In the study, the effect of acid and salt concentrations during the production of bio-ethanol from various brwon-algae raw materials was investigated. Especially, the possibility of the conversion of various polysaccarides contained in Laminaria japonica was studied. Bio-ethanol was produced by Saccharomyces cerevisiae KCCM1129 strains in Laminaria japonica. The maximum bio-ethanol production of 2.09 g/L using heat-treatment of Laminaria japonica was achieved. The optimum concentration for reducing sugar conversion by Laminaria japonica was found to be 3.95 g/L at the HCl concentration of 0.1 N. But bio-ethanol production was higher than the case without the non-acid pretreatment. Among the various polysaccharides, only mannitol produced maximum 3.09 g/L bio-ethanol. In case of laminaran, the ethanol was produced only at 0.15 g/L only in 0.1 N HCl pretreatment medium and cell growth was higher than other pretreatment.
This study was done to investigate the quality properties of beef jerky with soy sauce, red pepper paste, and soybean paste replacing salt. Sliced beef samples were cured in salt (control), soy sauce, red pepper paste, and soybean paste for 24 or 48 h and then dried at $70^{\circ}C$ for 8 h. Treatments showed higher final moisture content and lower $Na^+$ concentration than the control after drying for 8 h. The lightness and shear force values were lower in all treatment samples than in the control during 48 h of curing time. In particular, lower lipid oxidation was found in the jerky cured with red pepper paste than in the control. Sensory evaluation showed that color, flavor, and tenderness of jerky samples were improved by replacing salt with soy sauce, red pepper paste and soybean paste, and higher likeability scores of the beef jerky were obtained among those treatments after 48 h of curing time.
[ ${\Delta}^1$ ]pyrroline-5-carboxylate synthetase (P5CS) is a proline biosynthetic pathway enzyme and is known for conferring enhanced salt and drought stress in transgenics carrying this gene in a variety of plant species; however, the wild-type P5CS is subjected to feedback control. Therefore, in the present study, we used a mutagenized version of this osmoregulatory gene-P5CSF129A, which is not subjected to feedback control, for producing transgenic indica rice plants of cultivar Karjat-3 via Agrobacterium tumefaciens. We have used two types of explants for this purpose, namely mature embryo-derived callus and shoot apices. Various parameters for transformation were optimized including antibiotic concentration for selection, duration of cocultivation, addition of phenolic compound, and bacterial culture density. The resultant primary transgenic plants showed more enhanced proline accumulation than their non-transformed counterparts. This proline level was particularly enhanced in the transgenic plants of next generation ($T_1$) under 150 mM NaCl stress. The higher proline level shown by transgenic plants was associated with better biomass production and growth performance under salt stress and lower extent of lipid peroxidation, indicating that overproduction of proline may have a role in counteracting the negative effect of salt stress and higher maintenance of cellular integrity and basic physiological processes under stress.
This study investigated the sensory characteristics and antioxidative activity of the reduced salt dried bulgogi bibimbap containing AF-343, a dandelion-derived compound extract. A sample with a 30% reduced salt had a sodium amount of 659.01 mg, which was significantly lower than that of the sample before the reduction (p<0.05). The textural hardness of the SW (Standard sample with the AF-343), showed the lowest value of $1.03N/cm^2$, indicating that the AF-343-containing sample could have a slightly softer texture than the non-containing sample. In the analytical sensory test, the AF-343-containing samples showed a synergistic effect on the aroma and flavor of bulgogi, with values of 5.7 and 8.4, respectively, which were significantly higher than that of the non-containing sample group (4.3 and 4.4, respectively). The hardness by the sensory test was slightly hard with values of 7.2-8.1 in all samples, indicating that textural improvement was needed. In the acceptance test, all the samples scored with slightly lower values of 4-5 points, highlighting the need for future studies to improve the sensory characteristics of dried bibimbap. The flavonoid contents of AF-343-containing SW and RW sample groups were 68.21 and 64.31 mg GAE/100 g, respectively, which were significantly higher than those of the samples without AF-343 (49.06 and 44.82 mg GAE/100 g, respectively) (p<0.05). The ABTS and DPPH radical scavenging activities were similar to those of the flavonoid contents. As a result, in the production of AF-343-containing reduced-salt dried bulgogi bibimbap, more study on the textural improvement will be needed to achieve better palatability.
This study investigated the minimum salt concentration required for achieving the optimal quality characteristics of sauerkrauts made by adding 0.5, 1.0, 1.5, 2.0, and 2.5% (w/w) of sea salt to cabbage according to the fermentation period. For evaluating the quality characteristics, we measured the microorganisms (lactic acid bacteria, yeast, and coliform group), pH, total acidity, salinity, chromaticity, and hardness every 24 hours. The lactic acid bacteria were identified and analyzed, and acceptance test was carried out on the 4th day of fermentation. The results showed that the salinity of 0.5, 1.0, 1.5, and 2.0% sauerkrauts on the 4th day of fermentation was lower than the average salinity of Baechu-Kimchi. The 0.5, 1.0, 1.5, and 2.0% sauerkrauts had significantly higher lactic acid bacteria than the 2.5% sample, and the coliform group was not detected after the 5th day of fermentation. Among the microbes identified, Weissella cibaria JCM 12495 was found only in domestic sauerkraut, in addition to Lactococcus lactis NCDO 604, Leuconostoc citreum JCM 9698, and Lactobacillus sakei DSM 20017. The results of the acceptance test show that 1.0 and 1.5% sea salt sauerkraut had significantly higher overall acceptance compared to the other samples. In conclusion, sauerkraut with a salt concentration of 1.0 and 1.5% (w/w) had abundant lactic acid bacteria and excellent sensory properties, suggesting that the production of low-salted sauerkraut can be adopted to reduce consumer salt intake in the future.
Energy flow through the trophic levels was studied at a salt marsh ecosystem distinguished into low and high marsh. Gross primary productions of Suaedeto-Salicornietum and Artemisieto-Limonietum at low marsh were 8, 299 and 13, 154kca/$m^2$/yr, and those of Calama-grostetum and Sonchuso-Setaetum at high marsh were 17, 899 and 15, 177kca/$m^2$/yr, respectively. Efficiencies of solar energy utilization of plants were 1.7 and 2.6% at the former, and were 3.6 and 3.2% at the latter. Of gross productions, net primary productions were 3, 977 and 5, 280kca/m2/yr at low marsh and were 6, 354 and 5, 329kca/$m^2$/yr at high marsh, and the remainder, 52~67%, was consumed by respiration of plants. A small amount (0.03~0.04%) of the net primary production was flowed through grazing food chain and most amout was transferred into dead parts. Of dead parts, 40% was accuulated as litter and the rest was decomposed into detritus. In the detritus food chain, a little energy was utilized by detritus feeder, and a major by microorganism. The amounts of energy flowed through grazing and detritus feeders at high marsh were much more than those at low marsh, but tertary production as spider was Vice versa.
The composition rates of litters were studied at Camellia japonica forests in Koje-Do and Haenam, and at Quercus acuta, Quercus acutissima, Cryptomeria japonica, and Chamaecyparis abtusa forests in Haenam. Total amounts of nitrogen, phosphorus, potassium, calcium, and sodium in litter samples were measured and the relation between these amounts and decomposition rate was studied. Annual litter production were rarely different at each forest in Haenam. The amount of accumulated organic matter was about 4 times greater at C. japonica forest in Koje-Do than at the C. japonica forest in Haenam. These amounts were 5, 282.1 $\pm$ 1, 026.03g/m2 in Koje-Do and 1, 420.7 $\pm$ 384.77g/$m^2$ in haenam. The decomposition rate were rarely differnet at each forest in Haenam, but the rates showed great difference at C. japonica foreests in Koje-Do and Haenam. The rates were 0.093 and 0.313 at C. japonica forests in Koje-Do and in Haenam respectively. The sodium contents were 0.472% and 0.229% on L layer and on Css layer of C. japonica forest in Koje-Do, while they were 0.034% and 0.043% on L layer, and on Css layer of C. jpaonica forest in Haenam. It is sugested that much difference in the salt contents in the forest floor was present from the results of sodium content measured at each site, and that the decomposition rate was affected by the much concentration of salt in Koje-Do.
Proline accumulation has been shown to correlate with tolerance to drought and salt stresses in plants. We attempt to introduce the wild-type, mutant, and fusion proBA genes derived from Bacillus subtilis into Arabidopsis thaliana under the control of a strong promoter cauliflower mosaic virus 35S (CaMV35S). The transgenic plants produced higher level of free proline than control and the overproduction of proline resulted in the increased tolerance to osmotic stress in transgenic plants. Besides, the mutation in proBA genes, which were proved to lead $\alpha$-glutamyl kinase ($\alpha$-GK) reduces sensitivity to the end-product inhibition and the fusion of proB and proA also result in increasing proline production and confer osmotolerance in transgenic lines.

In many conditions, bacterial surface properties are changed as a result of variation in the growth medium and conditions. This study examined the influence of bile salt concentrations (0-0.1%) on colony morphotype, hydrophobicity, $H_2O_2$ concentration, S-layer protein production, and slpA gene expression in Lactobacillus acidophilus ATCC 4356. It was observed that two types of colonies (R and S) were in the control group and the stress condition. When the bile level increased in the medium, the amount of S type was more than the R type. A stepwise increment in the bile concentration resulted in a stepwise decline in the maximum growth rate. The results showed that hydrophobicity was increased in 0.01%-0.02% bile, but it was decreased in 0.1% bile. Treatment by bile (0.01%-0.1%) profoundly decreased $H_2O_2$ formation. S-Layer protein and slpA gene expression were also altered by the stress condition. S-Protein expression was increased in the stress condition. The slpA gene expression increased in 0.01%-0.05% bile and it decreased in 0.1% bile. However, we found that different bile salt concentrations influenced the morphology and some surface properties of L. acidophilus ATCC 4356. These changes were very different in the 0.1% bile. It appears that the bacteria respond abruptly to 0.1% bile.
Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica was studied. Maltose, yeast extract and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in flask culture were pH 5.0, $25^{\circ}C$ and 150 rpm in a meidum containing of 30 g maltose, 6 g yeast extract, 2 g polypeptone, 0.5 g $K_2HPO_4$, 0.2 g $KH_2PO_4$, 0.2 g $MnSo_4\;{\cdot}\;5H_2O$, 0.2 g $MgSO_4\;{\cdot}\;7H_2O$ in 1-L distilled water. Exo-biopolymer production and mycelial growth in the suggested medium were significantly increased in a 2.5-L jar fermentor, where the maximum biopolymer concentration was 8 g/1.
