Phytochemical Identification from Boehmeria nivea Leaves and Analysis of (-)-Loliolide by HPLC

  • Phytochemicals were isolated from leaves of the fiber crop, ramie (Boehmeria nivea, Bn), using open column chromatography and medium pressure liquid chromatography. Their structures were identified as ${\beta}$-sitosterol, (-)-loliolide, rutin, and pyrimidinedione by MS, $^1H$-, and $^{13}C$-NMR spectroscopic analysis. Among them, (-)-loliolide was isolated for the first time from B. nivea. A content analysis of (-)-loliolide in B. nivea collected from different regions and harvest times was conducted by HPLC. The highest content of (-)-loliolide was found in Bn-23 harvested in September. These results will be helpful to use the plant which harvest in September as a high content phytochemical additive in food, health supplements, and medicinal products.

  • Chemical characteristics of mulberry syncarp were investigated six mulberry varieties, including Subongppong, Daeryukppong, Kuksang 20, Chongilppong, Cataneo and a wild variety. Water content of mulberry syncarp was 78.5∼85.0%and sugar degree of Kuksang 20 was the highst among six varieties. Anthocyanin content of Chongilppong mulberry syncarp was 282.42mg% and Kuksang 20 was lower 13% than Chongilppong mulberry fruit. Also Daeryukppong and wild variety were higher 11% respectively. GABA content of wild variety was higher than other varieties. In flavonol glyconsides analysis, rutin content was the highest in Cataneo, isoquercitrin content was in a wild variety, guaijaverin and quercitrin content was the highst in Chongilppong respectively.

  • All ten compounds were isolated from the methanolic extract of the whole plants of Diodia teres through repeated silica gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as three iridoid glycosides, asperuloside, geniposidic acid and asperulosidic acid, a coumarin glycoside, scopolin, and six flavonoids, rutin, kaempferol-3-0-rutinoside, quercitrin, astragalin, isoquercitrin and quercetin by spectroscopic analysis.

  • Mulberry fruit is a new income product in Korea sericulture due to the increase of fruit consumption. However, flavonoids of Korean mulberry cultivar for fruit production did not reported yet. In this study, the typical mulberry cultivar, 'Daeshim' was analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique for flavonoids analysis. Nine flavonoids were isolated and analyzed from Daeshim using UPLC-DAD-QTOF/MS chromatogram. According to quantitative analysis, rutin (66.1 mg/100g DW) and quercetin 3-O-(6"-O-malonyl) glucoside (26.7 mg/100g DW) were abundant in mulberry fruit. Our results might be used as basic information for mulberry consumption.

Effect of active ingredients in the Citrus fruits on the proliferation of hepatocellular carcinoma cells (감귤 내 생리활성성분이 간암세포의 생존에 미치는 영향)

  • Previous studies have suggested that Citrus fruits might suppress the proliferation of various cancer cells. However, little is known about any specific ingredients in the extract of Citrus fruits to exert its anti-proliferative activity in cancer cells. The present study aimed to identify the active ingredients in Citrus fruits to suppress the proliferation of rat hepatocellular carcinoma cells. Among tested compounds, two polymethoxylated flavones (nobiletin and tangeritin) showed significant anti-proliferative activity whereas other compounds (synephrine, rutin, hesperidin) did not. Interestingly, nobiletin as well as tangeritin also decreased the protein amount of gluconeogenic enzymes, PEPCK and G6Pase. The possible involvement of gluconeogenic activity in the proliferation of hepatocellulacarcinoma cells are further to be investigated.

  • Abeliophyllum distichum Nakai belonging to Oleaceae is only species in Korean endemic genus, Abeliophyllum. Abeliophyllum distichum (AD) is divided into various types according to flower shape and color. AD is known to have various colors such as white, pink, and ivory. Recently, light yellow flowers have been registered as new varieties (Okhwang 1ho). To date, various ecological and morphological studies on AD have been carried out, but no studies have been made on the phytochemicals and activities according to various traits. In this study, we analyzed the phytochemicals and antioxidative activities of from four kinds of flowers (white, pink, ivory, light yellow) in full bloom. The contents of phytochemicals such as chlorogenic acid, Hirsutrin, Rutin, Acteoside and Isoacteoside were analyzed by HPLC. Antioxidant activity was evaluated by DPPH, ABTS. As a result, the content of each substance varied according to the flower color. These results will provide basic data for evaluating the usefulness of genetic resources in Korea and developing new functional materials in preparation for the Nagoya Protocol.

Physicochemical quality, antioxidant compounds, and activity of 'Beta Tiny' and 'TY Nonari' cherry tomatoes during storage

  • In this study, a comparative analysis was carried out between the 'Beta Tiny' and 'TY Nonari' cherry tomato cultivars harvested at the pink and red stages. Samples of the red stage were stored at room temperature for 9 days, during which physicochemical qualities, antioxidant compounds, and activities were measured. As cherry tomato ripening and storage progressed, firmness was reduced, whereas the lycopene content increased. Total phenolic content and antioxidant activity showed no significant changes as ripening and storage progressed; however, total flavonoid content of 'Beta Tiny' showed a significant increase (p<0.05). The main polyphenols in the two cultivars were identified as chlorogenic acid, rutin, and (-)-epigallocatechin gallate, among which chlorogenic acid showed a significant decrease (p<0.05) as ripening and storage progressed. A strong correlation was found between total phenolic and flavonoid content (R=0.744), and ABTS radical scavenging activity (R=0.975). Additionally, a negative correlation was shown by lycopene and chlorogenic acid (R= -0.934).

  • Phyllanthus ussuriensis Rupr. et Maxim. (Euphorbiaceae) has long been used in folk medicine to treat kidney and urinary bladder disturbances, intestinal infections, diabetes, and hepatitis. Reported chemical constituents of this species are one flavonoid (rutin), two gallotannins (gallic acid, methyl gallate) and two ellagitannins.An investigation of the n-BuOH fraction of P. ussuriensis led to the isolation of two new megastigmane glycosides, 10-hydroxy-4,7-megastigmadiene-3-one-9-O-b-D-glucopyranoside (1), 10- hydroxy-4,6-megastigmadiene-3-one-9-O-b-D-glucopyranoside (2) and two known compounds roseoside (3) and 3-oxo-a-ionol-9-O-b-D-glucopyranoside (4). Antioxidant compounds and activities of pedicel and sepals from twelve varieties of colored cherry tomatoes

  • This study analyzed the antioxidant contents and activities of the pedicel and sepals from 12 colored cherry tomatoes ('Green Joy', 'TY Item', 'Dotori Red TY', 'TY Sispen', 'KT Orange TY', 'White Joy TY', 'Dotori Norang TY', 'Beta Tiny', 'Blacklin', 'KT Red TY', 'KT Norang TY', 'Black Joy 200') for their potential use as bioactive ingredients. 'Green Joy' had a significantly higher content of total flavonoids (92.55±3.20 mg CE/100 g FW), total phenolics (261.94±8.32 mg GAE/100 g FW), and total antioxidant activity than the other varieties. The main polyphenols were rutin, chlorogenic acid, and methyl gallate. For all 12 samples, the total flavonoids content was highly correlated with the total phenolics content and the total antioxidant activities. Non-edible parts of cherry tomato have high potential as functional food materials because they contain similar or more antioxidants and antioxidant properties than the pulp of tomato and cherry tomato or other fruits.

  • Aegle marmelos (Rutaceae) is a deciduous shrub or tree and typically known as bael throughout Southeast Asia and has been used as a medicinal plant and a food ingredient. In this study eight compounds were determined to be O-(3,3-dimethylallyl) halfordinol (1), (R)-aegeline (2), (R)-marmeline (3), imperatorin (4), xanthotoxol (5), valencic acid (6), vanillic acid (7) and rutin (8). The chemical structures of the isolates were elucidated through spectroscopic evidence including 1D, 2D NMR, ESI-Q-TOF-MS and optical rotation.
