Production of Mycoplasma-Free Jujube Trees through In vitro Micrografting (기내미세접목(器內微細接木)에 의(依)한 대추나무의 마이코플라즈마 무병주(無病株) 생산(生産))

  • For the production of mycoplasma-free jujube trees from mycoplasma-infected trees in vitro micrografting visas carried out using apical meristems of in vitro grown plantlets as scions. The rootstocks were hypocotyl segments of in vitro germinated seedlings of Zizyphus jujuba Mill. which is widely used as rootstocks in the field grafting. Ten percent of scions showed normal growth and grew into plants. The presence of mycoplasma was tested using transmission electron microscopy and fluorescence microscopy. Mycoplasma was not found in the tissues of scion parts and seedlings, whereas it was found in in vitro grown plantlets. This suggests that the production of mycoplasmaa-free jujube trees is possible by the in vitro micrografting technique.

Effect of Different Budding Methods and Times on Grafting Success of Walnut

  • Vegetative propagation of walnut is difficult compared with that of other fruit and nut species. The present study assessed three methods of grafting (patch, shield, and chip) at various periods of walnut growth and with different timings of grafting in walnut. Early May was the best time for grafting, at which time the highest success rate was obtained by the patch method (96%), followed by chip-budding (75%), while shield-grafting showed the lowest efficiency (10%). Patch-grafting was also successful (75-80%) in early August and moderately successful in mid-June (51-55%), while the shield and chip methods had no success during these two times (0.00%). Patch-grafting was more efficient and also induced better callus formation and scion growth than the other two methods. The genotypes used did not affect grafting efficiency. The best results were obtained by patch-budding in both tested genotypes. The present findings show the potential value of patch-grafting in early May as a propagation method for walnut for establishment of guidelines for propagation.

Migration and Distribution of Graft-inoculated Jujube Witches'-broom Phytoplasma within a Cantharanthus roseus Plant

  • Periwinkle seedlings (Cantharanthus roseus) were inoculated with jujube witches'- broom (JWB) phytoplasma via grafting to analyze the migration of JWB phytoplasmas within the host plant. The phytoplasmas were detected using nested polymerase chain reaction (PCR) and fluorescence microscopy. Fluorescence microscopy was a simple and easy method of detecting phytoplasmas; however, it was not sufficiently sensitive to detect very low phytoplasma concentrations. Therefore, the migration of JWB phytoplasma was investigated through PCR. The first migration of JWB phytoplasma from an infected tissue to healthy tissues occurred late. After grafting, the phytoplasmas moved from the inoculated twig (or scion) to the main stem, which took 28 days. Afterward, the phytoplasma migrated faster and took less than 4 days to spread into the roots from the main stem. All twigs were then successively colonized by the JWB phytoplasmas from the bottom to the top. JWB phytoplasma was detected via nested PCR in all parts of the periwinkle seedling 82 days after inoculation. Based on these results, the inoculated JWB phytoplasma appeared to migrate downward to the roots along the main stem during the early stages, and then continued to move upward, colonizing twigs along the way until they reached the apex.

First Report on Bacterial Soft Rot of Graft-cactus Chamaecereus silvestrii Caused by Pectobacterium carotovorum subsp. carotovorum in Korea

  • A soft stem rot disease was observed on Chamaecereus silvestrii (Korean name: Sanchui), a scion of graft-cactus, in major growing areas of Suwon (National Horticulture Research Institute), Anseong, Eumseong, Cheonan, Daegu, and Goyang, Korea during 2000 and 2001. Typical symptoms were soft rots characterized by moist and watery decay of the whole cactus stem, which initiated as small water-soaked lesions and enlarged rapidly to the entire stem. The causal organism isolated from the infected stems was identified as Pectobacterium carotovorum subsp. carotovorum (Erwinia carotovora subsp. carotovora) based on its physiological and biochemical characteristics and confirmed by the cellular fatty acid composition and Biolog analyses. Artificial inoculation of the bacterium produced the same soft rot symptoms on the cactus stems, from which the same bacterium was isolated and identified. This is the first report of the P. carotovorum subsp. carotovorum in the graft-cactus C. silvestrii in Korea.

Relationship between Lignin Content and the Durability of Wood Pellets Fabricated Using Larix kaempferi C. Sawdust

  • This work was conducted to examine the relationship between lignin content and the durability of larch (LAR) pellets. LAR sawdust was immersed in tap water (TW), sulfuric acid (AC) and sodium hydroxide (AK) solutions, and then the immersed sawdust was used for pellet fabrication. Klason lignin (KL) content of the immersed LAR, contents of soluble lignin (SL) and monomeric sugars liberated from the immersion of LAR, and durability of LAR pellets were measured. KL content decreased as the concentration of AC and AK solutions increased, but glucose content increased with increase in AC and AK concentration. Durability of wood pellets fabricated using non-immersed LAR sawdust was the highest, followed by those made using TW-, AK- and AC-immersed sawdust. LAR pellets became more durable as the concentration of KL and SL increased, but a significant positive correlation was found only between pellet durability and KL content. Through the fluorescent microscopic observation and SEM-EDX analysis, it was verified that lignin content of non-immersed LAR pellets was higher than that of AC- and AK-immersed LAR pellets. These results suggest that lignin might contribute to an increase in inter-particle bonding in wood pellets.

Construction of Antibodies for Detection and Diagnosis of Cucumber green mottle mosaic virus from Watermelon Plants

  • We immunized BALB/c mice with purified Cucumber green mottle mosaic virus isolate HY1 (CGMMV-HY1). Through the selection of positive clones that were grown on the HAT medium, four sensitive monoclonal clones (CG99-01, CG99-02, CG99-03, and CG99-04) were selected from 500 Hypoxanthine-guanine phosphoribosyltransferase positive hybridoma cells. Four sensitive clones of CGMMV-HYI were determined as IgM type of the subclass of mouse immunoglobulins Ig group. The titer of monoclonal antiserum against CGMMVHY1 was estimated 1:12,800 by the indirect ELISA. Although monoclonal antibodies (MAbs) from CG99-01 and from CG99-04 cross-reacted with Zucchini green mottle mosaic virus and Kyuri green mottle mosaic virus, MAb from the cell line CG99-03 was highly specific to CGMMV. No MAbs cross-reacted with Cucumber mosaic virus-Fny. Only CG99-04 reacted with Pepper mild mottle virus weakly and CG99-02 reacted with both CGMMV and KGMMV. CGMMV was detected from the rind of watermelon fruit by DAS-ELISA of CGMMV-HY1, but not from the flesh of watermelon. Average seed transmission rate of CGMMV in watermelon was $24\%$ from symptomatic watermelon collected from 5 regions of Gyeongnam province. CGMMV was detected by DAS-ELISA with specific MAb of CGMMVHY1 periodically from root stock, during the sequential process for nursery seedling in Haman. Necrotic spots on cotyledons of root stock seedling progressed to reveal the typical symptomatology on the primary leaves of scion upon grafting. Here, we have established MAb based ELISA system, which could accurately detect CGMMV from watermelon seeds, nursery seedlings, transplants and field samples from greenhouse or open out door field as well.

  • The rootability of hardwood cutting of mulberry (Morus alba L.) depending upon the kinds of cutting beds, soil of cutting beds, effect of growth regulator (Rootone-F), pretreatments of cutting scion, parts of branches were investigated, The results were as follows ; 1. Rootability was the highest in cutting after callus formed on sand bed, whereas null in the treatment of rice seedling hot bed. 2. Optimum cutting season was around March 26, showed 100% rootability in all treatments except wooden box filled with sand. 3. The order of rootability related with cutting part of branch was upper$^{\circ}C$), and cutting after formation of callus increased rootability, whereas cuttings left in open air for 5 days showed null rootability.

Effects of Oral Administration of Ligigeopoongsan on Neuropathic Pain and c-Fos Protein Expression in Rats (이기거풍산(理氣祛風散)이 백서(白鼠)의 신경병리성(神經病理性) 동통(疼痛) 억제(抑制) 및 c-Fos 단백(蛋白) 발현(發顯)에 미치는 영향(影響))

  • Objective : We have studied to know effects of Ligigeopoongsan(LGS, 理氣祛風散) on mechanical allodynia, cold allodynia and c-Fos protein expression in the model of neuropathic pain of rats. Methods : The model of neuropathic pain was made by injured tibial nerve and sural never while common peroneal never was maintained. After 2weeks, we performed behavioral test for 7 days to try out mechanical allodynia using von frey filament and cold allodynia using acetone, which are calculated by counting withdrawal response on foot. Rat brains removed and sliced on 8th days. Serial sections were immunohistochemically reacted with polyclonal c-Fos antibody. The numbers of c-Fos protein immunoreactive neurons in the central gray were examined using scion image program. Results : 1. Mechanical allodynia in LGS-2, LGS-3 groups were significantly diminished compared with the control group. 2. Cold allodynia in LGS-3 group was significantly diminished compared with the control group. 3. c-Fos protein expression on the central gray LGS-2, LGS-3 groups were significantly lower than that of control group. conclusions : We have noticed that LGS(理氣祛風散) diminished mechanical and cold allodynia in the model of neuropathic pain compared with the control group. c-Fos protein expression in the central gray of that group was also decreased compared with the control group. Pain control group were LGS was accumulated time goes by. This study can be used as a basic resource on a study and a treatment of pain.

Studies on the Grafting of the Magnolia grandiflora L. (III) (태산목(泰山木)의 접목(接木)에 관(關)한 연구(硏究)(제(第)III보(報)))

  • This study was carried out to find out the proper method of grafting of Magnolia grandiflora L. Magnolia kobus DC. was used as stocks and the modified veneer grafting method in which a flap was attached to the cut surface of scion was adopted. And also the environment of the grafting beds was controlled for giving optimum humidity (90-100%) and temperature ($20-25^{\circ}C$). The results are summarized as follows: 1. Two year-old seedlings of M. kobus were best as stocks for grafting of M. gantiflora. 2. The modified veneer grafting which brought the successful result of 43 to 89.5 percent found to be very efficient method. 3. The environmental control made of vinyl tunnel and screen (lath) was very effective to attain to the excellent result of 89 percent. 4. For scions, one to two year-old branches with well developed winter buds collected just before the time of grafting gave the best results. 5. The optimum season for grafting in Kwangju districts was 15 to 20, April.

Evapotranspiration Rate of Grafted Seedlings Affected by Relative Humidity and Photosynthetic Photon Flux under Artificial Lighting (인공광하에서 접목묘의 증발산속도에 미치는 상대습도와 광합성유효광량자속의 효과)

  • To enhance the smooth graft-taking of grafted seedlings, the excessive evapotranspiration from grafted seedlings just after grafting should be avoided. A measurement system of the evapotranspiration rate of grafted seedling under artificial lighting was set up to investigate the effect of physical environment on the evapotranspiration and graft-taking characteristics of grafted seedlings quantitatively. The evapotranspiration rate of grafted seedlings affected by relative humidity and light intensity were analyzed using the measurement system. The hypocotyl of watermelons (Citrullus vulgaris cv. Sweetdew, Hungnong Seed Co.) was slantly cut and then inserted into a hole on the stem of rootstock (Lagenaria siceraria cv. FR-King, Hungnong Seed Co.). Grafted seedlings were healed and joined for 5 days under cool-white fluorescent lamps (FL20SEX-d/18, Keumho Electric Co.) with photoperiod of 12h$.$d$\^$-1/ except dark period for one day after grafting in a closed graft-taking enhancement system developed by Kim(2000). The evapotranspiration rate and graft-taking of grafted seedling at air temperature of 23$\^{C}$ and air current speed of 0.1m$.$s$\^$-1/ was highly affected by relative humidity. But light intensity showed higher effect on the stem length of scion than relative humidity. In conclusion, it was suggested that relative humidity should be controlled at higher than 90% with photosynthetic photon flux of 50$\mu$mol$.$m$^2$$.$s$\^$-1/ to increase the survival of grafted seedlings and to produce healthy seedlings.
