Changes in Phytoplankton Community Structure by Freshwater Input in the Cheonsu Bay, Korea (담수 유입에 따른 천수만 해역의 식물플랑크톤 군집 변화)

  • Environmental factors and changes in phytoplankton community structure before (August 5, 2017), during (August 18 and 25) and after (August 30 and September 15) freshwater input were analyzed to investigate the effects of freshwater input from Ganwol and Bunam lakes located in the upper part of Cheonsu Bay. Due to the large amount of freshwater input in the Cheonsu Bay, the surface salinity of the bay decreased by more than 8 psu, and the thermocline existing in the bay during August weakened. In addition, hypoxic phenomena occurred temporarily in the bay as the low oxygen water mass from the freshwater lakes flowed into the bay, and chemical oxygen demand, nutrients, and N/P increased with freshwater inflow. The density of phytoplankton during the freshwater inflow increased owing to their input from the freshwater lakes. Diatom species (Eucampia zodiacus) dominated the phytoplankton community in the bay before freshwater input; nanoflagellates, chlorophyta, cyanobacteria, and diatoms (Pseudonitzschia delicatissima, Chateocceros spp.) entered during freshwater input; and after freshwater inflow ended, diatoms (Chateocceros spp.) again became predominant indicating a return to previous conditions. The amount of phytoplankton standing crops increased sharply due to the inflow of freshwater species into the bay on the second day of discharge compared to before freshwater input; pre-discharge conditions were restored at most stations except at some sites close to the Bunam Lake three days after discharge. Therefore, the large amount of freshwater flowing into the bay affects not only the geochemical circulation in the bay but also the phytoplankton community structure. In particular, the high concentration of nutrients in the freshwater lake affect the marine ecosystem of the bay during August.

  • This study proposes a cost-effective method to monitor coastal environments using e-Navigation-implemented domestic and international ferries, and to analyze big data of records such as wind, temperature, salinity, waves, and currents that are gathered through e-Navigation system. First, we present the concept and architecture of e-Navigation operation system based on the General Information Center on Maritime Safety and Security. Then, the marine observation system that can be applied to ferries operating in our nation's territory is discussed. Analytical methods, such as spatio-temporal mixed effects model, ensemble method, and meshfree method, in handling real-time big data obtained by the e-Navigation observing system are then explained in detail. This study will support the implementation of the Korean e-Navigation project that focuses on the safety of small vessels such as coasters and fishing vessels.

A Study of The Effect of Corrosion on Heat Transfer in a Heat Exchanger (열교환기에서 부식이 열전달에 미치는 영향에 관한 연구)

  • Heat pump systems based on ocean thermal energy conversion (OTEC) systems use the temperature difference between deep ocean water and surface ocean water to operate. However, they may have heat transfer degradation due to corrosion on the heat exchanger surface due to the salinity of sea water. This study presents experimental results for the heat transfer decrease of corroded metal tubes with respect to corrosion time. In order to replace high-priced titanium, electro-deposition (ED) coating was performed on aluminum tubes. Aluminum tubes with ED coating thicknesses of 10, 15, and $20{\mu}m$ were tested for double-tube heat exchangers after performing accelerated corrosion for 6, 12, and 18 weeks. The effects of the coating thickness and the corrosion time on the heat transfer degradation were investigated. From the results, the aluminum tube with an ED coating of $20{\mu}m$ thickness can be suggested as a candidate for replacing titanium tubes.

A study on Spatiotemporal Variations of distribution characteristics in Artificial Rivers of the Brackish Water Zone (기수역 인공하천에서 시공간적 수질분포 특성 연구)

  • The purpose of this reaserch is to analyze the charateristics of water quality in space through the operation of ARA River in Artificial Rivers of the Brackish Water Zone. The spatial distribution measured water temperature and salt levels for the surface, middle and deep layers by dividing the four areas of Incheon, Sicheon, Gyeyang and Gimpo. Water temperature did not vary much by water depth and branch, and its purpose is to maintain stable water environment through correlation analysis and operation. To examine the temporal and spatial distribution patterns of the Arachon, we measured DO on the Incheon branch, Sicheon, Gyeyang and Gimpo branch twice a month, and on the surface, the temperature level, The water temperature did not vary much by depth and location, and the water temperature in January and March tended to rise from Incheon to Gimpo, with the average difference of 1.1 degrees during the same period. The salinity difference between Incheon and Gimpo sites was 3.3 psu deep and 5.4 psu deep. In particular, floodgates from July to September are found to be less than 10psu overall, which is considered to be a gas due to the effects of floods and the inflow of Gulpo Stream. D.O. is located in some areas due to summer rains. The hypoxic layer has been identified.Analysis of seasonal data shows that water temperature and DO are strongly correlated in autumn. It was found that the water temperature and salt levels in the fall showed a weak correlation.

Processing and Quality Analysis of Seasoned Low-salt Fermented Styela clava Supplemented with Fermentation Alcohol for Extended Shelf-life (발효주정 첨가 저염 미더덕(Styela clava) 양념젓갈의 제조 및 품질)

  • In order to develop value-added low-salt fermented seafood with a long shelf-life, we prepared seasoned low-salt fermented Mideoduck (Styela clava) supplemented with fermentation alcohol (SME). The SME was produced by washing and dewatering shelled Mideoduck, followed by cutting and salting for 24 h at 0℃. The salted Mideoduck was seasoned and fermented with ingredients, including garlic, ginger, monosodium glutamate, red pepper, sesame, sorbitol and sugar, for 7-8 days at 0℃. After adding 3-5% fermentation alcohol, the Mideoduck was packed in a polyester container. The salinity, volatile basic nitrogen, and amino nitrogen content of the SME was 4.5%, 20.9 mg/100 g and 92.0 mg/100 g, respectively. In comparison with the control, the addition of 3-5% fermentation alcohol showed inhibitory effects of decreased freshness, texture degradation, and growth of residual bacteria. Additionally, the SME had good storage stability and organoleptic qualities when stored at 4±1℃ for 40 days. Therefore, it is suitable for commercialization as a seasoned low-salt fermented product with a long shelf-life. The total amino acid content of the SME was 11,774.5 mg/100 g, majorly comprising glutamic acid, aspartic acid, lysine, arginine, and leucine, and the free amino acid content was 506.4 mg/100 g, majorly comprising hydroxyproline, taurine, and glutamic acid.

Enhanced Production of C30 Carotenoid 4,4'-Diaponeurosporene by Optimizing Culture Conditions of Lactiplantibacillus plantarum subsp. plantarum KCCP11226T

  • The rising demand for carotenoids can be met by microbial biosynthesis as a promising alternative to chemical synthesis and plant extraction. Several species of lactic acid bacteria (LAB) specifically produce C30 carotenoids and offer the added probiotic benefit of improved gut health and protection against chronic conditions. In this study, the recently characterized Lactiplantibacillus plantarum subsp. plantarum KCCP11226T produced the rare C30 carotenoid, 4,4'-diaponeurosporene, and its yield was optimized for industrial production. The one-factor-at-a-time (OFAT) method was used to screen carbon and nitrogen sources, while the abiotic stresses of temperature, pH, and salinity, were evaluated for their effects on 4,4'-diaponeurosporene production. Lactose and beef extract were ideal for optimal carotenoid production at 25℃ incubation in pH 7.0 medium with no salt. The main factors influencing 4,4'-diaponeurosporene yields, namely lactose level, beef extract concentration and initial pH, were enhanced using the Box-Behnken design under response surface methodology (RSM). Compared to commercial MRS medium, there was a 3.3-fold increase in carotenoid production in the optimized conditions of 15% lactose, 8.3% beef extract and initial pH of 6.9, producing a 4,4'-diaponeurosporene concentration of 0.033 A470/ml. To substantiate upscaling for industrial application, the optimal aeration rate in a 5 L fermentor was 0.3 vvm. This resulted in a further 3.8-fold increase in 4,4'-diaponeurosporene production, with a concentration of 0.042 A470/ml, compared to the flask-scale cultivation in commercial MRS medium. The present work confirms the optimization and scale-up feasibility of enhanced 4,4'-diaponeurosporene production by L. plantarum subsp. plantarum KCCP11226T.

Application of Lactic Acid Bacteria to Inhibit Fungal Contamination of Cured Cheeses (항곰팡이능 보유 유산균의 숙성치즈 적용 연구)

  • Lactic acid bacteria with antibacterial activity can be effectively used as probiotics to inhibit the growth of harmful bacteria that cause food spoilage or food poisoning. In this study, Pediococcus pentosaceus M132-2, isolated from soybean paste, was analyzed for its effects on three major contaminating fungi. M132-2 was confirmed to exert antifungal activity by inhibiting the growth of all three fungi tested. In addition, M132-2 displayed excellent salt resistance and low temperature tolerance. Thus M132-2 can survive at the salinity level in cheese and at the low temperatures used in the aging process. Finally, when supernatant from an M132-2 culture was applied to Gouda cheese, the growth of contaminating fungi was significantly inhibited. Consequently, M132-2 may be useful for the prevention of spoilage of various foods, including cheese.

Hydrogen Sulfide Alleviates Seed Germination Inhibition in Oilseed Rape (Brassica napus L.) Under Salt Stress

  • The germination process is critical for plant growth and development and it is largely affected by environmental stress, especially salinity. Recently, hydrogen sulfide (H2S) is well known to act as a signaling molecule in a defense mechanism against stress conditions but poorly understood regulating seed germination. In this study, the effects of NaHS (the H2S donor) pretreatment on various biochemical (hydrogen peroxide (H2O2) content and amylase and protease activity) and physiological properties (germination rate) during seed germination of oilseed rape (Brassica napus L. cv. Mosa) were examined under salt stress. The seed germination and seedling growth of oilseed rape were inhibited by NaCl treatment but it was alleviated by NaHS pretreatment. The NaCl treatment increased H2O2 content leading to oxidative stress, but NaHS pre-treatments maintained much lower levels of H2O2 in germinating seeds under salt stress. Amylase activity, a starch degradation enzyme, significantly increased over 2-fold in control, NaHS pretreatment, and NaHS pretreatment under NaCl during seed germination compared to NaCl treatment. Protease activity was highly induced in NaHS-pretreated seeds compared to NaCl treatment, accompanied by a decrease in protein content. These results indicate that NaHS pretreatment could improve seed germination under salt stress conditions by decreasing H2O2 accumulation and activating the degradation of protein and starch to support seedling growth.

Relationship between Pathogenic Vibrios and Zooplankton Biomass in Coastal Area, Korea (병원성 비브리오균과 동물성 플랑크톤과의 관계에 관한 연구)

  • An ecological study on pathogenic vibrios was done in the aquatic environments of southern coast of Korea during summer in 1995, to investigate the distribution and relationship between pathogenic vibrio and zooplankton. Furthermore, special emphasis was given to study on the efforts of zooplankton existence on the wintering of Vibrio cholerae in the aquatic region in Korea. During the study period, pathogenic vibrios were isolated from the samples such as V. parahaemolyticus, V. vulnificus, V. mimicus, and V. cholerae non O1, but V. cholerae O1 was not detected in any sample submitted in this study. Adsorption ratio of V. parahaemolyticus onto zooplankton was higher than that of E. coli. The efficiency of adsorption was found to be on the concentration of NaCl and other ions found in sea water. For example, adsorption ratio of V. parahaemolyticus were $75\%\;at\;5\%_{\circ}$ of NaCl solution and $55\%$ at same salinity of diluted sea water, but those were decreased as $20\%\;and\;7\%\;at\;15\%_{\circ}$ salinity of NaCl solution and diluted sea water, respectively. In addition, survival period of pathogenic vibrio was extended in the presence of live copepods at $25^{\circ}C$, but zooplankton existence has no significant effect on the survival rate at $5^{\circ}C$ in closed microcosm and also microalgae and dead copepods do not affect on the survival of V. parahaemolyticus. According to these experimental results, zooplankton has positive effects on the growth and survival rate of pathogenic vibrios in sea water during the summer season, but copepods have no significant effects on the growth and survival rate of them in winter season in Korea. Finally, authors suggest that V. cholerae is not able to over winter with zooplankton in adjacent sea water in Korea.

Quality Characteristics of Cabbage Kimchi by Different Packaging materials (포장재에 따른 양배추 김치의 품질특성)

  • In this study, cabbage ($Brassica$ $oleracea$ var. $capitata$) Kimchi was made packed into four kinds of packaging materials, PET vessel, PP tray, OPP/AL/PE film and Nylon/PE/LLDP film, and the effects of these packaging materials on Kimchi quality characteristics, such as lactic acid bacteria counts, salinity, sugar contents, pH, total acidity, electron donating ability were examine as well as their effects on the sensory qualities after storage at a temperature of $4^{\circ}C$. The pH change ranged from pH 6.24 to 6.43 shortly after manufacture, and did not significantly change until 7 days of storage. However, it began to decrease rapidly after 14 days. On the 35th day of storage, the acidity was 0.79% in the PET vessel and 0.83% in OPP/AL/PE. Therefore, the PET vessel and OPP/AL/PE were considered appropriate packaging materials for Kimchi storage. The salinity did not change significantly during the storage period, and the sugar content generally increased in the four kinds of packaging materials, but decreasing after the 7th day of storage. After 14 days of storage, the Kimchi stored in the OPP/AL/PE film showed the highest lactic acid bacteria counts. Although the electron donating ability was the highest after proper time for fermentation, it decreased in all the packaging materials after the proper time for fermentation. However, the OPP/AL/PE film had an antioxidant potential of up to 93.18%. In the sensory evaluation, fermented Kimchi was found to be superior unfermented Kimchi. In addition, the Kimchi stored in the OPP/AL/PE film for 14 days showed the high score of 6.70 and 6.60 in overall preference. Therefore, the results of this study provide basic knowledge on the fermentation level and packaging material's condition for commercialization of small packed cabbage Kimchi. Henceforth, industrialization must include a variety of studies under these conditions to increase the merchantability.
