Scopolamine으로 유도된 인지장애 마우스의 기억력 개선 효과에 대한 인삼, 생지황, 발효홍삼 추출물의 비교 연구 (Comparative Study of Ginseng Radix, Rehmanniae Radix and Fermented Red-ginseng Extracts Effects on Memory Improvement in Scopolamine-induced Memory Impaired Mice)

  • In this study, we investigated the effects of Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng extracts (FRG) on cognitive function in scopolamine-induced memory-impaired mice. We measured the effects of G, R, and FRG on the improvement of memory and cognition via behavior analysis. In addition, we measured the acetylcholinesterase (AChE) activity in the hippocampus of each group of mice. The expression of β-amyloid, Tau, and BDNF in the brain tissues were observed through immunohistochemical staining. Ginseng Radix (G) and Fermented red-ginseng (FRG) have effectively improved cognitive function in the water maze test. Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) have improved the willingness of mice to explore the new environment, as confirmed by Y maze test. In addition, immunohistochemical staining revealed that Ginseng Radix (G) decreased the expression of β-amyloid and Tau in the hippocampus. In addition, fermented red-ginseng (FRG) increased the expression of BDNF. Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) have decreased the concentration of acetylcholinesterase in the hippocampus as compared with the control group of mice. In conclusion, Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) are considered to have the potential for development as candidate drugs to control the progression of Alzheimer's disease (AD).

뇌 손상 환자의 침 흘림에 대한 전침 적용 1례 (A Case Report of Stroke Patient With Drooling Improved by Electro-acupuncture Therapy)

  • Drooling causes inconvenience and develops complications, therefore it can be very stressful to patients and caregivers. Drooling caused by neurological disabilities appears to be the consequence of dysfunction in the coordination of the swallowing mechanism. However, there is no explanation about drooling by brain injury in oriental medicine literature. This is the case report about a stroke patient with drooling improved by electro-acupuncture therapy. The patient already applied scopolamine patch, and improved drooling before admission. However, caregiver concerned about side effects, so we stopped using scopolamine patch and applied electro-acupuncture for drooling treatment. The patient was treated by 4 acupuncture points on either side of Yeomcheon(CV 23), and Jichang(ST 4), Hyeopgeo(ST 6) with mixed low frequency electro stimulation. Although we stopped using scopolamine patch, drooling maintained improved state without a change, futhermore after 16 days later, drooling had disappeared until the patient discharged. This result showed electro-acupuncture is useful method to improve drooling caused by brain injury.

발효유 산물인 LHFM의 인지기능 개선 효과 (Cognitive-enhancing Effects of a Fermented Milk Product, LHFM on Scopolamine-induced Amnesia)

  • 본 연구팀의 선행 연구 결과, L. helveticus IDCC3801의 발효유 주정 침전물이 BACE의 활성 억제를 통한 amyloid beta($A{\beta}$) 생성 저해효과를 갖고 있음을 확인하였다(15). 이러한 결과를 바탕으로 L. helveticus IDCC3801의 발효유 주정 침전물의 산업화를 위한 공정 개선연구를 진행하였고, 대량생산을 통해 LHFM을 확보하였다. 본 연구는 LHFM의 인지기능 개선 기능성 입증 연구로써 $A{\beta}$ 저해효과 외에 신경세포 보호효과 및 기억력 개선 효능을 확인하고 어떠한 작용 기전으로 개선 효과가 나타나는지 조사한 것으로 scopolamine으로 기억 손상을 유발한 mouse에서 학습 능력과 기억력 개선 효과, 신경세포 보호 효과, AChE 저해와 ACh 증가 효과 등을 평가하였다. LHFM은 신경세포 사멸을 유도하는 고농도의 glutamate가 존재하는 환경에서 농도 의존적으로 신경세포의 사멸을 억제하여 신경세포를 보호하였다. 또한 LHFM은 in vitro에서 농도 의존적으로 강력하게 AChE의 활성을 저해하였다. 이러한 in vitro 결과들을 근거로 scopolamine으로 기억 손상을 유발한 mouse에서 Morris water maze와 Y-maze 행동 시험을 통해 LHFM의 학습능력과 기억 개선 효과를 조사하였다. LHFM은 Morris water maze에서 실험 2일째부터 escape latency시간을 유의적으로 감소 시켰으며 5일째에서는 정상 mouse와 동등한 수준으로 감소되어 우수한 학습 효과를 보였다. 실험 6일째 platform을 치운 후 시행한 probe trial에서도 platform이 있던 위치에 머무는 회수가 scopolamine을 단독으로 처리한 것보다 유의적으로 높아 장기기억의 개선에도 효과가 있었다. Y-maze 시험에서도 LHFM은 scopolamine 단독 처리군과 비교하여 뚜렷하게 변경 행동력을 증가시켜 여러 행동 시험을 통해 LHFM의 인지기능 개선 효과를 확인하였다. 행동실험 후 hippocampus를 적출하여 ACh 및 AChE의 활성을 측정한 결과, donepezil과 마찬가지로 LHFM은 AChE의 활성을 유의적으로 억제하였으며 또한 ACh 함량도 증가시켰다. 결론적으로 LHFM은 신경세포 보호 효과와 신경전달물질인 ACh의 뇌 조직 내 농도를 높여 인지기능 개선 효과를 보이는 것을 확인하였다.

Ethanolic Extract of the Seed of Zizyphus jujuba var. spinosa Ameliorates Cognitive Impairment Induced by Cholinergic Blockade in Mice

  • In the present study, we investigated the effect of ethanolic extract of the seed of Zizyphus jujuba var. spinosa (EEZS) on cholinergic blockade-induced memory impairment in mice. Male ICR mice were treated with EEZS. The behavioral tests were conducted using the passive avoidance, the Y-maze, and the Morris water maze tasks. EEZS (100 or 200 mg/kg, p.o.) significantly ameliorated the scopolamine-induced cognitive impairment in our present behavioral tasks without changes of locomotor activity. The ameliorating effect of EEZS on scopolamine-induced memory impairment was significantly reversed by a sub-effective dose of MK-801 (0.0125 mg/kg, s.c.). In addition, single administration of EEZS in normal naive mouse enhanced latency time in the passive avoidance task. Western blot analysis was employed to confirm the mechanism of memory-ameliorating effect of EEZS. Administration of EEZS (200 mg/kg) increased the level of memory-related signaling molecules, including phosphorylation of extracellular signal-regulated kinase or cAMP response element-binding protein in the hippocampal region. Also, the time-dependent expression level of brain-derived neurotrophic factor by the administration of EEZS was markedly increased from 3 to 9 h. These results suggest that EEZS has memory-ameliorating effect on scopolamine-induced cognitive impairment, which is mediated by the enhancement of the cholinergic neurotransmitter system, in part, via NMDA receptor signaling, and that EEZS would be useful agent against cognitive dysfunction such as Alzheimer's disease.

황련해독탕 전탕액과 약침액이 치매유발생쥐의 인지와 기억에 미치는 영향 (Effects of Hwangryunhaedok-Tang Decoction & Hwangryunhaedok-Tang Pharmacopuncture Solution on the Cognitive & Memory Impairment Induced by Scopolamine in Mouse Model)

  • Objectives : This research was performed to investigate the effects of Hwangryunhaedok-tang decoction and Hwangryunhaedok-tang Pharmacopuncture at BL10 on cognition and memory impairment in a mouse dementia model induced by scopolamine. Methods : Fifty ICR mice were divided into 6 groups : Normal group (n=5), Control group (n=9), Positive control group for pharmacopuncture group (n=9, Donepezil 0.75 mg/kg), Positive control group for oral administration group (n=9, Donepezil 5 mg/kg), Pharmacopuncture group (n=9, Hwangryunhaedok-tang Pharmacopuncture undiluted solution 10 ml/kg), and Oral administration group (n=9, Hwangryunhaedok-tang 200 mg/kg). For a mouse dementia model, 1 mg/kg scopolamine was intraperitoneally administered to ICR mice. Hwangryunhaedok-tang pharmacopuncture was administered on BL10 for 4 weeks at intervals of 2 days. Hwangryunhaedok-tang decoction was given orally for 4 weeks every day. Morris water maze and passive avoidance test were conducted followed by measurement of acetylcholine concentration, acetylcholinesterase activity, and the amount of BDNF and p-CREB in the brain. Results : 1. In the Morris water maze test, the time spent staying around the platform significantly increased in the pharmacopuncture group and oral administration group than in the control group. 2. In the passive avoidance test, the time spent in the bright room significantly increased in the pharmacopuncture group and oral administration group than in the control group. 3. The level of acetylcholine in brains increased in the pharmacopuncture group and oral administration group than in the control group. Also, the activity of acetylcholinesterase decreased in the pharmacopuncture group and oral administration group than in the control group. 4. The expression of BDNF and p-CREB decreased in the control group, but increased in the pharmacopuncture group and oral administration group. Conclusions : These results suggest that Hwangryunhaedok-tang decoction and Hwangryunhaedok-tang pharmacopuncture at BL10 may have cognition and memory-enhancing effect in scopolamine-induced dementia in ICR mice via controlling the content of acetylcholine and the activity of acetylcholinesterase, and activating BDNF and p-CREB.

도라지 추출물 연양갱이 β-amyloid에 의한 세포독성 및 Scopolamine에 의해 유도된 인지능 저하 동물 모델의 개선효과 (Improving Effects of Platycodon Extracts Jelly on β-amyloid-induced Cytotoxicity and Scopolamine-Induced Cognitive Impairment Animal Models)

  • 본 연구에서는 도라지 추출물에 대한 세포 독성을 확인하였으며, $A{\beta}$에 의한 PC12 세포 독성에 대한 보호효과를 관찰하였다. 또한 도라지 추출물과 도라지 추출물 연양갱을 4주간 강제 경구 투여하여 Morris 수중미로시험에서 도달지점까지의 도달시간이 도라지 추출물 및 도라지 추출물 연양갱 투여에 의해서 모두에서 유의하게 감소하였다. 이와 유사하게 수동회피시험에서도 자극이 있는 어두운 방을 나오는 시간이 도라지 추출물 및 도라지 추출물 연양갱 투여에 의해서 현저하게 감소하였다. 따라서 도라지 추출물 및 도라지 추출물 연양갱은 인지능 개선에 도움을 줄 것으로 생각된다.

천연식물자원으로부터 Acetylcholine esterase 저해 활성 탐색 및 인지기능에 미치는 영향 (Screeening of Natural Plant Resources with Acetylcholine esterase inhibitory activity and Effect on Scopolamine-induced Memory Impairment)

  • 아세틸콜린 분해효소(acetylcholine esterase, AChE) 억제제는 아세틸콜린 함량을 높여 콜린성 neuron을 활성화함으로써 기억 능력의 개선 및 치매 개선을 가져와 현재 다양한 AChE 억제제들이 개발되어 사용되고 있다. 본 연구에서는 AChE에 대한 억제 활성을 갖는 천연물을 다양한 식물추출물 및 에센스오일로부터 탐색하였으며, 탐색한 추출물의 scopolamine으로 기억손상을 유발한 쥐의 기억력 개선 활성을 치매 치료제로 사용하고 있는 donepizil과 비교 분석하였다. 그 결과 자몽(Citrus paradisi) 유래의 에센스 오일이 AChE 억제 활성이 가장 높아 20 ug/ml의 농도로 처리하였을 때 90% 이상의 효소 억제 활성을 나타내었다. 수동회피 실험 결과, 자몽 유래의 에센스오일(100 mg/kg, p.o.)을 투여한 쥐는 치매 치료제로 사용하고 있는 donepizile (0.5 mg/kg)을 투여한 쥐와 유사한 latency time을 나타내어 인지기능이 개선되었다. 또한, 수중미로 시험 결과, 자몽 유래 에센스오일(100 mg/Kg, p.o.)을 투여한 쥐는 donepizile(0.5 mg/kg)을 투여한 쥐와 유사한 latency time을 나타내어 인지기능이 개선되었다. 이상의 결과로부터 자몽 유래 에센스오일은 매우 효과적으로 기억력을 개선하여 인지기능을 개선해 줄 수 있는 안전하고 효과적인 후보물질이라고 사료된다.

Comparative Study of Korean White, Red, and Black Ginseng Extract on Cholinesterase Inhibitory Activity and Cholinergic Function

  • This study evaluated cholineresterase inhibitory activity of Korean white ginseng extract (WGE), red ginseng extract (RGE), and black ginseng extract (BGE) and the cholinergic effect on scopolamine (SCOP)-induced amnesic mice. WGE, RGE, and BGE inhibited acetylcholineserase (AChE), as well as butyrylcholineserase (BuChE) in a concentration-dependent manner. BGE presented strong inhibition of AChE with an $IC_{50}$ value of 1.72 mg/mL, followed by WGE (5.89 mg/mL), RGE (6.30 mg/mL), respectively. The inhibitory activity of the three ginseng extracts on BuChE showed similar values among the groups. To better understand the mechanisms of the possible effect of ginseng extract on the cholinergic function, this study assessed the expression of the cholinergic markers of choline acetyltransferase (ChAT) and AChE using western blot and RT-PCR analysis in the brains of amnesic mice. Treatment with ginseng extracts led to inhibition of AChE expression and, the activation of ChAT expression in the hippocampus and the cerebral cortex of amnesic mice as induced by SCOP. The results suggest that ginseng extracts including BGE, appear to modulate the metabolism of acetylchoine (ACh), which would greatly increase synaptic ACh levels and most potently revert SCOP-induced amnesia.

총명탕(聰明湯)과 향부자총명탕(香附子聰明湯)의 추출물, 나노분말 제형을 이용한 치매에 관한 연구 (A Study of ChongMyungTang(CMT) and HyangbujaChongMyungTang(HCMT) on Dementia - Extract & Nano Powder Drug types)

  • Objective : This research aims to investigates the effects of CMT and HCMT on Dementia. and we also want to know the different effect of CMT, HCMT by drug types. Methods : The research is progressed by two types of experiment. one experiment is BV2 microglial cell line treated by LPS in vitro and another experiment is memory deficit mice induced by scopolamine in vivo. Results : The CMT and HCMT is effective in BV2 microglial cell line treated by LPS in vitro and in the serum of the memory deficit mice induced by scopolamine in vivo. But, there is no significant difference between CMT and HCMT extract&nano powder in experimental conclusion. Conclusions : These results suggest that the two drug types of CMT and HCMT may be effective for the prevention and treatment of Dementia. Investigation into the further study two drug types of the CMT and HCMT for Dementia is suggested for future research.
