  • The presence of saponin, terpenoid, steroid, anthraquinone and flavonoid were screened with the usual test methods in forty five(sixty four parts) Korean plants belonging to Urticaceae, Celastraceae, Rhamnaceae, Sterculiceae and Rubiaceae. Plants were extracted with methanol and fractionated with hexane, chloroform, ethyl acetate and butanol. Each fraction was tested for the components. The result showed that in saponin test, 21 plants were positive and 11 plants were weak positive; in terpenoid and steroid test, 26 plants were strong positive; in anthraquinone test, 20 plants strong positive; in flavonoid test, 31 plants were positive and 9 plants were weak positive.

  • Crude ginseng saponin fraction reduced the production of thromboxane $A_2$in the lipopolysaccharide-stimulated macrophages. Several kinds of crude saponins showed variant potency that might be caused by the compositional difference of ginseng saponins. From the metabolic labeling experimental data, this reduction of thromboxane $A_2$formation, at least in part, resulted from the reduction of protein synthesis of inducible isozyme of cyclooxygenase(COX-2). This activity may be resulted from the fact that ginseng saponins have steroidal moiety in their structures.

Liquid Membrane Permeation of Nitrogen Heterocyclic Compounds Contained in Model Coal Tar Fraction

  • We investigated the separation of nitrogen heterocyclic compound (NHC) contained in a model coal tar fraction comprising four kinds of NHC [indole (In), quinoline (Q), iso-quinoline (iQ), quinaldine (Qu)], three kinds of bicyclic aromatic compound (BAC) [1-methylnaphthalene (1MN), 2-methylnaphthalene (2MN), dimethylnaphthalene (DMN) mixture with ten structural isomers (DMNs; regarded as one component)], biphenyl (Bp) and phenyl ether (Pe) by liquid membrane permeation (LMP). A batch-stirred tank was used as the permeation unit. An aqueous solution of saponin and n-hexane were used as the liquid membrane and the outer oil phase, respectively. Yield and selectivity of individual NHC was much larger than that of BAC, Bp and Pe. Increasing the initial mass fraction of the saponin to the membrane solution ($C_{sap,0}$) and the initial volume fraction of O/W emulsion to total liquid in a stirred tank (${\phi}_{OW,0}$) resulted in deteriorating the yield of individual NHC, but increasing the stirring speed (N) resulted in improving the yield of each NHC. With increasing $C_{sap,0}$, the selectivity of each NHC based on DMNs increased. Increasing ${\phi}_{OW,0}$ and N resulted in decreasing the selectivity of individual NHC based on DMNs. At an experimental condition fixed, the sequence of the yield and selectivity in reference to DMNs for each NHC was Q > Qu = iQ > In. Furthermore, we compared LPM method with methanol extraction method in view of the separation efficiency (yield, selectivity) of NHC.

Further Evidence in Support of Psychotropic Action on Red Ginseng

  • Using an ethopharmacological technique, we demonstrated that saponin fraction from red ginseng root possessed a potent psychotropic actions on either intermale or maternal aggression models. A series of experiments clearly indicated that one of psychoactive ingredient is ginsenoside Rbl. Although a drug-induced debilitation of motor performance remains a possible cause of the antiaggressive effect of the drug, ginsenoside Rb 1 did not alter the locomotor activity of the mice during agonistic confrontations. Thus, one can eliminate the possibility that the psychoactive effect of ginsenoside Rbl might be concealed by a drugindulced impairment of motor performance. More recently, we developed a new model for copulatory disorder and introduced into the behavioral analysis of drug action. Male mice which has been housed individually from weaning for 5 weeks failed to manifest copulatory behavior when they encountered with the sexually-receptive females. Daily administration of crude ginseng saponin during isolation housing period prevented the development of copulatory disorder, whereas both ginsenoside Rbl and Rgl were ineffective.   • A macromolecular fraction from fresh ginseng root containing mainly polysaccharide showed mild mitogenic activity in human cord blood lymphocytes. 2) When lymphocytes were transformed by Con A or PHA, this fraction could greatly enhance the activity of these lectin mitogens, thus showing a potentiation effect. 3) Although this macromolerular fraction contains saponin and is susceptible to trypsin digestion, it is probably a peptido-glycan in nature on account of its important carbohydrate content and thermal stability. 4) This fraction could not support cancer cell (EAT, K562) growth : its inhibitory effect on these cells remains to be explored.

인삼 사포닌의 동물(쥐) 체내흡수에 관한 연구 (Absorption of Ginseng Saponin in Rats)

  • 인삼(Panax ginseng C. A. Meyer)뿌리 절편을 효소원으로 사용하여 $^14C$-acetate로부터 제조한 방사성 인삼사포닌을 함유한 인삼 사포닌 분획 조제물을 High performance liquid chromatography방법으로 분리하여 각 glycosides의 함량과 비 방사능을 측정하였다. 1mg의 $^14C$-표지사포닌을 함유한 인삼 사포닌을 쥐에게 구경 투여하고 1시간후 혈청 및 간장으로부터의 추출물을 분석한 결과 혈청이나 간의 경우 모두 사포닌 투여후 회수된 매탄올-물층의 방사능이 빠른 속도로 경감되었으나 방사능이 어느 수준 이하가 되면 방사능 소멸속도가 크게 저하되는 것으로 보아 일정수준의 인삼사포닌은 비교적 오래 혈청이나 간에 잠재하는 것으로 사료된다. 각 glycosides에 따라 흡수정도는 다르나 0.1%-1%정도 흡수된 것으로 확인되었고 간에서의 인삼 사포닌의 농도는 인삼 사포닌 투여후 상당기간 $10^{-6}$%-$10^{-5}$%수준을 유지할 것으로 예상된다.

  • The changes in aldehyde dehydrogenase(ALDH, E.C. 1.2.1.3.) activity in neurons and astrocytes isolated from rat brains were investigated after administration of ethanol and Korean red ginseng(Panax ginseng C.A. Meyer) saponln. The cerebral ALDH activity with acetaldehyde and Propionaldehyde was higher in the white matter than in the gray matter. However, using indole-3-a-cetaldehyde and 3,4-dihydroxyphenylacetaldehyde as substrates, there was no significant difference in activity between two regions in cerebrum. In ethanol treated group, ALDH activity with all the substrates in the gray and white matter was lower than in normal group. In ethanol-saponin treated group, the enzyme activity in the white matter remarkably Increased. The ALDH activity in neurons isolated from cerebral cortex in ethanol-treated group was lower than in normal group. In ethanol-saponin treated group, neuronal ALDH activity with propionaldehyde was significantly recovered but not with Indole-3-acetaldehyde. In astrocytes, although the ALDH activity with propionaldehyde in the ethanol-treated group was not changed as compared with normal group, considerable increase in activity was found in ethanol-saponin treated group. These results suggest that Korean red ginseng saponin may protect the neuronal functions from the toxic effects of acetaldehyde derived from ethanol by stimulation of ALDH activity in astrocytes surrounding nerve cells.

대두배아 사포닌의 유리기 생성 억제 및 세포독성 (Free Redical Scavenging and Cytotoxicity Activitives of Soybean Germ Saponin)

  • 대두 배아로부터 saponin을 분리.정제하여 유리기 억제효과 및 동물의 암세포에 대한 세포 독성을 조사하였다. 유리기 억제활성에 대한결과를 보면 대두 배아 saponin을 0.5 및 1.0%씩 첨가한 급여 식에서 4주 동안 사육한 후 간장을 절취하여 간장 획분의 활성산소 및 항산화 관련 효소에 대하여 평가하였다. 간장의 mitochondria 및 microtome 획분의 hydroxy radical(.OH)의 생성에 미치는 영향에서 급여식에 사포닌을 첨가한 급여군에서 .OH기의 생성억제 효과를 나타내었다. 간장의 microsome 획 분에서도 0.5 및 1.0% 투여군에서 $H_2O$$_2$생성억제 효과가 인정되었다. 또한 간장의 cytosol획분에서 $O_2$.$^{-}$의 생성은 대조군에 비하여 현저한 억제 효과를 나타내었다. 대두 배아 사포닌의 세포독성을 P338 (mouse Iyoupoid neoplasm)과 L1210 (mouse leukemia) 세포주를 사용하여 실시한 결과 200 $\mu$g/mL 농도에서 높은 성장억제효과를 나타내었다. 그리고 처리 농도에 따른 대두배아 추출물이 P338과 L1210에 대한 성장 효과 실험에서도 200 $\mu$g/mL에서 은 성장억제 효과가 나타났으며 농도가 높을 수록 완전히 억제되는 것을 볼 수 있었다.

The Effective Preparation of Protopanaxadiol Saponin Enriched Fraction from Ginseng using the Ultrafiltration

  • In this study, edible protopanaxadiol saponin enriched fraction were prepared by ultrafiltration (UF). Ginseng extract was prepared from mixtures of ginseng main root and rootlet (root: rootlet = 4 : 6). UF system was used the four-piston Diaphragm pump equipped with 5 kDa pore size Hydrosart Cassette made by regenerated cellulose acetate (CA) or 3 kDa pore size Hollow Fiber cartridge made by polyethersulfone (PES). Total ginsenoside contents of concentrated fraction by UF system was found to higher, compared to before those of untreated method. Especially, processing of UF showed the increase of PPD-type ginsenoside, while PPT-type ginsenoside was gradually decreased by both 3 kDa and 5 kDa membrane. After removal of 80% water by the 5 kDa Hydrosart Cassette and by 3 kDa Hollow Fiber cartridge, ginsenoside Rb1 content was higher 37.2 mg/g and 25.3 mg/g than 20.8 mg/g in untreated process. The ratio of Rb1 to Rg1 (Rb1/Rg1) and PPD- to PPT- type ginsenoside (PPD/PPT) were higher in inner fluid of ginseng extract after UF by 3 kDa cartridge (47.1 and 23.5, respectively) and 5 kDa Cassette (25.3 and 11.9, respectively) than those of before UF (5.7 and 3.7, respectively). PPD-type ginsenoside enriched fraction by UF system could be developed as a new ginseng material in food and cosmetic industrials.

Bifidus Fermentation Increases Hypolipidemic and Hypoglycemic Effects of Red Ginseng

  • Antihyperlipidemic and antihyperglycemic effects of Red Ginseng (RG, steamed and dried root of Panax ginseng C.A.Meyer, family Araliaceae), major component of which is ginsenoside Rg3, and Bifidodoterium-fermented RG (FRG), major component of which is ginsenoside Rh2, were investigated. Orally administered RG and FRG potently reduced the serum triglyceride levels in com-oil-induced hypertriglycemidemic mice as well as total cholesterol and triglyceride levels in Triton WR-1339-induced hyperlipidemic mice. Of the saponin and polysaccharide fractions of RG and FRG, the polysaccharide fraction inhibited postprandial blood glucose elevation of maltose- or starch-loaded mice and reduced the blood triglyceride levels in com-oil-induced hypertriglycemidemic mice. The saponin fraction and its ginsenosides Rg3 and Rh2 reduced blood triglyceride and total cholesterol levels in Triton WR1339-induced hyperlipidemic mice. The inhibitory effect of FRG and its main constituents against hyperlipidemia and hyperglycemia in mice were more potent than those of RG. These findings suggest that hypolipidemic and hypoglycemic effects of RG can be enforced by Bifidus fermentation and FRG may improve hyperlipidemia and hyperglycemia.
