근관 충전용 Sealer가 수종의 세포에 미치는 독성효과에 관한 연구 (A STUDY ON THE CYTOTOXICITY OF ROOT CANNAL SEALERS TO SEVERAL CELL LINES)

  • This study was performed to evaluate and compare the cytotoxic effects of five root canal sealers to several different cell lines. Five root canal sealers were AH-26, N2, Sealapex, Tubliseal, and Vitapex. Each sealers were mixed according to the manufacturer's instructions, and culture media were added to each sealers immediately after mixing (the immediate group) and after three days (the third day group) and seven days (the seventh day group) respectively. And every sealer solutions were diluted to 1:1, 1:2, 1:3 and 1:4. Three different permanent cell lines (HEp-2, McCoy, MRC-S) and human gingival fibroblasts and mononuclear cells were challenged by each sealer solution and the cytopathic effects were evaluated using MTT-ELISA, MTT-microscopy, and lactate dehydrogenase (LD) activity. The results were as follows: 1. In HEp-2 and MRC-5 cells, Vitapex was the least cytotoxic sealers. 2. AH-26 showed mild cytotoxic effects to HEp-2, gingival fibroblast and mononuclear cells. 3. N2 was the most toxic sealer to gingival fibroblast and it showed relatively strong cytotoxicity to HEp-2, McCoy and MRC-S cells. 4. Tubliseal showed strong cytotoxic effects to HEp-2, McCoy, MRC-S, and mononuclear cells. 5. Sealapex showed strong cytotoxic effect to HEp-2, McCoy, and gingival fibroblasts.

근관내(根管內) 근관충전제(根管充塡劑)의 유입성(流入性)에 관(關)한 실험적(實驗的) 연구(硏究) (AN EXPERIMENTAL STUDY ON THE FLOW OF CANAL SEALER BY MEANS OF LENTULO)

  • Artificial canals were prepared by means of engine reamer on the slab which was made of clear acrylic resin. The shape of this canal is similar to the natural canal, the size is almost same as Kerr File No. 70, and the length is about 20mm. Sealer, Zine oxide eugenol mixture was prepared as matched to spatula test. Lentulo filler was inserted as deep as middle portion and apical one-third portion of the canal and the dental engine was driven in the speed of 1,000 and 3,000 r.p.m. in 30 seconds and 60 seconds with feeding of the sealer. The author measured the excess flow of the canal sealer from Lentulo end. Following are the results obtained from this study. 1. Lentulo insertion at the middle portion 1,000 r.p.m. in 30 seconds case: 0.21mm Same as above in 60 seconds: 0.20mm 2. Lentulo insertion at apical-thirds. 1,000 r.p.m. in 30 seconds: 0.22mm. Same as above in 60 seconds: 0.20mm. 3. Lentulo insertion at middle portion. 3,000 r.p.m. in 30 seconds: 0.14mm Same as above in 60 seconds: 0.25mm. 4. Lentulo insertion at apical-thirds, 3,000 r.p.m. in 30 seconds: 0.11mm Same as above in 60 seconds: 0.27mm.

  • Recently, due to the aging of workers and the weakening of the labor base in the automobile industry, research on quality inspection methods through ICT(Information and Communication Technology) convergence is being actively conducted. A lot of research has already been done on the development of an automated system for quality inspection in the manufacturing process using image processing. However, there is a limit to detecting defects occurring in the automotive sunroof sealer application process, which is the subject of this study, only by image processing using a general camera. To solve this problem, this paper proposes a system construction method that collects image information using a infrared thermal imaging camera for the sunroof sealer application process and detects possible product defects based on the SVM(Support Vector Machine) algorithm. The proposed system construction method was actually tested and applied to auto parts makers equipped with the sunroof sealer application process, and as a result, the superiority, reliability, and field applicability of the proposed method were proven.

Influence of size and insertion depth of irrigation needle on debris extrusion and sealer penetration

  • Objectives: To determine the effect of size and insertion depth of irrigation needle on the amount of apical extruded debris and the amount of penetration depth of sealer using a confocal laser scanning microscope (CLSM). Materials and Methods: Twenty maxillary premolars were assigned to 2 groups (n = 10), according to the size of needle tip, 28 G or 30 G. Buccal roots of samples were irrigated with respective needle type inserted 1 mm short of the working length (WL), while palatal roots were irrigated with respective needle type inserted 3 mm short of the WL. Prepared teeth were removed from the pre-weighed Eppendorf tubes. Canals were filled with F3 gutta-percha cone and rhodamine B dye-labeled AH 26 sealer. Teeth were transversally sectioned at 1 and 3 mm levels from the apex and observed under a CLSM. Eppendorf tubes were incubated to evaporate the irrigant and were weighed again. The difference between pre- and post-weights was calculated, and statistical evaluation was performed. Results: Inserting needles closer to the apex and using needles with wider diameters were associated with significantly more debris extrusion (p < 0.05). The position of needles and level of sections had statistically significant effects on sealer penetration depth (p < 0.05 for both). Conclusions: Following preparation, inserting narrower needles compatible with the final apical diameter of the prepared root canal at 3 mm short of WL during final irrigation might prevent debris extrusion and improve sealer penetration in the apical third.

초음파 근관기구를 이용한 근관충전법의 근관폐쇄능에 관한 주사 전자현미경적 연구 (A SCANNING ELECTRON MICROSCOPIC EVALUATION OF THE ROOT CANAL FILLING WITH ULTRASONIC ENDODONTIC INSTRUMENT)

  • The purpose of this study was to evaluate the adaptation of root canal filling material to the dentinal wall of root canal and to compare the sealing ability of the root canal filling materials using ultrasonic endodontic instrument with injection-molded thermoplasticized gutta-percha filling method and lateral condensation method. Fifty fresh human single root exlracted for orthodontic treatment, were randomly selected, and instrumented by step-back technique. And then, the teeth were divided into 5 groups according to each root canal filling methods. In the experimental group 1 and group 2, the root canals were filled with gutta perdia cases using ultrasonic instrument with and without sealer. In the experimental group 3 and 4, using jection-moldeed thermoplasticized gutta-percha method by obtul$^{(R)}$ canals were filled with and without sealer. In the control group, the canals were filled with sealer by lateral candensation. And then, 5 teeth of each group were immersed in black Indian ink, decalcified and cleared. The depth of dye penetration into the root canal were evaluated with stereoscope (Reichert Ltd., USA). Among the 5 teeth remaining in each group, the single longituding grooves were made on the labial and lingual root surfaces and then immersed in the liquid nitrogen to fracture the teeth spontaneously without any distortions of gutta-percha. Each specimens were examined with X-650 Scanning Electron Microscope(Hitachi ltd, Japan) to show the adaptation to the canal wall, void, homogenicity of filling material and location of gutta-percha or sealer in the dentinal tubules of the root canal. The observations were as follows : 1. The experimental group 1 showed smaller mean dye penetration than control group, and showed the penetraton of sealer in the dentinal tubules of apical third of the root canal. 2. The experimental group 2 and group 4 showed the penetration of gutta-percha in the dentinal tubules of root canals. 3. The experimental group 1 and group 3 showed less mean dye penetration than the experimental group 2 and group 4. 4. The experimental group 1 and group 2 showed better adaptation of filling materials than control group.

Push-out bond strength and dentinal tubule penetration of different root canal sealers used with coated core materials

  • Objectives: The aim of this study was to compare the push-out bond strength and dentinal tubule penetration of root canal sealers used with coated core materials and conventional gutta-percha. Materials and Methods: A total of 72 single-rooted human mandibular incisors were instrumented with NiTi rotary files with irrigation of 2.5% NaOCl. The smear layer was removed with 17% ethylenediaminetetraacetic acid (EDTA). Specimens were assigned into four groups according to the obturation system: Group 1, EndoRez (Ultradent Product Inc.); Group 2, Activ GP (Brasseler); Group 3, SmartSeal (DFRP Ltd. Villa Farm); Group 4, AH 26 (Dentsply de Trey)/gutta-percha (GP). For push-out bond strength measurement, two horizontal slices were obtained from each specimen (n = 20). To compare dentinal tubule penetration, remaining 32 roots assigned to 4 groups as above were obturated with 0.1% Rhodamine B labeled sealers. One horizontal slice was obtained from the middle third of each specimen (n = 8) and scanned under confocal laser scanning electron microscope. Tubule penetration area, depth, and percentage were measured. Kruskall-Wallis test was used for statistical analysis. Results: EndoRez showed significantly lower push-out bond strength than the others (p < 0.05). No significant difference was found amongst the groups in terms of percentage of sealer penetration. SmartSeal showed the least penetration than the others (p < 0.05). Conclusions: The bond strength and sealer penetration of resin-and glass ionomer-based sealers used with coated core was not superior to resin-based sealer used with conventional GP. Dentinal tubule penetration has limited effect on bond strength. The use of conventional GP with sealer seems to be sufficient in terms of push-out bond strength.

Glass Ionomer Root Canal Sealer의 치근단 미세누출에 관한 연구 (A STUDY ON THE APICAL MICROLEAKAGE OF GLASS IONOMER ROOT CANAL SEALER)

  • The purpose of this study was to compare the degree of micro- leakage of new glass ionomer root canal sealer, Ketac-Endo(ESPE Co., Seefeld, Germany) with that of AH-26(De Trey Co., Ltd., U.S.A.). Root canal treatment using K -file, H -file, Gate Glidden drill was conducted on 49 extracted single-rooted teeth. 45 teeth were randomly divided into 3 experimental groups(15 teeth per group) and 4 teeth were used as the control group. Group 1 was used AH -26 sealer with the lateral condensation method for canal filling, group 2 was used Ketac-Endo with the single cone method and group 3 was used Ketac-Endo with the lateral condensation method. The control group was obturated with the single cone method without sealer. The teeth were covered with two coats of nail varnish after 48 hours of obturation. The teeth were immersed in India ink for 1 week and cleaned with methyl salicylate and then the degree of dye penetration were measured with stereomicroscope. The data were analyzed statistically by one-way ANOVA. The results were as follows: 1. 7 teeth in group 1, 5 in group 2, and 3 in group 3, were showed evidence of microleakage implying appropriate canal filling. 2, The mean average of microleakage was $0.17{\pm}0.32mm$ in group 1, $0.30{\pm}0.37mm$ in group 2. $0.10{\pm}0.21mm$ in group 3, showing that canal filling using the lateral condensation canal filling method with Ketac-Endo showed the least microleakage and using the single cone method with Ketac-Endo showed the largest amount of microleakage, 3. There were no statistically significant difference in the variation of microleakage among groups. From the results above, Ketac-Endo which has the advantage of glass ionomer, whether using the single cone method or the lateral condensation method, showed similar results as AH-26, but for clinical application it is thought that were studies on the properties of Ketac-Endo should be followed.

혐기성세균모델을 이용한 봉함제(Sealer)의 미세누출에 관한 연구 (LEAKAGE EVALUATION OF SEVERAL SEALERS USING ANAEROBIC BACTERIAL LEAKAGE MODEL)

  • The purpose of this in vitro study was to evaluate the sealing ability of three sealers(Sealapex, Pulp canal sealer, AH26) used with continuous wave method using an anaerobic bacterial leakage model. 53 extracted human teeth with straight and single canals were prepared with crown-down pressureless technique using .04, .06 taper Profile(Maillefer, Swiss). Master apical file was maintained as #35 K-file. All canals of the experimental teeth were obturated with continuous wave method using System B(Analytic technology, U.S.A.) The teeth were randomly divided into three experimental groups of 15 and two control groups of 4. Experimental group 1 was obturated with Sealapex and group 2 with Pulp canal sealer, and group 3 with AH26. A dual chamber anaerobic bacterial leakage model was assembled. Brain heart infusion with yeast extract, hemin, menadion, and the chromogenic indicator bromocresol purple was used as the culture broth for Fusobacterium nucleatum(VPI 10197), The specimens were incubated in anaerobic chamber at $37^{\circ}C$ and were observed every 2 to 3 clays, The coronal leakage was evaluated through the color change of culture broth in lower chamber for 60 days. The results were as follows: 1. The incidence of bacterial leakage in group 1 (Sealapex group was 80%, 53% in group 2 (Pulp canal sealer), 27% in group 3 (AH26). 2. There were statistically significant differences in leakage scores between group 1 and group 2, and between group 1 and group 3, respectively. (P<0.05) 3. There was no significantly difference in leakage score between group 2 and group 3. (P>0 05)

Effects of radiation therapy on the dislocation resistance of root canal sealers applied to dentin and the sealer-dentin interface: a pilot study

  • Objectives: This study evaluated and compared the effects of radiation therapy on the dislocation resistance of AH Plus and BioRoot RCS applied to dentin and the sealer-dentin interface. Materials and Methods: Thirty single-rooted teeth were randomly assigned to 2 groups (n = 15 each): AH Plus (Dentsply DeTrey) and BioRoot RCS (Septodont). Each group was subdivided into control and experimental groups. The experimental group was subjected to a total radiation dose of 60 Gy. The root canals of all samples were cleaned, shaped, and obturated using the single-cone technique. Dentin slices (1 mm) were sectioned from each root third for the push-out test and scanning electron microscopy (SEM) was done to examine the sealer-dentin interface. The failure mode was determined using stereomicroscopy. Bond strength data were analyzed by the independent t-test, 1-way analysis of variance, and the Tukey post hoc test (α = 0.05). Results: Significantly lower bond strength was observed in irradiated teeth than non-irradiated teeth in the AH Plus group (p < 0.05). The BioRoot RCS group showed no significant reduction in bond strength after irradiation (p > 0.05) and showed a higher post-irradiation bond strength (209.92 ± 172.26 MPa) than the AH Plus group. SEM revealed slightly larger gap-containing regions in irradiated specimens from both groups. Conclusions: The dislocation resistance of BioRoot RCS was not significantly changed by irradiation and was higher than that of AH Plus. BioRoot RCS may be the sealer of choice for root canal treatment in patients undergoing radiation therapy.

  • This study deals with the effect on penetration properties of siliceous ion througth the mortar applicated by the waterproofing coating materials of siliceous seler liquid type. The tests of properties for reinforcing effect in mortar substrate surface layer are five kinds of water permeability, absorption, compressive strength, surface layer strength, pH content and chemical attack effect. Water permeability of mortar coated siliceous sealer in very than that of plane mortar. compressive strength of mortar coated siliceous sealer in larger than that of plane mortar about 10%.
