Purpose: The purpose of this study was to investigate whether various saliva collection methods affect the observed salivary microbiome and whether microbiomes of stimulated and unstimulated saliva and plaque differ in richness and diversity. Methods: Seven sampling methods for unstimulated saliva, stimulated saliva, and plaque samples were applied to six orally and systemically healthy participants. Bacterial 16S ribosomal RNA genes of 10 major oral bacterial species, namely, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola, Fusobacterium nucleatum, Prevotella intermedia, Prevotella nigrescens, Streptococcus mitis, Streptococcus sobrinus, and Lactobacillus casei, were analyzed by real-time polymerase chain reaction. We comprehensively examined the dependence of the amount of bacterial ribosomal DNA (rDNA), bacterial-community composition, and relative abundance of each species on sample collection methods. Results: There were significant differences in the bacterial rDNA copy number depending on the collection method in three species: F. nucleatum, P. nigrescens, and S. mitis. The species with the highest richness was S. mitis, with the range from 89.31% to 100.00%, followed by F. nucleatum, P. nigrescens, T. denticola, T. forsythia, and P. intermedia, and the sum of the proportions of the remaining five species was less than 1%. The species with the lowest observed richness was P. gingivalis (<0.1%). The Shannon diversity index was the highest in unstimulated saliva collected with a funnel (4.449). The Shannon diversity index was higher in plaque samples (3.623) than in unstimulated (3.171) and stimulated (3.129) saliva and in mouthwash saliva samples (2.061). Conclusions: The oral microbial profile of saliva samples can be affected by sample collection methods, and saliva differs from plaque in the microbiome. An easy and rapid technique for saliva collection is desirable; however, observed microbial-community composition may more accurately reflect the actual microbiome when unstimulated saliva is assayed.

Authors surveyed the ground water near the waste disposed from a fiberglass production factory to confirm the presence of glassfiber in the water and to determine the effect of sampling conditions and storage on the recovery of fibrous materials in the ground water. Sample was collected at every 4 hours for 48 hours consecutively. After finishing the 48 hours sample, water sampling was done from each tap after repeated turning on and off the water for 30 seconds at each time. Sample was collected in the two 1.5 liter polyethylene bottle after vigorously shaking the bottle with the same water several times with the flowing tap water. At each paired sample, one bottle was stored stand still at room temperature, and the other sample was filtered immediately after sampling. Water was filtered on the Mixed Cellulose Ester filter with negative pressure. Each sample was divided into upper and lower layer. The other bottle was stored at room temperature standstill for 7 days and filtered in the same fashion as the other pair of sample did. Each MCE filter was divided into 4 pieces and one piece was treated with acetone to make it transparent. Each prepared sample was observed by two researchers under the light and polarizing microscopy, scanning electron microscopy and energy dispersive X-ra microanalysis. Fibers were classified by the morphology and polarizing pattern under the polarizing microscope, and count was done. 1. There was a significant fluctuation in number of the fibers, but there was no specific demonstrable pattern. 2. Non-polarizing fibers frequently disappeared after 7 days's storage. But cluster of fibers were found at the wall of the same container by scratching technique. 3. Polarizing fibers were usually found in between the filter and the manicure pasted area. Possible explanations for this phenomenon will be that either these fibers are very light or have electronic polarity. Hence, these fibers are not able to be attached on the surface of slide glass. 4. Under the scanning electron microscopic examination, the fibers which are not refractive under the light microscopy were identified as glassfiber. Other fibers which is refractive under the polarizing microscopy were identified as magnesium silicate fibers. It is strongly suggested that development of standardized method of sample collection and measurement of fibrous material in the water is needed.

Paint Spray is developed as a direct sampling ionisation method for mass spectrometric analysis of additives in polymer-based surface coatings. The technique simply involves applying an external high voltage (5 kV) to the wetted sample placed in front of the mass spectrometer inlet and represents a much simpler ionisation technique compared to those currently available. The capabilities of Paint Spray are demonstrated herein with the detection of four commercially available hindered amine light stabilisers; TINUVIN${(R)}$770, TINUVIN${(R)}$292, TINUVIN${(R)}$123 and TINUVIN${(R)}$152 directly from thermoset polyester-based coil coatings. Paint Spray requires no sample preparation or pre-treatment and combined with its simplicity requiring no specialised equipment makes it ideal for use by non-specialists. The application of Paint Spray for industrial use has significant potential as sample collection from a coil coating production line and Paint Spray ionisation could enable fast quality control screening at high sensitivity.
Three Holstein steers, fitted with ruminal and duodenal cannulas, were used in a replicated $3{\times}3$ Latin square design to determine the digestibility of dietary nitrogen in total digestive tract by three methods, 1) mobile nylon bag (MNB); 2) total fecal collection (TFC); and 3) washed fecal sample after freezing and thawing through a sieve with a pore size of $45{\mu}m$ (WFS). A basal diet of oaten hay-barley was supplemented with one of the following protein sources; soybean meal, fish meal or blood meal. Steers were fed at a level of 2% of body weight. The experimental diets were contained approximately 1.85% nitrogen. There were no differences (p>0.05) among the diets on DM, NDF and nitrogen disappearances, and the diet results were pooled to assess the methods. Total tract disappearances of dry matter and neutral detergent fiber were 61.6, 71.1 and 78.9 and 25.3, 63.2 and 64.6 for MNB, TFC and WFS methods, respectively. The lower digestibility of DM and NDF in the MNB method could be a result of low ruminal incubation time. The TFC method had the lower (p<0.05) determination of nitrogen disappearance in the total digestive tract than the MNB and WFS methods. On the other hand, nitrogen disappearance in the total digestive tract determined by the WFS technique was comparable to that in MNB technique, as there was no significant difference (p>0.05) between the methods. It is shown that the disappearance of dietary nitrogen in the total digestive tract could be estimated in the intact animals by using washed fecal sample prior to freezing and thawing.

The multivariate analysis techniques of cluster analysis are examined in this article. The theory and applications of the techniques and computer software concerning these techniques are discussed and sample jobs are included. A hierarchical cluster analysis algorithm, available in the IMSL software package, is applied to a set of data extracted from a group of subjects for the purpose of partitioning a collection of 26 attributes of a weapon system into six clusters of superattributes. A nonhierarchical clustering procedure were applied to a collection of data of tanks considering of twenty-four observations of ten attributes of tanks. The cluster analysis shows that the tanks cluster somewhat naturally by nationality. The principal componant analysis and the discriminant analysis show that tank weight is the single most important discriminator among nationality although they are not shown in this article because of the space restriction. This is a part of thesis for master's degree in operations research.
Genomics is providing targets faster than we can validate them and combinatorial chemistry is providing new chemical entities faster than we can screen them. Historically, the drug discovery cascade has been established as a sequential process initiated with a potency screening against a selected biological target. In this sequential process, pharmacokinetics was often regarded as a low-throughput activity. Typically, limited pharmacokinetics studies would be conducted prior to acceptance of a compound for safety evaluation and, as a result, compounds often failed to reach a clinical testing due to unfavorable pharmacokinetic characteristics. A new paradigm in drug discovery has emerged in which the entire sample collection is rapidly screened using robotized high-throughput assays at the outset of the program. Higher-throughput pharmacokinetics (HTPK) is being achieved through introduction of new techniques, including automation for sample preparation and new experimental approaches. A number of in vitro and in vivo methods are being developed for the HTPK. In vitro studies, in which many cell lines are used to screen absorption and metabolism, are generally faster than in vivo screening, and, in this sense, in vitro screening is often considered as a real HTPK. Despite the elegance of the in vitro models, however, in vivo screenings are always essential for the final confirmation. Among these in vivo methods, cassette dosing technique, is believed the methods that is applicable in the screening of pharmacokinetics of many compounds at a time. The widespread use of liquid chromatography (LC) interfaced to mass spectrometry (MS) or tandem mass spectrometry (MS/MS) allowed the feasibility of the cassette dosing technique. Another approach to increase the throughput of in vivo screening of pharmacokinetics is to reduce the number of sample analysis. Two common approaches are used for this purpose. First, samples from identical study designs but that contain different drug candidate can be pooled to produce single set of samples, thus, reducing sample to be analyzed. Second, for a single test compound, serial plasma samples can be pooled to produce a single composite sample for analysis. In this review, we validated the issue whether the second method can be applied to practical screening of in vivo pharmacokinetics using data from seven of our previous bioequivalence studies. For a given drug, equally spaced serial plasma samples were pooled to achieve a 'Pooled Concentration' for the drug. An area under the plasma drug concentration-time curve (AUC) was then calculated theoretically using the pooled concentration and the predicted AUC value was statistically compared with the traditionally calculated AUC value. The comparison revealed that the sample pooling method generated reasonably accurate AUC values when compared with those obtained by the traditional approach. It is especially noteworthy that the accuracy was obtained by the analysis of only one sample instead of analyses of a number of samples that necessitates a significant man-power and time. Thus, we propose the sample pooling method as an alternative to in vivo pharmacokinetic approach in the selection potential lead(s) from combinatorial libraries.
Image stacking technique is one of the key techniques for complex surface reconstruction. The process includes sample collection, image processing, algorithm editing, surface reconstruction, and finally reaching reliable conclusions. Since this experiment is based on laser scanning confocal microscope to collect the original contour information of the sample, it is necessary to briefly introduce the relevant principle and operation method of laser scanning confocal microscope. After that, the original image is collected and processed, and the data is expanded by interpolation method. Meanwhile, several methods of surface reconstruction are listed. After comparing the advantages and disadvantages of each method, one-dimensional interpolation and volume rendering are finally used to reconstruct the 3D model. The experimental results show that the final 3d surface modeling is more consistent with the appearance information of the original samples. At the same time, the algorithm is simple and easy to understand, strong operability, and can meet the requirements of surface reconstruction of different types of samples.

The importance of cooperative financial management is expected to be able to encourage the needs of the community especially towards the supervision of cooperatives and human resources. The study was conducted aiming to determine the effect of human resources, financial attitudes, and coordination on cooperative financial management. Research with quantitative methods. The study was conducted with multiple regression methods, while the sampling technique was based on a random sample of samples with the data collection method using a survey method in the form of a questionnaire that was measured with a Likert scale. The method of determining the research sample is done by calculating the Slovin formula, determining the research sample of 166 cooperatives in Buleleng Regency, Bali Province. The results of the research prove that partial human resources have a significant positive effect on cooperative financial management, financial attitude variables have a significant positive effect on cooperative financial management and coordination variables have a significant positive effect on cooperative financial management. The results of this study can be stated that the better the financial management of cooperatives, better will be the impact on cooperative growth so that the need for more competent resources in cooperative management.

This study aims to analyze the agreement of service innovation using contingency approach (manager personality, organizational structure) moderated by leadership agility. The study has been carried out on Islamic banking companies' managers in Indonesia, from East Java and Yogyakarta region using purposive sampling technique with questionnaire and interviews as the method of data collection. The total number of respondents in the sample is 184. This sample is then analyzed using Euclidience Distance Simple Regression and Simple Regression Moderation method. The results prove that: (1) there is a partial fit between incremental strategy with reactive personality and mechanical organizational structure, which increases the service performance; (2) there is a partial fit between radical strategy with proactive personality and organic organizational structure, which increases the service performance; (3) leadership agility is able to strengthen the fit of the relationship between incremental innovation strategy and reactive personality toward service performance; (4) leadership agility is able to strengthen the fit the relationship between radical innovation strategy and proactive personality toward service performance; (5) leadership agility is able to strengthen the fit of the relationship between incremental innovation strategy and mechanical organizational structure toward service performance; and (6) leadership agility is able to strengthen the fit of the relationship between radical innovation strategy and organic organizational structure toward service performance.

The library provides its services to satisfy the user's approach. Naturally, the development of library services may determine by considering the satisfaction of users. It traces users' perceptions regarding subject access highlighting problems in the retrieval of Bengali documents by subject. This study has designed to assess users' attitudes towards the retrievals of Bengali documents in OPAC through subject headings. For a collection of data, a representative sample has drawn from a large and heterogeneous population consisting of users in university libraries of West Bengal using a stratified sampling technique. Subsequently, under each of the universities, users' community was stratified into students, research scholars, and faculty members. Under each stratum, the sample selected on a random basis. The users met personally to collect relevant data, while they came to the library and went on to search OPAC. A structured schedule, prepared for the purpose, was presented before library users and consequently, interviews and interpretations recorded systematically. In this manner, several factors have identified concerning subject searching and retrieval performance for Bengali documents. This study explores the access using subject headings in multilingual information retrieval systems. Moreover, the suitability of subject headings for retrieval of Bengali resources has ascertained from the users' point of view. The findings demand standard principles and rules for the construction of Bengali subject headings to maintain uniformity and consistency.
