Identification of genes related to intramuscular fat content of pigs using genome-wide association study

  • Objective: The aim of this study is to identify single nucleotide polymorphisms (SNPs) and genes related to pig IMF and estimate the heritability of intramuscular fat content (IMF). Methods: Genome-wide association study (GWAS) on 704 inbred Berkshires was performed for IMF. To consider the inbreeding among samples, associations of the SNPs with IMF were tested as random effects in a mixed linear model using the genetic relationship matrix by GEMMA. Significant genes were compared with reported pig IMF quantitative trait loci (QTL) regions and functional classification of the identified genes were also performed. Heritability of IMF was estimated by GCTA tool. Results: Total 365 SNPs were found to be significant from a cutoff of p-value <0.01 and the 365 significant SNPs were annotated across 120 genes. Twenty five genes were on pig IMF QTL regions. Bone morphogenetic protein-binding endothelial cell precursor-derived regulator, forkhead box protein O1, ectodysplasin A receptor, ring finger protein 149, cluster of differentiation, tyrosine-protein phosphatase non-receptor type 1, SRY (sex determining region Y)-box 9 (SOX9), MYC proto-oncogene, and macrophage migration inhibitory factor were related to mitogen-activated protein kinase pathway, which regulates the differentiation to adipocytes. These genes and the genes mapped on QTLs could be the candidate genes affecting IMF. Heritability of IMF was estimated as 0.52, which was relatively high, suggesting that a considerable portion of the total variance of IMF is explained by the SNP information. Conclusion: Our results can contribute to breeding pigs with better IMF and therefore, producing pork with better sensory qualities.

Transition over 35 Years in the Incidence Rates of Primary Central Nervous System Tumors in Shanghai, China and Histological Subtyping Based on a Single Center Experience Spanning 60 Years

  • Background: Only few epidemiological data on primary central nervous system (CNS) tumors in Shanghai have been reported. Methods: All cases of primary CNS tumors that were registered at Center for Disease Control and Prevention (CDC) were collected (1973-2007: urban Shanghai; 2003-2007: whole Shanghai city). Trends were analyzed using joinpoint analysis and rates were stratified by age, gender and region. Histological data were collected from both CDC and Huashan Hospital. Results: From 1973 to 2007, the five-year average incidence rate in urban Shanghai increased in both genders, especially in the elderly population. Joinpoint analysis showed the age-adjusted incidence rate for males increased first but then plateaued, whilst rates for females continued increasing over the 35 years. For the five-year status quo (2003-2007), rural had a higher age-adjusted incidence rate than urban populations, and females higher than males, especially those with advanced age. According to CDC (2003-2007) and Huashan Hospital (1951-2011), the two most common histological subtypes were neuroepithelial tumors (with male predominance) and meningiomas (with female predominance). Conclusions: In Shanghai, a steadily increased incidence rate of primary CNS tumors was observed in general, and in the elderly and female population in particular.

Identification of Mating Type Loci and Development of SCAR Marker Genetically Linked to the B3 Locus in Pleurotus eryngii

  • In order to estimate how diverse the mating types in Pleurotus eryngii from different regions are, pairings between monokaryons derived from inter- and intra-groups were done. Sixteen and 15 alleles were identified at loci A and B from the 12 strains. In the P. eryngii KNR2312, widely used for commercial production, four mating loci, A3, A4, B3, and B4, were determined. Those loci, except A3, were found in 4 strains out of 12 strains. To improve breeding efficiency, especially in mating type determination, RAPD and BSA were performed to screen for a mating type specific marker. The SCAR marker 13-$2_{2100}$ was developed based on the RAPD-derived sequence typing B3 locus. The sequence analysis of 13-$2_{2100}$ revealed that it contained a conserved domain, the STE3 super-family, and consensus sequences like the TATA box and GC box. It seems likely that the SCAR marker region is a part of the pheromone receptor gene.

Isolation and Identification of Plant-Growth-Promoting Bacteria and Their Effect on Growth of Red Pepper(Capsicum annuum L.) (식물생육촉진(植物生育促進) 세균(細菌) 분리(分離), 동정(同定)과 고추에 대한 처리효과(處理效果))

  • This study was conducted to determine the effect of treatment with the plant-growth-promoting bacteria on the growth of red pepper(Capsicum annuum L.).The eight plant-growth-promoting bacteria were isolated from the humic soil in the forest region. The isolated bacteria(IB) was identified by the method of the biochemical test(API kit) and the composition of the fatty acid(MIDI system).The IBs were inoculated by spray of 17ml at 72 cell tray filled with peatmoss every week. respectively, with mixed liquid eulture of eight strains. The IBs were identified as Micrococcus sp.. Bacillus subtilis. Enterobacter agglomerans, Bacillus megaterium, Pseudomonas putida. Pseudomonas fluorescens, Xanthomonas maltophilia and Staphylococcus xylosus by API kit and MIDI system. The plant height number of leaves and leaf length of red pepper grown on peatmoss treated with the IB were better than those of nontreatment at the 10th day after inoculation.

Participation of the Women Covered by Family Physicians in Breast Cancer Screening Program in Kerman, Iran

  • Background: Mammography screening is a method for reducing breast cancer mortality in women over 40 years old. A participation rate of at least 70% is a prerequisite for screening programs. This study aimed at determining the participation rate of women in breast cancer screening in Iran. Materials and Methods: The study population in this prospective research consisted of 35 to 69 years old women in the villages and towns Kerman District, in 2013. The data were collected by a well-validated risk assessment questionnaire. The questionnaires were completed with the help of health workers and technicians in the health centers, who were trained on breast cancer screening program. Results: As a whole, 19,651 women were invited to complete the questionnaire, of whom 15,794 women (80.37%) completed it. In the urban region, of 3150 eligible women 2728 women (86.60%) participated in the study. The acceptance rates for mammography in rural and urban regions were 34.95% and 8.75%, respectively. Conclusions: Finally, 3.8% and 16.34% of 35 to 69 years old women in the urban regions were mammographed, respectively. Conclusion: The low participation of eligible women in breast cancer screening program alerts us against including the program in the health insurance package.

Infection State and Classification of Anisakid Larvae in Salmon (Oncorhynchus keta) which Caught from Taep'o Port, Kang-won-do (강원대 대포항에서 구입한 연어(Oncorhynchus keta)의 Anisakid 유충 감염상)

  • Seven specimens of salmon(Oncorhynchus keta) purchased directly, in Oct. 10. 1990, in the Taep'o port. Kang-won-do were examined for infection state of anisakid larvae and classification of extracted larvae according to morphological characters. The results were as follows. 1) From seven salmon 202 anisakid larvae were found, and mean infection number of anisakid larvae per individual salmon was 28.86. 2) From total extracted anisakid larvae 198 larvae(98%) were found in muscle. Therefore the distribution of anisakid larvae in intestine was extremely rare compared to in muscle. 3) The percentage of anisakid larvae in II, IV muscle region was 93% and from this we could surmise that most anisakid larvae in salmon did not penetrate to the very distanted muscle from intestine. 4) Three types of anisakid larvae(Anisakis Type I, Contracaecum Type B, Contracaecum Type D) were identified and, among them, Contracaecum Type B was the first recording type in Korea 5) Larvae of Contracaecum it genus were found only in intestine. Therefore it surmised that penetration neture to muscle of Contracaecum larvae was less than that of Anisakis Type I.

Genetic Analysis of Ancient Bones of Cervidae Animals from Archaeological Site in Jeju, Korea

  • DNA extracted from ancient bones of Cervidae animals was examined to identify the species and to determine the phylogenetic relationships to those from extant cervids. Abundant ancient bones were excavated from Kumsung archaeological site in Jeju Island, Korea, and were identified as Cervidae animals based on morphological features of their antlers and lower mandibles. Their mitochondrial DNA (mtDNA) control region (CR) was partially sequenced and subsequently compared with those previously reported in database. The results confirmed that the ancient sequences are lineage of Cervidae. On the phylogenetic trees constructed using the sequence diversity of the CR sequences of family Cervidae, the ancient DNA sequences were found on distinct clusters. The ancient sequences were located in the subfamily Capreolinae cluster, and six ancient sequences were closely related to those of extant Korean roe deer in Jeju Island and Korean Peninsula. Consequently, the results of this study suggest that the roe deer inhabited Jeju Island in ancient times. However, there is no evidence for the existence of subfamily Cervinae, including Sika deer, while it has been described in several historical records. The results suggest that this finding could contribute to understanding of the origin and phylogenetic relationships of extant and ancient roe deer on Jeju Island.

  • The purpose of this study is to investigate elementary school 6th grade children's preconception on body's digestion, circulation, breathing and excretion. By means of questionnaire, teacher is presented with misconception which students may have. On the basis of this way, firstly they draw up and put in free-concepts were investigated from objective test and sought after misconeption which student had and inquired whether the result of response made difference by sex and areas or not The result of study is as follows: 1. In the course of investigating and analyzing mis-preconecptios, teacher had misconceptions partially, as like students. 2. Many a student have misconceptions: nourishment in made for itself in body. 3. In circulation range most of students know that heart is making blood, and they don't make conception that blood is nourishment which was eaten from food. 4. They know the breath is only physiological function: they breathe to live. All air-breathing is necessary. 5. They are not relevant blood in course of urine and sweat formation they have non-scientific conceptions which water eaten is so. They know that all of sweat and urine and ordure is excretion. To sum up, many student have misconceptions as for digestion and circulation and breath and excretion of body especially. Circulation and excretion is more so. if inspection in a field of sex, they will know male had less misconceptions than female. in a field of region, student from rural are as had more misconceptions than the one from urban areas.

  • In general, a damage that occurs due to landslide or slope failure in urban areas is larger than that in rural areas. In order to reduce the damage, a program is necessary to categorize slopes based on properties and to manage them systematically. Based on the above necessity, a slope management program which is applicable to slope management in an urban area has been developed at Hwangryung mountain in Busan as a target area. The program has a function of systematic slope information constructed by slope ID number of each slope or sub-region of a mountain, making a slope data sheet, analysis and grouping of slope stability, and establishment of a data base. It can also be utilized practically by end users due to the convenient input, edition, printing, management and operation of slope data. For practical utilization of the developed program, a research related to construction of the slope management system for a regional area is demanded to be performed continuously. The supply and utilization of a web based slope management system would contribute to damage reduction.

The Factors Affecting on the Health Service Satisfaction of Health Center Visitors (보건소 이용자의 의료서비스 만족도에 영향을 미치는 요인)

  • The purpose of this study is to identify the nature of the patient service quality of health center, and based on that, to examine the relationship of health center visitor's perceived quality and overall satisfaction. Data were collected on the basis of 'the 3rd Regional Health Care Planning' operated by Rural Health Care Service Technology Aid Task-force Team(RHCSTAT), Ministry of Health and Welfare. Basically, the Manual of 3rd Regional Health Care Planning is composed of structure, process, and outcome measurement indicators. Of these indicators, 21 patient satisfaction questions as outcome indicators are used in this study. The samples are 3,530 patients who visited 80 health centers all of the nation. First, the reliability and validity of patient service quality items was evaluated. Second, the descriptive statistics of health service quality are calculated. Third, through the T-test and ANOVA, the differences of patient satisfaction by responder's general characteristics are compared. In final, to explore the relationship of service quality and overall satisfaction, multiple regression analysis was used. This study shows firstly, the dimension of patient service quality was categorized into 3 dimensions, that is, facilities and environment, staff kindness, and access convenience. Secondly, the reliability and validity of patient service quality items was satisfied. Thirdly, the mean score of staff kindness factors are high in contrast to the facilities and environment. And in regard to the overall satisfaction, the mean score is more higher in Daegu region, County type health center, male group, and upper 65 age group than comparison groups. Lastly, as a result of multiple regression analysis, the effect of patient's perceived service quality, especially, appropriateness of billing time(Beta=0.190) and of reservation time(Beta=0.132) are statistically significant(P<0.01). In conclusion, to satisfy the health need of community people and improve the service quality of the health center, the efforts for the access convenience might be needed. Additionally, the enlargement on health center space and supply of new medical equipment is urgent for the patient satisfaction.
