감성돔(Acanthopagrus schlegeli) 정자의 냉동보존에 미치는 희석액과 동해방지제의 영향 (Effects of Diluents and Cryoprotectants on Cryopreservation of Black Seabream (Acanthopagrus schligeli) Sperm)

  • 감성돔 (Acanthopagrus schlegeli) 정자의 냉동 보존시 정자의 생리활성과 보존효과를 비교하기 위하여, 순환여과 사육시스템에서 사육한 전장 $25.9{\pm}1.7$ cm, 체중$292.8{\pm}53.7$ g의 성어로부터 얻은 정액을 재료로 하여 희석액별, 동해방지제별 해동후 정자의 활성, 생존율 및 알에 대한 수정능력을 평가하였다. 희석액별 냉동보존에서 해동정자의 생존율은 3% sodium citrate에서 $80{\pm}1.4$%로 가장 높았으나, 수정률은 5.4% glucose에서 $63.4{\pm}4.4$%로 가장 높았다. 동해방지제별 냉동보존에서 해동정자의 수정률은 5~15% glycerol을 동해방지제로 사용하였을 때, 50.1~69.4%로 DMSO 보다 나은 효과를 보였다. 감성돔 정자의 냉동보존을 위한 적정 희석액과 동해방지제는 5% glucose와 5% glycerol이었다. 냉동하지 않은 감성돔 정자의 머리는 구형으로 직경 $1.5{\pm}0.1$ ${\mu}$m, 길이 $1.3{\pm}0.1$${\mu}$m였으며, 치밀한 핵질로 채워져 있었다. 편모는 전형적인 9+2 구조를 나타냈다. 그러나 냉동후 해동시킨 정자중에서는 염색질의 과립화, 세포막의 이탈에 의한 그 용적이 커지는 구조변화를 보였다.

Performance of Three Different Biofilter Media in Laboratory-Scale Recirculating Systems for Red Seabream Pagrus major Culture

  • Juvenile red seabream (mean body weight 29.0 g) were reared in recirculating culture systems with three different biofilter media, sand (SF), polystyrene microbeads (PF), and Kaldnes beads (KF). The efficiencies of the three different biofilter media were also tested. The SF was fluidized, and the PF and KF were trickled. All treatments were duplicated. The volumetric removal rates of total ammonia nitrogen by SF, PF, and KF were 193.8, 183.9, and 142.6 g $m^{-3}day^{-1}$, respectively, and those of nitrite nitrogen ($NO_2$-N) were 113.4, 105.9, and 85.8 g $m^{-3}day^{-1}$, respectively. The TAN and $NO_2$-N removal rates of KF were lower than those of SF and PF (P < 0.05), but there was no significant difference in these rates between SF and PF (P > 0.05). Among the biofilters used, only KF showed total suspended solid (TSS) removal capacity. The TSS removal efficiencies of SF and PF were negative. The growth rates of fish in SF were significantly higher than those in KF but not higher than those in PF. There was no difference in growth rate between fish in PF and KF. The specific growth rate and feed conversion efficiency of red seabreams in KF were lower than those in SF and PF, but there were no significant differences between SF and PF. These results indicate that sand and polystyrene microbeads are recommended for red seabream culture in a recirculating system.

연승어업에서 참돔의 섭식행동을 응용한 어획시스템의 성능 분석 (The Analysis of a Fishing System that Employs a Red Seabream Feeding Behavior in the Long Line Fishery)

  • In this study, we designed a fishing system to reproduce the underwater movement of the living Sword tip squid (Loligo edulis) used as bait in the red seabream long line fishery, and conducted the experiment of the fishing operation in 2 ways, i.e., a pole and line fishing method (fishermen) and a bait control fishing system used at fishing sites. Based on the catches in fishing operation, the experiment was conducted over a six times (2014 & 2015), and then 107 fishes were caught by the line fishing method while 57 fishes were caught by the bait control fishing system. The fishermen method actively controlled the speed of gear movement upward and downward while checking the reaction of red seabreams to the bite in the process which a jerk was transmitted to single line hook fishing gear manually. The bait control fishing system is a passive method which fishermen visually check only the movements at the end of fishing pole, enabled differentiation of bite reactions of red seabream during fishing operation. Thus, the difference between fishermen method and the bait control fishing system was found to about 53.3% in the catches. We confirmed the possibility of a site fishing operation based on the bait control fishing system designed newly as a result of this study. Improvement is in several areas for commercialization at the site. This fishing system is expected to be able to find wide-ranging applications as a new labor-saving method for the fishing red seabreams if it is applied to the fishing sites after aforesaid process.

저산소에 노출된 참돔(Pagrus major)의 개체 크기에 따른 내성 비교 (Comparison of tolerance of red seabream, Pagrus major exposed to hypoxia with body size)

  • 본 연구는 수온 24℃에서 체중 0.6-786 g 범위의 참돔 52개체의 SMR을 측정한 후 점진적인 저산소 조건에 노출시켜서 Scrit를 측정하여 개체 크기에 따른 저산소 내성을 평가할 목적으로 실시하였다. SMR (mg O2 kg-1hr-1)은 SMR=351.59·W-0.195 (r2=0.934, p<0.001)의 관계식으로 표시할 수 있었고, 체중(W) 증가에 따라 감소하는 경향을 보여주었다. 참돔의 체중과 Scrit 사이에는 Scrit (%)=2.316·logW+17.805 (r2=0.682, p<0.001)의 회귀직선식으로 나타낼 수 있었고, 체중 0.6-786 g인 개체의 Scrit는 17.3-24.4% 범위에서 개체 크기가 클수록 증가하여 작은 개체가 저산소 내성이 높은 것으로 나타났다.

조피볼락과 참돔의 표적 강도에 관한 연구 (Target Strength of Schlegel′s Black Rockfish (Sebastes schlegeli)and Red Seabream (Pagrus major))

  • 우리 나라 연안에서 주로 서식하고 있는 조피볼락(Schlegel's Black Rockfish, Sebastes schlegeli)과 참돔(Red Seabream, Pagrus major) 의 자원량을 추정할 때 보다 더 정확히 추정할 수 있는 기초 자료를 얻기 위하여 in situ TS 측정법과 유사한 환경 속에서 망과 3개의 주파수를 이용하여 어체의 크기와 자세별 및 주파수 특성에 따른 TS 에 대하여 조사하여 얻어진 결과를 요약하면 다음과 같다. 1. 전장(L, cm)에 따른 최대 TS 값을 이용한 조피볼락의 변환 계수(A) 값은 38kHz에서 -63.7dB, 120kHz에서 -64.4dB, 200kHz에서 -62.4 dB로 나타났으며, 참돔은 38kHz에서 -62.6dB, 120kHz에서 -65.4dB, 200kHz에서 -65..dB이었다. 또한, 평균 TS 값을 이용한 조피볼락의 변환 계수 값은 38kHz에서 -68.4dB, 120kHz에서 73.4dB, 200kHz에서 -70.8dB로 나타났으며, 참돔은 38kHz에서 -67.9dB, 120kHz에서 -72.7 dB, 200kHz에서 -73.4dB로 각각 나타났다. 2. 어체 1g당 최대 TS 값을 이용한 조피볼락의 변환 계수( B) 값은 38kHz에서 -52.0dB, 120 kHz에서 -52.7dB, 200kHz에서 -50.7dB로 나타났으며, 참돔은 38kHz에서 -50.9dB, 120kHz 에서 -53.7dB, 200kHz에서 -53.3dB이었다. 또한 평균 TS 값을 이용한 조피볼락의 변환 계수 값은 38kHz에서 -56.7dB, 120kHz에서 -61.7 dB, 200kHz에서 -59.IdB로 나타났으며, 참돔은 38kHz에서 -56.2dB, 120kHz에서 -61.0dB, 200 kHz에서 -61.6dB로 각각 나타났다. 3. 참돔 2마리를 대상으로 한 자세각 분포는 -26$^{\circ}$~25$^{\circ}$로 나타났으며, 그 때의 주파수에 따른 자세별 표적 강도는 38kHz에서 변화 폭이 비교적 적었으며, 120kHz에서는 Head down 되었을때 보다 Head up 되었을 때 3~6dB 정도 높게 나타났다.

여수 연안 승망 어장에서 어획된 어업생물의 계절별 분포특성 (Seasonal distribution characteristics of fishery creatures caught in funnel net fishing ground of the Yeosu coastal sea)

  • A study on the species composition and seasonal variations of fishery creatures caught by a funnel net was carried out in the coastal water off Dolsan Island, Yeosu from 2002 to 2003. During the study period, a total of 23 fishery creatures species were caught. Seabream(Acanthopagrus schlegeli), sea bass(Lateolabrax japonicus), mullet(Mugil cephalus), puffer(Takifugu niphobles) and rockfish(Sebastes inermis) predominated. These five species accounted for 85.1% of the total number of fishery creatures caught. Seasonal peaks of number of species occurred in summer, while those of number of individuals occurred in autumn. The lowest number of species and individuals were observed in winter. The large annual variation of diversity indices were observed from May to August. These large annual variation of diversity indices were mainly due to predominance of seabream, sea bass and mullet which accounted for most of all fishery creatures caught. The seasonal variations of fishery creatures showed that sea bass and mullet were caught mainly from spring to summer, seabream and puffer were caught mainly in autumn, and rockfish and brotula(Hoplobrotula armata) were caught mainly in winter.

Comparison of Growth Performance and Whole-body Amino Acid Composition in Red Seabream (Pagrus major) Fed Free or Dipeptide Form of Phenylalanine

  • This study was conducted to evaluate the efficacy of the dipeptide form of phenylalanine as a new source of amino acid in terms of growth performance and whole-body amino acid composition in comparison to the free form for red seabream (Pagrus major). Fish ($1.46{\pm}0.001g$) were fed four isonitrogenous and isocaloric experimental diets containing 0.7 or 1.4% phenylalanine either in free or dipeptide form. A feeding trial was carried out in three replicates and the fish were fed to apparent satiation for six weeks. At the end of the feeding trial, feed intake of fish was influenced by both phenylalanine form and level and significantly higher values were obtained at an inclusion level of 0.7% and by the use of dipeptide form. However, the other growth parameters did not significantly differ among treatments. Whole-body amino acid compositions revealed no significant changes in concentrations of both essential and non-essential amino acids regardless of the increase in phenylalanine levels or the use of its different forms. The finding in this study indicates that juvenile red seabream can utilize dipeptide phenylalanine as efficiently as free form without any undesirable effects on growth performance or whole-body amino acid composition.

Effects of Ammonia Concentration on Histological and Physiological Status in Black Seabream (Acanthopagrus schlegeli)

  • The histological changes of gill, liver, spleen and muscle, and respiration and blood variables and liver glycogen content were examined in black seabream, Acanthopagrus schlegeli. Fish were exposed to a high level of total ammonia nitrogen (10.4 mg/l) and recovered from exposure $(0.4{\pm}0.2mg/l)$ in a closed recirculating seawater system. In the process of exposure, mortality was $9\%$, and hyperplasia, necrosis or inflammation appeared in all tissues except for muscle. Oxygen consumption was decreased by $49\%$, and red blood cell (RBC) number, hematocrit and hemoglobin concentration were significantly decreased, while plasma glucose contents, activities of glutamate-oxaloacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT) increased. Liver glycogen content significantly increased from $6.6\%\;to\;10.4\%$. A large amount of hemosiderin was observed in the splenic tissue. During the recovery period, RBC number, hematocrit, hemoglobin concentration, GOT and GPT activities were returned to the normal status. Histological status of liver tissue was returned to the normal, but liver glycogen content was not recovered. During the recovery period, spleen melanin-macrophages temporarily increased, but subsequently decreased to the normal status.

질산성 질소가 감성돔(Acanthopagrus schlegelii) 치어에 미치는 급성 독성 영향 (Acute Nitrate Nitrogen Toxicity in Juvenile Blackhead Seabream Acanthopagrus schlegelii)

  • This study evaluated acute nitrate nitrogen ($NO_3-N$) toxicity in juvenile blackhead seabream Acanthopagrus schlegelii. Seventy juveniles (Trial A, $7.1{\pm}0.6g$) and nine juveniles (Trial B, $71.3{\pm}3.5g$) per 70 L tank were exposed to $NO_3-N$ concentrations of 0, 500, 1500, 2500, 3500, and 4500 mg/L and 0, 600, 1200, 1800, 2400, and 3000 mg/L, respectively, in triplicate for 7 days. In Trial A, all fish exposed to 3500 and $4500mg\;NO_3-N/L$ died within 48 h; in Trial B, all fish exposed to $3000\;NO_3-N/L$ died after 120 h. The $96\;LC_{50}$ and $168\;LC_{50}$ were 2505 and $1806mg\;NO_3-N/L$, respectively, in Trial A, and 2663 and $2377mg\;NO_3-N/L$ in Trial B. Large juveniles were more resistant to $NO_3-N$ than small juveniles. The results of acute $NO_3-N$ toxicity studies provide important data for subsequent chronic toxicity studies.

Effects of taurine supplementation in low fish meal diets for red seabream (Pagrus major) in low water temperature season

  • Background: Taurine is a conditional essential amino acid for fish. A study was conducted to investigate the compensating effect of supplemental taurine in diets for red seabream (Pagrus major) on impaired growth performance by fish meal (FM) replacement with soybean meal (SM) at low water temperature (14.15 ± 1.95 ℃). Methods: A FM-based diet was considered as a high FM diet and three other experimental diets were formulated to replace FM with SM by 20, 35, or 50% (HFM, SM20, SM35, or SM50, respectively) without taurine and other four diets were formulated by adding 1% taurine to the diets (HFM-T, SM20-T, SM35-T, or SM50-T, respectively). Triplicate groups of fish (108.9 ± 1.58 g/fish) were distributed into 24 polyvinyl circular tanks (215 L) with 20 fish per tank and fed one of the diets to satiation for 20 weeks. Results: Growth performance and feed utilization of red seabream were significantly improved by the dietary taurine supplementation. SM20-T and SM35-T diets increased fish growth that are comparable to HFM diet. Feed intake, feed conversion ratio, and protein efficiency ratio of fish fed SM20-T and SM35-T diets were not significantly different from those of HFM group. Dietary taurine supplementation in each FM replaced group numerically increased innate immunity of the fish. Lysozyme and superoxide dismutase activities were significantly decreased in fish fed SM35, SM50, and SM50-T diets compared to those of fish fed HFM diet while they were not significantly lower in SM20, SM20-T, SM35, and SM35-T groups. Glutathione peroxidase activity was significantly lower in fish group fed SM50 diet while SM50-T group did not significantly lower compared to that of HFM group. The relative expression level of hepatic IGF-1 mRNA was improved in fish fed taurine-supplemented diets compared to their respective SM diets. Conclusions: Growth performance and feed utilization of red seabream can be accelerated or restored by 1% taurine supplementation when they are fed high level of SM up to 35% in diets during low water temperature season.
