Background: Endophytic fungi play an important role in balancing the ecosystem and boosting host growth. In the present study, we investigated the endophytic fungal diversity of healthy Panax notoginseng and evaluated its potential antimicrobial activity against five major phytopathogens causing root-rot of P. notoginseng. Methods: A culture-dependent technique, combining morphological and molecular methods, was used to analyze endophytic fungal diversity. A double-layer agar technique was used to challenge the phytopathogens of P. notoginseng. Results: A total of 89 fungi were obtained from the roots, stems, leaves, and seeds of P. notoginseng, and 41 isolates representing different morphotypes were selected for taxonomic characterization. The fungal isolates belonged to Ascomycota (96.6%) and Zygomycota (3.4%). All isolates were classified to 23 genera and an unknown taxon belonging to Sordariomycetes. The number of isolates obtained from different tissues ranged from 12 to 42 for leaves and roots, respectively. The selected endophytic fungal isolates were challenged by the root-rot pathogens Alternaria panax, Fusarium oxysporum, Fusarium solani, Phoma herbarum, and Mycocentrospora acerina. Twenty-six of the 41 isolates (63.4%) exhibited activity against at least one of the pathogens tested. Conclusion: Our results suggested that P. notoginseng harbors diversified endophytic fungi that would provide a basis for the identification of new bioactive compounds, and for effective biocontrol of notoginseng root rot.
With transient flooding followed by poor or slow drainage plant roots may become reduction conditions because the root zone was fully filled with water. This study was examined the effects of calcium treatment in the early growth stage on biochemical changes in leaves and roots of melon (Cucumis melo L.) seedlings kept under flooding condition for 72 h. The activities of lactate dehydrogenase more gradually enhanced in the roots than those of leaves of melon seedlings treated with calcium. The activities of alcohol dehydrogenase associated with alcohol fermentation under low oxygen conditions continuously increased in the leaves and roots of seedlings untreated with calcium under flooding at least 72 h but those was constant within at least 12 h in treated with calcium. These results showed that calcium supplying in the early growth stage mitigated alcohol fermentation of melon seedlings kept under flooding condition for 72 h. Activities of nitrate reductase and acid phosphatase in the leaves and roots of seedlings in treated with calcium somewhat higher than those of non-treated with calcium. The activities of sucrose phosphate synthase and fructose-1,6-bisphosphatase of leaves of seedlings in treated with calcium more higher than those of non-treated with calcium. These results indicated that calcium nutrition mitigate the reduction of activities of some enzymes of melon seedling kept under flooding condition for 72 h.
Comparison of four phenolic compounds among three different parts of Panax ginseng C.A. Meyer was performed in this study. Contents of esculetin, p-coumaric acd, quercetin, maltol in the ethyl acetate fraction were determinated by HPLC analysis. The main root has a highest concentration of p-coumaric acid and the leave has a highest concentration of quercetin and esculetin. This ethyl acetate fraction of the leaves exhibited higher depigmenting activity and lower cell toxicity than the other parts of ginseng. In addition, it showed that quercetin and esculetin among phenolic compounds highly inhibited melanin biosynthesis in ginseng. It implies that leaves of ginseng may be used as skin whitening herb. Quercetin and esculetin proved to be active compounds in the leaves of ginseng.
This study was conducted to investigate the effect of simulated acid rain (SAR) on germination, growth, acid buffering capacity and nutrient leaching in Impatiens balsamina L. and Tagetes patula L.. In both species, germination was not inhibited at pH 4.0, but the germination rate decreased at the lower pH of 3.0, showing higher rate in Inpatiens balsamina L. than Tagetes patula L.. As the pH decreases, the growth of radicle was markedly decreased than that of hypocotyl in both species. The plant height, root length, leaf area, total dry weight, relative growth rate and net assimilation rate were inhibited by SAR. The acid buffering capacity in the leaves were increased at pH 4.0, on the other hand, it was shown a tendency to decrease at pH 2.0 in both species. As the pH levels decreased from 5.6 to 2.0, the nutrient leaching from leaves was significantly increased in both species. Based on the results, there are a great difference in the responses to SAR between the two species. In general, Tagetes patula L. represented a higher tolerance to SAR than Impatiens balsamina L.. These results suggested that interspecific variation in the acid buffering capacity and nutrient leaching from leaves may be responsible for the interspecific susceptibility to SAR.
Inhibitory substance in the water extracts from lilac(Syringa vulgaris) leaves was determined in terms of the allelopathic chemicals. The water extracts from S. vulgaris leaves inhibited the germination and root growth of Digitaria sanguinalis and L. sativa, indicating that a biological substances are presented in the lilac leaves. The phenolic acids were separated and tentatively identified from S. vulgaris leaves by gas chromatography and there were composed of higher contents of p-coumaric acid, salicylic acid, pyrogallol, and catechol. Polyphenolic compounds such as rutin (5.3%), scopoletin (3.3%), kaempferol (2.9%), and other polyphenolic compotmds were detected from lilac leaves. The mixtures of $10^{-6}M$ of pyrogallol with all the concentrations of catechol had high inhibition of the shoot growth on D. sanguinalis and E. crus-galli regardless of the catechol concentrations.
A kind of naphthoquinone, plumbagin was purified and identified from the leaves of Nepenthes $ventricosa\;{\times}\;maxima$ through solvent extraction, silica gel column chromatography, and recrystallization. The yield (0.51%) was higher than that of the root of Plumbago scandens (0.26%), P. capensis (0.15%), and N. thorelii (0.092%). It exhibited antifungal activity against all plant pathogenic fungi tested, Alternaria alternata, Aspergillus niger, Bipolaris oryzae, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani, Rhizopus stolonifer var. stolonifer and Sclerotinia sclerotiorum. The minimum inhibitory concentration values ranged from about 4.8 to $56.6\;{\mu}g/ml$ against the above eight fungi and R. solani was the most sensitive.
Root and basal stem rot disease occurred on moth orchid (Phalaenopsis spp.), Pung-nan (Neofinetia falcata) and Nadopung-nan (Aerides japonicum) grown in the farmers' greenhouses located in Namyangju Kyonggi province, Korea during 2005 to 2006. Wilting symptoms occurred on these orchard plants at initial stage and the infected plant leaves turned yellow to red. The discolored leaves were fallen down to lead to eventual death of the entire plant. A total of 59 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The cultural and morphological characteristics of isolated Fusairum spp. were identified as Fusarium oxysporum, F. proliferatum and F. solani. F. oxysporum and F. proliferatum were isolated from all these orchard plants but F. solani was isolated only from Phalaenopsis spp. Pathogenicity of the three Fusarium spp. was confirmed by artificial inoculation. Although F. oxysporum, F. proliferatum and F. solani cusing root rot disease in Phalaenopsis spp. have been reported in Korea, the pathogens in N. falcata and A. japonicum were not reported yet. Therefore, this is the first report on the root and stem rot of N. falcata and A. japonicum caused by F. oxysporum and F. proliferatum in Korea.
Effects of illumination with xenon or sulfur lamp on growth of tomato seedlings were studied. Results showed no obvious differences in net photosynthetic rate and apparent quantum efficiency. In comparison with xenon lamp, illumination with sulfur lamp slightly reduced ChI a and ChI b content in leaves, relatively shortened epicotyl and hypocotyl length, but significantly increased anthocyanin content of leaves and promoted root growth of tomato seedling plants.
Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.
Development of modern agricultural machinery and accompanying agricultural development cause soil compaction and reduce growth by stressing roots. Kalanchoe pinnata was used to investigate the impact of stress on rooting and changes in plant growth and reproduction. K. pinnata forms somatic embryos capable of asexual reproduction at the edge of leaves. Impact of root pressurization of K. pinnata on somatic embryogenesis and organ differentiation according to external stress factors was investigated by using a high concentration of agar and this phenomenon was studied histologically. Agar concentration in culture media ranged from 0.5%-1.5% to induce a compression effect on roots. The stem and leaf of K. pinnata were subjected to a microtechnique process to study changes in tissue. In vivo, K. pinnata produced 2nd and 3rd plantlets at edges of leaves from lack of water and excessive lighting conditions. In in vitro culture studies, the lower the concentration of agar, the higher the population and the higher the biomass, but plantlet did not occur in leaf bends. Conversely, as concentration of agar increased, increase in the number of individuals was low. Plantlet development occurred only in agar 1.5% medium. The difference in agar concentration was a stressor in the root of K. pinnata, and thus the pattern of asexual reproduction changed from the division method in root to a plantlet generation in leaf. This suggests root pressurization may act as stress and change in the plant reproduction pattern.
