We propose an effective deep learning model to denoise scanning transmission electron microscopy (STEM) image series, named Noise2Atom, to map images from a source domain 𝓢 to a target domain 𝓒, where 𝓢 is for our noisy experimental dataset, and 𝓒 is for the desired clear atomic images. Noise2Atom uses two external networks to apply additional constraints from the domain knowledge. This model requires no signal prior, no noise model estimation, and no paired training images. The only assumption is that the inputs are acquired with identical experimental configurations. To evaluate the restoration performance of our model, as it is impossible to obtain ground truth for our experimental dataset, we propose consecutive structural similarity (CSS) for image quality assessment, based on the fact that the structures remain much the same as the previous frame(s) within small scan intervals. We demonstrate the superiority of our model by providing evaluation in terms of CSS and visual quality on different experimental datasets.
Under Light and Scanning electron microscope, the pupal morphology of mulberry silkworm Bombyx mori Linn. revealed the prothorax and metathorax, well developed mesothorax, less defined last pair of sporacle, well exposed prothoracic femora and wing pads approaching the anterior margin of Ab III. The important sex separating characters viz, wegiht, antennal elevations, intersegmental lines and genings have discussed. Further, two separate openings bursa copulatrix and ovipositional opening were observed, performing different functions in abult moth.
Plant specimens for scanning electron microscopy (SEM) are commonly treated using standard protocols. Conventional fixatives consist of toxic chemicals such as glutaraldehyde, paraformaldehyde, and osmium tetroxide. In 1996, methanol fixation was reported as a rapid alternative to the standard protocols. If specimens are immersed in methanol for 30 s or longer and critical-point dried, they appear to be comparable in preservation quality to those treated with the chemical fixatives. A modified version that consists of methanol fixation and ethanol dehydration was effective at preserving the tissue morphology and dimensions. These solvent-based fixation and dehydration protocols are regarded as rapid and simple alternatives to standard protocols for SEM of plants.

본 강연에서는 방사광 연X-선 분광현미경학(spectro-microscopy) 중에서, 표면에서 방출되는 광전자를 이용하는 SPEM (Scanning Photoelectron Microscopy)과 PEEM (Photoemission Electron Microscopy)을 소개하고자 한다. SPEM은 입사하는 X-선을 작은 크기로 집속하여 특정의 작은 공간에서 광전자분광학(XPS) 데이터를 얻거나 특정 광전자에너지의 공간분포를 얻게 해주며, PEEM은 입사한 X-선에 의해 발생한 광전자를 전자렌즈 원리로 영상을 맺히게 하여 광전자의 발생 분포를 구하게 한다. 이들은 균일하지 아니한 이종의 표면 연구에 매우 유용한 측정기법들이지만, 그 원리 및 구성은 많은 차이점들을 가지고 있다. 예를 들어, SPEM은 시료를 scanning하면서 XPS에 보다 충실한 타입이고 PEEM은 full field imaging 타입으로 표면변화의 동역학 연구에 강점이 있다. 본 강의에서는 이들 각각의 원리, 장점들에 대해서 설명하고, 활용 예를 제시하고자 한다. 활용 분야에 있어서, SPEM의 경우는 포항가속기연구소의 SPEM으로 수행되었던 DMS, graphene, nano-lithography, OLED, 등 반도체 및 나노 소재, 소자에의 활용에 대한 예를 제시할 것이다. PEEM의 경우는 포항가속기연구소의 응용 예와 박막 형태의 magnetic material에 대한 예들을 제시할 것이다.
There are many kinds of microscopes suitable for general studies; optical microscopes(OM), conventional transmission electron microscopes (TEM), and scanning electron microscopes(SEM). The optical microscopes and the conventional transmission electron microscopes are very familiar. The images of these microscopes are directly formed on an image plane with one or more image forming lenses. On the other hand, the image of the scanning electron microscope is formed on a fluorescent screen of a cathode ray tube using a scanning system similar to television technique. In this paper, the features and some applications of the scanning electron microscope will be discussed briefly. The recently available scanning electron microscope, combining a resolution of about $200{\AA}$ with great depth of field, is favorable when compared to the replica technique. It avoids the problem of specimen damage and the introduction of artifacts. In addition, it permits the examination of many samples that can not be replicated, and provides a broader range of information. The scanning electron microscope has found application in diverse fields of study including biology, chemistry, materials science, semiconductor technology, and many others. In scanning electron microscopy, the secondary electron method. the backscattererd electron method, and the electromotive force method are most widely used, and the transmitted electron method will become more useful. Change-over of magnification can be easily done by controlling the scanning width of the electron probe. It is possible. to continuously vary the magnification over the range from 100 times to 1.00,000 times without readjustment of focusing. Conclusion: With the development of a scanning. electron microscope, it is now possible to observe almost all-information produced through interactions between substances and electrons in the form of image. When the probe is properly focused on the specimen, changing magnification of specimen orientation does not require any change in focus. This is quite different from the conventional transmission electron microscope. It is worthwhile to note that the typical probe currents of $10^{-10}$ to $10^{-12}\;{\AA}$ are for below the $10^{-5}$ to $10^{-7}\;{\AA}$ of a conventional. transmission microscope. This reduces specimen contamination and specimen damage due to heatings. Outstanding features of the scanning electron microscope include the 'stereoscopic observation of a bulky or fiber specimen in high resolution' and 'observation of potential distribution and electromotive force in semiconductor devices'.
This study was carried out through scanning electron microscopy to elucidate the mechanism of thickness swelling in wood particle-polypropylene composite which is a typical way of using wood and plastic materials. For this purpose, control particleboards and nonwoven web composites from wood particle and polypropylene fiber formulations of 100:0, 70:30, 60:40, and 50:50 were manufactured at target density levels of 0.5, 0.6, 0.7, and 0.8 g/cm3. Their water absorption and thickness swelling were tested according to ASTMD 1037-93 (1995). To elucidate thickness swelling mechanism of composite through the observation of morphological change of internal structures, the specimens before and after thickness swelling test by 24-hour immersion in water were used in scanning electron microscopy. From the scanning electron microscopy, thickness swelling of composite was thought to be caused by the complicated factors of degree of built-up internal stresses by mat compression and/or amount of wood particles encapsulated with molten polypropylene fibers during hot pressing. In the composites with wood particle contents of 50 to 60% at target densities of 0.5 to 0.8 g/cm3 and with wood particle content of 70% at target densities of 0.5 to 0.7 g/cm3, thickness swellings seemed to be largely dependent upon the restricted water uptake by encapsulated wood particles with molten polypropylene fibers. Thickness swelling in the composite with wood particle content of 70% at target density of 0.8 g/cm3, however, was thought to be principally dependent upon the increased springback phenomenon by built-up internal stresses of compressed mat.
The present study described with different microscopy approaches chloroplasts lobes in Laurencia sensu latu (Rhodophyta) species and found inter-specific differences among them. Chloroplasts were investigated using confocal laser scanning microscopy (LSM), transmission electron microscopy (TEM) and high resolution scanning electron microscopy (HRSEM). Using and TEM and HRSEM images we distinguished chloroplasts with lobes than chloroplasts without lobes in Yuzurua poiteaui var. gemmifera (Harvey) M. J. Wynne and Laurencia dendroidea J. Agardh cortical cells. The LSM images showed chloroplasts lobes (CLs) with different morphologies, varying from thicker and longer undulated projections in Y. poiteaui var. and L. dendroidea to very small and thin tubules as in Laurencia translucida Fujii & Cordeiro-Marino. The diameter and length of CLs from Y. poiteaui var. and L. dendroidea were significantly higher than L. translucida CLs (p < 0.01). Based on LSM observations, we suggest that lobes morphology has a taxonomic validity only to characterize L. translucida species.
Purpose: Laser treatment has become a popular method in implant dentistry, and lasers have been used for the decontamination of implant surfaces when treating peri-implantitis. This study was performed to evaluate the effects of an Erbium-doped:Yttrium-Aluminum-Garnet (Er:YAG) laser with different settings on machined (MA), sand-blasted and acid-etched (SA), and resorbable blast media (RBM) titanium surfaces using scanning electron microscopy and confocal microscopy. Materials and Methods: Four MA, four SA, and four RBM discs were either irradiated at 40 mJ/20 Hz, 90 mJ/20 Hz, or 40 mJ/25 Hz for 2 minutes. The specimens were evaluated with scanning electron microscopy and confocal microscopy. Result: The untreated MA surface demonstrated uniform roughness with circumferential machining marks, and depressions were observed after laser treatment. The untreated SA surface demonstrated a rough surface with sharp spikes and deep pits, and the laser produced noticeable changes on the SA titanium surfaces with melting and fusion. The untreated RBM surface demonstrated a rough surface with irregular indentation, and treatment with the laser produced changes on the RBM titanium surfaces. The Er:YAG laser produced significant changes on the roughness parameters, including arithmetic mean height of the surface (Sa) and maximum height of the surface (Sz), of the MA and SA surfaces. However, the Er:YAG laser did not produce notable changes on the roughness parameters, such as Sa and Sz, of the RBM surfaces. Conclusion: This study evaluated the effects of an Er:YAG laser on MA, SA, and RBM titanium discs using confocal microscopy and scanning electron microscopy. Treatment with the laser produced significant changes in the roughness of MA and SA surfaces, but the roughness parameters of the RBM discs were not significantly changed. Further research is needed to evaluate the efficiency of the Er:YAG laser in removing the contaminants, adhering bacteria, and the effects of treatment on cellular attachment, proliferation, and differentiation.
One of the most important factors determining the properties of a material is its grain size. However, unclear grain boundaries in the image hinder an accurate measurement of grain size. We demonstrate that grain boundaries existing in the images obtained by scanning transmission electron microscopy (STEM) can be clearly distinguished by applying a Sobel filter to a tilting series of STEM images of a hydrogenation-disproportionation-desorption-recombination processed Nd2Fe14B magnet sample.
A distinctive neuronal network in the brain is believed to make us unique individuals. Electron microscopy is a valuable tool for examining ultrastructural characteristics of neurons, synapses, and subcellular organelles. A recent technological breakthrough in volume electron microscopy allows large-scale circuit reconstruction of the nervous system with unprecedented detail. Serial-section electron microscopy-previously the domain of specialists-became automated with the advent of innovative systems such as the focused ion beam and serial block-face scanning electron microscopes and the automated tape-collecting ultramicrotome. Further advances in microscopic design and instrumentation are also available, which allow the reconstruction of unprecedentedly large volumes of brain tissue at high speed. The recent introduction of correlative light and electron microscopy will help to identify specific neural circuits associated with behavioral characteristics and revolutionize our understanding of how the brain works.
