  • 몇 가지 금속을 제거하기 위한 목적으로 건조된 해조류를 이용하였다. 금속 흡착제로 사용하기 위하여 40~60 mesh의 해조 분말을 칼럼에 충진시킨 후, 금속 용액을 1mL/min의 속도로 흘려 보내면서 금속이온을 흡착시켰다. Cu(II), Zn(II) 이온 모두 갈조류인 Sargassum horneri(Turner) C. Agarch보다 녹조류인 Ulva pertusa Kjellman에서 더 많은 흡착량을 보였고 두 해조류 모두 Zn(II) 이온보다 Cu(II) 이온이 더 많이 흡착하였다. 금속의 회수율은 산성 또는 중성일 경우에 켰으며 Zn(II) 이온보다 Cu(II) 이온의 회수율이 다소 높았다.

우뭇가사리와 괭생이모자반을 이용한 친환경 포장 필름 개발 및 특성 연구 (Development and characterization of an eco-friendly packaging film using Gelidium amansii and Sargassum horneri)

  • In this study, a biodegradable packaging film was developed using two marine algae, Gelidium amansii, and Sargassum horneri. The chemical properties and microstructure of the developed film were evaluated using field emission scanning electron microscope, Fourier transform infrared spectroscopy, gas chromatography-Mass spectroscopy, and thermogravimetric analysis. Furthermore, the mechanical properties and toxicity of the film were evaluated using the ISO 1924 and IEC 62321 methods, respectively. The biodegradability of the film was evaluated according to ISO 14855-1:2012 method. The film was primarily made of cellulose and had biodegradability that was about 17 times greater than that of PBS, a representative eco-friendly plastic. Moreover, the mechanical properties improved by approximately 40% compared to the seaweed-based film of the previous study. The virulence test revealed that the content of all of the toxic substances listed in IEC62321 was below the measurement limit. An egg carton that can be used in practice was manufactured in accordance with ISO 534, and its applicability was tested using the biodegradable packaging film prepared.

괭생이모자반(Sargassum horneri)으로부터 추출한 다당의 이화학적 특성 (Isolation and Characterization of Polysaccharides Purified from Brown Alga Sargassum horneri)

  • Physicochemical properties of fucoidan and alginate extracted from Sargassium horneri were investigated. The alginates were extracted and purified via three different routes (CaCl2, HCl and ethanol routes), and their rheological properties at various concentrations and temperatures were determined. The yield of fucoidan was 3.08%, and it was mainly composed of fucose and galactose. Fourier-transform infrared spectrum of fucoidan showed strong absorption band at 1,254 and 827 cm-1 corresponding to the sulfate group. Flow behavior of the alginate solution was characterized using the power-law model. The consistency index increased with increasing concentrations. The sodium alginate solution (1.5%) exhibited Newtonian behavior when extracted via the CaCl2 and HCl routes, while it exhibited pseudoplastic behavior when extracted via the ethanol route. The effect of temperature on the flow behavior was investigated in terms of the activation energy (Ea), which was obtained using the Arrhenius equation. The Ea value corresponding to the alginate solution decreased with increasing shear rates. The Ea values corresponding to the alginate solutions purified via the CaCl2, HCl, and ethanol routes were 13.54-18.64 kJ/mol, 13.42-19.21 kJ/mol, and 9.51-10.67 kJ/mol, respectively. The low Ea values corresponding to the solutions extracted via the ethanol route suggest that the flow behavior does not depend significantly on temperature.

Tracing the trajectory of pelagic Sargassum using satellite monitoring and Lagrangian transport simulations in the East China Sea and Yellow Sea

  • Northeastward drifts of massive Sargassum patches were observed in the East China Sea (ECS) and Yellow Sea (YS) by the Geostationary Ocean Color Imager (GOCI) in May 2017. Coverage of the brown macroalgae patches was the largest ever recorded in the ECS and YS. Three-dimensional circulation modeling and Lagrangian particle tracking simulations were conducted to reproduce drifting trajectories of the macroalgae patches. The trajectories of the macroalgae patches were controlled by winds as well as surface currents. A windage (leeway) factor of 1% was chosen based on sensitivity simulations. Southerly winds in May 2017 contributed to farther northward intrusion of the brown macroalgae into the YS. Although satellite observation and numerical modeling have their own limitations and associated uncertainties, the two methods can be combined to find the best estimate of Sargassum patch trajectories. When satellites were unable to capture all patches because of clouds and sea fog in the ECS and YS, the Lagrangian particle tracking model helped to track and restore the missing patches in satellite images. This study suggests that satellite monitoring and numerical modeling are complementary to ensure accurate tracking of macroalgae patches in the ECS and YS.

Screening for Angiotensin 1-Converting Enzyme Inhibitory Activity of Ecklonia cava

  • Seven brown algal species (Ecklonia cava, Ishige okamurae, Sargassum fulvellum, Sargassum horneri, Sargassum coreanum, Sargassum thunbergii and Scytosiphon lomentaria) were hydrolyzed using five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) and screened for angiotensin 1-converting enzyme (ACE) inhibitory activities. Most algal species examined showed good ACE inhibitory activities after the enzymatic hydrolysis. However, E. cava was the most potent ACE inhibitor of the seven species. Flavourzyme digest of E. cava exhibited an $IC_{50}$ of around $0.3\;{\mu}g/mL$ for ACE; captopril has an $IC_{50}$ of $\~0.05\;{\mu}g/mL$. The Flavourzyme digest was separated to three fractions by an ultrafiltration membrane (5, 10, 30 kDa MWCO) system according to the molecular weights. The active components were mainly concentrated in >30 kD fraction which are composed of the highest protein content $(27\%)$ and phenolic content (261 mg/100 mL) compared to the other two smaller molecular weight fractions. Therefore, the active compounds appear to be relatively high molecular weight complex molecules associated with protein (glycoprotein) and polyphenols. Therefore, E. cave is a potential source of antihypertensive compound.

괭생이 모자반 추출물의 비장세포 면역활성 증강 효과 (Splenocyte-mediated immune enhancing activity of Sargassum horneri extracts)

  • 본 연구는 괭생이 모자반 추출물의 추출방법에 따른 면역증강활성에 관하여 비교 관찰하였고, 추출물간의 혼합비율을 다르게 혼합하여 제조한 괭생이 모자반 추출물의 최적 면역활성을 갖는 혼합비를 평가하였다. 비장세포에 괭생이 모자반 열수 (HWE), 주정 (EE), 초임계 (SFE) 추출물을 각각 처리하였을 때 HWE 및 SFE 처리구에서 비장세포 증식능 및 사이토카인 (IL-2, IFN-γ) 분비능이 높게 나타나는 것으로 관찰되었고, 각각의 추출물을 50, 100 mg/kg BW의 농도로 2주간 마우스에 투여하고 비장을 적출하여 비장세포 활성능에 관하여 관찰한 결과, HWE 및 SFE 투여구에서 비장세포 증식능 및 사이토카인 분비능이 높게 나타나는 것으로 관찰되었다. 동물실험에서 높은 활성을 보인 HWE와 SFE의 추출물의 혼합 최적비를 평가하기 위하여 HWE에 SFE를 5, 10, 15 및 20%로 혼합하여 대식세포에 처리하여 NO 및 사이토카인 분비능에 관하여 관찰한 결과, SFE를 5%, 10% 혼합한 추출물 처리구에서 활성이 높게 나타나는 것으로 관찰되었으며 혼합추출물을 100 mg/kg BW으로 14일간 투여한 마우스의 비장세포 활성에 관하여 관찰한 결과, SFE를 5%의 비율로 혼합한 추출물 처리구 비장세포 활성이 높게 나타나는 것으로 관찰되어 HWE와 SFE의 최적 혼합 비율을 HWE에 SFE를 5% 첨가하는 것으로 결정하였다. 결정된 최적혼합비로 100 mg/kg BW의 농도로 14일간 투여한 마우스의 비장세포에서 NK 세포를 분리하여 NK 세포의 활성능을 평가한 결과, 대조구 대비 높게 증가하는 것으로 나타났다. 따라서 괭생이 모자반 추출물을 최적비율로 혼합하여 제조한 혼합추출물은 비장세포, 대식세포 및 NK 세포에 대한 높은 활성능을 나타내어 면역증진에 기여하는 것으로 판단된다.

  • This research dealt with the nutrient value of the seaweed, Sargessum horneri (Turner) C. Ag. which showed the highest nutrient value in the previous exporiment (Park 1968). Its objective was to find the most effective mixing ratio of ordinary chicken feed used for pullet. In order to investigate the most effective mixing ratio, seven kinds of sample feed with vary-ing amounts of the seaweed as shown in table 2 were prepared and were fed to seven groups of chickens, each group consisting of twenty, four week old, pullets. In addition, the effect of sodium glutaminate mixed in the feed was also investigated, and the following result was obtained. The average increase in body weight of each group during the experiment period was determined. The group fed with the feed containing $10\%$ powdered Sargassum horneri (Turner) C. Ag. showed the highest body weight increase. In this experiment the body weight increase was accelerated by mixing $0.5\%$ sodium glutaminate In the above feed.

수산 미이용자원 중에 존재하는 효소적 산화 억제제의 검색 -2. 감자 Lipoxygenase-II에 의한 효소적 산화에 대한 억제- (Screening for Potato Lipoxygenase-II Inhibitor in Unused Marine Resources by the Polarographic Method-)

  • 수산 미이용자원을 의약품화 등 고부가가치적으로 이용하기 위한 기초자료 수집을 위해 본 감자 lipoxygenase-II 활성을 조절해 줄 수 있는 천연 저해제의 존재유무를 8종의 수산 미이용자원을 대상으로 각각 순차용매 추출한 후 그들 용매 추출물들에 대해 검색 시도해 보았다. 시험 대상 시료는 별불가사리(Asterina pectinifera), 우렁쉥이(Holocynthia rotetzi)껍질, 오징어(Nototodarus sloani) 먹즙, 성게(Anthocidaris crassispina)껍질, 괭생이모자반(Sargassum horneri), 구멍쇠미역(Agarum cribrosum), 참빗풀(Odonthalia corymbifera) 및 산말(Desmarestia ligulata) 등이었다. 모자반은 물 추출구, 에테르 추출구, 아세톤 추출구 및 메탄올 추출구 모두에서 $IC_{50}$이 각각 320, 18, 9.5 및 $100\;{\mu}g/ml$로 나타나 낮은 농도의 첨가로도 높은 감자 lipoxygenase-II 저해능을 나타내었다. 또한, 불가사리 에테르 추출구와 오징어 먹즙 아세톤 추출구에서도 $IC_{50}$이 각각 29.5, $14.3\;{\mu}g/ml$로 비교적 높은 저해능을 보였으나, 전체적으로 모자반, 구멍쇠미역, 참빗풀, 산말 등 4종의 시험 대상 해조류에 불가사리, 성게, 우렁쉥이껍질, 오징어 먹즙 등 4종의 대상 수산 미이용동물자원에서 보다 저해제가 많이 내재해 있었다. 더욱이, 전반적으로 각 대상 시료의 에테르나 아세톤 등의 비극성용매 추출구쪽에서 물이나 메탄올과 같은 극성용매쪽보다 많은 유용물질의 용출경향을 나타내었다.

평생이모자반 젖산 추출물이 성장기 암컷흰쥐의 골 형성에 미치는 영향 (Effect of Lactic Acid Extracts of Sargassum horneri on Bone Formation in Female Sprage-Dawley Rats)

  • We investigated the effect of a lactic acid extract of Sargassum horneri (ExSL) as a calcium supplement on bone formation in 48 female Sprague-Dawley rats for 4 weeks of their growth phase. The rats were divided into four groups based on diet: two calcium-sufficient and two calcium-deficient diets. The normal control group (NC) was fed AIN-93G; the NCS group was fed the same diet containing 1% extract; the calcium-deficient control (DC) diet was based on AIN-93G; and the DCS group received the same calcium-deficient diet plus 1% extract. Bone formation in the rats was evaluated using the wet weight, length, diameter, and bone mineral density (BMD) of the femur. Serum parameters were also examined. The food intake among the groups did not differ significantly (P<0.05). The NCS group gained the most body weight, while the DC group gained much less weight than the other groups. The feeding efficiencies of the groups that received the extract (NCS and DCS) were slightly higher than those of the control groups (NC and DC). The calcium intakes of all groups depended on the amount of calcium in the feed; the NCS and DCS diets contained 12.15 mg more calcium than the NC and DC diets. The calcium absorption was lower in NCS than in DC and DCS, but significantly higher than in NC (P<0.05). The BMDs in the calcium-sufficient groups were not significantly different (P<0.05), while in the calcium-deficient groups the BMD was significantly higher in DCS than in DC (P<0.05). The serum calcium and phosphorus levels in all groups were not associated with markers of bone growth related to the extract. The osteocalcin content and alkaline phosphatase (ALPase) activity were higher in the calcium-deficient groups than in the normal groups (P<0.05). Ultimately, the osteocalcin content and ALPase activity were lower in DCS compared to DC. These results suggest that the addition of ExSL promotes bone formation and calcium absorption in growing rats.

수산 미이용자원 중에 존재하는 항균성 물질의 검색 (Screening for Antimicrobial Compounds in Unused Marine Resources by the Paper Disk Method)

  • 천연 생리활성물질의 개발과 수산미이용자원의 효율적 이용방안을 모색하기 위하여 별불가사리, 오징어먹즙, 성게껍질, 우렁쉥이 껍질, 구멍쇠미역, 산말, 모자반, 참빗풀 등을 대상으로 물, 에테르, 아세톤, 메탄올로 순차 용매추출하여 각 용매 추출구간별, 시료별 항균성을 검색하였다. 시료추출구간별 평균수율은 물 추출구, 에테르 추출구, 아세톤 추출구 및 메탄올 추출구에서 각각 20, 1.4, 2.8 및 1.1%로 나타나 물추출구의 수율이 가장 높았다. B. subtilis에 대한 항균력 검색 결과, 모자반 에테르 추출구와 메탄올 추출구에서 각각 MIC가 80 및 70 ${\mu}g/disk$로 나타나 비교적 강한 항균력을 가지는 것으로 나타났다. S. aureus에 대한 항균효과는, 모자반의 에테르 추출구, 아세톤 추출구 및 메탄올 추출구의 MIC가 각각 160, 180 및 70 ${\mu}g/disk$로 나타나 비교적 높은 항균성을 보였으며, 참빗풀의 에테르 추출구와 아세톤 추출구에서도 각각 MIC가 50 및 170 ${\mu}g/disk$로서 낮은 농도에서도 항균효과를 나타내었다. 추출구간별 항균효과를 보면, 비극성용매인 에테르 추출구에서 모든 시료의 MIC가 700 ${\mu}g/disk$ 이하로 나타나 에테르 추출구에 S. aureus에 대해 매우 강한 항균효과를 가지는 물질이 존재하는 것으로 나타났다.
