Pink Rot of Palms Caused by Gliocladium vermoseni (Gliocladium vermoeseni에 의한 야자 분홍썩음병(가칭))

  • Pink rot of Palm was occurred at Yeuju area in 2001 and 2003. Infected plants showed rotting at the leaf-stock bases and killing of the terminal bud. The first symptoms are dark brown necrotic areas on the stem. Bases of infected frond may be covered with pink spores and the spots produced oozing gum pockets. Oozing lesions occur on the stems, and leaves turn brown and droop. The causal agent were isolated from salmon-pink spores sporulating on the leaf sheaths and necrotic stem tissues. Pathogen were isolated from freshly infected tissues were identified as Gliocladium vermoseni based on mycological characteristics. Fungus were grown plenty on PDA culture. Temperature for mycelial grown was tested at 5 to $40^{\circ}C$ and optimal temperature was $25^{\circ}C$ and was not nearly grew at temperature above $35^{\circ}C$. Artificial pathogenicity were tested on 9 species of Palm family in the wound inoculation and symptoms showed similar to those observed in the field. This is the first report on pink rot of palm in Korea.

Expression of HSP90, HSP70 mRNA and Change of Plasma Cortisol and Glucose During Water Temperature Rising in Freshwater Adapted Black Porgy, Acanthopagrus schlegeli (담수 사육 감성돔, Acanthopagrus schlegeli의 수온 상승에 따른 HSP90, HSP70 mRNA의 발현 및 혈장 cortisol과 glucose 변화)

  • The objective of the present study was to investigate the expression of heat shock protein 90 (HSP90) and 70 (HSP70) mRNA as cellular stress responses, the levels of plasma cortisol with glucose as neuro-endocrine stress responses during water temperature rising in freshwater adapted black porgy, Acanthopagrus schlegeli. A cDNA fragment of 891 (HSP90) and 465 (HSP70) bp was cloned from black porgy testis by Reverse transcription-polymerase chain reaction (RT-PCR) with primers designed from the conserved regions of other teleost. The PCR product of HSP90 showed very high homology to red seabream (99%), rainbow trout (95%), Atlantic salmon (94%), zebrafish (94%) HSP90, HSP70 of black porgy was also highly similar to those of rainbow trout (96%), silver seabream (95%), zebrafish (95%) HSP70. Water temperature rising ($20{\sim}30^{\circ}C$) induced elevation of HSP90 mRNA in black porgy gonad, liver, brain, intestine and kidney, whereas it resulted in an induction of the HSP70 mRNA expression in gonad only. Plasma cortisol levels increased significantly at $30^{\circ}C$ in the fish compared to those at $20^{\circ}C$. Glucose levels of the fish showed a tendency of co-increase with cortisol during water temperature rising. These results suggest that increased HSP90 mRNA in liver with plasma cortisol following heat shock may be related to increasing glucose for homeostasis in this species.

Identification and chemotherapeutic effects of the fungi from three salmonid species and their eggs (3종의 연어과 어류와 수정난으로부터 분리한 물곰팡이병 원인 진균의 분류와 약물 효과)

  • Four strains of water mold, ChS-E0511, RaT-E0511, RaT-A0512 and MaS-F0512, were isolated from salmonid fish and/or their eggs taken from culture farms in Yangyang, Milyang and Pyeongchang, Korea in 2005. Descriptions of their morphological aspects, the results of the phylogenetic analysis conducted, and the sequence of the small sub-unit 18S rRNAs of the isolates confirmed that they all belong to the species Saprolegnia parasitica. Only one species, ChS-E0511, which was isolated from fertilized eggs of the chum salmon, was classified as part of the S. parasiticaGroup 1 according to its oogonia and gemmae production. The chemotherapeutic effects of various chemicals on the ChS-E0511 strain were assessed from the inhibitory effects of growth in GY media and the relative ratio of eyed eggs to fertilized eggs of the rainbow trout. Malchite green, a prohibited substance in food animals, was better than others, such as the Opuntia ficus-indicaextract, 2-bronopol, and sodium chloride. These results suggest that the fungi isolated from salmonids and/or their eggs identified as S. parasitica were composed of more than two groups. These isolates will be useful in an intensive evaluation of therapeutic agents.

Actions of a Gonadotropin-Releasing Hormone Antagonist on Gonadotropin II and Androgenic Steroid Hormone Secretion in Precocious Male Rainbow Trout

  • We used a mammalian GnRH antagonist, $[Ac-3,4-dehydro-Pro^1,\;D-p-F-Phe^2,\;D-Trp^{3.6}]$-GnRH, to examine the details of the salmon type gonadotropin-releasing hormone (sGnRH) and GnRH agonist analog $(Des-Gly^{10}$[d-Ala^6]-ethylamide GnRH; GnRHa) functions in the control of maturational gonadotropin (GTH II) secretion, in precocious male rainbow trout, in both in vivo and in vitro experiments. In the in vivo study, plasma GTH II levels increased by sGnRH or GnRHa treatment, but the response was more rapid and stronger in the GnRHa treatment group. The increase in GTH II was significantly suppressed by the GnRH antagonist, while the antagonist had no effect on basal GTH II levels in both groups. The GnRH antagonist showed stronger suppression of GTH II levels in the sGnRH treatment fish than in the GnRHa treatment fish. In addition, plasma androgenic steroid hormones (testosterone and 11-ketotestosterone) increased by the sGnRH or GnRHa treatment. The GnRH antagonist significantly inhibited the increases in plasma androgenic steroid hormone levels stimulated by the sGnRH or GnRHa, while the antagonist had no effect on basal androgenic steroid hormone levels in both groups. In the in vitro study, treatment with sGnRH or GnRHa increased GTH II release from the cultured dispersed pituitary cells, but the response was stronger in the GnRHa treatment group. The increase in GTH II release by GnRH was suppressed by adding the GnRH antagonist, dose­dependently. On the other hand, basal release of GTH II did not decrease by the GnRH antagonist treatment in both groups. These results suggest that the GnRH antagonist, $[Ac-3,4-dehydro-Pro^1,\;D-p-F-Phe^2,\;D-Trp^{3.6}]-GnRH$, used in this study is effective in blocking the action of GnRH-induced GTH II release from the pituitary gland both in vivo and in vitro.

Development of the Eggs and Pre-Leptocephalus Larvae by Natural Spawning of Artificially-Matured Japanese Eel, Anguilla japonica (인위적 성숙 유도된 뱀장어 Anguilla japonica의 자연산란에 의한 난발생과 Pre-leptocephalus 자어의 발달)

  • Embryonic and pre-leptocephalic larvae development of the eel, Anguilla japonica, are described following natural fertilization in the indoor tank of $23^{\circ}C$ water temperature. Following a routine hormone treatment technique for the brood stock, female eels were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE) at a dosage of 20 mg/kg body weight (BW) for a total of 10-14 doses to induce ovarian maturation, while male eels received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g BW for a total of 6-10 doses to induce testicular maturation in a separate aquarium and induced natural spawning. Fertilized eggs of about 1.0 mm in diameter were pelagic and showed a typical discoidal cleavage. Hatching occurs 38 hrs after fertilization at a water temperature of $23^{\circ}C$. The newly hatched larvae measured about 3.0 mm in total length and the number of myomeres averages 42. Their mouths and anuses were opened at 4.5 days and the yolk sacs of the pre-leptocephalic larvae were almost absorbed at 6.5 days after hatching. Pre-leptocephalic larvae survive for 14.5 days. At this time they are $5.87{\pm}0.25mm$ in total length and have about 98 myomeres. However, morphological characterization of embryonic and pre-leptocephalic larvae were not different between natural fertilization and artificial fertilization by the dry method.

A New Rhabdovirus (HRV-like) Isolated in Korea from Cultured Japanese Flounder Paralichthys olivaceus (양식산 넙치로부터 HRV-like Rhabdovirus의 분리)

  • In March 1997, a new rhabdovirus was isolated from moribund cultured Japanese flounder Paralichthys olivaceus in sea water tank and cage culture systems in Kyung-Nam and Chun-Nam province, Korea. At temperature $15^{\circ}C$ the virus replicated and induced cytopathic effects (CPE), which progressed to eventual cytolysis, in susceptible cell lines, including RTG-2 and EPC. The CHES-214 cell line was refractory. Virus particles were bullet-shaped and measured $70nm{\times}100$ to 150 nm in size. The isolate was sensitive to pH 3, to diethyl ether, and to heat ($50^{\circ}C$ 5 min, $60^{\circ}C$ 1 min). Viral replication was not inhibited by $10^{-4}$ M 5-iododeoxyuridine. Virus infectivity was reduced by anti-HRV (8401-H) rabbit serum, but can not reduced by antisera against infectious hematopoietic necrosis virus (IHNV), chum salmon reovirus (CSV), retrovirus of salmonid (RVS) and infectious pancreatic necrosis virus (IPNV). HRV virus antigen was detected by fluorescent antibody test (FAT) in the cytoplasm of infected EPC cell. Purified isolates virions were composed of: polymerase (L), glycoprotein (G), nucleoprotein (N) and 2 matrix proteins (M1 and M2). Based upon their relative mobilities, the estimated molecular weights of the proteins were: L, 160 kDa; G, 55 kDa; N, 45 kDa; M1, 26 kDa; and M2, 22 kDa.

  • Objective : There were few reports on the treatment of Adult-onset Still's disease and Diffuse cerebral dysfunction. This oriental medical treatment shows the possibility of healing Adult-onset Still's disease and Diffuse cerebral dysfunction, therefore we announce this. Methods : The acupuncture, herbal medicine, herbal acupucture(Jahageo, Nockyong) therapies were applied for treating this patient's chief symptom(chest discomfort, palpitation, rigidity, mentality change, global aphasia, dysphagia, coughing and sputum) Results : 1. Adult-onset Still's disease is a rare systemic inflammatory disorder of unknown etiology, characterised by salmon-colored rash, intermittent spiking high fever, arthralgia and variety of systemic features. This is one type of Juvenile arthritis. 2. Diffuse cerebral dysfunction is an anoxic-ischemic encephalopathy, and most cause of this is the hypoxia caused by hypotention or respiratory distress. 3. After oriental medical treatments, his chief symptoms were improved. Conclusion : The more study about oriental medical treatment on Adult-onset Still's disease and Diffuse cerebral dysfunction is needed.

Morphological and Genetic Stock Identification of Todarodes pacificus in Korean Waters (한국 주변해역에 서식하는 살오징어(Todarodes pacificus)의 형태 및 유전학적 계군분석)

  • Stock identification of Todarodes pacificus collected in the East Sea, Yellow Sea and East China Sea during the period from September to December in 2011 was analyzed by morphometric characters and mitochondrial DNA (mtDNA) cytochrome oxidase subunit I (COI) gene nucleotide variations. Frequency distributions of mantle length was analyzed by morphological method with measuring size of T. pacificus. Then each stock was estimated to confirm their maturation for mean mantle length comparing with mean mature mantle length 20-22 cm. According to morphologic stock identification, it is estimated that the northern part of East Sea is categorized as summer stock and the rest parts, including mid /southern part of the East Sea, northern part of the East China Sea and northern part of the West Sea were autumn stock. For genetic analysis, a total 49 haplotypes were defined by 33 variable nucleotide sites. From the extensive haplotype diversity, limited nucleotide diversity and star-like shape of haplotype network, T. pacificus appears to have undergone rapid population expansion from an ancestral population with a small effective population size. Although pair-wise Fst estimates which represent genetic difference among groups were low, there are relatively remarkable difference of Fst between middle and southern part of the East Sea. Although middle part of the East Sea and southern part of the East Sea were situated at the East Sea, genetically separated groups were appeared.

Determination of polycyclic aromatic hydrocarbons in processed foods (가공식품 중 다환방향족탄화수소 분석)

  • In this study, the following concentrations of some PAHs (Polycyclic Aromatic Hydrocarbons) were investigated; [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in processed foods (n=165) and cooked meats (n=45) and established the analytical method by characteristics of processed foods. The methodology involved extraction (alkali digestion, liquid-liquid extraction, microwave extraction), clean-up on Sep-Pak Florisil Cartridges and determination by HPLC/FLD (High Performance Liquid Chromatography/Fluorescence Detector). The recovery of overall method for 8 PAHs spiked into these products ranged from 92 to 103%. The mean level of detected foods was found to be benzo(b)fluoranthene $0.9\;{\mu}g/kg$ in smoked salmon, benzo(b)fluoranthene $1.0\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in dried banana, benzo(b)fluoranthene $0.2\;{\mu}g/kg$, benzo(k)fluoranthene $0.1\;{\mu}g/kg$, benzo(a)pyrene $0.2\;{\mu}g/kg$ in smoked chicken, benzo(b)fluoranthene $1.3\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in charcoal grilled pork, respectively.

Effect of the Treadmill Exercise and the Intake of DNA and Crude Catechin (from Puerariae thunbergiana Roots) on the Body Fat Composition and the Antioxidant Activity in Rats (Treadmill 운동과 DNA 및 칡 Catechin 섭취가 흰쥐 생체내 지방조성과 항산화 활성에 미치는 영향)

  • Rats(Sprague-Dawley) were randomly assigned to the following four groups, control, exercise only, exercise and the intake of DNA, exercise, and the intake of DNA plus crude catechin. 0.4% of DNA from salmon egg and 0.1% crude catechins from Puerariae thunbergiana roots were fed to the rats. The exercise group was exercised in a treadmill at 20 m/min speed for 6 wks. Body weight and body fat weight of 4 groups were investigated, and the body fat composition and antioxidant activity were evaluated by measuring the weight of organs and biochemical test. After 6 wks, body weight did not show any significant differences among those 4 groups, but body fat weight in exercised groups was significantly decreased. The weight of liver, epididymal adipose tissue(E.A.T) and perirenal adipose tissue(P.A.T) were significantly decreased in groups of exercise only, exercise and the intake of DNA, exercise and the intake of DNA plus crude catechin(p<0.05). Phospholipid, cholesterol and triglyceride levels of serum were decreased by exercise, but HDL-cholesterol level of serum was significantly increased(p<0.05). GOT, GPT and glucose levels in serum were slightly decreased by crude catechin, but serum NEFA levels were significantly increased by crude catechin(p<0.05). Results indicated that excercise with the intake of crude catechin would be helpful for the functional development of the compositions in blood lipid.
