Synthesis and Screening of Some Novel 2-[5-(Substituted phenyl)-[1,3,4]oxadiazol-2-yl]-benzoxazoles as Potential Antimicrobial Agents

  • A series of some novel 2-[5-(substituted phenyl)-[1,3,4]oxadiazol-2-yl]-benzoxazoles were synthesized by using benzoxazole-2-carboxylic acid on reaction with thionyl chloride in presence of ethanol solvent at room temperature gave benzoxazole-2-carbonyl chloride, which is turned into benzoxazole-2-carboxylic acid hydrazide on reaction with hydrazine hydrate in ethanol solvent under reflux. The subsequent treatment of benzoxazole-2-carboxylic acid hydrazide with an appropriate aromatic carboxylic acid in presence of polyphosparic acid under reflux afforded the title compounds. The chemical structures of the newly synthesized compounds were elucidated by their IR, $^1H$ NMR and Mass spectral data analysis. Further the compounds are used to find out their ability towards anti microbial and nematicidal activity.

Inhibition of the Calcineurin Pathway by Two Tannins, Chebulagic Acid and Chebulanin, Isolated from Harrisonia abyssinica Oliv.

  • In order to discover and develop novel signaling inhibitors from plants, a screening system was established targeting the two-component system of Cryptococcus neoformans by using the wild type and a calcineurin mutant of C. neoformans, based on the counter-regulatory action of high-osmolarity glycerol (Hog1) mitogen-activated protein kinase and the calcineurin pathways in C. neoformans. Among 10,000 plant extracts, that from Harrisonia abyssinica Oliv. exhibited the most potent inhibitory activity against C. neoformans var. grubii H99 with fludioxonil. Bioassay-guided fractionation was used to isolate two bioactive compounds from H. abyssinica, and these compounds were identified as chebulagic acid and chebulanin using spectroscopic methods. These compounds specifically inhibited the calcineurin pathway in C. neoformans. Moreover, they exhibited potent antifungal activities against various human pathogenic fungi with minimum inhibitory concentrations ranging from 0.25 to over $64{\mu}g/ml$.

7-Oxostaurosporine Selectively Inhibits the Mycelial Form of Candida albicans

  • In the course of screening for specific inhibitors against the mycelial form of Candida albicans from natural resources, we have isolated and identified A6792-1 from Streptomyces sp. A6792 by using several chromatographies. By spectral analyses, this compound was determined as 7-oxostaurosporine, having a structure of staurosporine aglycon noiety. 7-Oxostaurosporine exhibited a selective growth inhibitory activity against the mycelial form of Candida spp. up to $100\mu\textrm{g}/disc$ in bioassay. It also exhibited a specific antifungal activity against the mycelial form of Candida spp. including C. krusei, C. albicans, C. tropicalis, and C. lusitaniae with MICs ranging from 3.1 to $25\mu\textrm{g}/ml$ 7-Oxostaurosporine demonstrated no in vivo toxicity in SPF ICR mice. Therefore, this compound may have a considerable potential as an antifungal agent based on the preferential inhibition against growth of the mycelial form of Candida spp., dimorphic fungi.

Isolation of Sangivamycin from Streptomyces sp. A6497 and its Herbicidal Activity

  • During the screening for the inhibitors of cellulose biosynthesis as herbicides, we discovered a Streptomyces sp. A6497 with a selective antifungal activity against cellulose containing Phytophthora parasitica, but not against cellulose lacking Candida albicans. The inhibitor was isolated and identified, using a series of chromatographies. Based on structure analyses with UV spectrophotometry, mass and various NMR, the inhibitor was identified as sangivamycin. The compound exhibited strong antifungal activities against P. parasitica (MIC; 3.125 $\mu$g/ml). In particular, it showed strong herbicidal activities against various weeds in the greenhouse experiment. Taken together, these results suggest that sangivamycin is a useful lead compound for the development of new herbicides.

Isolation and Characterization of an Antifungal and Plant Growth-Promoting Microbe

  • Fungal diseases including anthracnose, stem rot, blight, wilting, and root rot of crops are caused by phytopathogens such as Colletotrichum species, Sclerotinia sclerotiorum, Phytophthora species, and Fusarium oxysporum and F. solani which threaten the production of chili pepper. In this study, to identify biological control agents (BCAs) of phytopathogenic fungi, potentially useful Bacillus species were isolated from the field soils. We screened out five Bacillus strains with antagonistic capacity that are efficiently inhibiting the growth of phytopathogenic fungi. Bacillus species were characterized by the production of extracellular enzymes, siderophores, and indole-3-acetic acid (IAA). Furthermore, the influence of bacterial strains on the plant growth promoting activity and seedling vigor index were assessed using Brassica juncea as a model plant. Inoculation with Bacillus subtilis SRCM 121379 significantly increased the length of B. juncea shoots and roots by 45.6% and 52.0%, respectively. Among the bacterial isolates, Bacillus subtilis SRCM 121379 showed the superior enzyme activities, antagonistic capacity and plant growth promoting effects. Based on the experimental results, Bacillus subtilis SRCM 121379 (GenBank accession no. NR027552) was finally selected as a BCA candidate.

Trifluoromethylated Dihydro-1,4-dithiin carboxanilide 유도체의 합성 및 살균활성 (Synthesis of trifluoromethylated dihydro-1,4-dithiin carboxamides and their antifungal activities)

  • 새로운 농약살균제의 개발을 목적으로 trifluoromethyl기 dihydro-1,4-dithiin기 포함된 ${\alpha},{\beta}$-불포화 carboxamide 유도체 12를 합성하였다. trifluoromethylated ${\beta}$-ketoester 유도체 4를 염화한 다음 1,2-ethandithiol과 반응시켜 중간체 1,4-dithiane 유도체 9를 얻었다. 중간체 9의 정제없이 hydroxy기를 염소로 치환하여 생성된 중간체 8을 triethylamine 존재하에서 탈염화하여 trifluoromethyl기가 포함된 dihydro-1,4-dithiin ethyl ester 7을 합성하였다. Ester 7을 가수분해하여 생성된 carboxylic acid 10의 hydroxy를 염소로 치환하여 활성화한 다음 여러 가지 amine 유도체와 반응시켜 trifluoromethyl기가 포함된 dihydro-1,4-dithiin carboxamide 유도체 12를 합성하였다. 합성된 화합물을 대표적이 6종의 식물병원균, 벼 도열병원균, 벼 잎집무늬마름병원균, 오이 잿빛곰팡이병원균, 토마토 역병원균, 밀붉은녹병원균, 보리 흰가루병원균 등에 대한 항균력을 시험 (in vivo) 하였다. 그 결과, 대부분의 화합물이 미약한 항균을 나타냈다. 페닐기의 meta 위치에 isopropy기가 치환된 화합물 12h는 250 ppm에서 밀 붉은녹병원균에 대한 항균력 (99%)을 나타냈다.

  • 신농약 개발을 목적으로 isosterism 이론을 근거로 하여 선도화합물인 2-imino-1,3-thiazoline 의 분자 수정을 통하여 분자내에 1,3-thiazoline과 urea기가 포함된 새로운 화합물 4를 디자인하였다. N-Alkylthiourea 5와 bromoacetonitrile을 에탄올 용액 중에서 반응시켜 위치 선택적으로 생성된 2,4-diimino-1,3-thiazolidine 브롬산 염 6을 isocyanate 유도체와 반응하여 화합물 4의 tautomer인 화합물 8을 얻었다. 합성된 화합물 13종을 대표적인 6종의 식물병원균, 벼 도열병, 벼 잎집무늬마름병, 오이 잿빛곰팡이병, 토마토 역병, 밀 붉은녹병, 보리 흰가루병 등에 대한 in vivo 항균력 시험을 하였다. 합성된 화합물 8의 벼 도열병원균에 대한 항균력은 2-phenylimino-1,3-thiazoline 1과 비교할 때 매우 낮았으며, 일부 화합물에서 밀붉은녹병에 대한 미약한 항균력을 나타냈다.

Candida albicans에 대한 생약의 항진균성에 관한 연구(I) (Antifungal Actions of Crude Drug Water Extracts on Candida albicans(I))

  • Some crude drugs in ancient literatures have been used as traditional therapeutic agent of leucorrhea mainly caused by Trichomonas vaginalis and Candida albicans. Sixty six kinds of crude drugs in ancient literatures and ten constituents were selected as sample drugs. Trichomycin standard was tested to compare with the above drugs. To determine the anti-fungal effect of these drugs on Candida albicans Yu 1200, a test organism, screening test was conducted. Antifungal activities of crude drug water extracts were observed by means of two test methods : firstly through the agar slant method and secondly the counting chamber method which was used for acknowledged drug agents upon the result of the agar slant method. And in order to improve the fungicidal effect, the organisms were stained with 0.02% methylene blue solution. The results of the above test indicated that Fritillariae Rhizoma has antifungal action in the concentration of 310mcg/ml, Coptidis Rhizoma in 620mcg/ml, Meliae Cortex, Scutellariae Radix both in 5,000mcg/ml. Baicalin, catechol among the pure isolated constituents inhibited in the range of 50mcg/ml. This score was based on 50% inhibition in comparison with amounts of control organisms. Rhei Rhizoma, Mori Radicis Cortex, Linderae Radix, and Amomi globosi Fructus showed the antifungal effect moderately in 5,000mcg/ml, and baicalein and pectolinarin in 50mcg/ml in the limit of between 35% and 50% antifungal activity. Staining with 0.02% methylene blue showed that any of the crude drug extracts was unable to stain the cells, but trichomycin in 0.86unit/ml able to stain 12% of the cells. This result means that crude drugs probably do not have fungicidal but fungistatic action.

Discovery of Chitin Deacetylase Inhibitors through Structure-Based Virtual Screening and Biological Assays

  • Chitin deacetylase (CDA) inhibitors were developed as novel antifungal agents because CDA participates in critical fungal physiological and metabolic processes and increases virulence in soil-borne fungal pathogens. However, few CDA inhibitors have been reported. In this study, 150 candidate CDA inhibitors were selected from the commercial Chemdiv compound library through structure-based virtual screening. The top-ranked 25 compounds were further evaluated for biological activity. The compound J075-4187 had an IC50 of 4.24 ± 0.16 µM for AnCDA. Molecular docking calculations predicted that compound J075-4187 binds to the amino acid residues, including active sites (H101, D48). Furthermore, compound J075-4187 inhibited food spoilage fungi and plant pathogenic fungi, with minimum inhibitory concentration (MIC) at 260 ㎍/ml and minimum fungicidal concentration (MFC) at 520 ㎍/ml. Therefore, compound J075-4187 is a good candidate for use in developing antifungal agents for fungi control.

Isolation, Screening, and Identification of Actinomycetes with Antifungal and Enzyme Activity Assays against Colletotrichum dematium of Sarcandra glabra

  • A serious leaf disease caused by Colletotrichum dematium was found during the cultivation of Sarcandra glabra in Jingxi, Rong'an, and Donglan Counties in Guangxi Province, which inflicted huge losses to plant productivity. Biological control gradually became an effective control method for plant pathogens. Many studies showed that the application of actinomycetes in biological control has been effective. Therefore, it may be of great significance to study the application of actinomycetes on controlling the diseases caused by S. glabra. Strains of antifungal actinomycetes capable of inhibiting C. dematium were identified, isolated and screened from healthy plants tissues and the rhizospheres in soils containing S. glabra. In this study, 15 actinomycetes strains were isolated and among these, strains JT-2F, DT-3F, and JJ-3F, appeared to show antagonistic effects against anthracnose of S. glabra. The strains JT-2F and DT-3F were isolated from soil, while JJ-3F was isolated from plant stems. The antagonism rate of strain JT-2F was 86.75%, which was the highest value among the three strains. Additionally, the JT-2F strain also had the strongest antagonistic activity when the antagonistic activities were tested against seven plant pathogens. Strain JT-2F is able to produce proteases and cellulase to degrade the protein and cellulose components of cell walls of C. dematium, respectively. This results in mycelia damage which leads to inhibition of the growth of C. dematium. Strain JT-2F was identified as Streptomyces tsukiyonensis based on morphological traits and 16S rDNA sequence analysis.
