화수소 fluoranthene의 축적과 배설)

  • Rockfish Sebastes schlegeli was exposed to fluoranthene, a ubiquitous polycyclic aromatic hydrocarbon, at 1 and 10 $\mu$g/L for 4 weeks followed by depuration period of 8 weeks. Although the fluoranthene in the p]asma reached only 1.8$\~$1.9 times seawater concentration, it was 6.5 $\~$ 15.7 times higher in the liver, spleen and bile indicating efficient accumulation in the lipid -containing body tissues. When the exposed fish were then maintained in clean water, rapid fluoranthene decline occurred in the initial 2 weeks followed by a rather slow phase. This result suggests that fluoranthene accumulates efficiently provided the existence in the culture medium, but the contaminant disappears rapidly once the chemical source is removed. The fluoranthne residue in fish tissues my be a good indifator for relent PAHs exposure.

Dissolution Behavior and Hydrate Effect on $CO_{2}$ Ocean Sequestration

  • $CO_{2}$ ocean sequestration is one of the promising options to reduce $CO_{2}$ concentration in the atmosphere because the ocean has vast capacity for $CO_{2}$ absorption. Therefore, in the present investigation, calculations for solubility and dissolution behavior of liquid $CO_{2}$ droplets released at 1000 m and 1500 m deep in the ocean from a moving ship and a fixed pipeline have been carried out in order to estimate the $CO_{2}$ dissolution characteristics in the ocean. The results show liquid $CO_{2}$ becomes bubble at around 500 m in depth, and the solubility of seawater is about $5{\%}$ less than of pure water. Also, it is shown that the injection of liquid from a moving ship is a more effective method for dissolution than from a fixed pipeline, and the presence of hydrate on liquid $CO_{2}$ acts as a resistant layer in dissolving liquid $CO_{2}$.

  • We have selected an valuable pmbiotic strain; Lactobacillus sakei BK19 which has wide antagonic spectrum against fish pathogens . Present study investigated cultural characterization of L. sakei BK19 including pH tolerance , susceptibility of antibacterial agents and growth pattern with different environment such as nutritions, temperature and salinity. L. sakei BK19 showed Significantly higher resistance at low pH(around pH 4) environment and relative high antibiotic tolerance . In the study of optimal culture condition, maltose and saccharose provided the optimal nutritional culture condition while lactose and mannitol were unable to supply its carbon source for the fermentation of L. sakei BK19. Moreover. L. sakei BK19 showed good growth at the temperature range of 15 to $45^{o}C$ und the NaCl concentration of 0 to 7%. Hence, this particular probiotic strain may be benificial both in seawater and fresh weter conditions.

Microarray-Mediated Transcriptome Analysis of the Tributyltin (TBT)-Resistant Bacterium Pseudomonas aeruginosa 25W in the Presence of TBT

  • The tributyltin (TBT)-resistant bacterium, Pseudomonas aeruginosa 25W, which was isolated in seawater from the Arabian Sea, was subjected to transcriptome analysis in the presence of high concentrations of TBT. Only slight effects were observed at TBT concentration of $50{\mu}M$, but exposure to $500{\mu}M$ resulted in the upregulation of 6 genes and the downregulation of 75. Among the 75 downregulated genes, 53% (40 out of 75) were of hypothetical function, followed by 14 transcriptional regulation- and translation-associated genes. The results of this study indicated that although the 25W strain was highly resistant to TBT, high concentrations of TBT result in toxic effect on the transcriptional and translational levels. The target genes likely belong to a specific category of transcription- and translation-associated genes rather than to other gene categories.

Prenconcentration and Determination of Mercury (II) and Methylmercury in Waters by Immobilized 1,5-Diphenylcarbazone and Cold Vapor Atomic Absorption Spectrometry

  • 1,5-diphenylcarbazone was immobilized on sodium dodecyl sulfate coated alumina. The alumina particle was effectively used for collection of mercury(II) and methylmercury cations at sub-ppb level. The adsorbed mercury was eluted with l mol $L^{-1}$ of hydrobromic acid solution. The mercury(II) was then directly measured by cold vapor atomic absorption spectrometry utilizing tin (II) chloride where as the total mercury was determined after the oxidation of methylmercury into the inorganic mercury. The methylmercury concentration was calculated by the difference between the value of total mercury and mercury (II). Mercury (II) and methylmercury cations were completely recovered from water with a preconcentration factor of 100 (for 1 L solution.) Relative standard deviation at Hg L ${\mu}gL^{-1}$ level 1.7%(n=8) and the limit of detection was 0.11 ${\mu}gL^{-1}$. The procedure was applied to spring water, well water and seawater and accuracy was assessed through recovery experiments.

  • When concrete structures are exposed under marine condition for a long time, the steel in concrete is corroded due to the ingression of chlorides in the seawater. Because the damages of corrosion resulting from the chloride ion are very serious, many researches have been performed. However evaluation on chloride attack of concrete structures are not fully examined, recently. Therefore, the objective of this paper is to study the applicability of colormetric method. For the purpose of this, reaction mechanism of colormetric method were investigated, and the colormetric method is applied for marine concrete structures. According to the test results, the diffusion coefficient by colormetric method is not so different to the value of chloride concentration profile test. It is confirmed that the colormetric method is useful tool for estimating the chloride of concrete structures in situ. The average chloride amount of colored parts indicates 0.9kg/$m^{3}$ per concrete unit weight.

Nitrification Efficiency in Fixed Film Biofilters using Different Filter Media in Simulated Seawater Aquarium System

  • Nitrification efficiency of fixed film biofilters with sand, loess bead, and styrofoam bead in biofilter columns of 1-m height and 30 cm width was studied. Synthetic wastewater was continuously supplied to the culture tank to maintain total ammonia nitrogen (TAN) concentration in the inflow water at around 8 mg/L. The hydraulic loading rate was set at 200 ㎥/$m^2$/day. TAN conversion was stabilized after about 90 day conditioning for all the selected filter media but with net accumulations of nitrite. On the volumetric basis, conversion rates of TAN and nitrite were the highest in styrofoam bead filter. Mean volumetric TAN conversion rates in the final samples were 682, 269, and 79 g TAN/㎥/day in the styrofoam bead, sand and loess bead filters, respectively. Low gravity and cost of styrofoam bead render the handling easier and more cost-effective.

  • For the effective treatment of shipboard sewage continuously, a non-diaphragm electrolytic treatment device using DSA type insoluble electrode, Ti/IrO2, anode and H-C metal cathode, was studied. The most effective electrolytic conditions were obtained when cell clearance, 6mm, pH 5-6 and the concentration of seawater, more than 20% as batch test results. The COD removal rate was varied in logarithmic function, showed as C=Coe-KE and the required current was E = A/QCo [A.min/mgCOD]. When the COD removal effeciency was more than 90%, the electrolytic reaction constant was 0.02.

  • Marine ecosystems are frequently exposed to a variety of chronic and acute pollutants derived from anthropogenic production and consumption activities. Mussels are sessile (can provide location-specific information), medium-sized (have enough tissue matrix for chemical analysis) filter-feeders (show accumulation of pollutant chemicals from seawater). These biological and ecological characteristics make mussels virtually ideal for pollution monitoring. In this study, Korean mussels (Mytilus coruscus) were collected from nine different sites situated along the coasts of the western sea to the eastern sea of Korea in 2017. Total mercury concentration was highest (mean ${\pm}$ standard error, $92.7{\pm}3.5ng/g\;dry$) on Baengnyeongdo Island in the western sea, and the sites in the southern sea showed the lowest mean concentrations (42.3 - 44.5 ng/g dry). These results were discussed in terms of possible pollution sources.

  • An experiment was conducted to evaluate the biochemical adverse effect of increased carbon dioxide in seawater on marine polychaete, Perinereis aibuhitensis. We measured the available energy reserves, Ea (total carbohydrate, protein, and lipid content) and the energy consumption, Ec (electron transport activity) of Perinereis aibuhitensis exposed for 7-d to a range of $CO_2$ concentration such as 0.39 (control =390 ppmv), 3.03 (=3,030 ppmv), 10.3 (=10,300 ppmv), and 30.1 (=30,100 ppmv) $CO_2$ mM, respectively. The cellular energy allocation (CEA) methodology was used to assess the adverse effects of toxic stress on the energy budget of the test organisms. The results of a decrease in CEA effect of increased carbon dioxide in seawater from all individual in Ea and Ec. Increase of carbon dioxide reduced pH in seawater, significantly. The chemical changes in sea- water caused by increasing $pCO_2$ might cause stresses to test organisms and changes in the cellular energy allocations. Results of this study can be used to understand the possible influence of $CO_2$ concentration increased by the leakage from sub-sea bed storage sites as well as fossil fuel combustion on marine organisms.
