Cultivation Characteristics of Sparassis crispa Strains Using Sawdust Medium of Larix kaempferi (낙엽송 톱밥을 이용한 꽃송이버섯 균주별 재배 특성)

  • Cultivation characteristics of 12 strains of cauliflower mushroom (Sparassis crispa) collected in Korea were investigated by growing the mushroom on sawdust medium of Larix kaempferi. As cultivation characteristics, incubation period for full growth of mycelium in a cultivation bottle, cultivation time period taken for first harvest, and mushroom color and yield were examined. S. crispa KFRI 723 showed the shortest for incubation period with 59 days while S. crispa KFRI 746 showed the longest with 94 days. The earliest mushroom harvesting was achieved by 29 days from S. crispa KFRI 746 and the latest was by 63 days from S. crispa KFRI 691. The colors of fruit body of the tested strains can be divided into three groups; S. crispa KFRI 700 was white, S. crispa KFRI 747 was yellow brown, and the others were light yellowish. KFRI 700 yielded the most as 163 g from 380 g sawdust media, while KFRI 746 and KFRI 747 were the lowest with 58 g and 35 g, respectively. As results of cultivation characteristics of 12 strains of cauliflower mushroom, we consider that three strains (KFRI 700, 723 and 724) of S. crispa are suitable for sawdust cultivation on L. kaempferi in the aspects of mycelial growth period, harvesting period and mushroom production, respectively.

Studies on the optimal conditions for the high yield in the production of Ganoderma lucidum (영지버섯 다수확 생산을 위한 최적조건에 관한 연구)

  • The results were as follows; 3 additives(rice bran, wheat bran and soybean bran) to the Quercus dentata sawdust were applied at 10, 20 and 30%. High yield was recorded at 20% of rice bran and wheat bran and 30% of soybean bran. Among various sawdust media, the cultivation with the Quercus dentata sawdust alone showed the most favored growth index, such as 22 days of full mycelial growth, 11 days of pinheading from spawning and 23.5g per $1,200m{\ell}$ bottle. In the size of bottles, the lowest infection ratio was 5 % at $1,200m{\ell}$ bottle. In the growth characteristics with the bottle size, Reishi at $1,600m{\ell}$ bottle showed best result when it is observed for 13~15 days in pinheading from spawing, 36~37 days in fruit bodies growing and 26.7~29.3g yield per bottle. The highest yield per area was 6.4kg at horizontal bed with 218 bottles per $3.3m^2$. The Reishi produced in $1,600m{\ell}$ bottle with horizontal cultivation had less number of fruit bodies and best yield, thus it is determined to be the most economic method.

  • We investigated the standard cultivation substrate for Sparassis latifolia "Neowul" bred in Jeollabuk-do Agricultural Research and Extension Services. Cultivation characteristics and yield were assessed by varying the kind of sawdust and additives, and the mixing ratio. The cultivation period in larch sawdust was the shortest at 97 days. The yield was excellent (143.6 g). The findings indicated that larch is a tree species appropriate for the cultivation of S. latifolia. When the additives were varied, the yield and productivity (53.1%) were highest (116.6 g) for the wheat bran treatment. Thus, wheat bran would be an additive appropriate for culturing S. latifolia. Concerning the use of different mixing ratios, larch sawdust:beet-pulp:wheat bran ratios of 80:15:5 and 85:10:5 resulted in yields of 114.4 g and 111.4 g, and productivity of 52.5% and 51.8%, respectively. These yield and productivity values were not statistically different. Thus, the standard cultivation substrate for S. latifolia can comprise larch sawdust, beet pulp, and wheat bran at a ratio of 80:15:5 or 85:10:5. The carbon/nitrogen (C/N) ratio assumed to be appropriate for the cultivation of S. latifolia was 184-223. Pinheading would be difficult below a C/N substrate ratio of 105. Thus, the C/N ratio of the media, as well as the pH, would be vital factors affecting pinheading during S. latifolia cultivation.

Mycelial Growth of Monokaryotic and Dikaryotic Strains of Lentinula edodes Cultivars for Sawdust Cultivation on the Agar and Sawdust Culture media (톱밥재배용 표고 품종의 단핵균사체와 2핵균사체의 한천과 톱밥배지에서의 균사생장)

  • In order to develop the culture material for breeding of Lentinula edodes, dikaryotic and monokaryotic mycelia were cultured in agar and sawdust medium. The cultivars were used Sanjo No. 701, Sanjo No. 705, Chamaram and Chuje No. 2 in this study. The mycelial growth of the cultivars were highest in PDA except for Sanjo No. 705, but Sanjo No. 705 showed the best mycelial growth in MCM. Regardless of the cultivars, the mycelial growth showed the most stable in MEA. There was no significant difference in mycelial growth among the cultivars in sawdust medium, but it was about 10% faster than that of Sanjo No. 701 and 705. Monokaryotic strains of Sanjo No. 701 and Chamaram showed less mycelial growth than dikaryotic mycelium. Dikaryotic mycelium of Chamaram showed better mycelial growth than that of Sanjo No. 701, but monokaryotic mycelium of Chamaram showed lower mycelial growth than Sanjo No. 701. The selected monokaryotic mycelium has a wide varience of mycelial growth, and the morphologies of the colonies are very diverse, so those are presumed that wide variences of monokaryotic are selected in genetically, and these monokaryotic mycelium are expected to be a good breeding materials.

Methods of Agrocybe cylindracea simultaneous cultivation for small scale bottle cultivation farmers of Pleurotus eryngii (소규모 큰느타리 병재배 농가에서 버들송이 동시재배 방법)

  • Small scale mushroom growers take a lot of the costs in the bottle cultivation than the large scale growers. Therefore, they are not competitive in the market. Mushroom cultivation in small scale mushroom farm is labor-intensive and this experiment was carried out to establish the method for the cultivation of various kinds of mushrooms together under the similar conditions in the production system. As a result, the common medium were selected based on the growing conditions of Pleurotus eryngii and Agrocybe cylindracea, and the level of medium moisture content were studied. The results were shown below. When the medium in the input stage for bottle cultivation was filled by using automatic machine, the range of the three state rates in the bottle is different depending on ingredients and the mixing ratio. The optimum moisture content of the medium for some mushroom production was a different trend based on the medium cluster of the raw materials. The optimum moisture content for P. eryngii in the medium was 70% (douglas fir sawdust, rice bran or sawdust, dried bean curd refuse). In the medium containing douglas fir sawdust, wheat bran was 65%, and the medium including douglas fir sawdust, wheat bran, dried bean curd refuse was 67%. The optimum moisture content of the above three media for A. cylindracea was 70%. The suitable medium for the bottle cultivation of P. eryngii was selected as douglas fir sawdust 75%, wheat bran 20%, dried bean curd refuse 5%, and moisture content 67%. The medium of A. cylindracea was selected as douglas fir sawdust 75%, wheat bran 25%, and moisture content 70%. The suitable medium for bottle cultivation of P. eryngii and A. cylindracea was shown as douglas fir sawdust 75%, rice bran20%, dried bean curd refuse 5%, and 70% moisture content to be used as a common medium of the growing. The incubation period, primordial formation, and growth environment conditions of P. eryngii and A. cylindracea were a similar trend. Therefore, the small scale farmers of P. eryngii can cultivate A. cylindracea together with P. eryngii.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.

Free sugar, amino acid, and beta-glucan content in Lentinula edodes strains collected from different areas (표고 수집종들의 유리당, 아미노산 및 β-glucan 함량)

  • In this study, we analyzed the nutrient composition and ${\beta}$-glucan content in Lentinula edodes fruiting bodies from different collection areas. Four types of free sugars were detected by HPLC, and the range of trehalose prevalence was 0.83% to 9%. The highest total amino acid content was observed from sawdust media cultivation of Lentinula edodes fruiting bodies, collected in China (JMI10050)The highest essential amino acid content, assessed by log cultivation of Lentinula edodes fruiting bodies, collected in Jangheung (JMI10059). Sixteen free amino acids were detected in Lentinula edodes fruiting bodies, and the major free amino acids were histidine, glutamic acid, and arginine. The highest essential amino acid including threonine, valine, methionine, isoleucine, leucine, phenylalanine, histidine, and lysine was fromsawdust media cultivation of Lentinula edodes fruiting bodies collected in China(JMI10052). The ${\beta}$-glucan content from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10059 and JMI10066) was higher than that from sawdust media cultivation of Lentinula edodes fruiting bodies collected in China. The highest ${\beta}$-glucan content was observed from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10066).

Grouping of Ganoderma strains based on cultural characteristics and fruitbody morphology (영지버섯 수집균의 배양적 특성 및 자실체 형태에 따른 구분)

  • To establish a genetic relationships of collected Ganoderma strains, mycelium growth according to media and temperature, colony morphology, chlamydospore formation and fruitbody morphology were investigated. For the identification of optimal growth conditions of the strains, five different growth media and four different temperature were tested. GCM (Ganoderma complete medium) at $30^{\circ}C$ was the most effective for mycelial growth of 68 strains with more or less variation. The strains were divided into 28 groups based on their colony shapes, and most of them belong to CM3 or CM8 group. Chlamydospores were observed in the mycelia of 16 strains including ASI 7022 on microscope, but not in most G. lucidum domestic strains, which showed relatively lagging growth on $35^{\circ}C$ in mycelial growth experiment. These results were not similar to those of G. lucidum but those of G. tsugae imported from USA. The strains were cultivated on oak sawdust media to see their fruit body formation. Ninety-seven among 115 strains formed fruitbodies in sawdust cultivation. They showed two forms of fruitbodies, 89.7% of flat type or 10.3% of antler type, although these shapes can be affected by $CO_2$ concentrations. These results suggest that the native strains formerly considered to belong to G. lucidum have to be re-classified with further study.

Growth and Rotting Characteristics of Lentinus edodes Isolates in Korea and Their Adaptability to Bag-Culture Using Oak Sawdust (우리나라 표고의 생장(生長), 부후특성(腐朽特性) 및 톱밥배양(培養)에 관(關)한 연구(硏究))

  • Studies were made to find out the mycelial growth and rotting characteristics of 91 Lentinus edodes isolates collected or hybridized in Korea and abroad, and their adaptability to bag-culture. The results are as follows. 1. There were little differences in mycelial growth at PDA and oak (Quercus mongolica) sawdust-based medium, and rotting among high-, mid- and low temperature types of L. edodes isolates. But, the fruit yield of high-temperature type was much higher than those of the other two types. The fruit yields of the isolates in the same strain group were apparently different. 2. From the sawdust-based culture of 91 isolates, FRI 221, FRI 208 and FRI 169 were selected as excellent strains in yield and Quality, showing fruiting of 157g, 152g and 119g per 1kg-medium, respectively. 3. Attempts to culture in various media with different substrates resulted in almost same fruit yields, and the yield was proportional to medium weights as 2.5kg-medium showed 2.5 times more yield than 1kg-medium. Fruit yields were different according to incubation periods and the period of little more than 100days was best. 4. When the correlation analyses among mycelial growth, rotting ability, yield and fruit-shape normality were made with the 91 isolates, mycelial growth at sawdust-based medium showed highly positive correlation with that at PDA, while fruit yield was negatively correlated with mycelial growth in PDA and sawdust-based medium.

The Effects of Temperature and Nutritional Conditions on Mycelium Growth of Two Oyster Mushrooms (Pleurotus ostreatus and Pleurotus cystidiosus)

  • The influences of temperature and nutritional conditions on the mycelium growth of oyster mushroom Pleurotus ostreatus (PO) and Pleurotus cystidiosus (PC) were investigated in laboratory experiment during the summer season of 2014. The results of the experiment indicated that potato dextrose agar (PDA) and yam dextrose agar (YDA) were the most suitable media for the mycelium growth of oyster mushroom PO while four media (PDA, YDA, sweet potato dextrose agar, and malt extract agar medium) were not significantly different in supporting mycelium growth of oyster mushroom PC. The optimal temperature for mycelium growth of both oyster mushroom species was obtained at $28^{\circ}C$. Mycelium growth of oyster mushroom PO was improved by carbon sources such as glucose, molasses, and at 1~5% sucrose concentration, mycelium colony diameter of mushroom PO was achieved the highest value. Whereas glucose, dextrose, and sucrose as carbon sources gave the good mycelium growth of oyster mushroom PC, and at 1~3% sucrose concentration, mycelium colony diameter of PC was achieved the maximum value. Ammonium chloride concentrations at 0.03~0.09% and 0.03~0.05% also gave the greatest values in mycelium colony diameter of mushroom PO and PC. Brown rice was found to be the most favourable for mycelium growth of two oyster mushroom species. In addition, sugarcane residue, acasia sawdust and corn cob were selected as favourable lignocellulosic substrate sources for mycelium growth of both oyster mushrooms.
