황금탕 및 황금추출물의 식중독 세균들에 대한 생육억제 효과 (Antimicrobial Activity of Whangkumtang Extract and Scutellariae Radix Extract on the Food-Borne Pathogens)

  • 각종 식중독의 원인균으로 중요시되는 11종의 균주에 대해서 황금탕 및 황금 추출물의 항균력을 검토한 결과, 황금탕 추출물은 Staphylococcus aureus, Bacillus cereus, Shigella sonnei, Shigella flexneri, Salmonella Typhimurium, Salmonella Enteritidis, Vibrio parahaemolyticus, Escherichia coli O111 및 Escherichia coli O126에서 항균력을 나타내었고, 황금 추출물은 Staphyoccus aureus, Bacillus cereus, Shigella sonnei, Shigella flexneri, Salmonella Typhimurium, Salmonella Enteritidis, Vibrio parahaemolyticus, Escherichia coli O111, Escherichia coli O126 및 Escherichia coli O157에서 항균력을 나타내었다. Listeria monocytogenes에 대해서는 황금탕 및 황금 추출물 모두 항균력이 없었고, Escherichia coli O157에 대해서는 황금탕 추출물은 항균력이 없었으나, 황금 추출물은 항균력을 나타내었다. 황금탕 추출물의 최소저해농도는Staphyloccus aureus와 Bacillus cereus에서 40 mg/mL, Shigella flexneri 와 Salmonella Enteritidis에서 100 mg/mL이었고, 황금 추출물의 최소저해농도는 Bacillus cereus에서 10 mg/mL, Staphylococcus aureus, Shigella flexneri 및 Salmonella Enteritidis에서 20 mg/mL이었으며, 황금탕 및 황금 추출물 모두 Staphyloccus aureus와 Bacillus cereus에 대해 48시간까지 배양하는 동안 최소저해농도 이상에서는 균이 거의 생육하지 못하였다. 이상에서 황금탕 추출물보다는 황금 추출물의 항균력이 더 강하며, 황금탕 추출물의 식중독 세균들에 대한 항균력은 황금에 의한 것으로 생각된다. 또한, 항균력이 확인된 세균으로 인한 각종 식중독에 황금탕의 사용 가능성을 고려할 만하며, 황금 추출물에 대한 천연 식품 보존료로서의 이용 가능성을 검토할 가치가 있다고 사료된다.

LC-MS를 이용한 황금추출물의 항균물질 검색 (Analysis of Antimicrobial Substance isolated from Scutellariae Radix Extract using LC-MS)

  • 황금의 천연항균제 개발을 위하여 황금의 일반성분 및 미량물질들을 분석하였으며 항균활성물질의 검색을 위하여 LC-MS를 이용하였고 검색되어진 성분들에 대해서는 항균력 비교시험을 하였으며 항균활성물질들에 대해서는 HPLC를 이용하여 그 양을 정량하였다. 황금의 일반성분으로는 수분 $8.93\%$, 탄수화물 $67.93\%$, 조회분 $4.13\%$, 조단백질 $15.30\%$ 및 조지방 $3.71\%$ 이었고 미량성분에 있어서는 칼륨 12,050mg/kg과 마그네슘4,296 mg/kg의 함량이 현저하게 높았다. LC-MS 분석결과 황금추출물에 있어서 분자량 445의 peak는 baicalin, 분자량 283의 peak는 wogonin, 분자량 269의 peak는 baicalein으로 확인되었다. disk method를 이용하여 항균력을 시험한 결과 baicalin이 주된 항균물질임을 추정할 수 있었으며, 메탄올 및 물추출물의 baicalin 함량을 분석한 결과 물추출물은 $0.75\%$이었고 메탄올추출물은 $0.82\%$로서 메탄올추출물에 있어서 baicalin 함량이 약간 높은 결과를 얻었다.

Quantitative Analysis for the Quality Evaluation of Scutellariae Radix by HPLC/UVD

  • In this study, quantitative analysis was developed using HPLC/UVD for the quality evaluation of Scutellariae Radix. For quantitative analysis, six major bioactive compounds were assessed. The separation conditions employed for HPLC/UVD were optimized using Phenomenex $C_{18}$ column ($250{\times}4.6$ mm, 5 ${\mu}m$) with a gradient of solvent A (1% acetic acid of $H_2O$) and solvent B (acetonitrile : methanol : acetic acid = 70 : 30 : 1) as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 275 nm. These methods were fully validated with respect to linearity, accuracy, precision and recovery. The HPLC/UVD method was applied successfully to the quantification of six major compounds in the extract of Scutellariae Radix. The results indicate that the established HPLC/UVD method is suitable for the quantitative analysis and quality control of multicomponents in Scutellariae Radix.

  • Ether-soluble flavonoid and neutral fraction (Fr. C) prepared from EtOAc extract of Scutellariae Radix showed inhibitory activities against bradykinin (BK) ; inhibited BK-induced ileum or uterus contractions, antagonized BK-induced plasma extravasation and protected mice from endotoxic shock, reduced acetic acid-induced writhing in mice.

Effect of Scutellariae Radix Extract on Human CYP450 Mediated-Drug Metabolism

  • Scutellariae Radix is widely used in the traditional herbal medicine for the treatment of fever, cough, dysentery, hepatitis and hypertension in Korea, China and Japan. In this study, we investigated the effects of 70% ethanolic extract of Scutellariae Radix (SRE) on CYP450-mediated drug metabolism in the in vitro systems using human liver microsomes and hepatocytes. The microsomal incubation assay showed that SRE inhibited the drug metabolism reactions catalyzed by CYP1A2, CYP2C8 and CYP2C9 in a dose-dependent manner. In particular, SRE was shown to strongly inhibit the metabolic activity of CYP1A2 with an $IC_{50}$ value of 4.6 ${\mu}g/mL$. When SRE was evaluated for its effect on the induction of CYP450 enzyme activities in cryopreserved human hepatocytes, SRE did not exhibit any effect.

The Effect of Chinese Herbs on Acne Pathogens

  • Chinese herbs have been used for a long period of time and less side effects than synthesized chemical drugs. Therefore, using Chinese herbs as natural additives in cosmetics becomes popular in recent years. The methanol extracts of Scutellariae Radix, Lithospermi Radix, Lonicerae Flos, Andrographitis. Herba, Angelicae Dahuricae Radix, Ligustici Rhizoma et Radix, Hedyotis Diffusae Herba, Isatidis Folium, Magnoliae Liliflorae Flos, Forsythiae Fructus, Anmarrhenae Rhizoma, Spirodelae Herba, Gardeniae Fructus, Sophorae Flavescentis Radix, Coptidis Rhizoma, Prunellae Spica, Equiseti Hiemalis Herba, Gentianae Radix, Moutan Radicis Cortex, Fraxini Cortex, Lycii Radicis Cortex, Violae Herba, Lophatheri Herba, Matricariae chamomillae Flos, Taraxaci Herba and Scutellariae Barbatae Herba are used to test the efficiency of inhibiting acne pathogens. Twenty-six Chinese herbs are extracted by methanol, and then condensed to dried powder. These extracts are divided into water-soluble part and DMSO soluble part. These two type solutions are tested for the effect on acne pathogens by paper disc diffusion method. The results show that the substances of water soluble part which are Coptidis Rhizoma, Moutan Radicis Cortex, Scutellariae Barbatae Herba have medium to high activity of inhibiting acne pathogents, and the substances of DMSO soluble part which are Coptidis Rhizoma, Ligustici Rhizoma et Radix, Sophorae Flavescentis Radix, Moutan Radicis Cortex, Scutellariae Radix, Scutellariae Barbatae Herba also have medium to high activity of inhibiting acne pathogens. Using Chinese herbs as natural additives in cosmetics is convenience and valuable application in cosmetceutical research and development. Therefore, it is worth that re-investigation and find out the potential of Chinese herbs being use in cosmetics.

황금(黃芩)이 사람 비만세포의 사이토카인 및 케모카인 분비에 미치는 영향 (Effect of Scutellariae Radix on Expression of Cytokines and Chemokines Levels in Human Mast Cells (HMC-1))

  • Objectives : Scutellariae Radix (Hwanggeum in Korean) is the root of Scutellaria baicalensis Georgi. Scutellariae Radix is well known to be used as a medicine for common cold, upper respiratory infections, and to strengthen and regulate the immune system and anemia etc. Little is understood about the roles of Scutellariae Radix in the cytokine and chemokine secretion by immune cells. This study was designed to find out the effects of Scutellariae Radix on the cytokine and chemokine secretion in human mast cells (HMC-1). Methods : We treated hwanggeum according to consistency on HMC-1 and measured cytokines and chemokines levels using flow cytometry CBA system. Results : In hwanggeum treated group, the expression of interferon-inducible protein 10 (IP-10), monocyte chemoattractant protein-1 (MCP-1), chemokine (C-X-C motif) ligand 9 (CXCL9, MIG), interleukin 8 (IL-8), interleukin 2 (IL-2), interleukin 4 (IL-4), interleukin 5 (IL-5), interleukin 10 (IL-10), and interferon ${\gamma}$ (IFN-${\gamma}$) were decreased significantly. Conclusion : These results suggest that hwanggeum may support some of immune diseases by means of amiliorating some chemokines or cytokines such as IP-10, MCP-1, MIG, IL-8, IL-2, IL-4, IL-5, IL-10, and IFN-${\gamma}$.

황금 추출물의 신경병증성 통증 유발 흰쥐에 미치는 영향 (Effects Study of Scutellariae Radix Extract on the Neuropathic Pain in Tibial and Common Peroneal Nerve Transected Rats)

  • TRPA1 and TRPV1 are members of the TRP superfamily of structurally related, nonselective cation channels. TRPA1 and TRPV1 are often co-expressed in sensory neurons and play an important role in mechanical hyperalgesia and allodynia during neuropathic pain. Scutellariae Radix was reported to possess anti-inflammation properties and similar patterns of therapeutic action against different diseases. also Baicalin(a known principal constituent of Scutellaria Radix) was shown to down-regulate the mRNA expression levels of TRPV1. In this study, we observed the effects of Scutellariae Radix extract(SRE) in neuropathic pain induced SD rats via modulation of TRPV1 and TRPA1. Oral administration of a Scutellaria Radix extract(in doses of 300mg/kg, SRE(300)) showed a meaningful increase in the withdrawal threshold of mechanical allodynia and showed a meaningful decrease in the expression of c-fos compared to the control group. SRE(100) and SRE(300) showed a meaningful decrease in the expression of TRPV1 level compared to the control group. These results suggest that Scutellariae Radix extract could decrease mechanical allodynia by down-regulate the TRPV1 on the model of neuropathic pain.

한방약물로부터 항고지혈증 치료약물개발(I) -수종 한약재의 항고지혈증 활성검색- (Studies on the Development of Antihyperlipidemic Drugs from Oriental Herbal Medicines(I) -Antihyperlipidemic Activities of Oriental Herbal Medicines-)

  • Twenty-four Oriental herbal medicines including Platycodi Radix and Scutellariae Radix, etc., which have been used for the cure of hyperlipidemia, coronary heart, disease were evaluated for the 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase inhibition and antioxidant effect on a free radical, 1,1-diphenyl-2-picrylhydrazyl(DPPH) in vitro, and antihyperlipidemic effect on hyperlipidemic rats induced by Triton WR-1339 in vivo. 80% MeOH extract of eight herbs including Scutellariae Radix, Coptidis Rhizoma, Paeoniae Radix Rubra, Moutan Cortex Radicis, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam, Cinnamomi Ramulus and Crataegi Fructus inhibited the HMG-CoA reductase activities and exhibited a radical-trapping action on a stable free radical, DPPH. On Triton WR-1339-induced hyperlipidemic rats, four herbs including Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus showed respectively the significant suppression of serum total cholesterol, triglyceride, phospholipid and LDL-cholesterol levels and serum transaminase(ALT and AST) activities. From these results, it is suggested that each 80% MeOH extract of Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus have effective antihyperlipidemic action against hyperlipidemia.
