Effect of Nutrient Concentrations and Leaching Percentage on Growth and Nutrient Uptake by Perilla Frutesens Britton var. Japonica Hara in Plug Culture (잎들깨의 플러그육모에서 용탈률 및 시비농도가 생육 및 무기원소 흡수에 미치는 영향)

  • Effect of Nutrient Concentrations, fertigation frequency, and learching percentage on crop growth and nutrient concentrations in root media were evaluated. The treatment of each irrigation with $50 mg.L^{-1}$ of nitrogen in stage 2 and increase to $80 mg.L^{-1}$ nitrogen in stage 3 had the highest crop growth at 34 days after sowing among treatments tested. Feeding with low nutrient concentrations and elevated frequency decreased crop growth. In treatments of each leaching percentage, feeding with low nutrient concentrations and elevated frequency resulted in increased tissue nutrient contents. The less tissue potassium content and higher calcium and magnesium contents were observed in treatment of 50% leach than those in 0% leach. All treatment tested had soil solution pH higher than 6.8. Electrical conductivity in treatments of 50% leach were lower than those of 0% leach. Feeding with low nutrient concentrations and elevated feeding frequency in each leaching percentage resulted in increased electrical conductivity in soil solution of root media. Trends of medium nutrient concentrations were similar to those of electrical conductivity.

Growth of Zoysiagrass and Seashore Paspalum on Volcano Eruption Sand and Clayey Soil with Organic and Inorganic Fertilizers in Indonesia

  • This study was carried out to determine the possible use of volcano eruption sand at Merapi mountain area compared with clayey soil, alfisol as turfgrass growing media. Indonesia has abundant source of eruption sand and clayey soil. Native zoysiagrass (Z. matrella) was collected from Sleman district and seashore paspalum (P. vaginatum) from Solo region. The experimental plots were treated with both organic and inorganic fertilizers. Zoysiagrass and seashore paspalum can grow on the mixture of eruption volcano sand and alfisol soil. Fertilizers application increased turf color, surface coverage rate and recovery rate of turfgrass. While fertilizer treated alfisol have no effect on the length and dry weight of zoysiagrass root. Mixing of volcano eruption sand on growing media resulted in longer root length, higher shoot dry weight of zoysiagrass than in alfisol soil. Type of fertilizer affected the pH, soil organic matter, total N, available P, available K and electrical conductivity of sand and alfisol after cultivation. Similarly to zoysiagrass, the recovery of seashore paspalum was increased by application of fertilizer, even the inorganic fertilizer showed faster recovery than organic fertilizer. Surface coverage of seashore paspalum was faster than that of zoysiagrass with or without fertilizer.

Chaos-Theoretical Approach to Media Convergence on Internet Media (인터넷미디어 중심의 매체융합 현상에 대한 카오스 이론적 접근)

  • Media convergence is prevalent all over the world, and is expected to change our lives dramatically in near future. Chaos theory is a new emerging theoretical approach to various social phenomena including organization management, economic system, stock market fluctuation, and so on. This thesis is intended to explain media convergence phenomena centered on internet media using basic concepts of chaos theory such as self-organization, co-evolution, and feedback loop. The implications of this tentative study are that we can apply chaos theory to media convergence as other social or natural phenomena, and that maintaining chaos state of media system is useful for accelerating creativities, and that developing media system as an organic system to cooperate with other relevant systems is helpful for promoting life qualities, and that establishing moral order or root value is desirable for implementing communicatopia.

Rooting Promotion in Cutting Propagation of Tea (다(茶)나무 (Camellia sinensis L.) 삽목(揷木)시 발근솔(發根率) 향상(向上))

  • These experiments were conducted to promote the rooting in tea cutting bymeans of several root media, growth regulators, cutting material and cutting condition The results are summarized as follows. The rate of rooting was higher in the softwood than in the hardwood, and the best cutting sea­son was about August 10 in the hardwood, about April 10 in the softwood. The most suitable root medium was the Masato at which the rate of rooting was 71%, 87% each in the softwood and in the hardwood. When growth regulators, such as oxyberon, rootone were sprayed upon the hardwood, the rate of rooting was promoted to 4 - 9 % better than that of no treatment. But such a good effect was not recognized significantly in the softwood.

  • Background: This study aims to describe the characterization of Paenibacillus polymyxa GBR-1 (GBR-1) with respect to its positive and negative effects on plants. Methods: The morphological characteristics of GBR-1 were identified with microscopy, and subjected to Biolog analysis for identification. Bacterial population and media optimization were determined by a growth curve. The potential for GBR-1 as a growth promoting agent, to have antagonistic activity, and to have hydrolytic activity at different temperatures was assessed. The coinoculation of GBR-1 with other microorganisms and its pathogenicity on various stored plants, including ginseng, were assessed. Results: Colony morphology, endospore-bearing cells, and cell division of GBR-1 were identified by microscopy; identification was performed by utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME). GBR-1 showed the strongest antagonistic activity against fungal and bacterial pathogens. GBR-1 cell numbers were relatively higher when the cells were cultured in brain heart infusion (BHI) medium when compared with other media. Furthermore, the starch-hydrolytic activity was influenced by GBR-1 at higher temperature compared to low temperatures. GBR-1 was pathogenic to some of the storage plants. Coinoculation of GBR-1 with other pathogens causes differences in rotting on ginseng roots. A significant growth promotion was observed in tobacco seedlings treated with GBR-1 suspensions under in vitro conditions, suggesting that its volatile organic compounds (VOCs) might play a role in growth promotion. Conclusion: The results of this study indicate that GBR-1 has both positive and negative effects on ginseng root and other stored plants as a potential biocontrol agent and eliciting in vitro growth promotion.

Effect of Dolomite Levels in Various Root Media Containing Micronutrient Mixes on Growth of Marigold Plug Seedlings (미량원소복합제가 혼합된 각종 상토에서 고토석회의 시비수준이 매리골드 플러그 묘의 생육에 미치는 영향)

  • Objective of this research was to determine the effect of application rate of dolomite in root media containing micronutrient mixes on growth of marigold 'Orange boy' in plug trays. To achieve this, three granular micronutrient mixes such as HF 1, MF 2, and MF 3 were produced and incorporated into three root media, peatmoss+composted pine bark (1:1, v/v, PB), peatmoss+composted rice-hull (1:1, v/v, PR), and peatmoss+composted saw-dust (1:1, v/v, PS), with a rate of $0.3g{\cdot}L^{-1}$. Elevated application rate of dolomite in PB medium containing MF 1 decreased crop growth at 35 days after sowing with the highest fresh and dry weight in the control treatment of dolomite. The treatments of 3.0 and $6.0g{\cdot}L^{-1}$ of dolomite in PB medium containing MF 2 produced 0.133g of dry weight and 0.686g of fresh weight, respectively, which were the highest among treatments of dolomite. But the treatments of 3.0 and $6.0g{\cdot}L^{-1}$ of dolomite in PB medium containing MF 3 produced the highest fresh and dry weight, respectively. Elevated application rate of dolomite in PR medium containing MF 1, 2, and 3 resulted in decrease of fresh an dry weight. The relative growth suppression induced by elevated application rate of dolomite was more severe when MF 3 were incorporated than MF 1 and 2. The treatments of 3.0 and $6.0g{\cdot}L^{-1}$ of dolomite in PS medium containing MF 1 and 3 showed good growth, but elevated application rate of dolomite in PS medium containing MF 2 and control resulted in increase of dry and fresh weight. These results indicated that the application rate of dolomite should be adjusted by kinds of root media and micronutrient mixes.

Growth and Mineral Contents of Spinach (Spinacia oleracea L.) and Radish (Raphanus sativus L.) as Related with a Low Dose Gamma Irradiation

  • This study was to evaluate the effects of gamma irradiation on the germination, nutrient concentrations and growth of spinach and radish. Both the spinach and radish seeds exhibited relatively higher germination rates in response to the low doses of gamma irradiation compared to the non -irradiated control. Leaf DW of the radish did not respond to gamma irradiation but that of the spinach increased significantly in response to a gamma radiation of 4 Gy (P< 0.05). Leaf growth parameters of the spinach including the leaf area and SLA (leaf area/leaf dry weight) also demonstrated increased responses to gamma irradiation. R/S (root dry weight/shoot dry weight), root DW and root length of the spinach exhibited a positive response to gamma irradiation while those of the radish did not. In contrast, SRL (root length/root dry weight) significantly decreased with gamma irradiation at 8 Gy for the spinach, but not for the radish. The tissue nitrogen concentrations of the spinach showed an increased response to gamma irradiation while that of the radish did not. Furthermore, higher concentrations of phosphorus, potassium, calcium and magnesium were found in the irradiated spinach, but not in the irradiated radish. It seems that the non-specific physiological and/or biochemical activities of spinach might be accelerated by gamma irradiation, possibly accounting for the stimulation of nutrient uptake from the root media and early biomass accumulation in the current study.

A Study on the Effects of Ramulus et Uncus Uncariae (REUU) on the Cultured Spinal Dorsal Root Ganglion Neurons Damaged by Oxygen Free Radicals (조구등(釣鉤藤)이 산소자유기(酸素自由基)에 의하여 손상(損傷)된 배영척수감각신경절세포(培養脊髓感覺神經節細胞)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • To study the effects of Ramulus et Uncus Uncariae (REUU) on oxygen free radical-mediated damage by hydrogen peroxide $(H_{2}O_{2})$ on cultured spinal sensory neurons, in vitro assays such as MTT assay, NR assay, neurofilament enzymeimmuno assay (EIA), sulforhodamine B (SRB) assay, assay for lactate dehydrogenase (LDH) activity and assay for lipid peroxidation were used in cultured spinal dorsal root ganglion neurons derived from mice, Spinal dorsal root ganglion neurons were cultured in media containing various concentrations of $H_{2}O_{2}$ for 5 hours, after which the neurotoxic effect of $H_{2}O_{2}$ was measured by in vitro assay. The protective effect of the herb extract, Ramulus et Uncus Uncariae (REUU) against H2O2-induced neurotoxicity was also examined. The results are as follows. 1. In NR assay and MTT assay, $H_{2}O_{2}$ significantly decreased the cell viability of cultured mouse spinal dorsal root ganglion neurons according to exposure concentration in these cultures. An additional time course study was done on these cultures. 2. Cultured spinal dorsal root ganglion neurons which were exposed to various concentrations of $H_{2}O_{2}$ showed a quantitative decrease of neuronal cells by EIA and of total protein by sulforhodamine B (SRB) assay, while they showed an increase of both lipid peroxidation and LDH activity. 3. The effect of Ramulus et Uncus Uncariae (REUU) on $H_{2}O_{2}$ induced neurotoxicity showed a quantitative increase in both neurofilament and total protein, but showed a decrease of lipid peroxidation and LDH activity. These results suggest that $H_{2}O_{2}$ has a neurotoxic effect on cultured spinal dorsal root ganglion neurons from mice and that the herb extract, Ramulus et Uncus Uncariae (REUU), was very effective in protecting $H_{2}O_{2}$ induced neurotoxicity by decreasing lipid peroxidation and LDH activity.

Plant Regeneration and Somatic Embryo Formation from Root-Derived Callus of Rice (벼 뿌리조직 유래의 캘러스로부터 체세포배 형성과 식물체 재분화)

  • The competence of callus formation and plant regeneration from root derived callus was higher in japonica cultivars than those of Tongil-type cultivars of rice. A japonica type cultivars Yeongdeogbyeo, showed the highest capacity (13%) for plant regeneration from root calli of 6 cultivars tested. The callus induced from seed and root tissues maintained higher capacity for plant regeneration during 7 passages of subculture on N$_{6}$ solid media at 2-week intervals. The maximum frequency (2 x 10$^{5}$ mL) of round cells and their cell colonies showed about 24 days after suspension culture of root-derived callus in N$_{6}$ medium with lmg/L 2,4-D, 300mg/L casein hydrolysate, 10mM L-proline, 20g/L sucrose and 30g/L sorbitol. The frequency of somatic embryo formation in suspension cultures of root-derived callus increased with prolonged advance of subculture time from 30 to 90 days, but their regenerative capacities decreased.

COMPARISON OF THE SEALING ABILITY OF VARIOUS RETROGRADE FILLING MAIERIALS (수종의 역충전 재료의 치근단 밀폐력 비교)

  • This study was performed to evaluate the sealing ability of various retrograde filling materials by using bacterial penetration and dye penetration test. One hundred and forty extracted human teeth with single, straight canals and mature apiece were collected and used for this study. All canals were instrumented using an engine driven Ni-Ti file (ProFile). After removing 3mm from the apex of tooth, a standardized 3mm root end cavity was prepared using an ultrasonic instrument. The 70 teeth were randomly divided into 7 groups : 6 groups for retrograde filling using Super-EBA, ZOE, Chelon-Silver, IRM, ZPC and amalgam. The 7th group was used as a negative control. Nail varnish was applied to all external root surfaces to the level of the reseated root ends to prevent lateral microleakages. The specimens were then sterilized in an ethylene oxide sterilizer for 24 hours. 2 mm of the reseated root was immersed in a culture chamber containing a Tripticase Soy Broth with a phenol red indicator. The coronal access of each specimen was inoculated every 72 hours with suspension of Proteus vulgaris. The culture media were observed every 24hours for color change indicating bacterial contamination. The specimens were observed for 4weeks. The remaining 70 teeth were submitted to a dye penetration test. The canals of all teeth were first sealed with AH26 and obturated using an Obtura II system. Root resection, root end preparation and retrograde filling was performed as above. All specimens were suspended in 2% methylene blue dye for 72 hours before being ion gitudinally split. The degree of dye penetration was then measured using a stereomicroscope at 10 magnification and evaluated. The results were as floows : 1. In the bacterial penetration, the degree of leakage was the lowest in the Super-EBA, followed by, in ascending order, ZOE, Chelon-Silver IRM and ZPC. The amalgam showed highest bacterial leakage of all(p<0.01). 2. In the dye penetration, the degree of microleakage was the lowest in the Chelon-Silver and Super-EBA, followed by, in ascending order, IRM, ZPC. The ZOE and amalgam showed the highest microleakage of all (p<0.05). These results suggested that the eugenol based cement, Super-EBA, have excellent sealing ability as a retrograde filling material.
