Analysis of UV Filters in Water using Stir Bar Sorptive Extraction (SBSE) and GC/MS-MS (교반막대 추출법과 GC/MS-MS를 이용한 수중의 자외선 차단제 분석)

  • A highly sensitive analytical method based on stir bar sorptive extraction (SBSE) technique and gas chromatography/tandem mass spectrometry (GC/MS-MS) has been developed, allowing the simultaneous multi-analyte determination of seven UV filters in water samples. The stir bar coated with polydimethylsiloxane (PDMS) was added to 40 mL of water sample at pH 3 and stirred at 1,100 rpm for 120 min. Other SBSE parameters (salt effect and presence of organic solvent) were optimised. The method shows good linearity (coefficients > 0.990) and reproducibility (RSD < 12.9%). The extraction efficiencies were above 84% for all the compounds. The limits of detections (LOD) and limits of quantification (LOQ) were 2.1~8.6 ng/L and 6.8~27.5 ng/L, respectively. The developed method offers the ability to detect 8 UV filters at ultra-low concentration levels with only 40 mL of sample volume. Matrix effects in tap water, river water, wastewater treatment plant (WWTP) final effluent water and seawater were investigated and it was shown that the method is suitable for the analysis of trace level of 7 UV filters except of benzophenone (BP). The method developed in the present study has the advantage of being rapid, simple, high-sensitive and both user and environmentally friendly.

  • The humidity sensor containing ammonium salt was prepared from the copolymer of 2-methacryloxyethyl dimethyl 2-hydroxyethyl ammonium bromide (MDHAB)/MMA/DAEMA = 6/3/1. The humid membrane was fabricated on the gold/alumina electrode by dipping. The impedances were $298k{\Omega},\;11k{\Omega}$, and $2.3k{\Omega}$ at 40%RH, 70%RH and 90%RH, respectively, at $5^{\circ}C$ and the humidity-sensitive characteristics were suitable for low temperature humidity sensor. The temperature-dependent coefficient between $5^{\circ}C$ and $20^{\circ}C$ was found to be $-0.80%RH/^{\circ}C$ and the hysteresis falled in the ${\pm}2%RH$ range. The response time was found to be 38 sec for the relative humidity ranging from 34%RH to 88%RH at $20^{\circ}C$. The reliabilities such as temperature cycle, humidity cycle, high temperature and humidity resistance, electrical load stability, stability of long-term storage and water durability were measured and evaluated for the application as a humidity sensor.

  • Salt stress is one of major environmental factors influencing plant growth and development. To identify salt tolerance determinants in higher plants, a large-scale screen was conducted with a bialaphos marker-based T-DNA insertional collection of Arabidopsis ecotype C24 mutants. One line for salt stress-sensitive mutant (referred to as ssm1) exhibited increased sensitivity to both ionic (NaCl) and nonionic (mannitol) osmotic stress in a root growth assay. This result suggests that ssm1 mutant is involved in ion homeostasis and osmotic compensation in plant. Molecular cloning of the genomic DNA flanking T-DNA insert of ssm1 mutant was achieved by mutant genomic DNA library screening. T-DNA insertion appeared in the first exon of an open reading frame on F3M18.7, which is the same as AtSYP61. SSM1 is SYP61/OSM1 that is a member of the SNARE superfamily of proteins required for vesicular/target membrane fusions and factor related to abiotic stress.

Adaptations and Physiological Characteristics of Three Chenopodiaceae Species under Saline Environments (명아주과 3종 식물의 염 환경에 대한 적응특성의 비교)

  • Three species of Chenopodiaceae, i.e. Suaeda japonica, Salicomia herbacea, Beta vulgaris var. cicla, were investigated to compare the physiological characteristics through inoic balances and osmoregulations under different environmental salt gradients. Plats were harvested in two weeks from treatments with salt gradients (0, 50, 100, 200 and 400 mM NaCl) and mineral nutrition gradients(1/1, 1/5, 1/10 dilutions of Hoagland solution). Plants were analyzed for growth responses, ionic balances, osmolalities, conductivities, glycinebetaine and proline contents quantitatively. Three plants of Chenopodiaceae accumulated slats into tissues unlike some salt sensitive species, and showed unique adaptation patterns to overcome saline environments, i.e. strong growth stimulation for Salicomia herbacea, growth negative tolerance for Suaeda japonica, and growth positive tolerance for Beta vulgaris var. cicla. The absorption of inorganic Ca/sup 2+/ ions was inhibited remarkably due to the excess uptake of Na+ with increasing salinity. The K+ content in plants was significantly reduced with increasing salinity. Total nitrogen content was reduced as mineral nutritions and salinity increased. Conductivity and osmolality increased with increasing salinity regardless of mineral nutritions. The ranges of glycinebetaine and proline contents were 0.2∼2.5 μM/g plant water and 0.1∼0.6μM/g plant water, respectively.

Effect of Calcium Chloride Concentration on Roadside Ground Cover Plant Growth (염화칼슘 처리농도가 가로변 지피식물의 생장에 미치는 영향)

  • The purpose of this study was to provide information on management and apply it to a roadside ground cover plant understanding the capacity of calcium chloride in the plant. The experimental group was composed of the ratio control group of calcium chloride, 0.5%, 1.0%, and 3.0% in 500g of soil. Plant materials were selected and measured according to their ecological characteristics such as ground cover plant, Pachysandra terminalis, Hosta plantaginea, Trachelospermum asiaticum, Vitex rotundifolia, Euonymus japonica and Callicarpa japonica. The acidity of the amended soil was increased gradually depending on the treatment and conductivity was continually decreased. The EX-Ca increased after the treatment, but decreased in the middle of the experiment. Pachysandra terminalis, Trachelospermum asiaticum and Euonymus japonica were able to grow and survive at the ratio of 0.5%. Hosta plantaginea and Vitex rotundifolia were able to survive at the ratio of 1.0%. Hosta plantaginea, the possible state can absorb salts due to moisture and, can be applied to ground cover plants in the roadside. The growth and development of Callicarpa japonica was poor and the leaves were open to grow for calcium chloride treatment except the control group. It was concluded that Callicarpa japonica was very sensitive to calcium chloride.

Electromechanical impedance-based long-term SHM for jacket-type tidal current power plant structure

  • Jacket-type offshore structures are always exposed to severe environmental conditions such as salt, high speed of current, wave, and wind compared with other onshore structures. In spite of the importance of maintaining the structural integrity for an offshore structure, there are few cases to apply a structural health monitoring (SHM) system in practice. The impedance-based SHM is a kind of local SHM techniques and to date, numerous techniques and algorithms have been proposed for local SHM of real-scale structures. However, it still requires a significant challenge for practical applications to compensate unknown environmental effects and to extract only damage features from impedance signals. In this study, the impedance-based SHM was carried out on a 1/20-scaled model of an Uldolmok current power plant structure in Korea under changes in temperature and transverse loadings. Principal component analysis (PCA)-based approach was applied with a conventional damage index to eliminate environmental changes by removing principal components sensitive to them. Experimental results showed that the proposed approach is an effective tool for long-term SHM under significant environmental changes.

Studies on the RK-temperate phage of bacillus cereus (Bacollis cereis의 RK-용원파아지에 관한 연구)

  • The RK-temperate phage which infected with Bacillus cereus was isolated and the characters were investigated. The induction of RK-temperate phage from host bacterium attained by ultraviolet light irradiation (15W, 30cm, 30-120sec) and mitomycin C treatment (0.2-2 ug/ml). The host range of RK-temperate phage was not revealed with lysogenic and related strains of B. cereus. But B. cereus(PS) 352 which obtained by N-nitrosoguanidine treatment (1,000{$\mu}g/ml)$ to phage infected with host bacteria was sensitive bacteria of RK-temperate phage. RK-temperate phage was stabilized at the condition of nutrient broth (pH 7-8), Tris-buffer (pH 7-8) and ammonium buffer (pH 8-9) and Sorensen's phosphate buffer (pH 6-7), but unstabilized at other salt solutions and pH range. Also, thermostability was to $45^{\circ}C$ but unstabilized at above $50^{\circ}C$. At RK-temperate phage, the measurment values of head, neck, mid tail and end tail were 59nm, $9{\times}16nm,\;10{\times}189nm,\;and\;10{\times}14nm$ respectively. The morphology of head was regular polyhedron, and the end tail was coneate form. On the one hand, the number of capsid protein layer of tail were consist of 4, 35, and 1 at neck, mid tail, and end tail, respectively. RK-temperate phage was identified with DNA phage and G+C contents were 38.63. The latent time of RK-temperate phage was 30 minutes and the burst size was 70-80. And the host bacteria was lysed in case of multi-infection, above moi 1.

Purification and Characterization of Extracellular Poly(3-hydroxybutyrate) Depolymerase from Penicillium simplicissimum LAR13

  • An extracellular PHB depolymerase was purified from P. simplicissimum LAR13 cultural medium by Sepharose CL-6B chromatography. When the fungus was grown in a basal salt medium with poly(3-hydroxybutyrate) (PHB) as the sole carbon source, PHB depolymerase production reached maximum at its stationary phase. The mycelial growth rate was higher at 37$^{\circ}C$ than at 30$^{\circ}C$ and even higher than at 25$^{\circ}C$, However, the enzyme production was lower at 37$^{\circ}C$ than 30$^{\circ}C$ or 25$^{\circ}C$. The isolated enzyme is composed of a single polypeptide chain with a molecular mass of about 36 kDa as determined by SDS-PAGE. The optimum conditions for the enzyme activity are pH 5.0 and 45$^{\circ}C$. The enzyme was stable for 30 min at a temperature lower than 50$^{\circ}C$, and stable at pH higher than 2.0 but it was unstable at pH 1.0.1 mM Fe$\^$2+/ reduced the enzyme activity by 56% and the enzyme was inhibited almost completely by 4 mM Fe$\^$2+/ . The enzyme was partially inhibited by phenylmethylsulfonyl fluoride and was very sensitive to diazo-DL-norleucine methyl esters dithiothreitol and mercuric ion. However, N-p - tosyl - L - Iysinechloromethyl ketone, p -hydroxymercuricbenzoate and N- acetylimidazole had no influence upon its activity.

Preparation and Characteristics of Phosphatidylcholine Liposomes Covalently Coupled with Immunoglobulin Fragments (이뮤노글로부린 토막과 결합된 포스파티딜콜린 리포솜의 제조와 특성)

  • The characteristics and stabilities of phosphatidylcholine liposomes covalently coupled with immunoglobulin fragments prepared by the REV method were investigated by the dynamic light scattering, absorbance and calcein release. Using a sulfhydryl-reactive phospholipid derivative of N-[4$({\rho}-maleimido-phenyl)$ butyl] phosphatidylethanolamine (MPB-PE), Fab' antibody fragments were covalently combined with preformed large unilamellar vesicles (LUV), Coupling ratio was $250\;{\mu}g$ of $Fab'/{\mu}mol$ of phospholipid in vesicles, From dynamic light scattering, it was found that the size of the vesicles increases as the ratio of cholesterol to lipid increases, but that apparently, the size of liposomes was not sensitive to the existence of Fab' fragments. Regardless of inserting Fab' fragments, the absorbance of liposomes decreased as the amounts of bile salt (BS) added. At very low BS concentrations, BS/lipid aggregates would be formed in the outer vesicles monolayer, while, at the high BS concentrations, mixed micelles would be preferred. The vesicles incorporated with Fab' fragments, however, are more resistant to the bile salts than the MPB-PE vesicle are. The absorbance of vacant liposomes and calcein release resulted in that the Fab' vesicles and MPB-PE vesicles by the REV method are very stable, but that those by the sonication method sufferred the significant change of turbidities.

Accelerated life testing of concrete based on stochastic approach and assessment

  • This study aimed to design the accelerated life testing (ALT) of concrete, which stimulating the special natural environment maximumly. Its evaluation indexes, such as dynamic elastic modulus, mass and ultrasonic velocity were measured, and the variation of relative mass and relative dynamic elastic modulus of concrete were studied. Meanwhile, the microanalysis method was used. Moreover, an exploratory application of the stochastic approach, the Weibull distribution and the lognormal distribution, were made to assess the durability of concrete structures. The results show that the ALT for simulating natural environment is more close to the service process of concrete structure under actual conditions; The relative dynamic elastic modulus can be used as the dominant durability evaluation parameters, because it is more sensitive to the environmental factors compared with the relative quality evaluation parameters; In the course of the concrete deterioration, the destruction of the salt freezing cycle is the dominant factor, supplemented by other factors; Both of those two stochastic approaches can be used to evaluate the reliability of concrete specimens under the condition of ALT; By comparison, The lognormal distribution method is better to describe the reliability process.
