Isoquercitrin의 세포 보호 작용과 피부 흡수 증진을 위한 리포좀 제형 연구 (Cellular Protective Effect and Liposome Formulation for Enhanced Transdermal Delivery of Isoquercitrin)

  • 본 연구에서는 $H_2O_2$와 rose bengal로 처리된 HaCaT 세포에 있어서 isoquercitrin의 세포 보호 효과를 조사하였다. Isoquercitrin의 피부 전달시스템으로 에토좀 및 탄성 리포좀을 제조하고 입자크기, 포집효율 및 피부 흡수 증진 능력을 평가하였다. Isoquercitrin은 HaCaT 세포에 대해 50 ${\mu}M$의 농도에서 독성을 나타내지 않았다. 5 mM의 $H_2O_2$ 및 25 ${\mu}M$의 rose bengal로 HaCaT 세포를 처리하였을 때 isoquercitrin은 산화적 손상에 대항하여 농도 의존적(6.25 ~ 50 ${\mu}M$)으로 세포 보호 효과를 나타내었다. 0.03 % Isoquercitrin을 담지한 에토좀의 입자 크기는 222.85 nm, 포집효율은 82.26 %였다. 0.03 % isoquercitrin 함유 에토좀은 제조 후 2주일 동안 안정하였고, 일정한 입자 크기를 유지하였다. 피부 투과 실험 결과 에토좀은 일반 리포좀이나 에탄올 용액에서 보다 우수한 피부 투과능을 보여주었다. 0.1 % Isoquercitrin을 담지한 탄성 리포좀의 최적의 제형은 입자 크기(341.2 nm), 가변형성(59.89), 포집효율(54.3 %) 및 피부투과능 (초기 적하량 대비 54.4 %) 확인을 통해 인지질 대 계면활성제의 비율이 85 : 15인 제형이 가장 우수한 탄성 리포좀 제형임을 나타내었다.

  • $^{131}I$$^{125}I$를 함유(含有)하는 Hippuran, L-Thyroxine, Triiodothyronine, Rose Bengal, RISA, MAA, Triolein, Oleic acid 및 주사용(注射用) 옥소액(沃素液_의 합성방식(合成方式)과 $^{203}Hg$을 함유(含有)하는 Neohydrine의 합성방식(合成方式)을 각각(各各) 연구(硏究)하여 표지수율(標識收率) $100{\sim}60%$의 좋은 결과(結果)를 얻었다. 합성방식(合成方式)에서는 교환법(交換法), 옥화법(沃化法)을 사용(使用)하였고 특(特)히 Chloramin-T를 이용(利用)한 저온(低溫) 옥화반응(沃化反應)을 이용(利用)하였다. 합성품(合成品)의 제제법(製劑法) 및 pyrogen free 시험결과(試驗結果)를 기술(記述)하였으며 당연구소(當硏究所)에서의 제품분배(製品分配) 상황(狀況)을 보고(報告)하였다.

Serological prevalence of brucellosis of cattle in selected dairy farms in Bangladesh

  • This study was conducted to investigate the status of brucellosis in dairy cattle from five selected dairy farms in the Mohammadpur Beribadh area of Bangladesh. A cross-sectional study was carried out from October 2010 to March 2011 in which a total of 334 serum samples from cattle in five herds were screened by the Rose-Bengal plate-agglutination test (RBPT) and the positives were confirmed using an indirect enzyme-linked immunosorbent assay (I-ELISA). A structured questionnaire was used to collect epidemiological information describing the animals. Overall, 4.20% of the animals were RBPT positive, while subsequent confirmatory tests with I-ELISA revealed that the overall animal-level prevalence derived from the samples was 1.20%. Additionally, the prevalence was relatively higher in females than in males. A significant association was found between abortion, age of the animals, and the occurrence of brucellosis (p < 0.05). Considering the overall low prevalence of brucellosis in the selected farms in the present study, a brucellosis eradication program for dairy farms using a test-and-slaughter policy would be possible.

Differential Effects of herbicidal Compounds on Cytoplasmic Leakages of Green- and White-Maize Leaf Segments

  • Using maize green- and white-leaf tissue, we have examined the effect of various chemicals on cytoplasmic leakage with respect to the light requirement or chloroplast targeting for their activities. Oxyfluorfen, oxadiazon, diuron, and paraquat, which are known as representative herbicides acting on plant chloroplasts, caused the electrolyte leakage only in the green tissues, whereas 2, 4-dinitrophenol, rose bengal (singlet oxygen producing chemical) and methyl-jasmoante (senscence-stimulating chemical) play a role both in green- and white-tissue. Benzoyl(a) pyrene, generating superoxide radical upon light illumination, functions only in white tissues. Tralkoxydim, metsulfuron-methyl and norflurazon showed no effect in two tested plant samples. In terms of light requirement in electrolyte leakage activity, diuron, oxyfluorfen, oxadiazon, rose bengal, and benzoyl(a) pyrene absolutely require the light for their functions, but other chemicals did not. based on these results, we could classify into four different response types according to whether chemicals require light or chlroplasts for their action. This classification is likely to be applied to simply and rapidly identify the requirement of light and chlroplasts for the actions of chemicals, thereby it makes easy to characterize many new herbicides that their action mechanisms are unclear, and to elucidate the mode of action of them.

마이크로 에멀젼 계면을 통과하는 광유발전자의 양자수득률 (Quantum Yield of Photoinduced Electron Transfer Across Microemulsion Interfaces)

  • 태양에너지의 한 저장방법으로 녹색식물의 광합성을 모방한 계, 마이크로 에멀젼(microemulsion)을 고안하였으며, 이때 광에 의해서 유발된 전자가 계면을 통하는 능력을 측정하였다. 광증감제로 류테니늄비피리딘 착물$[Ru(bipy)_3]^{2+}$을 사용, 전자공여체 EDTA와 함께 물층에, 전자수용체 $HV^{2+}$(Hexadecyl violagan)이 계면에 각각 존재할 때 광에 의한 전자전이에 따르는 $HV^+$ 형성수득률은 0.12이였다. 또 계면에 $BNA^+$(Benzyl nicotinamide)를 넣고 유층에 아조(azo) 화합물을 넣었을 때는 azobenzene이 환원되었는데, 이때 양자수득률이 줄었다. (${\Phi}$ = 0.0016) 양이온 마이크로 에멀젼과 음이온 마이크로에멀젼의 광유발 전자전이 능력을 비교하였다. 광증감제로 유기염료인 로즈벤갈(Rose bengal)을 시험하였는데, 류테니늄착물보다 낮지 않았지만 광유발 전자가 계면에 전이되는 것을 알았다.

소 브루셀라병의 혈청학적 진단법 비교실험 (Comparison of a new ELISA with other serodiagnostic tests for bovine brucellosis)

  • A novel enzyme linked immunosorbent assay (ELISA) is described and compared with other established serologic tests for bovine brucellosis, namely the rose bengal test (RBT), the complement fixation test (CFT), and the tube agglutination test (TAT) approved and used in Korea. A total of 109 bovine serum samples were tested using all the 4 assays and analyzed as to specificity, sensitivity, reproducibility and predictive value. The ELISA showed 100% agreement with the CFT. The least agreement between ELISA was observed with the TAT. The agreement between the ELISA and RBT was not significantly different from that observed between the CFT and the ELISA. It is concluded that the new assay would be a good candidate for routine serologic survey for brucellosis in Korea. A protocol combining the ELISA and the CFT would increase the power for detection of serologically positive individuals and herds.

Seed and Root Rots of Ginseng (Panax quinquefolius L) Caused by Cylindrocarpon destructans and Fusarium spp.

  • Ginseng (Panax quinquefolius L.) has become one of the most valuable herb crops grown in North America. However, traditional cropping practices are favourable to disease and significant losses due to root disease are common, despite frequent use of fungicides. Seedlots are often contaminated with pathogens, however, little is known about the causes of seed decay and the role of seed pathogens as incitants of root rots. It was shown that both Fusarium spp. and Cylindrocarpon destructans were able to rot seeds and that C. destructans was more virulent than Fusarium spp. on seedling roots. A modified rose bengal agar MRBA) medium (1 g KH$_2$PO$_4$; 0.5 g MgSO$_4$; 50 mg rose bengal; 10 g dextrose; 5 g Bacto peptone; 15 g Bacto agar; 30 mg streptomycin sulfate; 250 mg ampicillin; 10 mg rifampicin; 500mg pentachloronitrobenzene; 500 mg dicloran; and 1 L distilled water) was superior to potato dextrose agar in detecting C. destuctans in diseased roots. Isolation of C. destructans from diseased seedlings arising from seeds sown in replant soil supported the hypothesis that this pathogen is a cause of ginseng replant failure in North America.

  • Co-sensitized photooxygenation of 1,1-diphenyl-2-vinylcyclopropane (VCP-DPh) with 9,10-dicyanoanthracene and biphenyl in oxygen-saturated acetonitrile solution produced 3,3-diphenyl-5-vinyl-1,2-dioxolane as the major product. The same photoproduct was obtained by acetone sensitized photooxygenation in oxygen-saturated acetone solution. However, VCP-DPh remained intact when directly irradiated with DCA or irradiated with Rose Bengal to generate singlet oxygen. A mechanism involving a cosensitizer radical cation and sensitizer radical anion is proposed.

  • Copidognathus bengalensis n. sp. is described from Visakhapatnam coast, Bay of Bengal, India. The present species is characterized by posterodorsal plate (PD) with four costae made up of po rose panels, subdivided posterior cornea of ocular plate (OC), and two pairs of basirostral setae in female. This species is related to C. pulcher group, but the nature of po rose panels on anterior areolae of anterior dorsal plate (AD) and the setal ornamentation of legs were different between the members of C. pulcher group and the present new species.

국내 가축에서 브루셀라병에 대한 혈청학적 모니터링 (Serological monitoring on brucellosis in livestock of Korea)

  • In Korea, brucellosis has been reported periodically in cattle and rarely in dogs; however, it has not previously been screened in domestic animals such as elk, pigs and goats. To investigate the serological prevalence, serum samples were taken from the aforementioned animals annually during 2007-2013 and screened by the rose-bengal test (RBT) or modified RBT, after which positive sera were evaluated by the standard tube agglutination test (STAT). Finally, RBT and STAT-positive sera were confirmed by competitive-ELISA. Brucella abortus biovar 1 was isolated from three elk that were shown to be positive serologically in 2008. There was no evidence of brucellosis in pigs. Based on serological monitoring and investigation of etiological agents, there is no evidence of outbreak of brucellosis in elk, pigs or goats of Korea since 2008. However, the possibility for brucellosis from cattle to affect these other livestock exists; therefore, extensive and continuous serological monitoring is required to maintain their brucellosis-free status.
