유묘특성(幼苗特性)을 이용한 참당귀(當歸) 계통(系統)의 조기선발(早期選拔) (Selection of Angelica gigas Nakai Lines Using Seedling Characteristics)

  • 참당귀(當歸) 묘상기(苗床期)에서의 유묘특성(幼苗特性)과 추태율(抽苔率) 및 뿌리 수량(收量)과의 관계(關係)를 밝혀 유식물(幼植物) 시기(時期)에 조기검정(早期檢定)의 선발체계(選拔體系) 확립(確立)을 위한 기초자료(基礎資料)를 얻고자 시험을 실시한 결과는 다음과 같다. 1. 공시계통간(供試系統間) 묘초장(苗草長), 엽장(葉長), 엽병장(葉柄長) 등 지상부(地上部) 유묘특성(幼苗特性)에 유의적(有意的)인 차이가 나타나 묘상(苗床)에성의 조기검정(早期檢定) 가능성을 보였다. 2. 성숙기(成熟期)의 추태율(抽苔率)은 $0.6{\sim}75.6$% 범위였고, 뿌리수량(收量)은 $350{\sim}3,893kg/ha$으로 계통간(系統間) 차이가 컸으며, 추태율(抽苔率)이 5%미만으로 낮을 때 뿌리수량(收量)이 높았다. 3. 유전력(遺傳力)은 묘상(苗床)에서 엽병장(葉柄長), 묘초장(苗草長), 엽장(葉長) 등과 성숙시(成熟時)의 추태율(抽苔率), 뿌리수량(收量)등에서 92%이상으로 높았고, 엽폭(葉幅), 근경(根徑), 근장(根長) 등에서 낮게 나타났다. 4. 유묘특성(幼苗特性)과 추태율(抽苔率) 및 뿌리수량간(收量間)에는 관계(關係)는 묘초장(苗草長), 엽장(葉長), 엽폭(葉幅), 엽병장(葉柄長), 엽수(葉數), 근장(根長) 등은 뿌리수량(收量)과 부(負)의 상관(相關)을 보인 반면, 근경(根徑)은 정상관(正相關)을 나타내었다. 추태율(抽苔率)과 뿌리수량간(收量間)에는 부(負)의 상관(相關)으로 고도(高度)의 유의성(有意性)이 인정되었다. 5. 본 결과로 볼 때 묘상기(苗床期)에 묘초장(苗草長), 엽장(葉長), 엽병장(葉柄長) 등 유묘특성(幼苗特性)을 검정(檢定)하므로서 추태율(抽苔率)이 낮고 다수성(多收性)인 우량개체(優良個體)를 조기(早期)에 선발(選拔)할 수 있을 것으로 기대된다.

  • Agorobacterium rhizogenes에 의하여 유도된 당근(Daucus carota L. )의 hairy root를 배양하였으며 opine 유무에 대한 형질전환 확인 및 배지조성을 달리하여 성장률에 따른 색소함량을 비교하였고, 재분화된 식물체의 형태적 차이를 관찰하여 몇가지 결론을 얻었다. Hairy root는 균 접종 2-4주 후에 형성층 부위를 중심으로 유도되었다. 유도된 hairy root의 초기 배양에는 R.C.M 배지가 적합하였으며 MSO(2, 4-D $10^{-4} ml/ l, pH6, sucrose 5%, 질소원 0.03M 등)에서 최대 성장을 보여주었고 성장의 증가에 따른 색소의 형성은 비교적 안정하였다. 재분화된 식물체는 정상 식물체에 비하여 형태적으로 차이를 나타내었으며 형질전환된 hairy root 및 재분화된 식물체에서 mannopine 분석으로 Ri-plasmid에 의한 형질전환이 이루어졌음을 확인하였다.

차광율 및 육묘용기 크기에 따른 갯기름나물 유묘의 생장 및 뿌리 발달 특성 (Characteristics of Growth and Root Development of Peucedanum japonicum Seedling by Shading Rate and Container Size)

  • This study was carried out in order to investigate the early growth and root development characteristics of Peucedanum japonicum container seedlings. The experiment was performed by containers (128 and 200 cavities containers) and shading levels (0%, 35%, 50% and 75% shading). Germination rate of P. japonicum seeds was getting higher in the lower shading level and the highest in the full sunlight (71.9%). Plant height was the highest under 50% shading of all containers. Fresh weight and dry weight of the shoot (leaves + stem) were the highest under 50% shading of all containers, too. Meanwhile, fresh weight and dry weight of the root per plant were the highest under the full sunlight of 128 cavities container such as 0.34 g and 0.03 g, respectively. Total root length, root project area and root surface area were higher under the full sunlight of 128 cavities container such as 234.5 cm, $6.6cm^2$ and $20.8cm^2$, respectively and the next higher was under 35% shading such as 201.7 cm, $5.9cm^2$ and $18.4cm^2$, respectively. A case of root volume was the highest with $0.15cm^3$ under the full sunlight. As a result of the surveying the whole experiment, it is concluded that the shoot and root of P. japonicum seedling grow nicely by maintaining 35% shading.

Distribution of Fatty Acids in Newly Developed Tissues of Soybean Seedlings

  • The objective of this study was to determine the fatty acid composition of newly developed tissues of germinated soybean seeds. Five soybean accessions with varied fatty acid composition were allowed to germinate in sand under greenhouse conditions. Seedlings were picked up after 4, 6, 8, 10 and 12 days of germination and freeze dried. The fatty acid composition of the newly developed tissues was analyzed by gas chromatography. Significant variation in fatty acid composition was observed between accessions, days of germination, and variety ${\times}$ day of germination in whole and the cotyledons. In the case of newly developed five tissues, significant variation in fatty acid composition were observed between days of germination except oleic acid for root, hypocotyl and epicotyl stem and except stearic acid for hypocotyl and unifoliate leaves while all the parameters were significantly different for accession. Significant interactions of accession and days of germination were observed for palmitic, linoleic and linolenic acid in all tissues; only for oleic acid in hypocotyl, epicotyl and unifoliate leaves; and only for stearic acid in root, hypocotyl, epicotyl and unifoliate leaves. During germination, the fatty acid composition of newly developed tissues changed dramatically but whole seedlings and cotyledons changed slightly. These tissues contained five major fatty acids as found in original seeds, but compositions were totally different from that of the seed: higher in palmitic, stearic and linolenic acid and lower in oleic and linoleic acid. New tissues conserved their fatty acid compositions regardless of genotypic variation in the original seeds.

예취주기의 상호전환에 따른 White clover 품종의 예취수량 및 생장 (Harvest Yield and Growth Response White Clover Cultivars to Infrequent , Frequent Defoliation and Their Alternation)

  • Pasture composed of about 30% botanical composition of white clover (Trijolium repem L.) is desirable in its productivity and quality. To get information on maintaining its productivity in the pasture, the experiment was canied out to determine the effect of infrequent, frequent defoliation or their alternation on change of its harvest yield and growth. Individual plants of Regal, Louisiana S-1, Grasslands Huia and Aberystwyth S184 were grown in 22cm plastic pot containing a 2 : 1 : 1 soil : sand : Peat moss mixture for 27 days after transplanting 50day seedlings raised on 3cm pots, and then all their fully expanded leaves are removed. Defoliation treatments were forced every 1 (CC, frequent), 4 week (RR, infrequent) or their alternations (CR, RC) after 8 weeks from the removal. To analyze the treatment effects, plants were sampled on 0 (the removal day), 4, 8, 12 and 16 weeks after the removal and seperated to leaves, petioles, stolons and roots. Each harvest yield of infrequent defoliation (RR) was higher than that of frequent defoliation (CC). Their alternations (RC, CR), however, forced fluctuation of the yield according to defoliation interval. Fraction weights from their altemations showed intermediate ones of infrequent and Frequent defoliation whose weights did the similar result to their harvest yields. Fraction and total dry weights per plant, shoot/root ratio were changed by relative span of defoliation interval, which resulted from the weights of leaves and petioles, removed parts. Root and total dry weight per plant of Regal were greater than the other cultivm in infrequent defoliation interval regardless of continuous or alternations. Productivity of white clover in pasture, therefore, can be controlled by alternation of infrequent and frequent defoliation or reverse.

해바라기의 붕소결핍(硼素缺乏) (Boron deficiency of sunflower (Helianthus annuus.))

  • 해바라기의 생육불량(生育不良) 원인(原因)을 토양(土壤) 및 식물체(植物體) 분석(分析)에 의(依)하여 본 결과 붕소결핍(硼素缺乏)으로 나타났으며 다음과 같은 결과를 얻었다. 1. 붕소결핍(硼素缺乏)은 토양(土壤)의 유효붕소함량(有効硼素含量)(열수침출(熱水浸出))이 적은데 기인(基因)했으며 유효붕소(有効硼素) 임계농도(臨界濃度)는 0.17ppm으로 나타났다. 2. 식물체(植物體)에서의 임계농도(臨界濃度)는 화부(花部)(두부(頭部))에서 15ppm 근(根)에서는 10ppm으로 나타났다. 상위엽(上位葉)으로 갈수록 B함량(含量)은 상당히 감소하였다. 3. 붕소(硼素)가 적은 토양(土壤)은 Ca가 많았으며 유기물(有機物)이나 산도(酸度) 기타 양분(養分)과의 관계가 불확실했다. 4. 붕소결핍(硼素缺乏)된 식물체(植物體)는 Ca와 P가 두부(頭部)에는 높고 근부(根部)에 낮으며 N는 높고 K는 낮은 편이며 Mg와 Fe는 관계가 불확실했다. 5. 붕소결핍(硼素缺乏)은 줄기와 상부(上部)의 갈변고사(褐變枯死) 및 줄기와 뿌리가 부분적으로 파열(破裂)되고 흑변(黑變)하였다.

Energized 기능수 처리에 따른 후지사과의 잎, 뿌리 및 과실특성 (Characteristics of Leaves, Roots, and Fruit as Influenced by Energized-Functional Water Supply in Fuji Apple Trees)

  • Energized 기능수는 미약 에너지 발생 신소재로서 다음과 같은 공정을 거쳐서 경원생명과학연구소에서 제조되었다; 지하수 순수화 처리촉매제 첨가 에너지 imprinting in platinum column 재료혼합 energized water + 특수처리된 zeolite + 광합성 세균균주 발효 (25C) 여과 최종산물 energized 기능수 (여과액) + energized powder (잔유물), 후지사과 나무가 발아하기 전에 주당 3kg의 energized power를 수관하부에 살포하여 토양과 혼합하고 energized 기능수를 생육기 동안 3회에 걸쳐 토양관주한 결과 수확된 과실의 과피와 과육에 Ca 함량이 증가된 반면 잎과 신초 수피조직중의 Ca 및 N함량은 감소되었다. Energized 기능수 처리는 사과나무 잎의 순광합성율과 뿌리활성을 촉진시켰다. 또한 과실특성에 있어서 가용성 고형물과 안토시아닌 함량이 현저히 증가되었고, 과실의 저온저장중 과실경도가 높게 유지되었으며 과심부 갈변현상이 감소되었다. 과실의 유기산 함량은 처리간에 차이가 없었다.

Serratia plymuthica에 의한 인삼 세균뿌리썩음병 발생 (First Report of Bacterial Root Rot Caused by Serratia plymuthica on Panax ginseng)

  • 2011년 8월 경상북도 봉화군 재산면 농가포장 5년생 인삼포장에서 잎이 푸른색을 띈 채 급격하게 마르는 증상이 발견되었다. 잎이 시든 직후 인삼의 뿌리를 채취하였으나, 뿌리나 줄기에서 특별한 병징은 없었으며, 뿌리의 수분이 약간 빠져나간 상태였다. 이러한 증상이 발생된 지 10일이 경과된 뿌리에서는 뿌리가 물러지면서 썩는 증상이 나타났다. 이러한 병징과 건전부의 경계부위에서 세균이 분리되었으며, 이 세균에 의한 뿌리썩음병이 의심되었다. 세균을 배양한 후 건전 인삼에 접종하자, 동일한 썩음 증상이 발현되었으며, 병반으로부터 동일한 세균이 재분리되었다. 이 세균을 BioLog system에 의한 탄소원 이용여부, Vitek 2 system을 이용한 생화학적 반응, GC-MIDI Sherlock system을 이용한 지방산 조성, 16S rRNA 염기서열을 분석한 결과, 모두 Serratia plymuthica로 동정되었다. 이에 따라 인삼 뿌리에 발생하는 이 병을 Serratia plymuthica에 의한 인삼 세균뿌리썩음병으로 명명하여 보고하고자 한다.

Comparison of Glucosinolate Contents in Leaves and Roots of Radish (Raphanus spp.)

  • Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.

Pinus rigida Allelochemicals가 차풀종자의 발아과정에서 동위효소의 활성에 미치는 영향 (Effects of Pinus rigida Allelochemicals on Isozyme Activities during Seed Germination of Cassia mimosoides var.nomame)

  • Eleven phenolic compounds including caffeic acid were identified through analyzing the aqueous extracts of Pinus rigida by HPLC. Among them, protocatechuic acid was the maximum amount of 6.84 ppm. Seed germination of Cassia mimosoides var. nomame was significantly stimulated by the extract of P. rigida leaves in the proportion ot concentration. However, root growth was elevalted at a threshold concentration below 25%, but it was inhibited at high concentrations. In 50% extract of P. rigida, upward root tip of C. mimosoides var. nomame showed negageotropism which the root end showed necrosis. New isozyme bands were induced indicating concentration activity of peroxidase from the extract of C. mimosoides var. nomame, especially in the cathodic region. Although it reduced the mumber of isozyme bands of esterase, esterase activities were stimulated in the anodic region of C. mimosoides var. nomame. The activity of amylase was not remarkably different between control and treatment.
