Identification of the Orchid Mycorrhizal Fungi Isolated from the Roots of Korean Native Orchid

  • The orchid symbiotic fungi were isolated from the roots of Korean native orchid (Cymbidium goeringii) collected and Chinese orchid (C. sinense) obtained from greenhouses. They were identified as a species of Rhizoctonia, based on the sequences of 18r rDNA, the microscopic observations of mycelia, and the symbiotic relationships with commercial orchids. The isolate collected from Chinese orchids was revealed to be a species of Ceratobasidium endophytica, and to be different from the other isolates at the thickness of the mycelia stained in the root cells of Korean native orchids. The other isolates collected from the Korean native orchids were considered to be a species of Tulsanella repens (anamorphic: Epulorhiza repens) or its related one. The physiologic or microscopic variations were oftenly observed among them, but the tendency of grouping these in the 18s rDNA sequences were observed to be consistent with those of the localities collected. The further taxonomical segregating for Korean symbiotic fungi was not made because the information concerned were limited in this moment, but was recognized as based on the sequences of 18s DNA.

  • We have fabricated a single flux quantum 4-stage shift register with interface-controlled $Y_1$$Ba_2$$Cu_3$$O_{7-x}$(YBCO) Josephson junction. The YBCO Josephson junctions showed RSJ-like current-voltage(I-V) curves at temperatures 45~80K. We tested load and shift operation of shift register with binary data sequences “1000”, “1010”, “1011”, and “1111” at 58K. For all the binary data sequences, the shift register operated successfully. By operating the circuit with proper current pulses, we observed no errors during at least 12 hours operation for all the data sequences.s.

  • An epizootic of calf diarrhea occurred in a Korean native cattle farm located in Chonbuk province. The calves that had either bloody or watery diarrhea were 1 to 30 days old. Some of these animals died during the acute course of the disease. Five calves with predominant clinical signs were examined in more detail. Hematological and serum chemical findings were suggestive of dehydration and nutritional insufficiency. Fecal material from the calve was cultured on/in brilliant green agar (BGA), xylose-lysine deoxycholate (XLD) medium, MacConkey agar, eosin methylene blue (EMB) agar and triple sugar iorn (TSI) A bacteria was isolated. which was subsequently identificed as belonging to Salmonella spp. To differentiate Salmoenlla serotype, rfbs gene of S. dublin was amli- find (720 bp) by multiplex (PCR). The rfbS gene sequences of S, dublin ficld isolate(SDC-1) was com- pared with that off S. dublin(S-37) S, dublin(Ahn et al, 1996), S enteritidis(Ahn et al 1996)and S. typhi (Generbak accession No M29682). The identities of nucleotide sequences were 100%. 99.6%, 99.6%, 97.5% respectively.

nrDNA-ITS 분자마커를 이용한 오미자(五味子) 종 감별 및 기원분석 -ITS 염기서열을 이용한 오미자(五味子) 감별- (Molecular Authentication of Schisandrae Fructus and Analysis of Phylogenetic Relationship based on nrDNA-ITS sequences)

  • Objectives : The original plant species of Schisandrae Fructus (O-mi-ja) is prescribed as Schisandra chinensis $B_{AILL.}$, in Korea, but S. chinensis $B_{AILL.}$ and S. sphenanthera $R_{EHD.}$ et $W_{ILS.}$ in China. Moreover, fruit of several other species in genus Schisandra also have been used as the same herbal medicines. To develop a reliable method for correct identification of Schisandrae Fructus and to evaluate the phylogenetic relationship of S. chinensis and its related species, we analyzed internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA (nrDNA). Methods : Twenty-four plant samples of three Schisandra species and one Kadsura species, S. chinensis $B_{AILL.}$, S. spenanthera $R_{EHD.}$ et $W_{ILS.}$, S. nigra $M_{ax.}$ and Kadsura japonica $D_{UNAL}$ were collected from each different native habitate and farm in Korea and China. The nrDNA-ITS region of each samples were amplified using ITS1 and ITS4 primer and nucleotide sequences were determined after sub-cloning into the pGEM-Teasy vector. Authentic marker nucleotides were estimated by the analysis of ClastalW based on the entire nrDNA-ITS sequence. Results : In comparative analysis of the nrDNA-ITS sequences, we found specific nucleotide sequences including indels (insertions and deletions) and substitutions to distinguish C. chinensis, S. spenanthera, S. nigra, and K. japonica. These sequence differences at corresponding positions are avaliable nucleotide markers to determine the botanical origin of O-mi-ja. Moreover, we evaluated the phylogenetic relationship of four plant species by the analysis of nrDNA-ITS sequences. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by the providing of definitive information that can identify each plant species and distinguish it from unauthentic adulterants for O-mi-ja.

Identification and Characterization of Novel Sequences of ev21-K Locus for Feather-Sexing in Chickens

  • 본 연구는 조우성과 만우성 닭을 식별하기 위한 유전자 마커를 발굴하고자 한 것으로 만우성과 관련된 ev21-K라는 새로운 좌위를 발견하고 이의 특성을 구명하였다. 더불어, 본 좌위의 유전적 전이 양상을 조사하여 자가 성감별 라인 조성의 이용 가능성도 살펴보았다. 본 시험을 위해 5개 품종의 닭 707수를 공시하고 이를 대상으로 유전자 마커 발굴 및 유전적 전이 시험을 수행하였다. ev21-K 특이 좌위 발굴은 깃털 발육과 연관된 ev21 유전자와 만우성 유전자인 K 유전자를 탐색하고 이들 간 염기서열을 비교 분석하여 획득하였다. 확인된 좌위의 분석을 위해 대상 서열에 대한 특정 프라이머를 제작하고 중합효소연쇄반응(PCR)을 수행하여 결과물을 획득한 후 이들의 염기 서열을 분석하였다. 발굴된 염기 서열의 유전적 전이 양상을 조사하기 위하여 조우성과 만우성 닭 간의 교배조합시험을 수행하였다. 시험결과, 발굴된 230 bp ev21-K 유전자 좌위를 ev21-related K specific sequences라 명명하였고, 이는 기존 ev21 유전자와 99%의 상동성을 나타내었다. PCR 분석을 통해 해당 서열이 만우성 닭에만 존재하는 것으로 확인되었다. 본 서열은 조직, 품종 및 연령에 관계없이 만우성 닭에만 존재하는 일관된 결과를 보여주었다. 교배 시험을 통하여 본 서열의 전이 양상을 살펴본 결과, 반성 유전을 하며 깃털 표현형과 일치하는 분리결과를 보였다. Ev21-related K specific sequences의 유전적전이 양상은 본 서열이 우성으로써 전형적인 멘델 유전에 따르는 것으로 나타났다. 결론적으로, ev21-K 특이 좌위의 새로운 서열은 품종에 관계없이 조우성과 만우성 닭을 식별하기 위한 신뢰할 수 있는 분자 마커로 확인되었다.

ON THE SOLUTION OF NONLINEAR EQUATIONS CONTAINING A NON-DIFFERENTIABLE TERM

  • We approximate a locally unique solution of a nonlinear operator equation containing a non-differentiable operator in a Banach space setting using Newton's method. Sufficient conditions for the semilocal convergence of Newton's method in this case have been given by several authors using mainly increasing sequences [1]-[6]. Here, we use center as well as Lipschitz conditions and decreasing majorizing sequences to obtain new sufficient convergence conditions weaker than before in many interesting cases. Numerical examples where our results apply to solve equations but earlier ones cannot [2], [5], [6] are also provided in this study.

  • 이상혁(李尙爀)(익산(翼算))의 퇴타술중 삼각타, 사각타 계열에 관한 부분을 조사하고, 익산(翼算)의 결과를 부분합 복수열의 성질로 재해석한다. 유한생성 부분합 복수열의 개념을 도입하고 삼각타, 사각타 계열에 의한 부분합 복수열이 유한생성 부분합 복수열임을 보인다. 단위 부분합 복수열이 부분합 복수열의 연구에 핵심적 역할을 함을 보인다. 또한, 부분합 복수열이 유한생성이 되기 위한 필요충분조건을 구한다. 그리고, 교초적에 대한 곱셈법칙에 대응하는 삼각타적, 삼각낙일적(三角落一積)에 대한 곱셈법칙을 구한다.

Nitrifying Bacterial Community Structure of a Full-Scale Integrated Fixed-Film Activated Sludge Process as Investigated by Pyrosequencing

  • Nitrifying bacterial community structures of suspended and attached biomasses in a full-scale integrated fixed-film activated sludge process were investigated by analyzing 16S rRNA gene sequences obtained from pyrosequencing. The suspended biomass had a higher number of ammonia-oxidizing bacterial sequences (0.8% of total sequences) than the attached biomass (0.07%), although most of the sequences were within the Nitrosomonas oligotropha lineage in both biomasses. Nitrospira-like nitrite-oxidizing bacterial sequences were retrieved in the suspended biomass (0.06%), not in the attached biomass, whereas the existence of Nitrobacter-like sequences was not evident. The suspended biomass had higher nitrification activity (1.13 mg N/TSS/h) than the attached biomass (0.07 mg N/TSS/h). Overall, the results made it possible to conclude the importance of the suspended biomass, rather than the attached biomass, in nitrification in the wastewater treatment process studied.

Functional Annotation and Analysis of Korean Patented Biological Sequences Using Bioinformatics

  • A recent report of the Korean Intellectual Property Office(KIPO) showed that the number of biological sequence-based patents is rapidly increasing in Korea. We present biological features of Korean patented sequences though bioinformatic analysis. The analysis is divided into two steps. The first is an annotation step in which the patented sequences were annotated with the Reference Sequence (RefSeq) database. The second is an association step in which the patented sequences were linked to genes, diseases, pathway, and biological functions. We used Entrez Gene, Online Mendelian Inheritance in Man (OMIM), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Gene Ontology (GO) databases. Through the association analysis, we found that nearly 2.6% of human genes were associated with Korean patenting, compared to 20% of human genes in the U.S. patent. The association between the biological functions and the patented sequences indicated that genes whose products act as hormones on defense responses in the extra-cellular environments were the most highly targeted for patenting. 북한강 수계 조류대발생 원인종 남조 Anabaena의 분자계통학적 검토 (Molecular Identification of the Bloom-forming Cyanobacterium Anabaena from North Han River System in Summer 2012)

  • 2012년 북한강수계 전역에 걸쳐 대발생을 일으킨 남조 Anabaena의 분자생물학적 특성을 검토하였다. 시료는 2012년 7월 13일에 남조 Anabaena 밀도가 높았던 3개 호소- 의암호, 청평호, 팔당호에서 각각 채집하였다. 분석은 선행연구 문헌을 근거로 하는 형태학적 분석, 16S rRNA 염기서열 분석을 통한 분자계통학적 분석을 동시에 실시하였다. 결과를 종합하면 북한강수계 3개 호소에서 조류 대발생을 일으킨 남조 Anabaena는 모두 동일종 Anabaena crassa (Lemmermann) Komark.-Legn. & Cronberg이며, 동시에 출현하고 본 종과 형태 및 분자생물학적으로 매우 유사한 A. circinalis는 환경요인에 따라 형태변이가 쉽게 일으키는 동일종으로 밝혀졌다.
