Cultural Characteristics and Fruiting-body Productivity of New Cultivar 'Hwadam' (Lentinula edodes) by Punching Treatments on the Sawdust Medium (톱밥배지 침공처리에 따른 표고 '화담' 의 균사배양 및 자실체 생산성)

  • The effect of punching treatment on bags containing sawdust medium for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was investigated. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached 27.0-28.8℃ on the 23rd day of incubation, and 14.0-15.6% on the 16th day of incubation, respectively, decreasing thereafter. The O2 concentration showed an opposite pattern to the CO2 concentration. As the number of punching treatments on the medium increased during the incubation period, the lightness value decreased, and conversely, the color difference and weight loss rate increased. There was no difference in the cultivation period according to the punching treatment. As the punching treatment increased on the sawdust medium, the fruiting body yield increased; a three-time punching treatment produced the best yield with 605.7 g. Thus, proper punching treatment on a sawdust medium during the incubation period can increase the number of fruiting bodies and yield.

  • Six strains of Sparassis crispa such as S. crispa DUM-01, DUM-02, DUM-03, DUM-04, DUM-05, and DUM-06 were tested for their mycelial growth on 6 different kinds of sawdust media and primordial formation on 10 different compositions of larch sawdust media. The highest mycelial growth was recorded on the larch sawdust. Of the 6 strains of S. crispa, S. crispa DUM-04 recorded the favorable formation of primordia. The primordial formation of S. crispa DUM-04 was more favorable on L-3 medium than 9 kinds of larch sawdust media. When S. crispa DUM-04 was cultured on the media of larch sawdust+pine sawdust, the formation of its fruiting bodies was more outstanding on the media of larch sawdust+pine sawdust than those of larch sawdust.

Comparison of Productivity According to Sawdust Size and Effect of Additives for Sawdust Cultivation of Shiitake (표고톱밥재배시 톱밥 크기 및 첨가제의 혼합 여부에 따른 생산성 비교)

  • Studies were processed to confirm the differences of productivity according to sawdust size and effect of additives for sawdust cultivation of shiitake. In results of investigation until shiitake mycelia fully spread on surface of sawdust medium, mycelial growth in treatment of sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$, and treatment of sawdust sizw $3{\sim}5\;mm$ without $CaCO_3$ and $KNO_3$ was the fastest. And, in investigation of rate of weight reduction, treatment of sawdust size below 1 mm without $CaCO_3$ and $KNO_3$ was the highest. The tendency of fruiting was somewhat different according to treatments. In case of sawdust size below 1 mm with $CaCO_3$ and $KNO_3$, yield produced from 1st to 2nd flushing period was over 80%. However, in case of sawdust size below 1 mm plus $5{\sim}7\;mm$ without $CaCO_3$ and $KNO_3$, yield produced from 3rd to 4th flushing period was more than it from 1st to 2nd flushing period and rate of yield produced from 1st to 2nd flushing period to total yield was 34%. Results of investigation on productivity, yield and the number of fruit-bodies were remarkably different according to treatments. Sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$, whose total yield and the number of fruit-bodies were 568 g and 67, respectively, was the highest. And, total yield of medium of sawdust size below 1 mm plus $2{\sim}3\;mm$ without $CaCO_3$ and $KNO_3$ was lowest as 227 g and the number of fruit-bodies of medium of sawdust size below 1 mm plus $5{\sim}7\;mm$ without $CaCO_3$ and $KNO_3$ was lowest as 24. In investigation on amount of fruit-bodies over 10 g, medium of sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$ was highest as 397 g.

Reutilization of Enokitake Cultural Waste as Lentinus edodes Cultivation Substrate

  • The availability of enokitake cultural waste for Lentinus edodes cultivation was investigated, although hardwood sawdust has traditionally been used as a substrate for this fungus. Firstly, physiochemical characteristics of cultural waste were analysed. Secondly, mycelial growth characteristics and fruiting yields of L. edodes on waste treated in some methods were determined. Physiochemical characteristics of enokitake cultural waste showed that the millwaste complex was a little degraded by enokitake fungus and suggested the probability that most component lost by enokitake could be rice bran. Mycelia of L. edodes grew and fruited well on waste supplemented by fresh rice bran and Quercus sawdust although didn't on waste only. Mycelial growths of these fungi on waste were accelerated when supplemented by rice bran to the percent of 40(w/w) but decreased or suppressed at above ratios(30, 40%, w/w). Supplementations of oak sawdust at above 40%(w/w) of the waste and rice bran at 20%(w/w) of the sawdust allowed such a good mycelial growth as to be selected as a pertinent mixing ratio for fruiting medium. A fruiting yield on enokitake cultural waste supplemented by oak sawdust (at 40% of the waste, w/w) and rice bran (at 20% of the sawdust, w/w) was not inferior to that on oak sawdust supplemented by rice bran only (at 20% of the sawdust, w/w). These results indicated strongly the potentiality of enokitake cultural waste as raw materials for shiitake cultivating substrates.

Methods of Agrocybe cylindracea simultaneous cultivation for small scale bottle cultivation farmers of Pleurotus eryngii (소규모 큰느타리 병재배 농가에서 버들송이 동시재배 방법)

  • Small scale mushroom growers take a lot of the costs in the bottle cultivation than the large scale growers. Therefore, they are not competitive in the market. Mushroom cultivation in small scale mushroom farm is labor-intensive and this experiment was carried out to establish the method for the cultivation of various kinds of mushrooms together under the similar conditions in the production system. As a result, the common medium were selected based on the growing conditions of Pleurotus eryngii and Agrocybe cylindracea, and the level of medium moisture content were studied. The results were shown below. When the medium in the input stage for bottle cultivation was filled by using automatic machine, the range of the three state rates in the bottle is different depending on ingredients and the mixing ratio. The optimum moisture content of the medium for some mushroom production was a different trend based on the medium cluster of the raw materials. The optimum moisture content for P. eryngii in the medium was 70% (douglas fir sawdust, rice bran or sawdust, dried bean curd refuse). In the medium containing douglas fir sawdust, wheat bran was 65%, and the medium including douglas fir sawdust, wheat bran, dried bean curd refuse was 67%. The optimum moisture content of the above three media for A. cylindracea was 70%. The suitable medium for the bottle cultivation of P. eryngii was selected as douglas fir sawdust 75%, wheat bran 20%, dried bean curd refuse 5%, and moisture content 67%. The medium of A. cylindracea was selected as douglas fir sawdust 75%, wheat bran 25%, and moisture content 70%. The suitable medium for bottle cultivation of P. eryngii and A. cylindracea was shown as douglas fir sawdust 75%, rice bran20%, dried bean curd refuse 5%, and 70% moisture content to be used as a common medium of the growing. The incubation period, primordial formation, and growth environment conditions of P. eryngii and A. cylindracea were a similar trend. Therefore, the small scale farmers of P. eryngii can cultivate A. cylindracea together with P. eryngii.

Comparison of Productivity according to Different Quantity of Sawdust and Liquid Spawn for Sawdust Cultivation of Shiitake (표고톱밥재배시 톱밥종균과 액체종균의 접종량 차이에 따른 생산성 비교)

  • Studies were made to investigate the difference of the shiitake productivity according to the use of sawdust and liquid spawn on middle-temperature type strain. Treatments inoculated with sawdust spawn have shown that yields were dispersed according to flushing periods and fruit-bodies occurred sporadically on medium. However, treatments inoculated with liquid spawn have shown that yield was relatively concentrated at 1st flushing period and fruit-bodies were densely occurred on medium. At total yield and the number of fresh fruit-bodies until seventh flushing, treatments inoculated with liquid spawn were more than those inoculated with sawdust spawn. Also, biological efficiency (B.E.) of inoculation with 20 ml liquid spawn which produced 30% of the medium weight was highest as 69%, but biological efficiency of inoculation with 10 g sawdust spawn was lowest as 42%. And, although rate of fruit-bodies over 10 g in total yield was high on treatments inoculated with sawdust spawn, amounts of fruit-bodies over 10 g were not different among treatments.

Effects of Sterilization and Cultivation Temperatures of Oak Sawdust Medium on Lentinula edodes Hyphal Growth (참나무 톱밥배지의 살균 및 배양온도가 표고 균사생장에 미치는 영향)

  • Sterilization of oak sawdust at $65^{\circ}C$ for Lentinula edodes bed cultivation can be efficient in sterilization facility cost, but its effect on the mushroom production is uncertain due to high contamination probability. The effective conditions for L. edodes hyphal growth in the low temperature sterilized oak sawdust were investigated with combinations of three sterilization temperatures ($65^{\circ}C$, $100^{\circ}C$ and $121^{\circ}C$) and four cultivation temperatures ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$). L. edodes inoculation density effect was also tested with 1 cm, 2 cm, and 4 cm distance in the sawdust (4%, 11% and 25% inoculation rate by surface area). L. edodes hyphal growth in the sawdust sterilized at $65^{\circ}C$ was as much as at those $100^{\circ}C$ and $121^{\circ}C$ when the fungus cultured below $25^{\circ}C$, but it was greatly reduced when cultured at $30^{\circ}C$. And the sawdust medium with 1cm distance inoculation density was fully occupied with L. edodes hyphae, but those with 2~4cm distance inoculation were contaminated by 4~33%. Therefore, we conclude that low temperature sterilized oak sawdust needs to be cultured under $25^{\circ}C$ after sufficient inoculation by 25% for successful bed cultivation of L. edodes.

Effect of Sawdust Culture on Oak Mushroom, Lentinula edodes (Berk.) Pegler by Inoculation of the Liquid Spawn (액체종균(液體種菌) 접종(接種)에 의한 표고톱밥재배(栽培) 효과(效果))

  • For cultivation on sawdust-bed of oak-mushroom until present time, inoculation of spawn on sawdust bed has been performed by sawdust spawn. But, liquid spawn may have advantages for rapid mass production of spawn, and now, sawdust-cultivation by liquid spawn inoculation should be applied instead of sawdust spawn. Therefore, investigations were performed to evaluate the effect of sawdust-cultivation by liquid spawn inoculation. The results were as follows: 1. When 11 kinds of liquid media were applied, the oak-mushroom culture medium was the most excellent in growth. Most suitable temperature at PDA was $25^{\circ}C$, and $22.5\sim27.5^{\circ}C$ in range were optimal for liquid culture. In liquid culture, amount of mycelial growth increases rapidly up to 40 days of cultivation. Incubation at fermentor brought yield of 106mg dry mycelia per 40ml media after 17 days. 2. In 1l-spawn bottle, growth of mycelium by inoculation of 20ml-liquid spawns were faster than 6g-sawdust spawn in spread of mycelia. On 2kg-bag culture, inoculations of 10ml-, 20ml- and 30ml-liquid spawns were all slower than 20g-sawdust spawn in mycelial spread. So, amount increasement in ampunt of liquid spawn should be discussed. Yields of mushrooms until third sproutings of 2kg-bag culture were 580g in 30ml-liquid spawn inoculation, but 510g, 486g and 470g from 20g-sawdust spawn, 20ml-liquid spawn and 10ml-liquid spawn, respectively. Thus, 30ml-liquid spawn inoculation was highest in yield.

The effects of biomaterials in growing medium on the response of Zelkova serrata in a containerized production system

  • Changes in the physical and chemical properties of soil materials during the nursing process have a great influence on the quality of containerized seedlings and on growth and survival after planting. In this study, the effect of biomaterials and their mixed ratios on the growth of Zelkova serrata seedlings in a containerized seedling production system was investigated. Mushroom sawdust, pine bark, and carbonized rice husk were used as biomaterials. The mixed ratios were 10% and 20% volume ratio of the growing medium volume, including the untreated controls. There was no significant difference in the height growth of the Zelkova serrata seedlings according to the biomaterials. The root collar diameter was the highest with the 20% carbonized rice husk and the lowest with the mushroom sawdust. The difference between the highest quality index and the lowest quality index was 30% in the order of the carbonized rice husk, pine bark, control, and mushroom sawdust, but there was no statistical significance. In this study, if the growing medium mixed with biomaterials does not reduce the seedling growth compared with the control, it is considered that the biomaterial can replace a part of the growing media. Therefore, the results show that some of the growing media can be replaced with carbonized rice husk or pine bark when producing Zelkova serrata seedlings.

Effects of difference in medium composition on the growth of Lentinula edodes (배지조성에 따른 표고 톱밥재배의 생육 효과)

  • This study was carried out to select the suitable for bag cultivation of Lentinula edodes. We investigated the optimal major materials and its mixing ratio in bag cultivation of L. edodes, 'Sanjo701ho' and 'Nongjingo'. The Suitable substrates for L. edodes bag cultivation were oak sawdust and douglas fir sawdust and rice bran mixed ratio 40:40:20(v/v). At the result, the period mycelial incubation shortened up to 13~18 days compared to the control. And Yield of commercial fruiting bodies was 17~19% higher than that in control. We expect that the cultivation period of L. edodes will be shortened, and the yield increased in the medium replaced half of oak sawdust by douglas fir sawdust.
