Inflammation inhibitory effect of water extract from pumpkin's tendril (호박 덩굴손 물 추출물의 염증제어 활성)

  • Pumpkin has long been used as traditional health materials in oriental medicine, pharmacy, medicine, and food industries in many countries. In this study, the water extract and two active components from tendril of young C. moschata Duch. were investigated on inflammation inhibitory activity. The water extract of young C. moschata Duch. showed high cell viability over 95% and it decreased the production of interlukin 6 (IL-6) and tumor necrosis factor (TNF-${\alpha}$) in the capacity of mouse bone marrow-derived macrophages (BMDM) upon lipopolysaccharide (LPS) stimulation. Also, isolated fraction (B4) suppressed the secretion of IL-6 and TNF-${\alpha}$. Among the domestically cultivated pumpkins, B4 fraction contained in the tendril part of them and it comprised of the order of tendril from Cucurbita pepo var. cylindrica, old of C. moschata Duch., and young of C. moschata Duch. These results suggest that water extracts of C. moschata Duch. and purified active compound, rutin, show anti-inflammation activity by suppression of the secretion of IL-6 and TNF-${\alpha}$. It can be applicable as pharmaceutical materials.

Composition of Buckwheat (Fagopyrum esculentum Moench) Cultivars from Korea (한국산 메밀의 성분)

  • To clarify the values and varieties of the buckwheats as a dietary source of nutritional and functional components, thirteen different samples of buckwheat were analyzed for this investigation. Six developed seeds were given by RDA, Korea or RDA branch of Kangwondo, and seven land race seeds were collected from a farmhouse. Amino acid analysis showed that glutamate, arginine and asparagine were major amino acids, whereas tryptophan, methionine and cysteine were minor ones of buckwheat. In addition, tryptophan content of buckwheat cultivars from Korea was 195 mg% on average. The content of rutin tended to be higher in developed cultivars than land races. On the other hand, the contents of phytic acid in buckwheats were in the range of 7.0 to 13.6 mg/g. In the tocopherol homologues of the buckwheats analyzed by HPLC, mean ${\gamma}-tocopherol$ contents were 6.16 mg/100 g with the actual range of $4.67{\sim}8.58\;mg/100g$, whereas ${\beta}-form$ was very low or zero. There were a big variations in the iron content of the buckwheats of the minerals. SDS-PAGE showed that total proteins from buckwheats exhibited a relatively similar electrophoretic patterns on the whole. The results show that CV Suwon 1 has good quality, judged from the distribution of the components of buckwheats analyzed.

Anti-oxidation and Anti-thrombosis Activities of Polyphenol-enriched Fraction of Acorn Pollen Extract (도토리 화분 추출물의 폴리페놀 고함유 분획물의 항산화 및 항혈전 활성)

  • Pollen corrected from Quercus species is abundant in Korea and has been used to treat anemia and inflammation of the prostate gland. It is also used for hemostasis in oriental medicine. In this study, the ethanol extract of the acorn pollen and its subsequent organic solvent fractions using hexane, ethylacetate, butanol, and water residue were prepared, and their antioxidant, anti-thrombosis, and hemolysis activities were evaluated. The ethylacetate fraction of acorn pollen (EF-AP) showed the highest polyphenol content (225.0 mg/g) and strong antioxidant activity among the solvent fractions. The RC50 of EF-AP against DPPH, ABTS, and nitrite radicals was 72.2, 27.7, and 62.6 ㎍/ml, respectively. The blood coagulation activities of the all-solvent fractions determined by thrombin time, prothrombin time, and activated partial thromboplastin time were negligible up to 5 mg/ml. Platelet aggregation inhibitory activities were observed in the EF-AP, butanol fraction, and water residue, whereas hexane fraction induced strong platelet aggregation. The EF-AP has no hemolysis activity against human RBC up to 1 mg/ml. UPLC/MS/MS analysis of the EF-AP revealed that rutin, isoquercitrin, and astragalin are major compounds for antioxidant and anti-thrombosis activities. Our results suggest that EF-AP could be developed as a noble antioxidant and anti-thrombosis agent.

Components and Pharmaceutical Effect of Beverage Extracted from Sugar-treated Tartary buckwheat (타타리메밀 당절임 추출음료의 성분 및 약리효능)

  • Tartary buckwheat is one of specialized plants in Pyeongchang county, Korea and contains rutin much more than common buckwheat. Rutin is a kind of flavonoid (polyphenol compound) that has effects on blood vascular disease, strengthen capillary, and anti-inflammatory effect. This study was conducted to determine the possibility of development of beverage extracted from sugar-treated plants and sprouts of tartary buckwheat. By using two types of undiluted solution extracted from plant and sprouts of tartary buckwheat, we analyzed their nutrition components and did experiment on mice to find out pharmaceutical effects. In an experiment on mice, we administered various concentration of buckwheat to induced diabetic mellitus mice for 1 weeks. As a result, the buckwheat effected finely on lowering blood sugar and decreased LDL-cholesterol and total lipid level but increased HDL-cholesterol level.

  • Sevral flavonoids were isolated from the ethanol extract of Carthami flos which has been used in treatment of uterin congestion and also as analgesic and antiinflammatory. They were elucidated as kaempferol, quercetin, 6-hydroxy kaempferol, kaempferol 3-glucoside (Astragalin), quercetin 3-glucoside (isoquercitrin), quercetin 7-glucoside (quercimeritrin), kaempferol 3-rutinoside and quercetin 3-rutinoside (rutin). The structures of the isolated compounds were established by spectroscopic and chemical methods.

  • Seven compounds, euscaphic acid (1), nepehinol (2), ursolic acid (3), phytol (4), ${\beta}-amyrin$ (5), scopoletin (6), and rutin (7) were isolated from the aerial parts of Synurus excelsus which has been used as Korean traditional medicine to treat rheumatic arthritis and dysmenorrhea. Their structures were established by chemical and spectroscopic methods. All compounds were isolated for the first time from this plant.

  • Two phenolcarboxylic acids. five flavonoids and one hydrolysable tannin were isolated from the leaves of Acer ginnala Maxim. On the basis of chemical and spectroscopic evidence, the strutures of these compounds were established as gallic acid, ethylgallate, acertannin, quercetin, quercitrin, isoquercitrin, rutin, $quercetin-3-O-{\alpha}-_L-rhamnopyranosyl-2'-gallate$.
