인공숙주에서 증식된 Tetrastichus sp.의 생물학적 특성 (Biological Characteristics of Tetrastichus sp. reared on Artificial Host)

  • Tetrastichus sp.는 미국흰불나방의 번데기 기생벌로서, 1개의 숙주에 여러마리의 자손을 번식하는 집단 기생벌이며, 일년에 여러 세대 발생한다. 작잠, 신생아용 분유, 난황, 로얄젤리 등을 주요성분으로 기생벌의 인공먹이를 조성하였으며, polypropylene필름으로 번데기형태를 만들어 인공숙주로 사용하였다. 인공숙주에서 발육한 기생벌은 자연숙주에서 자란 기생벌보다는 크기가 작았으나, 계대 사육으로 번식 가능한 자손을 생산하였다. 인공숙주에서 발육한 in vitro 2세대의 생물학적 특성은 in vitro 1세대보다 훨씬 개선 었다. 평균 자손 수, 산란기간, 수명 등은 in vitro 에서 45.7,7.8, 13.8일 이었고 암컷 편중의 성비 76.9%를 나타내었다. 이러한 결과는 Tetrastichus sp.가 in vitro 대량사육에 적합한 기생벌임을 보여준다.

효모 자가분해물의 새로운 제조방법 개발 및 품질검사 (Development of a New Processing Method and Quality Evaluation of Yeast Autolyzate)

  • 이 연구는 재래방법과 비교하여 수율이 높고 품질이 향상된 효모 자가분해들의 제조방법을 연구하는데 그 목적을 두었다. 제빵용 생 효모에 glucanase와 protease의 복합효소를 작용시킨 다음, 10,000 psi의 고압으로 균질하면 약 85.3%의 효모 세포벽이 파손되었다. 세포벽이 파손된 효모를 사용하여 자가분해 시키는데 가장 적당한 조건을 반응 표변방법(RSM)을 이용하여 조사한 결과는 pH 7.0, 온도 $30^{\circ}C$ 에탄올 4%, 소금 3%이었다. 세포벽이 파손된 효모를 위와 같은 최적조건에서 3일간 분해시켜 얻은 효모 자가분해물의 수율은 2.6배 향상되었으며, 총 아미노산의 함량도 현저히 증가(2.8배)하였다. 그리고 관능적인 품질도 재래방법으로 제조된 시료보다 고기 향미가 많고, 이미-이취가 현저하게 적어서 전체인 기호도에서도 상당히 우수한 것으로 평가되었다.

  • Potato (Solanum tubersosum L.) is susceptible to various environmental stresses such as frost, high temperature, and drought. Enhancement of potato drought tolerance can reduce yield loss under drought that has negative effect on potato tuber growth. Genetic engineering can be utilized to achieve this goal, but such approaches using endogenous potato genes have rarely been applied. Since unpredictable global weather changes cause more severe and frequent water limiting conditions, improvement of potato drought tolerance can minimize such adverse effects under drought and can impact on sustainable potato production. Genetic engineering can be utilized to improve potato drought tolerance, but such approaches using endogenous potato genes have rarely been applied. We were obtained AtbZIP28 gene transgenic potato plants. It is identified transcript levels at various stress conditions, polyethylene glycol (PEG), NaCl, abscisic ${\underline{acid}}$ (ABA). Also, For identification to regulate ER stress response genes in AtbZIP28 gene transgenic potato plant, we screened seven potato genes from RNA-seq analysis under TM treatment. Five and two genes were up- and down-regulated by TM, respectively. Their expression patterns were re-examined at stress agents known to elicit TM, DTT, DMSO and salt stress.

Analysis of metabolites in wheat roots in response to salinity stress

  • Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.

Improved growth and development in Suaeda glauca through exogenous treatment with indole-3-carboxylic acid

  • Mechanistic studies of halophytes are urgent areas of agricultural research due to the increase in saline-contaminated and irrigated land worldwide. The halophyte Suaeda glauca (S. glauca) has advantages in terms of biomass and saline elimination due to its large mass and well-developed phenotype on seashores, although its mechanistic features and growing specificities still require systematic investigation. In this study, S. glauca was cultivated under various saline concentrations (0-400 mM) in Hoagland's solution in the absence or presence of indole derivatives to elucidate physiological features. The results confirmed the optimal growth and development of S. glauca in 50 mM NaCl, and morphologies such as the number of branches, shoot length, and fresh and dry weights were improved by indole-3-carboxylic acid (ICA) treatment. The cation concentrations in roots, shoots and leaves were investigated to examine the ionic imbalances in response to saline treatment, and the results demonstrated that sodium ions accumulated to high concentrations in leaves. The levels of calcium and potassium ions in roots were maintained or slightly decreased in the presence of 50 mM NaCl and proline concentration was increased significantly in roots at optimal concentrations. These results demonstrate that the concentrations of ions and metabolites are key regulators of optimal growth by regulating the physiology of halophytes.

MVR해수담수화플랜트의 고농도 농축수를 활용한 미네랄 제어 및 제염기술 연구 (A Study on the mineral control and salt manufacture technology using higher concentration in the MVR desalination plant)

  • 일반적으로 해수담수화플랜트는 크게 두 가지의 문제점을 가지고 있는데, 에너지 비효율성과 해수담수화 후 부산물인 농축수의 해결방안이다. 전자는 태양에너지 등 신 재생에너지원의 활용과 새로운 기술에 대한 꾸준한 연구개발로 상당히 그 효율성이 개선되었으나, 농축수 해결방안에 대한 연구는 아직 미미한 실정이다. 본 논문에서는 많은 에너지를 투입하여 담수와 함께 생산된 농축수의 활용과 관련하여 제염이 가능한 설비의 구축과 그 효용성을 제고하기 위한 미네랄(Mg) 성분 제어에 관하여 제주 MVR해수담수화 플랜트를 중심으로 그 연구 결과를 소개하고자 한다. 플랜트의 정상 작동으로 생산된 농축수에 본 연구의 제염 설비를 적용하여 단계별 소금의 성분 함량을 분석했을 때 Mg 성분 수치가 줄어드는 것으로 보아 미네랄(Mg) 제어가 정상적으로 진행되고 있음을 알 수 있었다. 아울러, 본 MVR해수담수화 플랜트의 에너지 이용의 효율성과 담수 생산 및 농축수 활용에 관하여 그 활용성을 확인할 수 있었다.

Carcass Characteristics and Chemical Composition of the Longissimus Muscle of Nellore, Caracu and Holstein-friesian Bulls Finished in a Feedlot

  • This work was carried out to study the carcass characteristics, the chemical composition and the fatty acid profile in Longissimus muscle (LM) of bull breeds Nellore, NEL (11), Caracu, CAR (12) and Holstein-Friesian, HFR (12) finished in a feedlot. The bulls were fed twice a day with corn silage, cotton meal, cracked corn, urea, limestone and mineral salt. NEL and CAR bulls had similar (p>0.05) final weight and hot carcass weight. However, NEL and CAR bulls had higher (p<0.05) final weight and hot carcass weight than HFR bulls. Carcass hot dressing, carcass conformation, cushion thickness, Longissimus muscle area and texture were similar (p>0.05) among NEL, CAR and HFR bulls. NEL and HFR bulls had higher (p<0.05) carcass length in comparison to the CAR breed. Nellore breed had higher (p<0.05) leg length in comparison to CAR and HFR breeds. Leg length was similar (p>0.05) between CAR and HFR breeds. Thickness fat, color and marbling score were lower (p<0.05) in NEL breed in comparison to CAR and HFR breeds. LM of NEL bulls had higher (p<0.05) meat moisture content in comparison to CAR and HFR bulls. In contrast, lipid content was lower (p<0.10) in HFR bulls. LM ash and crude protein contents were similar (p>0.05) among breeds. Saturated fatty acids (SFA) were higher (p<0.10) in HFR animals. Monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), n-6, n-3 and PUFA/SFA ratio were similar (p>0.05) among the different breeds. N-6/n-3 ratio was higher (p<0.05) in CAR animals.

Growth in the Sultanate of Oman of Small Ruminants Given Date Byproducts-Urea Multinutrient Blocks

  • This study investigated the use of multinutrient blocks (MNB) composed of 35% date syrup, 35% date syrup byproducts, 10% date fronds, 7% urea, 7% cement and 6% common salt for growing small ruminants, as partial substitute for the roughage component of the diet (Rhodes grass hay) and its effect on feedlot performance and economics of feeding. Eight growing local Omani goats and 8 sheep (each about one year old) were used in this study. Mean body weights for goats and sheep (kg), respectively were $21.1{\pm}4.5$ and $25.5{\pm}4.1$. The goats and sheep were subdivided into two sub-groups of more or less equal body weights for each species. Each sub-group in both species was either fed on 0.5 kg concentrate+ad libitum Rhodes grass hay or the same diet+restricted hay (about 0.2 kg/head/day) and ad libitum amount of MNB. Sheep significantly (p<0.05) consumed greater amounts of MNB ($36{\pm}17$g/head/day) than goats ($6{\pm}2.5$ g/head/day). Feeding of the MNB was effective in sparing about 40% of the roughage Rhodes grass hay for goats (from 240 to 140 g) and about 42% for sheep (from 252 to 146 g) or approximately 100 g/head/day for both goats and sheep. This reduction (or sparing) in the consumption of Rhodes grass hay was coupled by an improvement in daily liveweight gain (g/head/day) in both goats (from 29 to 46 by 58.6%) and sheep (from 26 to 39 by 50%) and also by an improvement in the feed conversion efficiency (g feed/g gain) of both goats (from 25 to 13.8 by 45%) and sheep (from 28.7 to 17.2 by 40%). Cost of daily consumed feeds as well as cost/kg gain (or cost of meat) were both reduced due to feeding of MNB. They were both respectively reduced by 7.5% (from 53 Baisa/day to 49) and 38% (from 1,828 Baisa/kg to 1,140). It was economically viable to feed MNBs containing date by-products and urea to small ruminants in the Sultanate of Oman.

  • This work was conducted to study the performance, carcass characteristics, chemical composition and fatty acid profile of the Longissimus muscle (LM) of first-generation Purunã (n = 7, PUR1), second-generation Purunã (n = 9, PUR2), and 1/2 Puruna vs. 1/2 Canchin (n = 13, PUCA) bulls finished in a feedlot. The animals averaged 18 months of age at the beginning of the experiment. The animals were kept in a feedlot from 18 to 22 months of age. They were fed twice a day with corn silage ad libitum, along with soybean meal, cracked corn, urea, limestone and mineral salt. PUR1 bulls had greater (p<0.05) final weight (496.0 kg) than PUCA bulls (449.2 kg). However, there was no difference (p>0.05) between PUR1 and PUR2 (472.0 kg), and between PUR2 and PUCA. Hot carcass weight was similar (p>0.05) among the different genetic groups. Hot carcass dressing percentage was similar (p>0.10) between PUR1 (50.4%) and PUCA (53.8%), and higher (p<0.10) than PUR2 (48.6%). Cushion thickness was greater (p<0.05) in PUCA bulls (27.12 cm). The other parameters were similar (p<0.05) among the three genetic groups. Total lipid and cholesterol contents were higher (p<0.01) in PUR2. Moisture, ash and crude protein contents were similar (p>0.05) among the three genetic groups. Saturated fatty acid (SFA) levels were higher (p<0.10) in PUR1 and PUR2 (51.58% and 50.41%, respectively). Polyunsaturated fatty acid levels were higher (p<0.05) in PUR1 and PUCA (6.50% and 8.29%, respectively). N-6, n-3 and PUFA/SFA ratios were higher (p<0.05) in PUCA. MUFA and n-6/n-3 ratios were similar (p>0.05) among the different genetic groups.

Transgenic Expression of MsHsp23 Confers Enhanced Tolerance to Abiotic Stresses in Tall Fescue

  • Tall fescue (Festuca arundinacea Schreb.) is an important cool season forage plant that is not well suited to extreme heat, salts, or heavy metals. To develop transgenic tall fescue plants with enhanced tolerance to abiotic stress, we introduced an alfalfa Hsp23 gene expression vector construct through Agrobacterium-mediated transformation. Integration and expression of the transgene were confirmed by polymerase chain reaction, northern blot, and western blot analyses. Under normal growth conditions, there was no significant difference in the growth of the transgenic plants and the non-transgenic controls. However, when exposed to various stresses such as salt or arsenic, transgenic plants showed a significantly lower accumulation of hydrogen peroxide and thiobarbituric acid reactive substances than control plants. The reduced accumulation of thiobarbituric acid reactive substances indicates that the transgenic plants possessed a more efficient reactive oxygen species-scavenging system. We speculate that the high levels of MsHsp23 proteins in the transgenic plants protect leaves from oxidative damage through chaperon and antioxidant activities. These results suggest that MsHsp23 confers abiotic stress tolerance in transgenic tall fescue and may be useful in developing stress tolerance in other crops.
