Serum Stress Responses during Seawater Acclimation in Coho Salmon, Oncorhynchus kisutch (은연어의 해수 순치에 따른 스트레스 반응의 변화)

  • Stress response to the seawater acclimation in coho salmon (Oncorhynchus kisutch) smolt was investigated. Salt concentration of rearing water was gradually increased for 3 days from freshwater to seawater (30 ppt salt level). The changes of serum concentrations of cortisol as a primary stress indicator, and as secondary indicators, glucose (GLC), lactate (LAC), triglyce.ide (TG), cholesterol (CHOL), sodium ion $(Na^+)$, chloride ion $(Cl^-)$ and enzyme activities (alanine aminotransferase, ALT: aspartate aminotrasferase, AST; lactate dehydrogenase, LDH) were quantified during the acclimation experiment. Among them, cortisol, LAC, TG, CHOL, ALT, AST concentrations showed rapid increase at the first exposure to the 10ppt salt level (day 1), and began to decrease to the constant values after day 2 of adaptation at 20ppt salt level. However, LDH concentration tended to decrease during the whole experimental period. $Na^+\;and\;Cl^-$ showed slight decrease at day 1, and increased to a little bit higher values after day 2 rather than those in freshwater. All the fishes started on taking a food after day 4 of seawater adaptation. From these results, to reduce osmotic shock inducible stress to fish in seawater acclimation, gradual increase of salt levels is recommended.

Taste Preference and Taste Perception of Korean Elderly (한국노인의 맛 선호도와 맛 감지도에 관한 연구)

  • To investigate the changes of taste perception by aging, sixty one healthy elderly people and sixty five young adults were participated in the study. Most preferred levels of salt sour and sweet taste were chosen in bean sprout soup radish salad and yaksing respectively. Threshold levels of each taste were also chosen in NaCl solution for salt taste in citric acid solution for sour taste and in sucrose solution for salt taste, in citric acid solution for sour taste and in sucrose solution for sweet taste. The results are as follow: 1.Most preferred salt concentration in bean sprout soup was significantly higher in the elderly than in the young adults(p$\leq$0.001). There was no difference in sour taste preference of radish salad in both age groups. Most preferred sweetness in yacksig was significantly higher in both elderly men and women(p$\leq$0.001) 2. Threshold levels of salt and sweet taste were significantly higher in the elderly (p$\leq$0.001) However there was no difference in threshold level of sour taste in both age groups.

Salt Taste Acuity and Menstruation (월경중 염미감각의 변화에 관한 연구)

  • It was the purpose of this study to determine if the changes in the sense of taste occur with the periods of the menstrual cycle in 14 women aged 20 to 22 years. Results showed that the perceived intensity responses to different suprathreshold salt concentrations and the ad libitum salting level in soybean sprout soup did not differ significantly according to the different periods of the menstrual cycle. But women in the three or five days period previous to menstruation were sensi\ulcornertive at the lower salt concentration of 0.25%, but, on the contrary, were insensitive at the higher concentration of 1.25%, with increasing ad libitum salt preference in soybean sprout soup. Also, they were more or less high in the intensity slopes of perceived saltiness on the linear regression. The data suggests that a physiological mechanism for increasing salt intake may develop during the three or five days right before menstruation.

Soil salinity shifts the community structure and diversity of seed bacterial endophytes of salt-sensitive and tolerant rice cultivars

  • Soil salinity due to accumulation of salts particularly sodium chloride affects agricultural lands and their vegetation. Generally, rice is a moderately sensitive plant with some cultivars with varying tolerance to salinity. Though there are physiological differences between salt-sensitive and salt-tolerant rice cultivars, both are still affected especially during high salinity and prolonged exposure. This also ultimately affects their indigenous bacterial endophytes particularly those that inhabit the rice seed endosphere. This study investigates the dynamic structure of seed bacterial endophytes of salt-sensitive and tolerant rice cultivars grown in different levels of soil salinity. Endophytic bacterial diversity was studied Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Results revealed a very interesting pattern of diversity and shifts in community structure of bacterial endophytes in the rice seeds. There is a general decrease in diversity for the salt-sensitive rice cultivar, IR29 as soil salinity increases. For the salt-tolerant cultivars, IC32 and IC37, diversity interestingly increased at moderate salinity then decreased at high soil salinity. The patterns of community structure is also strikingly different for the salt-sensitive and salt-tolerant rice cultivars. IR29 has a more even distribution of abundance, but under soil salinity, the community shifted where Curtobacterium, Pantoea, Flavobacterium and Microbacterium become the more dominant bacterial communities. For IC32 and IC37, the dominant bacterial groups under normal stress conditions were also the dominant bacterial groups during salt stress conditions. Their seed bacterial community is dominated by endophytes belonging to Microbacterium, Flavobacterium, Pantoea, Kosakonia and Enterobacter. Stenotrophomonas and Xanthomonas have not changed in terms of abundance under different salinity stress level in the salt-sensitive and salt-tolerant rice cultivars. This study showed that soil salinity greatly influenced the seed bacterial communities of rice seeds irrespective of their physiological tolerance to salinity.

Processing of Low Salt Mackerel Fillet and Quality Changes during Storage (저염 고등어 Fillet의 제조 및 저장중 품질변화)

  • The processing conditions of low salt mackerel (Scomber japonicus) fillet was investigated, in which fresh mackerel was filleted, salted in brine until the expected salt concentration reached, dried with cool air (3 m/sec, $10{\sim}20^{\circ}C$), and finally packed individually in polyvinyl chloride film. Salting time and salt concentration of brine decided the final salt level penetrated into the fillet. As the final salt level was fixed to $0.8{\sim}1.0%, salting for $15{\sim}20 hours with 5% or 10% brine at $5^{\circ}C$ was enough to get that level of salt. Formation of histamine during salting was negligible. Changes in VBN, salt soluble proteins, and histamine formation of salted mackerel fillet during the storage occurred more rapidly in cases of storage at $5^{\circ}C than af $-2^{\circ}C and $-20^{\circ}C. Oxidation of lipid during the storage progressed, however it was delayed longer then 100 days in case of storage at $-20^{\circ}C. Addition of sodium erythrobate or ginger extracts could provide some extent of browning retardation. The shelf-life of the salted mackerel fillet based on panel scores of brown color and rancidity appealed to be 14 days when stored at $5^{\circ}C, and more than 28 days in case of storage at $-2^{\circ}C and about 3 months stored at $-20^{\circ}C.

Effect of Mineral-rich Salt Intake on Diabetic Goto-Kakizaki Rats (미네랄이 풍부한 천일염이 Goto-Kakizaki Rat에 미치는 영향)

  • The objective of this study was to determine the hyperglycemic effects of mineral-rich salt in type 2 diabetic Goto-Kakizaki (GK) rats and normal Wistar rats. Animals were divided into five groups, including a normal group, fed three different experimental salts [purified salt (PS), mineral-rich salt (WS1 and WS2), and bamboo salt (BS)] in the form of 1% salt solution for 12 weeks. Liver, kidney, and spleen weights were significantly increased in GK rats of salt groups as compared to Wistar normal group without salt. However, there was no difference among the salt groups. For serum lipids, total cholesterol level in the BS group and triglyceride level in the WS group were significantly reduced compared to those of the PS group. The concentration of blood glucose in the GK-PS group increased continuously during the experimental period, whereas that in the GK-WS group was significantly reduced at 12 weeks. In GK rats, glucose levels among the salt groups in OGTT by glucose were not significantly different compared to normal rats. Insulin and glucagon levels in blood were not significantly different among the groups, and no such association was observed for insulin. Pancreatic lslets of Langerhans in the PS group showed irregular formation compared to those of the normal, WS, and BS groups.

Probabilistic Structure Design of Automatic Salt Collector Using Reliability Based Robust Optimization (신뢰성 기반 강건 최적화를 이용한 자동채염기의 확률론적 구조설계)

  • This paper deals with identification of probabilistic design using reliability based robust optimization in structure design of automatic salt collector. The thickness sizing variables of main structure member in the automatic salt collector were considered the random design variables including the uncertainty of corrosion that would be an inevitable hazardousness in the saltern work environment. The probabilistic constraint functions were selected from the strength performances of the automatic salt collector. The reliability based robust optimum design problem was formulated such that the random design variables were determined by minimizing the weight of the automatic salt collector subject to the probabilistic strength performance constraints evaluating from reliability analysis. Mean value reliability method and adaptive importance sampling method were applied to the reliability evaluation in the reliability based robust optimization. The three sigma level quality was considered robustness in side constraints. The probabilistic optimum design results according to the reliability analysis methods were compared to deterministic optimum design results. The reliability based robust optimization using the mean value reliability method showed the most rational results for the probabilistic optimum structure design of the automatic salt collector.

The Effect of Salt Concentrations on the Production of Volatile Organic Acids by Zygosaccharomyces rouxii, a Soy Sauce Yeast (간장에서 분리한 Zygosaccharomyces rouxii의 휘발성 유기산 생성에 미치는 식염농도의 영향)

  • By using a synthetic medium simulated on the amino acid composition of soybeam, the effect of salt concentrations on the production of volatile organic acid by the strains of Zygosaccharomyces rouxii So-3101, a soy sauce yeast, was studied at the concentrations of 12.5, 18.0, 22.0 and 28.5% NaCl. The growth, consumption of glucose, and production of alcohol, total acid and volatile organic acid, showed the highest values at a concentration of 12.5% NaCl, and those values were decreased with an increase in the salt concentration. The ratio of volatile organic acid to total organic acid was remained at approximately the same level within the range of salt concentrations between 12.5~22.0%, whereas the ratio was decreased at a salt concentration of 28.5%. After incubation for 16 days, 8 volatile organic acids, i.e. acetic, propionic, n-butyric, isobutyric, isovaleric, isocaproic, n-caproic, and heptanoic acids, were detected by gas chromatography. Among the volatile organic acids, acetic acid was produced in the appreciable amiunt and its ratio to the other volatile acids was increased with an increase in the salt concentration.A small amount of isocaproic, propionic, isobutyric and isovaleric acids were produced, and n-caproic, n-butyric and heptanoic acids were detected only at the lower salt concentration.

Chitooligosaccharides in Korean Commercial Salt-Fermented Shrimps, Determined by Enzyme-Linked Immunosorbent Assay

  • In this study, we determined the content of chitooligosaccharides (COS) in Korean commercial salt-fermented shrimps by competitive direct enzyme-linked immunosorbent assays (cdELISAs), using anti-COS mixture (COSM) antibody and COSM horseradish peroxidase (HRP) conjugate. When COS6 was spiked into salt-fermented shrimps at the level of $10-300\mu{g/g,}$ the average recovery was $120\pm19%$ ($mean\pmS.D.$). The COS contents of the 92 samples of Korean commercial salt-fermented shrimps collected during February 2000 and August 2002 were $36.3\pm20.7\mug$ COS6 equivalent/g (expressed as "$\mug/g$" hereafter). Among the samples, the COS contents of yuk-jeot ( $40.3 \pm 22.5 \mug/g, n=27$) and buksaewoojeot ($40.2 \pm 21.6 \mug/g, n=5$) were higher than the others. The COS contents of salt-fermented shrimps produced at Gwangcheon ($47.1 \pm 20.7 \mug/g, n=18$) and Gomso ($44.1 \pm 21.8 \mug/g, n=6$) areas were higher than those produced at the other areas. This is the first report to determine COS of salt-fermented shrimps by cdELISA.

Relationship between halophyte distribution and soil environmental factors in the west coast of South Korea

  • Background: The objective of this study was to determine the relationship between soil environmental factors and halophyte distribution in the west coast of South Korea. Soils of our study sites were categorized into two groups: salt marsh and estuary marsh. Results: Salinity was higher in the salt marsh group than that in the estuary marsh group. However, total nitrogen, silt, and clay contents were higher in the estuary marsh group than those in the salt marsh group. Although altitude had a wider range in the salt marsh group, the mean altitude was higher in the estuary marsh group than that in the salt marsh group. Annual halophytes of seed propagation species were distributed parallel to the coast line on salt marsh. Higher coverage of vegetation was found in the area closer to the coast line. Plant density was higher near dead parental plants in estuary marsh, showing less difference in area that was more distant from the coast line. Conclusions: Results of canonical correspondence analysis (CCA) for vegetation distribution and sediment environmental factors and germination analysis in the coast line showed significant relationship with halophyte distribution. Therefore, they can be used as an indicator of coastal plant movement due to sea level rise.
