Anti-oxidant and Anti-inflammatory Effects of Rutin and Its Metabolites

  • Rutin is one of the major flavonoids found in buckwheat (Fagopyrum esculentum Moench). While rutin is already known to exhibit anti-oxidative, anti-inflammatory, and anti-carcinogenic activities. However, the health beneficial function of rutin metabolites is not well understood. In DPPH radical scavenging assays, the present study found that 3,4-dihydroxyphenyl acetic acid had the highest total anti-oxidant activity, followed by 3,4-dihydroxyphenylacetic acid, rutin, homovanillic acid, and 3-hydroxyphenyl acetic acid. Further, 3,4-dihydroxyphenylacetic acid strongly reduced LPS-induced IL-6 production in RAW 264.7 cells, compared with other metabolites. Therefore, these results suggest that rutin metabolites have potential to be utilized as food ingredients with anti-oxidant and anti-inflammatory activities.

Interleukin-1으로 유도된 급성폐손상에서 rutin의 효과 (Rutin Ameliorates Neutrophilic Oxidative Stress-Induced Acute Lung Injury by Intratracheal IL-1 Insufflation in Rats)

  • 흰쥐에서 Interleukin-1 (IL-1)으로 유도된 급성폐손상에서의 group II phospholipase $A_2$ ($PLA_2$) 억제제인 rutin의 효과를 알아보기 위하여 본 연구를 시행하였다. Rutin은 IL-1에 의해 증가한 폐장내의 myeloperoxidase의 활성도를 감소시키지는 못하였으나 폐포세척액 내의 호중구의 수 및 모세혈관의 손상지표로 알려져 있는 폐장 모세혈관에서의 단백질 누출양을 감소시켰다. 동시에 rutin은 IL-1에 의하여 증가한 폐장의 염증조절효소인 $PLA_2$의 활성도를 감소시키고 결과적으로 호중구에서의 산소기의 생성을 감소시켰다. Rutin 뿐만 아니라 manoalide, scalaradial 같은 group II $PLA_2$의 억제제도 호중구의 respiratory burst를 감소시킴을 확인하였다. IL-1에 의하여 증가한 폐포세척액 내에서의 cytokine induced neutrophil chemoattractant의 농도는 rutin에 의해 영향을 받지 않았다. 형태학적으로는 IL-1에 의한 폐장조직에서의 산소기의 형성이 관찰되었고 rutin은 이러한 산소기의 생성을 현저히 감소시켰다. 이러한 결과로 미루어 group II $PLA_2$ 억제제인 rutin은 호중구에서의 활성 산소기의 생성을 효과적으로 억제함으로써 IL-1에 의한 급성폐손상의 감소를 가져 오는 것으로 결론지을 수 있다.

뽕나무 계통별 오디함유 rutin 분석 및 품종변이 (Quantification and Varietal Variation of Rutin in Mulberry Fruits)

  • Rutin은 GABA(γ-aminobutyric acid)와 더불어 콜레스테롤 저하, 혈당상승 억제, 고혈압 치료, 동맥경화 및 중풍 예방 등 다양한 뽕잎의 기능성을 나타내는 생리활성 물질로서 인식되고 있다. 뽕잎과 더불어 기능성 및 천연색소 자원으로서 유망시되고 있는 오디를 뽕나무 계통별로 수확하여 rutin 분석을 실시하고 계통간 특성을 살펴봄으로써 품종육종 효율을 높임은 물론 오디의 기능성 및 이용성을 증대시키고자 하였다. 그 결과를 요약하면 다음과 같다. 1. 공시계통의 평균함량은 0.14±0.050% DW이었으며, '사방소'는 0.29% DW로 가장 높은 함량을 나타낸 반면, '심설'은 가장 낮은 함량인 0.05% DW를 나타냈다. 2. 오디 중의 생육시기에 따른 rutin 함량의 경시적인 변화를 살펴본 결과, 뽕잎의 경우와 마찬가지로 '암뢰상', '청노상', '강선', '만생백피노상' 및 '사방소' 5계통 모두 수확시기가 이른 것의 오디에서 함량이 높았다. 3. Rutin 고함유 계통에 대한 오디로서의 이용가능성을 검토하기 위해 '사방소', '휘카스', '강선', '중호상', '만생백피노상' 및 '팔청시평 ' 6계통의 과실적 특성을 조사한 결과. '휘카스', '강선' 및 '팔청시평 ' 3계통은 수량, 과중, 당도 등 과실적 특성도 양호한 성적을 나타냈으므로 금후 오디생산과 기능성을 동시에 고려한 품종선택시 활용할 가치가 있는 우수계통으로 선발하였다. 4. 건조방법 및 저장방법이 식물체의 rutin 함량에 영향을 미치므로 오디 건조 및 저장시 지나친 고온과 저온을 피해야 하며, 식품으로 가공시에도 이 점에 주의하여 생리활성물질인 rutin의 섭취량을 늘리도록 하여야 할 것이다.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01). 익모초의 계절 및 지역별 Rutin 함량변화 연구 (Study on the Seasonal and Regional Variation of Rutin Content of Leonuri Herba)

  • In a pattern analysis of GC-MS of 21 Leonuri herba samples harvested in China and Korea, characteristic differences were observed between Chinese and Korean samples. specifically, Leonuri herba form Hanllasan showed unusual retention time. Rutin was employed as a standard compound of Leonuri herba. All samples had 0.32${\pm}$0.18(%) rutin contents in average, but Korean samples had more rutin than Chinese one, especially the sample from Hanllasan of Jeju showed high-est rutin content (3.2%). In rutin contents of seasonal variation, there were more rutin contents in after blooming, 2years aged Leonuri herba than before.

쓴 메밀에서의 루틴 추출 최적 공정 개발 (Development of Optimum Rutin Extraction Process from Fagopyrum tataricum)

  • The rutin content of Fagopyrum tataricum is 100-fold higher than that of Fagopyrum esculentum. For the development of a rutin-containing beverage, a suitable method to extract rutin from buckwheat (Fagopyrum tataricum) with high rutin yield was investigated. A roasting temperature range of $310/240^{\circ}C$ (Ed-confirm that this is indeed a range; otherwise perhaps, 'Roasting temperatures ranging from 310 to $240^{\circ}C$ were considered$\ldots$') was considered to be the best as the basic color reference. Rutin content varied according to the roasting time and heating temperature; i.e., it decreased with increasing roasting time and temperature. (Ed- this sentence is unnecessarily complicated and should be simplified to 'Rutin content decreased with increasing roasting time and heating temperature.') The optimal extraction temperature and processing time were obtained as $80^{\circ}C$ and 10 minutes to maximize the rutin concentration in the extract.

메밀의 rutin 생합성과 수량에 대한 토양개량제의 효과 (The Effect of Soil Conditioners on the Rutin Biosynthesis and the Yield of Buckwheat(Fagopyrum esculentum moench))

  • 메밀의 rutin생합성과 수량에 대한 토양개량제의 효과를 구명 하기 위하여 전남농업기술원 시험포장에서 3요소, 3요소+석회, 3요소+붕사, 3요소+계분퇴비, 3요소+유박을 시용 하여 시험을 수행한 결과 메밀 식물체중의 인산 함량은 개화전 보다 개화기에 증가 하였고, 질소, 가리, 석회, 고토는 감소 하였다. 메밀중의 rutin은 줄기보다 잎에 다량함유되어 있으며 개화전보다 개화기에 많이 함유 하고 있었다. 잎중의 rutin과 아미노산 함량과는 정의 상관 이었으며, 줄기중의 아미노산 함량과는 부의 상관 이었다. 수량은 3요소+붕사 시용구에서 $1,092kg\;ha^{-1}$로 3요소 시용구 보다 21% 증수 하였다.

Evaluation for Protective Effect of Rutin, a Natural Flavonoid, against HCl/Ethanol-Induced Gastric Lesions

  • In this study we investigated the protective effects of rutin, a natural plant flavonoid, on HCl/ethanol-induced gastric lesions in rats. Rutin showed the antioxidant activities, the acid-neutralizing capacities, and the inhibitory effects on the growth of Helicobacter pylori (H. pylori.), which are equivalent to control compounds. In addition, rutin significantly inhibited HCl/ethanol-induced gastric lesions. Antigastritic action of rutin may be associated with the antioxidant activities, acid-neutralizing capacities, anti-H. pylori action, and the stimulation of mucus secretion. From these results, we could suggest that rutin may be useful for the treatment and/or protection of gastritis.

  • This study was carried out to investigate the chemical change of rutin and fatty acids of buckwheat during germination. The content of rutin in buckwheat before germination was 31.5mg% and that after 7 days was increased to 1, 660.3mg%. The content of stearic acid was increased slightly after 3 day during germination but those of other fatty acids were decreased gradually.

Rangium koreana Ohwi의 성분 연구 (Study on the component of rangium ohwi)

  • In order to find a resouces of "Rutin", the chemical component of flower of Rangium Ohwi is studied in this paper. Rutin is isolated and identified from f of rutin in flower of this family plants followcetin derived from the isolated substance. The lower of this plant confirming acetate and quering the paper of J. Naghski and W. L. Portes, and no one has reported on the presence of rutin in flower of this plant.
