Fermentation characteristic of Yeongdeok Bobsikhae to which a natural substance (Bellflower) was added (길경을 첨가한 영덕밥식해의 발효 특성)

  • Bobsikhae and Bellflower tea-added Bobsikhae samples were prepared. Their general ingredients and useful ingredients were analyzed. Among the general ingredients of the two samples, the crude protein, crude fat, crude ash, and carbonhydrate contents, but not the water, were found to have been higher in the Bellflower tea-added Bobsikhae. To summarize all the results of this study by comparing Bobsikhae with Bellflower tea-added Bobsikhae that used the Bellflower root with edible and medicinal values, their general ingredient contents were similar, but the lactic acid bacteria all tended to be higher in the Bellflower tea-added Bobsikhae. The results of the sensory evaluation showed that the overall taste and general acceptability were better in the Bellflower tea-added Bobsikhae than in the Bobsikhae. Thus, the Bellflower tea-added Bobsikhae is considered ideal for further commercialization. However, it was determined that further studies will be required to show if the bioactive substance would be influenced by the fermentation in the production progress of the Bellflower tea. Such findings and their application to the product development are expected to contribute much to the popularization of Bobsikhae.

In vitro Callus and Somatic Embryo Induction of Six Hosta Species Native to Korea

  • Hosta is a genus of the family Asparagaceae and distributed in East Asia. There are six Hosta species (Hosta capitata (Koidz.) Nakai, H. clausa Nakai, H. jonesii M.G.Chung, H. minor (Baker) Nakai, H. venusta F.Maek., and H. yingeri S.B.Jones) native to Korea and among them, four species (H. minor, H. jonesii, H. venusta and H. yingeri) are endemic to the Korea peninsula. Hosta is generally propagated by seed, crown division or tissue culture. However, tissue culture is a more efficient method to mass proliferation, a new cultivar development and disease-free plantlet production in a limit time. Hence, we conducted this study to evaluate the influence of various plant growth regulators (PGRs) treatments on the induction of callus and somatic embryo of the six Hosta species. Leaf, petiole and root were used to select optimum tissue culture explants. Petiole explants thus only were used for callus induction and somatic embryogenesis with TDZ (0.1, 0.5 or 1.0mg/L) and NAA (0.1 or 0.5 mg/L) combinations. After 12 weeks of culture, the highest rate of somatic embryogenesis was achieved on modificated MS medium containing 1.0 mg/L TDZ and 0.1 mg/L NAA in H. capitata and H. minor (15.5%, respectively), 0.1 or 0.5 mg/L TDZ and 0.1 mg/L NAA in H. jonesii (22.2%), 1.0 mg/L TDZ and 0.5 mg/L NAA in H. yingeri (26.7%), and 0.1 mg/L TDZ and 0.5 mg/L NAA in H. venusta (53.3%). H. clausa showed very low effect on somatic embryogenesis by PGRs; 2.2%. There was interspecies difference to PGRs respond for callus and somatic embryo induction. Regenerated multiple shoots and plantlet of H. minor, H. jonesii, H. venusta and H. yingeri were obtained via somatic embryogenesis.

Spontaneous bone regeneration after surgical extraction of a horizontally impacted mandibular third molar: a retrospective panoramic radiograph analysis

  • Background: The mandibular third molar (M3) is typically the last permanent tooth to erupt because of insufficient space and thick soft tissues covering its surface. Problems such as alveolar bone loss, development of a periodontal pocket, exposure of cementum, gingival recession, and dental caries can be found in the adjacent second molars (M2) following M3 extraction. The specific aims of the study were to assess the amount and rate of bone regeneration on the distal surface of M2 and to evaluate the aspects of bone regeneration in terms of varying degree of impaction. Methods: Four series of panoramic radiographic images were obtained from the selected cases, including images from the first visit, immediately after extraction, 6 weeks, and 6 months after extraction. ImageJ software® (NIH, USA) was used to measure linear distance from the region of interest to the distal root of the adjacent M2. Radiographic infrabony defect (RID) values were calculated from the measured radiographic bone height and cementoenamel junction with distortion compensation. Repeated measures of analysis of variance and one-way analysis of variance were conducted to analyze the statistical significant difference between RID and time, and a Spearman correlation test was conducted to assess the relationship between Pederson's difficulty index (DI) and RID. Results: A large RID (> 6 mm) can be reduced gradually and consistently over time. More than half of the samples recovered nearly to their normal healthy condition (RID ≤ 3 mm) by the 6-month follow-up. DI affected the first 6 weeks of post-extraction period and only showed a significant positive correlation with respect to the difference between baseline and final RID. Conclusions: Additional treatments on M2 for a minimum of 6 months after an M3 extraction could be recommended. Although DI may affect bone regeneration during the early healing period, further study is required to elucidate any possible factors associated with the healing process. The DI does not cause any long-term adverse effects on bone regeneration after surgical extraction.

  • Tooth impaction is a frequently observed eruption anomaly in pediatric dental practice. Young patients with impacted or unerupted teeth have more prediction for dentigerous cyst formation. Dentigerous cyst presents radiographic features, unilocular or multilocular radioluscency. Cysts occur most frequently in the premolar region except third molar. Dentigerous cysts can grow to a considerable size, and large cysts may be associated with a painless expansion of the bone in the involved area. Extensive lesions may result in facial asymmetry, osseous destruction, root resorption of proximal teeth and displacement of associated tooth. The nature of the causative tooth influences the type of surgical treatment required for the dentigerous cyst. If the cyst is associated with a supernumerary or wisdom tooth, complete enucleation of the cyst along with extraction of tooth may be the first treatment choice. Otherwise, preservation of the associated teeth should be considered to prevent a young patient from psychological and mental trauma because of the loss of tooth. We should consider the degree of tooth displacement, osseous destruction and growth pattern of oromaxillofacial area when planning treatment. Thus a proper and logical treatment planning can help a proper growth and development of oromaxillofacial area and can save the patient from a psychological and mental trauma. This report describes 4 cases of the management of impacted premolars and molars associated with dentigerous cysts in children.

Synthesis and evaluation of PDLs22 recombinant protein (PDLs22 재조합 단백질의 합성과 평가)

  • Periodontal ligament (PDL) is the connective tissue located between the tooth root and alveolar bone. In a previous study, PDLs22 was isolated as a PDL-specific gene by using subtractive hybrid-ization between cultured PDL fibroblasts and gingival fibroblasts. It was also suggested that PDLs22 plays important roles in the development, differentiation and maintenance of periodontal tissues. However, little is known about functional study of PDLs22 using recombinant protein in PDL fibroblast differentiation and periodontium formation. In this study, in order to produce the PDLs22 recombinat protein, PDLs22 expression vector were constructed and expressed its protein in various host cell and temperature conditions. The results were as follows: 1. PDLs22 protein was not strongly expressed In the induction system using pRSET-PDLs22 construct. 2. When the BL21(DE3) pLysS was used as a expression host, PDLS22 protein was strongly ex-pressed in the induction system using pHCEIIBNd-PDLs22 construct. 3. The PDLs22 protein was recognized at a molecular weight of 28 kDa in western blots. 4. Almost of the expressed PDLs22 protein was not soluble and observed like as inclusion body. 5. The protein solubility was not improved after modification of induction time and temperature during PDLs22 protein production. In this study, the system for the PDLs22 protein production was connstructed. However, the re-results suggest that further studies will be needed to produce the considerable amount of PDLs22 re-combinat protein, which can use for the periodontal regeneration.

REGIONAL ODONTODYSPLASIA : CASE REPORT (국소적 치아 이형성증(Regional odontodysplasia)에 대한 증례)

  • Regional odontodysplasia is a relatively rare condition in which both enamel and dentin are hypoplastic and hypocalcified. The result is localized arrest in tooth development. The etiology of regional odontodysplasia is uncertain, but, disturbance in vascular supply, somatic mutation, latent virus infection, trauma, hyperpyrexia, irradiation, nutrition, metabolic disorders and hereditary transmission are supported to be etiologic factors. Females are more often affected than males. (1.4 : 1). The maxillary arch is more often affected than the mandibular arch with the maxillary left quadrant being the most commonly involved. Affected teeth are hypoplastic, typically discolored yellow or yellowish brown, smaller in size and display a variety of surface marking including pitting and grooving. Radiographically, the teeth affected have been described to have a "ghost like" appearance or "fuzzy" appearance. Pulp calcification and denticles may be present within the pulp chambers of the affected teeth. In it's case, a 2 years old male visited for a treatment of uneruption of lower right teeth. Partial eruption of lower right deciduous central incisor and unerupted deciduous lateral incisor, deciduous canine and deciduous first molar showed severly delayed eruption state. On radiographic appearance, "Ghost like appearance", shortened root and opened apexes on lower right region were observed. It was suspected regional odontodysplasia with clinical and radiographic condition.

  • Upper and lower cuspids are very important teeth in point of function as well as esthetic. However, failure of eruption of the canine sometimes happens and is frequently encountered clinical problems. Upper cuspids are more frequently impacted as compared with lower cuspis and palatally impaction is more frequent event than labially impaction. Impaction of the mandibular canine is unusual event, especially crossing the midline (transmigration) is very rare. Because of, in most cases, there is no signs and symptoms, impacted teeth can be detected by periodic radiograph examination. The etiology of impaction is unclear and multifactorial. The treatment of impacted canine is influenced by presence of obstacles, location and direction of impaction, space for alignment and development of root. There are several treatment options; (1) Surgical removal and/or observation, (2) Window opening and orthodontic traction, (3) Autotransplantation. The first patient (impaction of upper right cuspid) is treated with window opening and orthodontic traction. The second patient (bilaterally impaction of lower cuspids) is treated with autotransplantation, because of transmigration.

Morphological Development of Larvae and Juveniles of Codlets, Bregmaceros atlanticus in the East China Sea (동중국해에 출현한 대서양날개멸(Bregmaceros atlanticus Geode et Bean) 자치어의 형태 발달)

  • The study focuses on the morphology of larval and juvenile codlets, Bregmaceros atlanticus, collected from the East China Sea in May 2001 and September 2002. A total of 47 individuals were collected by using an IKMT, and the range of their body lengths was $2.41{\sim}17.18$ mm. Specimens can be categorized into two groups: larvae and juvenile. The first group, in which the yolk is completely absorbed, had a body length of 2.41m. They had big heads, and large and thick melanophores were sparsely scattered about the head, bottom of the pectoral fins, rear parts of the trunk and ends of the notochords. The specimens had a body length of 2.54mm and over, and thin and elongated body trunks. Their heads were significantly smaller, including the body depth, and melanophores were coarsely developed all over their bodies. Occipital rays, the first and unique-ray dorsal fin, were observed on individuals of body length greater than 4.01 m, and the rays grew proportionally with body length, but didn't reach the root of the second dorsal fin. The second group, juveniles, had an body length of 9.82mm and over, and their fin rays satisfied the completed numbers of rays and vertebrae for adults. Melanophores were densely scattered all over the body, where thick colors were observed on the rear parts of the dorsal and anal fins and front part of the caudal fin.

A Study on Development of Modified Hydroponic System - On the Intermittent Soaking System of Soilless Culture - (새로운 수경재배방식 개발에 관한 연구 -간헐침지식 수경재배시스템에 관하여 -)

  • This experiment was carried out to investigate the possibility of practical use of intermittent soaking system(ISS) in hydroponicaly growing tomato. There were four treatments which were NFT, Aeroponics, and ISS with 10 and 15 minute soaking intervals. 1. Plant height is highest in NFT, but main root length is longest in ISS. While stem diameter and leaf number were not significantly difference. 2. Fresh and dry weight were heavier in NFT than those of other systems at early growth stage, but the heavier in ISS system, the growth more develop. 3. In 43 days after treatment, the number of flowerlet was largest in aeroponics, and then ISS and NFT in order. Fruit weight per plant was also heaviest in aeroponics. 4. Average yield per plant were 1929.1g in aeroponics, 1475.2g in ISS with 10 minute soaking interval, 1276.2g in NFT and 1084.8g in ISS with 15 minute soaking interval. 5. Average fruit number per plant were 11.0 in aeroponics, 9.7 in NFT, 8.3 in ISS with 15 minute soaking interval, and 7.7 in ISS with 10 minute soaking interval. Average fruit weight were 192.9g in aeroponics, 172.4g in ISS with 10 minute soaking interval, 134.0g in NFT and 126.0g in ISS with 15 minute soaking interval. 6. Average fruit length was longer in aeroponics and ISS with 10 minute soaking interval than in the NFT and ISS with 15 minute soaking interval. Average fruit width was similarly wider in NFT, aeroponics and ISS with 10 minute soaking interval than in ISS with 15 minute soaking interval.

  • A multi line electric tube consisted of XL pipes contained with 2~4 hot wires and water in it. The specification of one meter length multi-line electric tube was investigated and the proper number in the multi-line electric tube was determined. A multi line electric tube with three hot wires were found to be the most efficient for the media heating control system. Temperature rise of medium in the rice hulls media was faster than that in the perlite media, showed better insulation effect of rice hulls media. Temperature rise of medium with mulching on the top of the bed was faster than without mulching, resulted in the beneficial effect of temperature rise with mulching. The regression model for the rice hulls media with mulching air temperature of $5^{\circ}C$ were a = -0.1458 and b = -0.1088. Using the model, the temperature rise of medium during low temperature season can be predicted for the various media according to the different depths.
