Multivariate Analysis of Pyrolysis Mass Spectra of Scutellaria baicalensis to Identify its Origin (열분해질량스펙트럼에 의한 황금의 원산지 판별법 연구)

  • To overcome the limit of morphological method for classification of herbal drug, a novel method to discriminate its origin using pyrolysis mass spectrometry-multivariate analysis was developed. This method was applied successfully to Scutellaria baicalensis Georgi, one of the most popular herbal drug in oriental countries. The ethylacetate soluble fractions were prepared by sonication from pulverized roots of S. baicalensis which were collected from various regions including Korea and China, and subjected to direct insertion probe (DIP) mass spectrometry to achieve mass spectra of pyrolizates of extracts. The probe temperature was elevated from $30^{\circ}C$ to $320^{\circ}C$ at increasing rate $64^{\circ}C/min$, and the average mass spectrum calculated from total ion chromatography (TIC) was obtained. The relative peak intensities versus m/z were subjected to SAS program, and the training set (9 from Korea origin and 22 from China origin) was clustered two groups as its origin. In the test set, 11 samples among total 13 test sample were successfully classified according to their origin by developed method with accuracy of 85%.

Antioxidant and Antiallergic Activity of Compounds from the Aerial Parts of Scutellaria baicalensis Georgi (황금 지상부의 항산화 및 항 알러지 활성 성분)

  • Roots of Scutellaria baicalensis have been used for fever remedy; diuresis, antiphlogistic. For the investigation of the active component from aerial parts of Scutellaria baicalensis, MeOH extracts from aerial parts of Scutellaria baicalensis were suspended with $H_2O$, and partitioned by $CHCl_3$. In order to investigate the efficacy of antioxidative activity the activity guided fraction and isolation of physiologically active substance were peformed. Its $H_2O,\;30\%,\;60\%$ MeOH and MeOH fractions were examined on antioxidative activity using DPPH method and TBARS assay; It was revealed that $30\%\;and\;60\%$ MeOH fractions have significant anti-oxidative activity. its fractions testing type I allergy, compound 48/80 induced systemic anaphylaxis was applied. As a result, compared with reference (cromolygate), these fraction significantly inhibited systemic anaphylaxis by $71\%\;and\;57\%$, respectively. From $30\%,\;60\%$ MeOH fraction, five compounds were isolated and elucidated apigenin 6-C-${\alpha}$-L-arabinopyranosyl-8-C-${\beta}$-D-glucopyranoside (isoschaftside, I), scutellarein 7-O-${\beta}$-D-glucuronopyranoside (scutellarin, II), apigenin 7-O- ${\beta}$-D-glucuronopyranoside (III), isoscutellarein 8-O-${\beta}$-D-glucuronopyranoside (IV), kaempferol 3-O-${\beta}$-D-glucopyranoside (V) through their physicochemical data and spectroscopic methods. We measured radical scavenging activity with DPPH method and anti-lipid peroxidative efficacy on human LDL with TBARS assay. [$I] showed antioxidant activities in order. Type I allergy compound 48/80 induced systemic anaphylaxis was applied. $[V inhibited systemic anaphylaxis in order.

Inhibition effect of neuronal death by Scutellaria baicalensis GEORGI Water-Extract in a Hypoxic Model of Cultured Rat Cortical Cells (흰쥐 대뇌세포의 저산소증 모델에서 황금(黃芩)에 의한 신경세포사 저해 효과)

  • Scutellaria baicalensis GEORGI(SB) is used in oriental medicine for the treatment of incipient strokes. Although it has been reported that SB is neuroprotective in a hypoxia model, its mechanism is poorly understood. Here, we investigated the effect of SB on the modulation of retinoic acid receptor a (RARa). Rat cerebrocortical cells were grown in neurobasal medium. On DIV12 cells were treated with SB $(20{\mu}g/ml)$ and given a hypoxic shock $(2%\;O_2/5%\;CO_2,\;3hr)$ on DIV14. In situ hybridization using cRNA probe revealed that RARa mRNA punctae are distributed, in addition to nucleus, throughout neuronal dendrites, where SB upregulated its density by 69.8% (p=0.001) and 129.8% (p=0.001) in both normoxia and hypoxia, respectively. At the protein level, SB upregulated RARa in the neuronal soma by 78.8% (p=0.004) and 23.6% (p=0.001) in both normoxia and hypoxia, respectively. These results indicate that SB upregulates RARa in both normoxia and hypoxia, which might contribute to the neuroprotection.

Anti-inflammatory Effect of Ethyl alcohol Roasted Scutellaria baicalensis Georgi on Croton Oil-induced Mice Ear Edema

  • Scutellaria baicalensis Georgi (SB) has been widely studied to treat inflammatory diseases in east Asia. In the recent years, many studies have focused on modifying herbs to increase the pharmacological effects. Roasting alcohol absorbed SB is one of the traditional methods to increase the therapeutic effects. Currently there are no reports on the pharmacological effects of roasted SB. This study investigated the anti-inflammatory effects of roasted 30% ethyl alcohol absorbed SB extract (SR) on mice ear edema. After intra-gastric injection of dexamethasone (for positive control, 2 mg/kg) and SR (50, 100, 400 mg/kg), ear edema was provoked by croton oil (5% v/v in acetone, 10 ul/ear). Ear thickness was measured with a digital caliper to quantify the change in swelling. For histological study, we made paraffin sections and performed Phloxine-Tartrazine staining and Masson's trichrome staining to observe epidermis, dermis and subcutaneous region and collagen fiber of mice ear tissues. Ear thickness decreased dose-dependent manner in SR treated groups. Histological analysis compared with dexamethasone treated group, SR treated groups demonstrated a similar reduction in hypoplasia of epidermis and influx of inflammatory cells. Increase of subcutaneous layer and decrease of collagen fibers were significantly recovered in SR treated group (400 mg/kg) and dexamethasone treated group. In conclusion, treatment with SR ameliorates auricular inflammation induced with croton oil in mice. Experiments are now underway to understand the molecular mechanisms underlying anti-inflammatory activities of SR.

Quantitative analysis of the marker compounds in the decoctions of Coptis chinensis-Scutellaria baicalensis at different proportion produced by 'Mixed decoction' and 'Single decoction mixture' (배합 비율에 따른 황련과 황금의 혼합 전탕액 및 개별 전탕 혼합액 내 성분 함량 분석)

  • Objective : The present study aimed to evaluate the change of the content of 7 active components in decoctions produced by various proportional pairs of Coptis chinensis Franch and Scutellaria baicalensis Georgi in 'Mixed decoction (MD)' and 'Single decoction mixture (SDM)'. Methods : The samples of MDs were prepared by decocting C. chinensis : S. baicalensis with the ratios of 10 g:10 g, 10 g:20 g, and 20 g:10 g. Those of SDMs were prepared by blending each single decoction from C. chinensis and S. Baicalensis with the ratios of 1:1, 1:2, and 2:1. The samples were evaluated by high-performance liquid chromatography with statistical analyses. Results : The analytical methods, which were optimized and validated, were reliably applied to present research. The content of all components in both MDs and SDMs at C. chinensis : S. baicalensis = 1:1 ratio were reduced compared with single herb decoction. The components from each compositional herb in MDs were proportionally increased with the ratio of original herb increased, but inversely proportional to paired herb. The contents of components in MDs were significantly lower than those in SDMs at all ratios, except for high content of baicalin at C. chinensis : S. baicalensis = 2:1. Conclusion : It was concluded that MDs and SDMs as well as the proportions of herbs could affect the contents of the components from original herbal medicines. These results provide the information for the quality control of herbal medicine combined C. chinensis with S. baicalensis.

Effect of Scutellaria Baicalensis Georgi Extract on Oxidant-Induced Apoptosis in Renal Epithelial Cells (Renal epithelial cells에서 oxidant에 의한 apoptosis에 미치는 황금(黃芩)의 영향)

  • 목적 : 黃芩(황금)과 黃芩(황금)의 주요 flavonoid 성분인 baicalein이 신장세뇨관 상피세포에서 산화제에 의한 apoptosis에 미치는 효과를 살펴보고자 한다. 방법 : 신장세뇨관 상피세포주인 opossum kidney (OK) 세포를 유기산화제인 t-butylhydroperoxide (tBHP)에 노출시켜 apoptosis를 일으킨 후 관련된 변화를 관찰하였다. 결과 : tBHP는 농도에 의존하여 apoptosis를 유발시켰는데, 이러한 효과는 黃芩(황금)과 baicalein에 의해 농도 의존적으로 방지 되었다. tBHP에 의한 OK 세포사는 항산화제인 Trolox와 N-acetylcysteine에 의해 방지 되었다. tBHP는 mitogen-activated protein kinase의 subfamily인 extracellular signal-regulated kinase (ERK)를 활성화시켰다. ERK 억제제인 PD98059와 U0126은 tBHP에 의한 세포 사망을 방지하였다. tBHP에 의한 ERK 활성화는 U0126에 의해 억제되었으나 黃芩(황금)과 baicalein에 의해서는 영향을 받지 않았다. 철착염제인 deferoxamine은 tBHP에 의한 세포 사망과 ERK 활성화를 방지하였다. tBHP에 의한 세포 사망은 casopase 억제제인 BOD-U-FMK와 zDEVD-FMK에 의해 방지되었다. 결론 : 黃芩(황금)은 산화제에 의한 세포 사망을 방지하는데, 이는 kinase 억제, 항산화제 역할 및 철착염제의 작용에 기인하지 않았다. 黃芩(황금)의 이러한 효과는 산화제에 의관 신부전 예방 및 치료제로 개발하는데 이용될 수 있는 가능성을 보였다.

Cryopreservation of Scutellaria baicalensis Cells by Two-step Cooling Method

  • A two-step cooling technique has been developed for cryopreservation of suspension cultured Scutellaria baicalensis cells. Efficient regrowth of cryopreserved cells was obtained in cryoprotected cells with a mixture of 1.5 M glycerol and 0.4 M sucrose in Schenk and Hildebrandt medium without pretreatment in high osmotic medium. Optimum freezing conditions were found to be a cooling rate of $0.5^{\circ}C$ min from $4^{\circ}C$ to $-40^{\circ}C$, and then retaining samples at $-40^{\circ}C$ for 30 min prior to plunging into liquid nitrogen. A regrowth rate of approximately 95$%$ was obtained after three month storage in liquid nitrogen. Callus cultures established from the cryopreserved cells were found to produce the same patterns of flavonoid accumulation and retain their baicalin producing activity.

  • A medicinal herb, Scutellaria baicalensis G., can be a potential cash crop if proper cultural practices are established with proper genotypes. In this report the current status and future measure in cultivation of Scutellaria baicalensis G. in the southern parts of Korea are illustrated. We hope that this report stimulates research to investigate for the establishment of not only cultural practices such as sowing period after harvesting of barley, fertilizer application, planting density, weed control, and disease and insect control but also ideotype and disease resistant genotypes for the high yield of the herb.

The Anxiolytic-like Effects of Scutellaria baicalensis Using Elevated Plus-Maze in Rats (Rat에서 Elevated plus-maze를 이용한 황금의 항불안 효과)

  • Scutellaria baicalensis Georgi is one of most important medicinal herbs in traditional chinese medicine. The object of this study was to determine the effects of the water extracts of Scutellaria baicalensis (SB) on the anxiolytic-like activities in the elevated plus-maze (EPM) test. The watεr extracts of SB (100, 200, or 400 mg/kg) were orally administered to male SD rats for 3 days, and behavioral tests for the anxiolytic activity were performed. SB (100, 200, or 400 mg/kg) significantly increased in time-spent and arm entries into the open arms of the EPM compared with the control group. Futhermore, those anxiolytic-like activities of SB were antagonized by flumazenil (a $GABA_A$ antagonist, 3 mg/kg), but not by pindolol (a $5-HT_{1A}$ antagonist, 10 mg/kg). SB did not cause myorelaxant effects in the horizontal wire test at any dosage regimen. Therefore, these findings suggest that SB promote the anxiolytic-like activity mediated by GABAergic nervous system in rats.

Antioxidant and Xanthine Oxidase Inhibitory Activities of Hot Water Extracts of Medicinal Herbs (한약재 열수추출물의 항산화 활성 및 Xanthine Oxidase 저해 활성)

  • In our study, as many as 29 edible medicinal herbs were selected for testing their ability in the effective treatment of gout based on oriental medicine theory. We extracted each medicinal herb (135 g) with 4 L of distilled water at $100{\sim}105^{\circ}C$ for 210 min. Thereafter, we evaluated both the antioxidant and xanthine oxidase inhibition activities of the extracts obtained. Among all the edible medicinal herbs used in our study, only the extract from Scutellaria baicalensis Georgi (Korean name: hwang-geum) showed (1) the maximum total phenolic content (TPC) (2.25 mg gallic acid equivalent/mL), (2) DPPH radical scavenging activity (94.04%), and (3) xanthine oxidase inhibition activity (87.75%). We also observed that TPC was relatively highly correlated with both the DPPH radical scavenging activity (r=0.63) and xanthine oxidase inhibition activity (r=0.77). Our results suggest that S. baicalensis G. may be a potent antioxidant source for the extraction and development of nutraceuticals that may be utilized for effective treatment of gout.
