Total soluble proteins from three developmental stages of induced root nodules of Canavalia lineata were compared with those of non-nodulated roots by SDS-PAGE and two dimensional (2-D) gel electrophoresis. Thirteen nodule-specific protein (nodulin) bands were identified by the former and 30 nodule specific protein spots were detected by the latter method respectively. Some of the nodulins were detected differentially depending on the nodule's developmental stages. For example, only three leghemoglobin (Lb)-like protein spots appeared at stage I (d<2 mm), but two additional Lb-like protein spots appeared at stage II (d <4-5 mm). pI value and molecular weight of nomomers of Lb-like protein were narrower and greater than those of soybean, ranging from 4.4 to 5.0 and 15.7 kd respectively. Northern blot hybridization of total RNAs from roots and root nodules using soybean Lb cDNA as a probe made it clear that Lb gene was expressed tissue-specifically only in the root nodules.odules.
To research the effect of chemotaxis of Rhizobia toward the root exudate on nitrogen fixing ability in soybean Rhizobia symbiosis system. Root exudate from seedlings of Glycine max. L was collected aseptic conditions. B. japonicum KCTC 2422 induced the formation of symbiotic nitrogen fixing nodules on the root of soybean plant and possessed motility and chemotaxis toward the 2mM proline. LPN-100 mutant was $Nod^-$, $Che^+$, and LPN-101 was $Che^-$, $Nod^+$ strains. Physiological properties of mutants were similar to parent strain. The crude root exudate was tested for its chemotactic ability using the capillary tube method. Chemotactic responses of RCR 3407 toward crude root exudate were 2.2, 2.6, 2.9, those of KCTC 2422 were 2.3, 2.9, 3.0, respectively. The crude root exudate was fractionated into neutral, cationic and anionic fractions. Chemotactic responses of KCTC 2422 was least with anionic fraction, most with neutral and intermediate with cationic fraction. B. japonicum KCTC 2422 was attracted by carbohydrates, amino acids and carboxylic acid. Carbohydrates and amino acids were good chemoattractants and carboxylic acids were intermediate chemoattractants. The peak concentration was $10^{-3}M$ for ribose, glucose, glutamine, aspartic acid and carboxylic acids, with exception of xylose, arabinose, tryptophan, which elicited maximum responses at $10^{-4}M$. The formation of nodules and nitrogenase activity of soybean inoculated with KCTC 2422 was determined in 7days after inoculation, and those of LPN-101 was detected in 15days after inoculation, but LPN-100 didn't form of nodules in soybean plants.
Pygmaeopremna herbacea (Roxb.) Mold. (Verbenaceae) is a small herb or sometimes an undershrub arising from a perennial rootstock. The dry roots are dark muddy brown in colour having root nodules. Its extensively developed roots are widely used in tribal medicine. They are used as an aphrodisiac and for the treatment of gout, rheumatism and ulcers. This study deals with the detailed pharmacognostical evaluation of the dried roots and root nodules of P. herbacea which includes macro and microscopic studies, determination of physicochemical parameters and chemoprofiling of the extract using HPTLC fingerprint profiles. It was observed that the roots consist of a well developed cortical region consisting of tangentially elongated thin walled parenchymatous cells and contain polygonal stone cells as well as compound starch grains. Also the pith was parenchymatous. The HPTLC fingerprint profile of the methanolic extract showed the presence of seven major bands. Such an analysis may thus be utilized in identifying P. herbacea and in differenciating it from other species which are similar to it or are used as its adultrants/substitutes under the same vernacular name of Bharangi.
The responses of six mungbean [Vigna radiata (L.) Wilczek] varieties to Rhizobium inoculation on number and dry weight of nodules, nitrogenase activity of root nodule bacteria, dry weight of shoot and root, nitrogen content, and uptake by shoot were investigated. The mungbean varieties were BARI Mung-2, BARI Mung-3, BARI Mung-4, BARI Mung-5, BINA Moog-2, and BU Mung-1. Two-third seeds of each variety were inoculated with Rhizobium inoculant and the remaining one-third seeds were kept uninoculated. Rhizobium strains TAL 169 and TAL 441 were used for inoculation of seeds. Inoculation of seeds with Rhizobium strains significantly increased nodulation, nitrogenases activity, dry matter production, nitrogen content, and uptake by shoot of the crop compared to uninoculated control. There was positive correlation among the number and dry weight of nodules, nitrogenase activity, dry weight of shoot and root, nitrogen content, and uptake by shoot of the crop. It was concluded that BARI Mung-4 in association with Rhizobium strain TAL 169 performed best in recording nodulation, nitrogenase activity, dry matter production, and nitrogen uptake by shoot of mungbean.
Legumes are important agricultural and commercial crops characterized by root nodules formed as a result of the symbiotic relationship with nitrogen-fixing rhizobia. Due to the economic significance the chemistry of some of these species has been well recognized. Soybeans and processed soy products, which contain isoflavonoids and saponins, are of wide interest for their multifacted biological effects. (omitted)
Dry matter production, leaf area growth and total nitrogen changes were studied in Glycine max. soybean communities, which were grown in sandy loam soils controlled to provide various moisture levels, i.e., 5-7%(level 1), 8-10%(level 2), 11-13%(level 3), 14-15%(lev디 4), 17-20%(level 5) and 22-24%(level 6). A summary of the results is shown. The maximum dry matter production of leaves, stems and nodules and the maximum leaf area per unit area were at level 5, but the maximum of root dry matter production was at level 4. Total nitrogen content of the soybean plant decreased with growth, but each level of soil moisture content also showed a little difference. Water content of the plant decreased with plant age and soil water deficiency, especially in roots and nodules. Nodule formation increased in proportion to soil moisture content. total nitrogen content of the soil on which the soybeans grew, increased from 0.23% before sowing to 0.30% at 100 days after sowing. It seems that soil water content acts as a linear factor in the elongation or dry weight increase of shoots and roots until increasing to level 5. Considering the pattern of plant growth through analysis of the shoot and root dry weight ratio, or the photosynthetic organ and non-photosynthetic organ dry weight ratio, the asymptote of plant growth at a high soil water content exceeded that at a low soil water content.
Earliness of the nodule formation was significantly different among soybean varieties. The numbers of the nodules were much various due to the origins of the soybean varieties and the date of observation. The significant positive correlations were shown between the weight of nodule and soybean yield and also between the weight of nodules and number of nodules but the correlations among the other characters were not significant.
The activity of symbiotic -fixation and environmental factors of Robinia pseudo-acacia L., bearing root nodules, were quantitatively analyzed during the growing period. Among changes of total nitrogen and phosphorus contents of each organ, leaves showed prominent decreases from the highest quantity of the early growing period to the lowest of the late period. The rhizosphere showed acidic pH and low level of nitrogen, phosphorus and organic matter contents during the growing period. -fixation activity of nodules initiated from April and showed the maximum value of 190 $\mu$/g DW/hr in late June and than decreased to 50$\mu$M/g DW/hr during the rainy and dry season. Legume plants develop root nodules to recruit nitrogen-fixing bacteria called rhizobia. This symbiotic relationship allows the host plants to grow even under nitrogen limiting environment. Since nodule development is an energetically expensive process, the number of nodules should be tightly controlled by the host plants. For this purpose, legume plants utilize a long-distance signaling known as autoregulation of nodulation (AON). AON signaling in legumes has been extensively studied over decades but the underlying molecular mechanism had been largely unclear until recently. With the advent of the model legumes, L. japonicus and M. truncatula, we have been seeing a great progress including isolation of the AON-associated receptor kinase. Here, we summarize recent studies on AON and discuss an updated view of the long-distance control of nodulation.
