Simulated Moving Bed [SMB] (모사 이동층 크로마토그래피)

  • Chromatography has been a method of choice in the separation of complex biological mixtures for the analytical purpose in particular for the last half of century. In current years, chromatographic method extends its use to the preparative separation where the productivity per resin amount and solvent use become a matter of concern. Recently, simulated moving bed (SMB) method which claims high separation efficiency of the ideal counter-current moving bed chromatography has become a workhorse of preparative separation. SMB technology was developed in the early 1960s for large-scale hydrocarbon separation by UOP and approximately 120 Sorbex units have been licensed to date. Recently, SMB separation technology has been successfully extended from hydrocarbons and sugars to fine chemicals, particularly biochemicals, from laboratory to pilot to production plant. In this paper, the current status of SMB and its modifications were reviewed.

Degradation of Volatile Organic Compound Mixtures Using a Biofiltration System (생물여과 시스템을 이용한 다성분계 휘발성 유기화합물의 분해)

  • A bench-scale air biofilter was evaluated for the removal of volatile organic compounds (VOCs) from a gas stream. Compost and peat were used as the biological attachment media. Biofilter operating parameters such as incoming VOCs concentrations, temperature, and packing materials were examined. After 26 days of acclimation periods, at 25$^{\circ}C$ and 45$^{\circ}C$, the biofilter removed more than 90% of 30 to 72 mg/㎥ of total VOC. After 40 days of operation, the concentrations of isoprene, toluene, and m-xylene were reduced to 96∼99, 91∼93, and 91∼93% of the original concentrations. VOC removal efficiency was not affected by the temperature. The medium pH was maintained near neutral (pH 6.5∼7.1). After 37 days of operation, the total bacteria count in the biofilter media increased to 1.12${\times}$10(sup)8 cells/g of medium.

  • A thermal decomposition of the tert-butoxy radical has been studied in the gas phase over the pressure range of 1-200 torr at $413_{\circ}K$ using di-tert-butyl peroxide + trimethylsilane mixtures. The relative rate constants were obtained by studying the competitive reactions between tert-butoxy radical decomposition 4(t-BuO·→ CH_3COCH_3 + CH_3·)$ and hydrogen abstraction reaction from trimethylsilane $(t-BuO·+ HSi(CH_3)_3 → t-BuOH + Si(CH_3)_3)·).$ The conventional RRKM calculations were carried out to compare the observed fall-off behavior of the decomposition rate constant $({\kappa}_d)$ with the theoretical predictions using reasonable values of input parameters. In all cases the calculated half-rate pressure $(P_{1/2})$ were significantly higher than those observed. The failure of RRKM to reproduce the fall-off behavior led us to suggest that not all of vibrational modes contribute to excitation (leading to decomposition) on the same time scale.

  • Fine particles of water-soluble pharmaceuticals were prepared using a new micronization method, Carbon Dioxide Assisted Nebulization in a Bubble Dryer(CAN-BD). The process utilized mixtures of CO$_2$ in aqueous solution at supercritical conditions to form an emulsion. The aerosols were dried with pre-heated nitrogen, and the drying chamber was operated at near atmospheric pressure. The dry particles were collected on membrane filter at the bottom of the drying chamber. Several processing parameters such as flow rate, temperature, pressure, solid concentration and processing scale were accessed using NaCl, human serum albumin, and granulocyte-colony stimulating factor as model pharmaceuticals. Nano-scale Proteomics Approach Using Two-dimensional Fibrin Zymography Combined with Fluorescent SYPRO Ruby Dye

  • In general, a SYPRO Ruby dye is well known as a sensitive fluorescence-based method for detecting proteins by one-or two-dimensional SDS-PAGE (1-DE or 2-DE). Based on the SYPRO Ruby dye system, the combined two-dimensional fibrin zymography (2-D FZ) with SYPRO Ruby staining was newly developed to identify the Bacillus sp. proteases. Namely, complex protein mixtures from Bacillus sp. DJ-4, which were screened from Doen-Jang (Korean traditional fermented food), showed activity on the zymogram gel. The gel spots on the SYPRO Ruby gel, which corresponded to the active spots showing on the 2-D FZ gel, were analyzed by a matrix-assisted laser desorption ionization time of flight (MALDI-TOF) mass spectrometric analysis. Five intracellular fibrinolytic enzymes of Bacillus sp. DJ-4 were detected through 2-D FZ. The gel spots on the SYPRO Ruby dye stained 2-D gel corresponding to 2-D FZ were then analyzed by MALID TOF MS. Three of the five gel spots proved to be quite similar to the ATP-dependent protease, extracellular neutral metalloprotease, and protease of Bacillus subtilis. Also, the extracellular proteases of Bacillus sp. DJ-4 employing this combined system were identified on three gels (e.g., casein, fibrin, and gelatin) and the proteolytic maps were established. This combined system of 2-D zymography and SYPRO Ruby dye should be useful for searching the specific protease from complex protein mixtures of many other sources (e.g., yeast and cancer cell lines).

  • Alkali-activated slag concretes are being extensively researched because of its potential sustainability-related benefits. For such concretes to be implemented in large scale concrete applications such as infrastructural and building elements, it is essential to understand its early and long-term performance characteristics vis-a'-vis conventional ordinary portland cement (OPC) based concretes. This paper presents a comprehensive study of the property and performance features including early-age isothermal calorimetric response, compressive strength development with time, microstructural features such as the pore volume and representative pore size, and accelerated chloride transport resistance of OPC and alkali-activated binder systems. Slag mixtures activated using sodium silicate solution ($SiO_2$-to-$Na_2O$ ratio or $M_s$ of 1-2) to provide a total alkalinity of 0.05 ($Na_2O$-to-binder ratio) are compared with OPC mixtures with and without partial cement replacement with Class F fly ash (20 % by mass) or silica fume (6 % by mass). Major similarities are noted between these binder systems for: (1) calorimetric response with respect to the presence of features even though the locations and peaks vary based on $M_s$, (2) compressive strength and its development, (3) total porosity and pore size, and (4) rapid chloride permeability and non-steady state migration coefficients. Moreover, electrical impedance based circuit models are used to bring out the microstructural features (resistance of the connected pores, and capacitances of the solid phase and pore-solid interface) that are similar in conventional OPC and alkali-activated slag concretes. This study thus demonstrates that performance-equivalent alkali-activated slag systems that are more sustainable from energy and environmental standpoints can be proportioned.

Rate-Product Correlations for the Solvolysis of 5-Nitro-2-Furoyl Chloride

  • The solvolysis rate constants of 5-nitro-2-furoyl chloride (5-$NO_2(C_4H_2O)$-2-COCl, 1) in 27 different solvents are well correlated with the extended Grunwald-Winstein equation, using the $N_T$ solvent nucleophilicity scale and YCl solvent ionizing scale, with sensitivity values of $1.20{\pm}0.05$ and $0.37{\pm}0.02$ for l and m, respectively. The activation enthalpies (${\Delta}H^{\neq}$) were 5.63 to $13.0kcal{\cdot}mol^{-1}$ and the activation entropies (${\Delta}S^{\neq}$) were -25.9 to $-43.4cal{\cdot}mol^{-1}{\cdot}K^{-1}$, which is consistent with the proposed bimolecular reaction mechanism. The solvent kinetic isotope effect (SKIE, $k_{MeOH}/k_{MeOD}$) of 2.65 was also in accord with the $S_N2$ mechanism and was possibly assisted using a general-base catalysis. The product selectivity (S) for solvolysis of 1 in alcohol/water mixtures was 1.2 to 11, which is also consistent with the proposed bimolecular reaction mechanism.

  • Mixtures of hog manure slurry and sawdust were composted by an intermittent aeration method to verify the performance evaluation of pilot-scale reactor vessels during composting high rate (decomposition) process. Instrumentation was designed to measure temperatures in compost, oxygen and carbon dioxide concentration, air flow rates, and ammonia gas emitted. It was found that ammonia concentration during composting high rate decreased more quickly to the allowable range of 34-40 ppm after 14days at near the optimal levels (II) than in the case of lower levels (I). The influence of the optimal levels (II) such as moisture content (55-65%), C/N ratio (20-40), pH (7-8) and temperature in compost (<$60^{\circ}C$) on the reduction of ammonia gas was considerable for commercial composting.

  • A comparison was made between workers exposed to organic solvents and those not exposed to them regarding stress perception and psychopathology, using GARS scale and SCL-90-R. The exposed group scored significantly higher than the non-exposed group in stress perception relevant to job-wort changes in relationship, sickness or injury, financial, and overall global area. The exposed group was significantly higher in somatization scale than the non-exposed group. However, there was no significant correlation between duration of exposure and psychopathology. In each of the groups. female workers showed more psychopathology than male ones. Age showed significantly nagative correlation with psychopathology. These results suggest that the exposed group shows higher stress perception and more psychopathology than the non-exposed group. Thus. it is emphasized that health policy makers as well as employers should pay more attention to health. espcially mental health for the workers exposed to organic solvents.
