Characterization of the Maize Stalk Rot Pathogens Fusarium subglutinans and F. temperatum and the Effect of Fungicides on Their Mycelial Growth and Colony Formation

  • Maize is a socioeconomically important crop in many countries. Recently, a high incidence of stalk rot disease has been reported in several maize fields in Gangwon province. In this report, we show that maize stalk rot is associated with the fungal pathogens Fusarium subglutinans and F. temperatum. Since no fungicides are available to control these pathogens on maize plants, we selected six fungicides (tebuconazole, difenoconazole, fluquinconazole, azoxystrobin, prochloraz and kresoxim-methyl) and examined their effectiveness against the two pathogens. The in vitro antifungal effects of the six fungicides on mycelial growth and colony formation were investigated. Based on the inhibition of mycelial growth, the most toxic fungicide was tebuconazole with 50% effective concentrations ($EC_{50}$) of < $0.1{\mu}g/ml$ and $EC_{90}$ values of $0.9{\mu}g/ml$ for both pathogens, while the least toxic fungicide was azoxystrobin with $EC_{50}$ values of 0.7 and $0.5{\mu}g/ml$ for F. subglutinans and F. temperatum, respectively, and $EC_{90}$ values of > $3,000{\mu}g/ml$ for both pathogens. Based on the inhibition of colony formation by the two pathogens, kresoxim-methyl was the most toxic fungicide with complete inhibition of colony formation at concentrations of 0.1 and $0.01{\mu}g/ml$ for F. subglutinans and F. temperatum, respectively, whereas azoxystrobin was the least toxic fungicide with complete inhibition of colony formation at concentrations > $3,000{\mu}g/ml$ for both pathogens.

  • 자몽씨 추출물, 키토산, 인산용액을 콩나물 재배용수로 사용하여 콩나물 부패를 억제할 수 있는 재배방법을 검토하였다. 키토산과 인산용액은 콩나물의 생장에 특별한 저해를 일으키지 않으면서 재배수율을 증가시켰으며 특히 인산용액의 수율 증가와 부패억제 효과가 우수하였다. 인산용액의 관수회수가 증가함에 따라 대조구에 비해 12.3% 까지 수율이 증가하였으며 $4{\sim}8$회의 적은 관수회수에서도 높은 수율 증가와 뚜렷한 부패억제 효과를 확인하였다.

Fungicide selections for control of chili pepper stem rot caused by Sclerotium rolfsii using an agar dilution method

  • Sclerotium rolfsii causing southern blight on numerous vegetable and fruit crops was isolated from stems of chili peppers showing wilting symptoms. The pathogen was identified by morphological observation and DNA sequencing analysis of ITS region. To select an effective fungicide for control of southern blight, we investigated the inhibition efficacy of thirty fungicides included in nine groups of fungicides with different mechanisms of action. A fungal growth inhibition assay was conducted through an agar dilution method by using mycelial discs and sclerotia of the pathogen as inoculum, respectively. When mycelial discs were used as an inoculum, several fungicides showed good inhibitory activity against the mycelial growth of S. rolfsii 12-6. All DMI fungicides tested had a good inhibition except for prochloraz which had low inhibitory effect. All strobilurin fungicides tested except for kresoxim-methyl and all SDHI fungicides tested except for boscalid and fluopyram, had a good inhibition. Also, fludioxonil, a protective fungicide and fluazinam had a good inhibitory effect. Interestingly, when sclerotia were used as an inoculum, inhibition efficacy was increased for fluopyram, a SDHI fungicide, and for some protective fungicides such as propineb, chlorothalonil, dithianon, and folpet. All the fungicides selected in this study should be tested in the field for their control activities against stem rot for practical use in chili pepper cultivation.

The Effect of Fungicides on Mycelial Growth and Conidial Germination of the Ginseng Root Rot Fungus, Cylindrocarpon destructans

  • Ginseng root rot caused by Cylindrocarpon destructans is the most destructive disease of ginseng. Six different fungicides (thiophanate-methyl, benomyl, prochloraz, mancozeb, azoxystrobin, and iprodione) were selected to evaluate the inhibitory effect on the mycelial growth and conidial germination of C. destructans isolates. Benomyl and prochloraz were found to be the most effective fungicides in inhibiting mycelial growth of all tested isolates, showing 64.7% to 100% inhibition at a concentration of $10{\mu}g/mL$, whereas thiophanate-methyl was the least effective fungicide, showing less than 50% inhibition even at a higher concentration of $100{\mu}g/mL$. The tested fungicides exhibited less than 20% inhibition of conidium germination at concentrations of 0.01, 0.1, and $1{\mu}g/mL$. However, the inhibition effect of mancozeb on condium germination of C. destructans was significantly increased to 92% to 99% at a higher concentration of $100{\mu}g/mL$, while the others still showed no higher than 30% inhibition.

  • The disease development of root-rot [pathogen:Cylindrocarpon destruction (Zinssm.) Scholten] occurred in 2-year old ginseng (Panax ginseng C.A. Meyer) was investigated in the continuous (the first cultivation: 1978∼1982, 2nd cultivation: 1990∼1993 and abolished in 1993 due to replanting problem) and replanted cultivation (period of cultivation: 1980∼1984). In the continuous cultivation, incidences of root-rot were 0.7% on May 3, 48.6% on May 24, and 95.8% on June 14, respectively. In the replanted cultivation, no symptom was observed on May 3 and disease incidence was generally lower compared with the continuous cultivation. On the aerial part of the ginseng infected by C. desiccates, the end and/or margin of leaves were changed to dark reddish color that appeared for behind the root-rot symptom. In this field, the longitudinal growth of lateral root was more inhibited than in the case of the replanted cultivation by C. destmctans. The inhibition rate of rootlet growth was 37.3% in the continuous cultivation as compared with that of replanted cultivation at June 14. Though lesions of root-rot were formed all over the roots, 61.2% of the lesions was positioned within 6 cm under the rhizome. Effect of Rosemary Essential Oil and Trichoderma koningiopsis T-403 VOCs on Pathogenic Fungi Responsible for Ginseng Root Rot Disease

  • Rosemary essential oil was evaluated for antifungal potentiality against six major ginseng pathogens: Sclerotinia sclerotiorum, Sclerotinia nivalis, Cylindrocarpon destructans, Alternaria panax, Botrytis cinerea, and Fusarium oxysporum. The in vitro fungicidal effects of two commonly used fungicides, namely mancozeb and fenhexamid, and the volatile organic compounds (VOCs) of Trichoderma koningiopsis T-403 on the mycelial growth were investigated. The results showed that rosemary essential oil is active against all of the pathogenic strains of ginseng root rot, whereas rosemary oil displayed high ability to inhibit the Sclerotinia spp. growth. The highest sensitivity was S. nivalis, with complete inhibition of growth at 0.1% v/v of rosemary oil, followed by Alternaria panax, which exhibited 100% inhibition at 0.3% v/v of the oil. Minimum inhibitory concentrations (MICs) of rosemary oil ranged from 0.1 % to 0.5 % (v/v). Chemical analysis using GC-MS showed the presence of thirty-two constituents within rosemary oil from R. officinals L. Camphore type is the most frequent sesquiterpene in rosemary oil composition. Mancozeb and fenhexamid showed their highest inhibition effect (45% and 30%, respectively) against A. panax. T. koningiopsis T-403 showed its highest inhibition effect (84%) against C. destructans isolate. This study may expedite the application of antifungal natural substances from rosemary and Trichoderma in the prevention and control of phytopathogenic strains in ginseng root infections.

채소연부병균 Erwinia herbicola의 생육억제균 분리 및 특성 (Isolation and Characterization of Antagonistic Bacteria for Biocontrol of Erwinia herbicola Causing Vegetable Soft Rot)

  • For the selection of powerful antagonistic bacterium for biological control of Ewinia sp. causing vegetable soft rot, two excellent strains (54, 565) were selected from 1, 196 strains of bacteria which were isolated from rhizospere in vegetable root rot suppressive soil. Selected 2 strains were identified to be a species to Pseudomonas fluorescens S4 and pseudomonas fluorescens S65 (PS65). The highest of inhibitory activity was produced in 523 synthetic broth medium at pH 7.0 and 25t during 3 ethyl-Al-folpet, and the antibiotics such as vancomycin, perucillin and lincomycin, only PS4 was resistant to erythromycin.

Screening of Antagonistic Bacillus against Brown Rot in Dendrocalamus latiflorus and Preparation of Applying Bacterial Suspension

  • The aim of this study was to isolate biocontrol bacteria that could antagonize brown rot of Dendrocalamus latiflorus, optimize the culture conditions, and develop an effective biocontrol preparation for brown rot of D. latiflorus. This study isolated a bacterium with an antagonistic effect on bamboo brown rot from healthy D. latiflorus rhizosphere soil. Morphology, molecular biology, and physiological biochemistry methods identified it as Bacillus siamensis. The following culturing media and conditions improved the inhibition effect of B. siamensis: the best culturing media were 2% sucrose, 1.5% yeast extract, and 0.7% potassium chloride; the optimal culturing time, temperature, pH, and inoculation amount were 48 h, 30℃, 6, and 20%. The optimum formula of the applying bacterial suspension was 14% sodium dodecyl benzene sulfonate emulsifier, 4% Na2HPO4·2H2O, 0.3% hydroxypropyl methylcellulose thickener, and 20% B. siamensis. The pot experiment results showed the control effect of applying bacterial suspension, diluted 1,000 times is still better than that of 24% fenbuconazole suspension. The applying bacterial suspension enables reliable control of brown rot in D. latiflorus.

Effect on Colony Growth Inhibition of Soil-Borne Fungal Pathogens by Available Chlorine Content in Sodium Hypochlorite

  • Our study investigated the available chlorine content, contact time and difference among strains of each pathogen for sodium hypochlorite (NaOCl) to control chemically against soil-borne fungal pathogens, such as Phytophthora rot by Phytophthora cactorum, violet root rot by Helicobasidium mompa, and white root rot by Rosellinia necatrix, causing die-back symptom on apple trees. As a result, the colony growth of Phytophthora cactorum was inhibited completely by soaking over 5 s in 31.25 ml/l available chlorine content of NaOCl. Those of H. mompa and R. necatrix were inhibited entirely by soaking over 160 s in 62.5 and 125 ml/l available chlorine content in NaOCl, respectively. Also, inhibition effect on available chlorine in NaOCl among strains of each soil-borne pathogen showed no significant difference and was similar to or better than that of fungicides.

Effect of Cadium Ions on the Activity of Fungal Laccase and Its Decolorization of Dye, RBBR

  • The effect of cadmium ions on ligninolytic and decolourizing activities in cultures of two white-rot fungi, Cerrena unicolor and Trametes versicolor, were examined. Cadmium was added to the shallow stationary cultures growing on a liquid mineral medium. Both examined strains sorbed Cd ions in the first 24 hr of incubation. An appreciable stimulation of the activity of extracellular laccase (LAC) and inhibition of the extracellular manganese-dependent peroxidase (MnP) were simultaneously observed when 25 mgL-1 and 50 mgL-1 of cadmium ions were added to the cultures. On the other hand, the addition of cadmium ions also resulted in stimulating the decolorization activity of C. unicolor to decolorize Remazol Brilliant Blue R (RBBR) in the cultures, but decreasing it in the culture of T. versicolor, which is compared to the inhibition of MnP activity in this fungus. Our data indicate that the presence of Cd(II) ions can affect the ligninolytic activity of white-rot fungi. It was found that C. unicolor is a strain resistant to the presence of Cd ions in the liquid culture media, and has a potential to use this strain for bioremediation of sites contaminated with both heavy metals and aromatic pollutants.
