Effect of Mixed Oxidants and Sodium Hypochlorite on Pathogenic Microorganisms in Olive flounder Paralichthys olivaceus Aquaculture on Jeju Island (제주도 양식 넙치(Paralichthys olivaceus)에서 분리한 병원균 3종에 대한 Mixed Oxidant 및 차아염소산나트륨 살균효과)

  • Marine pathogenic bacteria, such as Streptococcus parauberis, Edwardsiella tarda and Vibrio harveyi, can cause lethal infections in farmed fish, ozone and antibiotics, are employed to sterilize waters used for rearing fish to mitigate this threat. The most widely used method is treatment with sodium hypochlorite solution. However, the maintenance of a constant concentration of chlorine in rearing waters can be difficult. We investigated the potential of a mixed oxidant (MO) solution generated by electrolysis of sea water to improve water quality. We measured the survival rates of fish pathogenic bacteria exposed to different concentrations of MO (0.5, 1.0, 1.5 and 2.0 MO) and sodium hypochlorite (0.5, 1.0, 1.5 and 2.0 ppm) for various lengths of time (0, 5, 10, 15, 20, 25 and 30 min). We found a time-dependent decrease in the survival rates of the tested pathogenic microorganisms. The sterilization effect of the MO solution on pathogenic organisms was greater than that of sodium hypochlorite for gram-negative and gram-positive bacteria. We conclude that MO solution produced by electrolysis could be used to maintain a constant chlorine concentration in aquaculture systems.

Detoxification of PSP and relationship between PSP toxicity and Protogonyaulax sp. (마비성패류독의 제독방법 및 패류독성과 원인플랑크톤과의 관계에 관한 연구)

  • The purpose of this study was to investigate the detoxifying effect on PSP-infested sea mussel, Mytilus edulis, by heating treatment and correlation between the PSP toxicity and the environmental conditions of shellfish culture area such as temperature, pH, salinity, density of Protogonyaulax sp. and concentration of inorganic nutrients such as $NH_4-N,\;NO_3-N,\;NO_2-N\;and\;PO_4-P$. This experiment was carried out at $Suj\u{o}ng$ in Masan, Yangdo in Jindong, $Hach\u{o}ng\;in\;K\u{o}jedo\;and\;Gamch\u{o}n$ bay in Pusan from February to June in $1987\~1989$. It was observed that the detection ratio and toxicity of PSP in sea mussel were different by the year even same collected area. The PSP was often detected when the temperature of sea water about $8.0\~14.0^{\circ}C$. Sometimes the PSP fox of sea mussel was closely related to density of Protogonyaulax sp. at $Gamch\u{o}n$ bay in Pusan from March to April in 1989, but no relationship was observed except above duration during the study period. The concentration of inorganic nutrients effects on the growth of Protogonyaulax sp., then effects of $NO_3-N$ was the strongest among them. When the PSP-infested sea mussel homogenate was heated at various temperature, the PSP toxicity was not changed significantly at below $70^{\circ}C$ for 60 min. but it was proper-tionaly decreased as the heating temperature was increased. For example, when the sea mussel homogenate was heated at $100^{\circ}C,\;121^{\circ}C$ for 10 min., the toxicity was decreased about $67\%\;and\;90\%$, respectively. On the other hand, when shellstock sea mussel contained PSP of $150{\mu}g/100g$ was boiled at $100^{\circ}C$ for 30 min. with tap water, the toxicity was not detected by mouse assay, but that of PSP of $5400{\mu}g/100g$ was reduced to $57{\mu}g/100g$ even after boiling for 120 min.

Reduction of Organic Sludge using High Efficiency Aeration System(HEAS) (고효율 포기 시스템을 이용한 유기성 슬러지의 감량화)

  • In Korea, ocean dumping has been widely used as the ultimate disposal of sewage sludge. However, ocean dumping of food wasted and sewage sludge from 2013 is expected to legally restricted as London convention on marine pollution prevention has been effective in 2009. This research aims to examine the effect of HEAS in treating the environmental pollution load caused by organic high concentrated sludge. Thus, onsite laboratory scale treatability test using HEAS was adopted to treat the high concentrated organic sludge from sewage and industrial wastewater treatment plant. The research results showed that the HEAS is useful to reduce the environmental pollution caused by organic high concentrated sludge. Specific results are as follows. 1. The organic removal after the sludge digestion using the high efficiency aeration system was 55.2-85.8%. Although these results were lower than those from the general sewage treatment, the high efficiency aeration system could be evaluated as efficient, considering the object sludge contained the industrial waster water. 2. The average removal efficiency was about 25.2%. 3. It was revealed that sludge digestion by the high efficiency aeration system could effectively contribute to the sludge treatment cost. Especially, the high efficiency aeration system is more applicable to the onsite treatment of small sewage and wastewater treatment plant that contains high solid content sludge, industrial wastewater sludge, high fixed solid sludge.

Effects of a Carbohydrase Mixture, Ultrasound, and Irradiation Treatments on the Physical Properties of Defatted Mustard Meal-based Edible Films (탈지 겨자씨로 제조한 가식성 생고분자 필름의 물리적 특성에 대한 탄수화물 가수분해 효소 혼합체, 초음파, 그리고 방사선 처리의 효과)

  • Effects of depolymerization treatments of a carbohydrase mixture (CM), ultrasound, and irradiation on the physical properties of defatted mustard meal-based edible films (DMM films) were investigated. DMM hydrocolloids were added to CM (0.42% (w/w solution)), treated by ultrasound (500-700 W, 10-30 min) or ${\gamma}$-ray (40-100 kGy) to prepare film-forming solutions. Films were formed by drying. The CM treatment at 0.42% (w/w), pH 5.5, and 40-$50^{\circ}C$ with a 0.5 hr incubation time resulted in the highest colloidal stability in the film-forming solution. The depolymerization treatments did not dramatically change the water vapor permeability of the films. The solubility of the film decreased up to 53.1% by the CM treatment. The ultrasound treatment (700 W-30 min) decreased tensile strength and elongation. The ultrasound treatment (600 W-20 min) resulted in more compact and uniform structures of the films. Flavor profiles were differentiated by the power level and the time of the ultrasound treatment.

Operating Status and Improvement Plans of Ten Wetlands Constructed in Dam Reservoirs in Korea (국내 10개 댐저수지 인공습지의 운영현황 및 개선방안)

  • To propose the improvement and management plans to strengthen the pollutant removal efficiency of dam reservoir's constructed wetlands(CWs), the operation status and configuration of CWs (including water depth, operational flow, water flow distribution, residence time, and pollutant removal efficiency, aspect ratio, open water/vegetation ratio etc.) were analyzed in 10 major wetlands constructed in dam reservoirs. The pollutant concentrations in the inflows of the studied CWs were lower than those of American and European constructed wetlands. Especially, organic matter concentrations in all of inflows were below 3 mg/L(as BOD) due to advanced treatment of sewage disposal plant and an intake of low concentration water during dry and normal seasons. The average removal efficiency of total nitrogen(TN) and total phosphorus(TP) for 10 CWs ranged from 7.6~67.6%(mean 24.9%) and -4.9~74.5%(mean 23.7%), respectively, showing high in wetlands treating municipal wastewater. On the other hand, the removal efficiency of BOD was generally low or negative with ranging from -133.3 to 41.7%. From the analysis of the operation status and configuration of CWs, it is suggested that the low removal efficiency of dam reservoir's CWs were caused by both structural (inappropriate aspect ratio, excessive open water area) and operational (neglecting water-level management, lack of facilities and operation for first flush treatment, lake of monitoring during rainy events) problems. Therefore, to enable to play a role as a reduction facility of non-point source(NPS) pollutants, an appropriate design and operation manuals for dam reservoir's CW is urgently needed. In addition, the monitoring during rainy events, when NPS runoff occur, must be included in operation manual of CW, and then the data obtained from the monitoring is considered in estimation of the pollutant removal efficiency by dam reservoir's CW.

Viability Test and Bulk Harvest of Marine Phytoplankton Communities to Verify the Efficacy of a Ship's Ballast Water Management System Based on USCG Phase II (USCG Phase II 선박평형수 성능 평가를 위한 해양 식물플랑크톤군집 대량 확보 및 생물사멸시험)

  • The type approval test for USCG Phase II must be satisfied such that living natural biota occupy more than 75 % of whole biota in a test tank. Thus, we harvested a community of natural organisms using a net at Masan Bay (eutrophic) and Jangmok Bay (mesotrophic) during winter season to meet this guideline. Furthermore, cell viability was measured to determine the mortality rate. Based on the organism concentration volume (1 ton) at Masan and Jangmok Bay, abundance of ${\geq}10$ and $<50{\mu}m$ sized organisms was observed to be $4.7{\times}10^4cells\;mL^{-1}$and $0.8{\times}10^4cells\;mL^{-1}$, and their survival rates were 90.4 % and 88.0 %, respectively. In particular, chain-forming small diatoms such as Skeletonema costatum-like species were abundant at Jangmok Bay, while small flagellate ($<10{\mu}m$) and non chain-forming large dinoflagellates, such as Akashiwo sanguinea and Heterocapsa triquetra, were abundant at Masan Bay. Due to the size-difference of the dominant species, concentration efficiency was higher at Jangmok Bay than at Masan Bay. The mortality rate in samples treated by Ballast Water Treatment System (BWMS) (Day 0) was a little lower for samples from Jangmok Bay than from Masan Bay, with values of 90.4% and 93%, respectively. After 5 days, the mortality rates in control and treatment group were found to be 6.7% and >99%, respectively. Consequently, the phytoplankton concentration method alone did not easily satisfy the type approval standards of USCG Phase II ($>1.0{\times}10^3cells\;mL^{-1}$ in 500-ton tank) during winter season, and alternative options such as mass culture and/or harvesting system using natural phytoplankton communities may be helpful in meeting USCG Phase II biological criteria.

A Tracer Study on Mankyeong River Using Effluents from a Sewage Treatment Plant (하수처리장 방류수를 이용한 추적자 시험: 만경강 유역에 대한 사례 연구)

  • We investigated the possibility of using effluents from a municipal sewage treatment plant (STP) as tracers a tracer for hydrologic studies of rivers. The possibility was checked in a 12-km long reach downstream of Jeonju Municipal Sewage Treatment Plant (JSTP). Time-series monitoring of the water chemistry reveals that chemical compositions of the effluent from the JSTP are fluctuating within a relatively wide range during the sampling period. In addition, the signals from the plant were observed at the downstream stations consecutively with increasing time lags, especially in concentrations of the conservative chemical parameters (concentrations f3r chloride and sulfate, total concentration of major cations, and electric conductivity). Based on this observation, we could estimate the stream flow (Q), velocity (v), and dispersion coefficient (D). A 1-D nonreactive solute-transport model with automated optimization schemes was used for this study. The values of Q, v, and D estimated from this study varied from 6.4 to $9.0m^3/sec$ (at the downstream end of the reach), from 0.06 to 0.10 m/sec, and from 0.7 to $6.4m^2/sec$, respectively. The results show that the effluent from a large-scaled municipal STP frequently provides good, multiple natural tracers far hydrologic studies.

STUDIES ON LABORATORY CULTURE OF FREE-LIVING CONCHOCELIS OF PORPHYRA AND METHODS OF MONOSPORE LIBERATION (김의 선발육성을 위한 기초조사 1. 김의 유리계상체 배양 및 유체의 단포자방출)

  • In order to find effective seed collection method from cultivated Porphyra, benthic diatom elimination, culturing conditions of Conchocelis, liberation of conchospores and treatment of the fronds to obtain monospores have been studied. Elimination of benthic diatoms from Porphyra fronds is successfully performed by careful brushing the fronds in sea water and freshwater alternatively. For the culturing of Conchocelis Schleiber's solution enriched with only soil extracts, vitamins and Fe-EDTA was satisfactory. Growth under 16 hours illumination is 1.29 times faster than those under 10 hours illumination. When the culturing water was airated the growth was $1.41\~l.50$ times faster than the growth in stagnant water. Total amount of conchospores liberated from Conchocelis which has been cultured under airation was much more than those of conchospores under stagnant condition. Effective liberation of monospores was observed in the fronds which have been dried in air for 6 hours $(21.23\~24.19\%\;water\;content)$.

Eutrophication and Freshwater Red-tide Algae on Early Impoundment Stage of Jeolgol Reservoir in the Paikryeong Island, West Sea of South Korea (백령도 절골저수지의 부영양화와 담수적조)

  • A systematic water quality survey was conducted in August, 2005 for a drinking water supply reservoir (the Jeolgol reseuoir located in an island), which is at an early stage of impoundment, to investigate the causes of water color deterioration of the reservoir and the clogging of filter beds of a water treatment plant. The reservoir shape was simple and its average depth was 5.5 m, increasing from upreservoir toward the downreservoir end near the dam. Dissolved oxygen (DO) and chloropllyll-a (chi-a) showed a large variation while water temperature had a smaller range. Transparency ranged from 0.6 to 0.9 m (average 0.7 m). The average value of turbidity was 9.3 NTU, ranging from 8.0 ${\sim}$ 12.1 NTU. The transparency and the turbidity appear to be affected by a combination of biological and non-biological factors. The poor transparency was explained by an increase of inorganic colloids and algal bloom in the reservoir. The blockage of the filter bed was attributed to the oversupply of phytoplanktons from the reservoir. The range and the average concentration of chi-a within the reservoir were 31.6 ${\sim}$ 258.9 ${\mu}g\;L^{-1}$, 123.6 ${\mu}g\;L^{-1}$ for the upper layer, and 17.0 ${\sim}$ 37.4 ${\mu}g\;L^{-1}$, 26.5 ${\mu}g\;L^{-1}$ for the bottom layer, respectively. A predominant species contributing the algal bloom was Dinophyceae, Peridinium bipes f. occultatum. The distribution of Peridinium spp. was correlated with chi-a concentrations. The standing crop of phytoplankton was highest in the upreservoir with $8.5\;{\times}\;103\;cells\;mL^{-1}$ and it decreased toward the downresevoir. Synedra of Bacillariophyceae and Microcystis aeruginosa of Cyanophyceae appeared to contribute to the algal bloom, although they are not dominated. It is mostly likely that sloped farmlands located in the watershed of the reservoir caused water quality problems because they may contain a significant amount of the nutrients originated from fertilizers. In addition, the aerators installed in the reservoir and a shortage of the inflowing water may be related to the poor water quality. A long-term monitoring and an integrated management plan for the water quality of the watersheds and the reservoir may be required to improve the water quality of the reservoir.

Functional Magnetizing Treatment of Natural Quartz and Volcanic Lava Scoria (내추럴 퀄쯔와 화산암재 스코리아의 기능성 마그네타이징 처리)

  • The non-magnetic materials with non-conductive showing high structure dispersity were developed on the base of natural quartz and lava-scoria which was collected from Je-ju island in Korea, and treated by methane-chemical technology those were obtained novel properties of magnetization through the analyzing. Depending on the processing conditions and subsequent applications the materials produced by strong methane-chemical reaction (MCR) in alcohol solution showed concurrently magnetic, dielectric and electrical properties. The obtained magnetic-electrical powders classified by aggregate complex of their features as segnetomagnetics, containing a dielectric material as a carrying nucleus, particularly the quartz on that surface one or more layers of different compounds were synthesized having thickness up to 10~50 nm and showing magnetic, electrical and other properties. It was confirmed in magnetizing process that powders of quartz and lava-scoria produced by MCR were better oil adsorbent as of oleophilic and floating matter on water surface although their specific gravities are comparably more than 1 in quartz or less than unity, as that of water, in lava-scoira. Therefore, it will be Possible and very useful to remove low density and light gravity oil spillage in difficult recovery from sea and inland water contamination spread on water surface, by marine accident and ship sinking accident occurring frequently in recent years, by way of magnetic adsorbent conveyer system in continuous, if it could be built up the mass Production system of water-floating magnetizable oleophilic adsorbent materials with use of iow cost and good Qualify lava-scoria spread on volcano district in Je-ju island. And, there will also be urgent advent of necessity with strong possibility to develop useful applications of various magnetic functional materials include oleophilic adsorbent for removal of sea oil-contaminants and maritime pollutants, and other kinds of various utilities in industrial applications and practical uses of novel functional materials in the fields of environments and health care applications with in deep expectation.
