Analysis of Penicillin Antibiotics in Aquatic Products (수산물 중 penicillin계열 항생제의 분석)

  • A simple, rapid method for determining amoxicillin (AMO) and ampicillin (AMP) in aquatic products (flatfish, salmon, shrimp, tilapia, and yellow croaker) was evaluated. For quantification, the AMO and AMP ions at m/z 348.9 and 105.9, respectively, were selected. The limit of detection (LOD) and limit of quantification (LOQ) for detecting AMO were 0.09 and $0.25\;{\mu}g/L$, respectively. The respective values for AMP were 0.02 and $0.05\;{\mu}g/L$. After $100\;{\mu}g/L$ AMO treatment, the level decreased 10% after 7 days at $5^{\circ}C$, while it decreased 20% at $25^{\circ}C$. After 7 days, 94.9.100% of $100\;{\mu}g/L$ AMP remained after storage at $5^{\circ}C$, while 62.3.100% remained after storage at $25^{\circ}C$. Using the food code method, the recovery of AMO ranged from 64.1.92.0% and that of AMP from 44.8.86.2%. With a protein centrifugation method, the recovery of AMO ranged from 39.8.87.9% and that of AMP from 78.0.98.1%. With liquid-liquid extraction, the recovery of AMO ranged from 36.5.88.3% and that of AMP from 31.8.75.1%.

  • We experienced one rare case of Diphyllobothrium latum infection in childhood. It was proved by collection of worm after praziquantel treatment. Human diphyllobothriasis is chiefly caused through eating raw or unproperly cooked fish such as trout or salmon. This case was a 5-year old girl residing in Seoul, from whom a kind of pseudophyllidean tapeworm was detected in the feces. She would often eat sushi(raw fish) and had abdominal pain, but not revealed any sign of anemia. Obtained chain of worm segments that sized $0.25{\times}1.25cm$ was compatible with D. latum. Including the present 31 cases, total number of human D. latum infections proven by worms in Korea becomes 32 cases, and total number of child D. latum infections proven by worms in Korea becomes 4 cases.

Study on the Temperature Adaptation of Contractile Myofibrillar Proteins from Fish Species (어류의 수축성 근섬유단백질의 온도 적응성에 관한 연구)

  • To investigate on the biochemical characteristics of myofibrillar proteins between cold(pollack, salmon) and warm current fish (yellow corbina, shark), myofibrils and actomyosin were prepared, and their biological activities, effect of temperature on the myofibrillar ATPase activities and SDS-polyacrylamide gel electrophoretic patterns of myofibrils were compared. SDS-polyacrylamide gel electrophoretic analysis showed that electrophoretic patterns of myofibril vary from fish to fish. Difference in KCl concentration dependency of myofibrillar ATPase activities and ATPase activity- pH curve were found among fish species. Myofibrillar proteins from cold current fish showed higher specific activity at low temperature $(5^{\circ}C-10^{\circ}C)$ than those from warm current fish.

  • This paper presents an echo-hydraulic experimental approach to assessment of ascending capacity of migratory fish in a baffled fishway. Sweetfish, Plecoglossus altivelis, trout, Oncorhynchus mykiss, are used in these experiments. Ascending environment of the migratory fish in rivers is analyzed through the results of taco-hydraulic experiments. Important factors affecting the ascending capacity of migratory fish in baffled fishways are the discharge and slope of fishways and the fall height between the fishway entrance and the river bed. In these fishways the migratory capacity of sweetfish for a given fishway discharge is mainly affected by fishway slope, whereas the ascending capacity of trout for a given fishway slope was influenced by the change of fishway discharge. Loss of ascending properties of landlocked salmon, Oncorhynchus masou var ishikawai, was confirmed by these experiments. An ascending hydraulic criteria of the baffled fishway is defined by means of dimensionless factor F$_{f}$ and falling head H$_{f}$. Ascending capacity of migratory fish in existing baffled fishways in river of eastern coastal region is poor because the slope of fishways is verb steep.eep.

  • Regional foods is influenced by weather, local and social environment etc. The purpose of this study was to understand that regional foods in the middle of Chosun Dynasty through Domundaejac. Some of representative dishes of the Hansung and Kyonggi-do Area is duck(rice cake) of seasonally celebrated days, hangua(korea confectionary) and rice wine. Typical foods in Chungchong-do Area is included persimmon, jujube, watermelon and wax gourd. Some of the well-known food in Kangwon-do Area is pear, bangpungchuk(gruel), sukebyung(rice cake), woongjijunggua(korea confectionary) and sanat-kimchi(picked mustard leaves and stem) that is seemed characteristics of mountainous section. Some of the famous dishes in Cholla-do Area is backsanja(korea confectionary), tea, chuksun kimchi(bamboo shoot pickle), citrus fruits and ear shell. Kyongsan-do Area ia famous persimmon, bamboo fruit, dasik(korea confectionary) and yakban(glutinous rice cake). Hwanghae-do Area is famed pear and choshi as assumed the eatly form of gochujang(thick soypaste mixed with red pepper). Representative dishes are deljjuk(blue berry), herring, sangat-kimchi(picked mustard leaves and sterm) in Hamgyong-do Area and daemandu(big dumpling) in Pyongan-do Area. The coast area is famous for using lots of sea food. Fresh seafood is consumed raw, and clams, fish and seaweed are liberally added to soups and other dishes. The East coast is catched salmon, flatfish, codfish, sandfish and mackerel The West coast is catched yellow corvenia, lobster, tiny shrimp and large-eyed herring. The South coast is catched sea mussel, codfish and laver.

  • History of Instream Flow Incremental Methodology (IFIM) Following the large reservoir and water development era of the mid-twentieth century in North America, resource agencies became concerned over the loss of many miles of riverine fish and wildlife resources in the arid western United States. Consequently, several western states began issuing rules for protecting existing stream resources from future depletions caused by accelerated water development. Many assessment methods appeared during the 1960's and early 1970's. These techniques were based on hydrologic analysis of the water supply and hydraulic considerations of critical stream channel segments, coupled with empirical observations of habitat quality and an understanding of riverine fish ecology. Following enactment of the National Environmental Policy Act (NEPA) of 1970, attention was shifted from minimum flows to the evaluation of alternative designs and operations of federally funded water projects. Methods capable of quantifying the effect of incremental changes in stream flow to evaluate a series of possible alternative development schemes were needed. This need led to the development of habitat versus discharge functions developed from life stage-specific relations for selected species, that is, fish passage, spawning, and rearing habitat versus flow for trout or salmon. During the late 1970's and early 1980's, an era of small hydropower development began. Hundreds of proposed hydropower sites in the Pacific Northwest and New England regions of the United States came under intensive examination by state and federal fishery management interests. During this transition period from evaluating large federal reservoirs to evaluating license applications for small hydropower, the Instream Flow Incremental Methodology (IFIM) was developed under the guidance of the U.S. Fish and Wildlife Service (USFWS).

Effects of Gonadotropin-Releasing Hormone on in vitro Gonadotropin Release in Testosterone-Treated Immature Rainbow Trout

  • The control mechanism of gonadotropin-releasing hormone (GnRH) on gonadotropin (GTH) release was studied using cultured pituitary cell or cultured whole pituitary obtained from Testosterone (T) treated and control immature rainbow trout. The release of FSH was not changed by salmon type GnRH (sGnRH), chiken-II type (cGnRH-II), GnRH analogue ([des-$Gly^{10}D-Ala^6$] GnRH ethylamide) and GnRH antagonist ([Ac-3, 4-dehydro-$Pro^1$, D-p-F-$Phe^2$, D-$Trp^{3,6}$] GnRH) in cultured pituitary cells of T-treated and control fish. Indeed, FSH release was not also altered by sGnRH in cultured whole pituitary. All tested drugs had no effect on the release of LH in both culture systems of control fish. The levels of LH, in contrast, such as the pituitary content, basal release and responsiveness to GnRH were increased by T administration in both culture systems. In addition, the release of LH in response to sGnRH or cGnRH-II induced in a dose-dependent manner from cultured pituitary cells of T-treated fish, but which is not significantly different between in both GnRH at the concentration examined. Indeed, LH release was also increased by sGnRH in cultured whole pituitary of T-treated fish. GnRH antagonist suppressed the release of LH by sGnRH ($10^{-8}\;M$) and GnRH analogue ($10^{-8}\;M$) stimulation in a dose-dependent manner from cultured pituitary cells of T-treated fish, and which were totally inhibited by $10^{-7}\;M$ GnRH antagonist. These results indicate that the sensitivity of pituitary cells to GnRH is elevated probably through the T treatment, and that GnRH is involved in the regulation of LH release. GnRH-stimulated LH release is inhibited by GnRH antagonist in a dose-dependent manner. The effects of gonadal steroids on FSH levels are less clear.

Gametogenic Cycle and the Size at 50% of Group Sexual Maturity in Male Chlamys (Azumapecten) farreri nipponensis (Kuroda, 1932) (Bivalvia: Pectinidae) in Western Korea

  • We investigated the gametogenic cycle and spawning seasons of the male Chlamys (Azumapecten) farreri nipponensis by qualitative and quantitative analyses, and also the size at 50% of group sexual maturity was calculated by the data of first sexual maturity. In this study, the male gametogenic cycle of this species by qualitative analysis was divided into five successive stages: early active stage (January to March), late active stage (March to April), ripe stage (April to August), partially spawned stage (July to September), and spent/inactive stage (August to January). The male gametogenic cycle showed similar patterns with monthly changes in the gonadosomatic index and condition index. Particularly, spawning in male scallop occurred once a year from July to September, unlike the spawning period of this species (from June to August) reported by the previous researchers. In quantitative statistical analysis using an image analyzer system, the patterns of monthly changes in the percent (%) of the areas occupied by spermatogenic stages to the testis areas in males showed a maximum in June, and then sharply dropped from July to September, 2006. From these data, it is apparent that the spawning season of C. (A.) farreri nipponensis occurred once per year from July to early September, indicating a unimodal gametogenic cycle during the year. Shell heights at 50% of group sexual maturity (RM50) fitted to an exponential equation were estimated to be 49.90 mm in males (considered to be one year old), and it was 100% for male scallops over 61.0 mm (considered to be two years old).

  • 면역효소법을 이용하여 3종의 한국산 개구리 참개구리(Rono nigromaculuto), 옴개구리(R. rugosa), 북방산개구리(R. 겨대bowskii)의 뇌에서 GnRH 뉴우런의 분포 부위와 GnRH의 종류 등을 연구하였다. 1차 항체로는 anti-rat GnRH, anti-salmon GnRH anti-chicken 11 GnRH 항체를 사용하였다. 3종의 개구리에서 mGnRH cGnRH 11와 sGnRH가 이둔 동정되었으나 3가지 항체에 대한 각 종의 면역 반응성은 종에 따라 달리 나타났다 mGnRH는 옴개구리와 참개구리에서, sGnRH는 북방산개구리에서 강한 면역 반응을 나타냈으며 cGnRH 11는 3종의 개구리에서 중간 정도의 면역 반응을 나타냈다. 각각의 GnRH의 상대적인 양에는 차이가 있으나 일부 경우를 제외하고는 뇌의 동일한 지역에 분포하였다. 참개구리에서는 GnRH가 중격 내측핵(NMS), Broca band 핵(NDB)에 집단으로 분포하였다. 북방산개구리에서는 GnRH가 중격 내측핵, Broca bnad 핵에서 등쪽에서 배쪽으로 길게 선상으로 가장 협소하게 분포하였으며, 번식기와 직전(1월-3월)에만 면역 반응을 나타냈다. 옴개구리의 뇌에서 가장 광범위한 지역, 즉 종뇌의 중격 내측핵, Broca band 핵, 아래 교차 지역(SCA)과 간뇌에 GnRH 신경세포가 분포하였으며. 제3뇌실 맥락얼기에서 mGnRH 신경세포가 처음으로 동정되었다. 3종에서 공통적으로 중격 내측핵과 Broca band 핵에서 유래한 신경섬유는 복측 시상하부를 거쳐 정중융기에 이르렀다. 이러한 결과는 GnRH가 뇌하수체에서 생식소 자극 호르몬의 분비 조절에 밀접한 관계가 있음을 뜻한다.

Purification and Characterization of Carboxypeptidase B from Wild Salmon (Salmo Salar) Eggs (연어알중의 Carboxypeptidase B 분리정제 및 그 특성에 관한 연구)

  • Carboxypeptidase B from Salmo Salar eggs was purified by CM-cellulose, 0.5 ammonium sulfate saturation, DEAE-cellulose, and Sephadex G-75 gel filtration and its enzymatic properties were investigated. Optimum temperature was 55$^{\circ}C$, pH optima were 4.0 and 7.0 at 37$^{\circ}C$, and the enzyme was stable at pH 2.0∼3.0 and 5.5∼7.0 for 1.5h. This enzyme showed substrate specificity hydrolyzing the peptide bond of glycyl-L-arginine. Its K$_m$ values was 0.21mM, and the enzyme activity was stimulated by Cu$^{2+}$ and Fe$^{3+}$, while inhibited by Zn$^{2+}$. The lysine was found to be competitive inhibitor and its K$_i$ value was determined to be 4.3mM. Molecular weight of this enzyme was determined to be 36,400 daltons by SDS-PAGE and the enzyme was monomeric protein composed of 19 kinds of amino acid residues.
