This study was carried out to screen 11 peony varieties collected for resistance to northern root-knot nematode, Meloidogyne hapla. The larval population of Meloidogyne halpa to the Uisongjakyak, Taebaekjakyak, Doseo, EP368, EP512 peony were detected high denisties with individuals of 98-173 per 300 ml soil, but Sabeol, Yongsu, Kimchon, Pl23 peony showed low number of larvar with 21-60. The root rot were lowered in Yongsu, Kimchon peony than Uisongjakyak, Taebaekjakyak, EP368, EP512. It seems to involved with densities of 2nd juvernile population in that rot rate of root was increased as increase of 2nd stage larvae. Two peony, Uisongjakyak, Taebaekjakyak were susceptable to M. hapla with high egg mass formation which showed 49,28 in each root. And Yongsu, Doseo, EP368, EP512 were Mid resistance with 1-15 egg mass of each root. Whereas Sabeol, Kimchon, Pl23, EP337, EP425 were found to be resistant to M, hapla with few egg mass formation of 4-7 each root.

A lot of SSRs (simple sequence repeats) in peach and pear from enriched genomic libraries and in peach from a cDNA library were developed. These SSRs were applied to other related species, giving phenograms of 52 Prunus and 60 pear accessions. Apple SSRs could also be successfully used in Pyrus spp. Thirteen morphological traits were characterized on the basis of the linkage map obtained from an $F_2$ population of peach. This map was compiled with those morphological markers and 83 DNA markers, including SSR markers used as anchor loci, to compare different peach maps. Molecular markers tightly linked to new root-knot nematode resistance genes were also found. A linkage map including disease-related genes, pear scab resistance and black spot susceptibility, in the Japanese pear Kinchaku were constructed using 118 RAPD markers. Another linkage map, of the European pear Bartlett, was also constructed with 226 markers, including 49 SSRs from pear, apple, peach and chewy. Maps of other Japanese pear cultivars, i.e., Kousui and Housui, were also constructed. These maps were the first results of pear species.

Effects of the root-knot nematode, Meloidogyne hapla, on the growth and yields of hot-pepper (var.: Cheongyong gochu) and tomato$(var.:\;Bogsu\;\#)$ were studied with inoculating 500, 2,500, 5,000, 7,500, 10,000 and 20,000 nematodes per plant in pots. Results were analyzed with comparing weight of fruits harvested throughout the season from both hot-pepper and tomato, and with mineral contents in dried stoots and roots of pepper plant 20 weeks after the inoculation. No significant difference was found on the plant growth at all levels of inoculation until the 8th week after the inoculation. However, the plant growth was significantly depressed from the 12th week in the pots inoculated with over 10,000 nematodes, and the yields were reduced by $16\%$ in hot-pepper and $14\%$ in tomato respectively when 10,000 nematodes were inoculated.

A preliminary survey on the incidence and intensity of root disease complex (association of Meloidogyne incognita and root rot pathogens) was carried out in the sericultural areas of Karnataka. A total of 280 mulberry gardens were surveyed in 14 districts of Karnataka belonging to different types of soil (red sandy, red loamy and black cotton), farming systems (irrigated and rainfed), varieties (V-1, K-2, Local and S-13) and age of the plants (0-5, 5-10 and 10-15 years). It was observed that the association of M. incognita with Botryodiplodia theobromae and Fusarium solani causes the root disease complex in mulberry. Of the 280 gardens visited, 94 were infested with the disease complex and incidence was recorded as 33.6%. The higher intensity of root disease complex was observed when the root system had more than 100 galls/plant with infection of mixed population of B. theobromae and F. solani in sandy soil under irrigated farming. The 5-10 years old mulberry plantation with V-I variety was found to be most susceptible to root disease complex. Districts like Mysore, Kolar, Mandya, Tumkur, Chitradurga and Bangalore were observed as sensitive areas. Further, the wounds caused by M. incognita in mulberry roots favour the easy entry of root rot pathogens, which increased the severity of the disease very fast.

Southern root-knot nematode (SRKN, Meloidogyne incognita) is known to be responsible for annual economic losses of 38.2 million USD in cucumber cultivation in Korea. Nematicides are frequently used to manage SRKN in plastic greenhouses. A field experiment was conducted to assess the effect of a soil fumigant (dimethyl disulfide, DMDS) and a non-fumigant (fluopyram, FL) against SRKN during the winter fallow season in a greenhouse from October to December. Nematicidal efficacy was assessed at 43 days after treatment, and the root gall index was measured 4 months after transplanting cucumber seedlings. DMDS effectively reduced second-stage SRKN juvenile density with 91% control efficacy, while FL showed no control efficacy against SRKN. Root gall index values were significantly different(p=0.020) in DMDS treatment and controls at 1.0±0.00 and 4.3±0.58, respectively. This study showed that DMDS could be considered an effective nematicide for controlling SRKN in the fallow period in winter.

Root-knot nematodes (Meloidogyne spp.) are parasites that attack many field crops and orchard trees, and affect both the quantity and quality of the products. A root-colonizing bacterium, Pseudomonas chlororaphis O6, possesses beneficial traits including strong nematicidal activity. To determine the molecular mechanisms involved in the nematicidal activity of P. chlororaphis O6, we constructed two mutants; one lacking hydrogen cyanide production, and a second lacking an insecticidal toxin, FitD. Root drenching with wild-type P. chlororaphis O6 cells caused juvenile mortality in vitro and in planta. Efficacy was not altered in the fitD mutant compared to the wild-type but was reduced in both bioassays for the mutant lacking hydrogen cyanide production. The reduced number of galls on tomato plants caused by the wild-type strain was comparable to that of a standard chemical nematicide. These findings suggest that hydrogen cyanide-producing root colonizers, such as P. chlororaphis O6, could be formulated as "green" nematicides that are compatible with many crops and offer agricultural sustainability.

A neem based product, Suneem-G (granules) alone as well as in combination with urea was tested against root-knot nematode, Meloidogyne incognita, on okra (Abelmoschus esculentum) in clay pots of 20-cm-diameter, containing 2 kg sterilized soil, inoculated with 2,000 freshly hatched second stage juveniles (J$_2$) of M. incognita. Application of different dosages of Suneem-G with or without urea significantly reduced nematode population and incidence of galling on okra roots and increased vegetative plant growth compared with the untreated control. Suneem-G with urea was the most effective and its effect increased with the increase of dosage. Suneem-G 6 g+urea 25 g per pot gave maximum plant growth and root gall reduction, as plants shoot and root lengths were increased by 84 and 58% and root galling was reduced by 58%, respectively. The lowest dosage of Suneem-G alone, 2 g per pot increased shoot and root lengths of plants by 25.7 and 17%, respectively, and reduced root galling by 24%. The lowest effect was recorded in the pots treated with 25 g urea alone; shoot and root lengths were increased and root galling was decreased by 14% each.
One hundred seventy five red pepper varieties were bioassayed for selecting resistant varieties to northern rootknot nematode, Meloidogyne hapla. Fifteen native varieties (IT 102794, 104806, 105516, etc) and two imported varieties were proved to be resistant while the currently cultivated varieties such as Hongtap, Kangsan, Hongsil, and Bookang were moderately resistant to the nematodes. Resistant varieties resulted in less nematode infection and development than did the susceptibles. Roots of the resistant strains had significantly higher esterase and peroxidase activities than did those of the susceptibles.
Poria cocos, a famous traditional Chinese medicine, was found to have nematicidal activity in experiments searching for nematicidal fungi. The experiment showed it could kill 94.9% of the saprophytic nematode, Panagrellus redivivue, 92.6% of the root-knot nematode, Meloidogyne arenaria, and 93.5% of the pine nematode, Bursaphelenchus xylophilus, on PDA plate within 12 hours. According to the nematicidal activity, three new compounds, 2, 4, 6-triacetylenic octane diacid, 2, 4, 5, 6-tetrahydroxyhexanoic acid and 3, 4-dihydroxy-2-keto-n-butyl 2,4,5,6-tetrahydroxyhexanate, were isolated from submerged cultures of Poria cocos. Of these, 2, 4, 6-triacetylenic octane diacid could kill 83.9% Meloidogyne arenaria and 73.4% Panagrellus redivivus at 500 ppm within 12 hours. Here, it is reported for the first time that Poria cocos has nematicidal activity.
