The experiments were carried out to determine the optimum conditions for the induction of hairy roots in ginseng(Panax ginseng C.A. Meyer) by Agrobacterium spp. We were examined the antibiotics resistance of Agrobacterium spp and various ginseng parts, and the media for induction of hairy roots. The optimum concentration of NaOCl for sterilization of ginseng root segments without tissue damage with reduce of contamination was 7% NaOCl for 15-20 min and 9% NaOCl for 5 min, respectively. The more ginseng ages, the more contamination of ginseng root segment by sterilized in 7% NaOCl for 20 min, and especially in ginseng root segments with epidermis in six-year old roots. The growth of Agrobacterium spp were inhibited, but ginseng root segments was death in 30mg/L tetracycline. In 500mg/L cefotaxime or 500mg/L carbenicillin, the growth of Agrobacterium sup were inhibited, and root segments was grown normally. The optimum conditions for induction of hairy roots were using the root segments of three-year old ginseng cultured in 1/2MS medium supplemented with 500mg/L cefotaxime, and inoculation of Agrobacterium to root segments were better co-culture than smear method. After 2 weeks co-culture, the callus induced in cambium of root segments cultured in 1/2MS solid medium with 500mg/L cefotaxime. And then after 2 weeks, ginseng hairy roots were induced in callus of root segments. PCR analysis of rot C gene fragment confirmed that hairy roots were transgenic tissues.
The root of Astragalus membranaceus is a traditional folk medicine that has been used for many therapeutic purposes in Asia. It reportedly acts as an immunostimulant, tonic, hepatoprotective, diuretic, antidiabetic, analgesic, expectorant, sedative, and anticancer drug. In this study, metabolomic profiling was applied to the roots of A. membranaceus of different ages using NMR coupled with two multivariate statistical analysis methods: such as principal components analysis (PCA) and canonical discriminant analysis (CDA). This allowed various metabolites to be assigned in NMR spectra, including $\gamma$-aminobutyric acid (GABA), aspartic acid, succinic acid, glutamic acid, glutamine, N-acetyl aspartic acid, acetic acid, arginine, alanine, threonine, lactic acid, and valine. The score plot from PCA and also CDA allowed a clear separation between samples according to age.

This study tested 25 lightweight aggregate concrete (LWAC) mixtures using the expanded bottom ash and dredged soil granules to examine the compressive strength gain of such concrete with different ages. The test parameters investigated were water-to-cement ratios and the natural sand content for the replacement of lightweight fine aggregate. The compressive strength gain rate in the basic equation specified in fib model code was experimentally determined in each mixture and then empirically formulated as a function of the water-to-cement ratio and oven-dried density of concrete. When compared with 28-day compressive strength, the tested LWAC mixtures exhibited relatively low gain ratios (0.49~0.82) at an age of 3 days whereas the gain ratios (1.16~1.41) at 91 days were higher than that (1.05~1.15) of the conventional normal-weight concrete. Thus, the fib model equations tend to overestimate the early strength gain of LWAC but underestimate the long-term strength gain. The proposed equations are in good agreement with the measured compressive strength development of LWAC at different ages, indicating that the mean and standard deviation of the normalized root mean square errors determined in each mixture are 0.101 and 0.053, respectively.
Hair loss affects both men and women of all ages and often significantly affects social and psychologic health. Recent therapeutic approach for hair loss such as finasteride and minoxidil focused on regulation of hormonal system blood flow. However, long-term use of these drugs caused adverse effects. To develop herbal medicine for therapeutic effect on hair growth, here we screened the 10 medicinal herbs (Red ginseng, Licorice, Ulmus, Barberry root, Lycium root, Rehmanniae radix crudus, Sophora root, Sweet flag, Polygala root, Achyranthes) based on oriental medicine literature. We measured cytotoxicity, anti-oxidant activity, and $5-{\alpha}$ reductase inhibitory effect of the herbal medicine on human dermal papilla (DP) cells to investigate therapeutic effect of the herbal medicine. Treatment of the 1% herbal medicine did not show any cytotoxic effects, and cell growth was increased by treatment of the 0.1% herbal medicine. In addition, the herbal medicine showed stronger antioxidant activity than resveratrol and comparable inhibitory activity of $5-{\alpha}$ reductase with finasteride. Furthermore, when applied to in vivo mouse model, we also observed increases in the number and length of hair of the herbal medicine-treated group. These results suggest that the herbal medicine promotes hair growth by its antioxidant activity and inhibitory activity of $5-{\alpha}$ reductase and might therefore be a promising hair growth-promoting agent.
The objective of this study was to examine effects of stand age on soil $CO_2$ efflux in plantation Pinus koraiensis, and to elucidate what extent plant (fine) root and soil microbial biomass contribute to the whole soil $CO_2$ efflux. In three age classes (20-yr-old. 40-yr-old, 70-yr-old) of plantation Pinus koraiensis, in-situ soil respiration, plant fine root biomass and soil microbial biomass were measured from April to November in 2004. Regardless of stand age, soil temperature and soil $CO_2$ efflux increased until July then slowly decreased. Soil respiration was higher in 70-yr-old stand than in 20- and 40-yr stands. Fine root biomass and soil microbial biomass was also higher in 70-yr-old stand. Root exclusion decreased soil respiration in 40-yr stand, but not in 70-yr stand. Soil microbial biomass was higher in 70-yr stand, but there was no monthly variation between July and November. The results suggest that soil respiration may increase as plant stand ages and microbial contribution could play more roles in older stands.
Background: Ginsenosides are the unique and bioactive components in ginseng. Ginsenosides are affected by the growing environment and conditions. In New Zealand (NZ), Panax ginseng Meyer (P. ginseng) is grown as a secondary crop under a pine tree canopy with an open-field forest environment. There is no thorough analysis reported about NZ-grown ginseng. Methods: Ginsenosides from NZ-grown P. ginseng in different parts (main root, fine root, rhizome, stem, and leaf) with different ages (6, 12, 13, and 14 years) were extracted by ultrasonic extraction and characterized by Liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry. Twenty-one ginsenosides in these samples were accurately quantified and relatively quantified with 13 ginsenoside standards. Results: All compounds were separated in 40 min, and a total of 102 ginsenosides were identified by matching MS spectra data with 23 standard references or published known ginsenosides from P. ginseng. The quantitative results showed that the total content of ginsenosides in various parts of P. ginseng varied, which was not obviously dependent on age. In the underground parts, the 13-year-old ginseng root contained more abundant ginsenosides among tested ginseng samples, whereas in the aboveground parts, the greatest amount of ginsenosides was from the 14-year-old sample. In addition, the amount of ginsenosides is higher in the leaf and fine root and much lower in the stem than in the other parts of P. ginseng. Conclusion: This study provides the first-ever comprehensive report on NZ-grown wild simulated P. ginseng.
This study was conducted to investigate the relationship of development of root and soil condition and also to elucidate the difference between stands for mechanical and physical stability according to distribution of roots in the Norway Spruce (Picea abies [L.] Karst.) stands of 30 to 40 year of ages with different soil condition. In the root structure, remarkably different root pattern was shown among the three stands; Eberg$\ddot{o}$tzen stand showed large number f roots with much fine roots which deeply in vertical direction while Barbis stands had thicker root and small number of root than Eberg$\ddot{o}$tzen stand. The Weidenbrunnen stand showed differently half-vertical and plate root structure and the root developed not to be deeply compared to the other two stands. In the total number of root, Eberg$\ddot{o}$tzen stand had the most number of 74 and Barbis stand 33, Weidenbrunnen stand 57 respectively. Eberg$\ddot{o}$tzen stand had much more number of vertical root with 57 than the other two stand; Barbis 21 and Weidenbrunnen 36. In the total length of root, Weidenbrunnen stand showed to be longer than the other two stands and the length of horizontal root also showed longer. Regarding to the rates of horizontal root against vertical root, Eberg$\ddot{o}$tzen stand showed comparatively higher value as 1,2 than other two stands(0.5). In the total weight of root, Weidenbrunnen stand showed very high value compared with the other stands and ratio of vertical root against horizontal root was 0,16 which is very lower than the other two stands(0,4). The remarkable differences of root pattern of Norway spruce stands appeared in this study are through to be caused more by chemical property of soil such as acidity den the mechanical or physical factor of soil conditions.
The activities of the antioxidative enzymes in the roots of Codonopsis lanceolata have been compared depending on the cultivated environments - wildness, cultivate paddy fields and cultivate dry fields - and the parts of the root. In the Codonopsis lanceolata raised in cultivate paddy fields, the activity of SOD was higher in 2 yrs old than 1 yr old, but the activity in 1 yr old was higher than in 2 yrs old for the plants raised in the cultivate dry fields. The specific activity of SOD in wildness plants 86.069unit/mg protein was the highest among plants studied. The tissue distribution of the SOD activity showed differences depending on the enviroment. The highest activity of SOD was shown in the upper part of the root for the cultivate paddy fields, the loewr parts for the cultivate dry fields and middle parts for the wildness. The specific activity of POD was increased with ages of the plants, and that in the wildness was the highest 68 unit/mg protein among the plants studied. The activity of POD in the parts of the roots was shown as middle>lower>upper. The activity of POD in the middle part of the root, rasied in Soebick province was 85 unit/mg protein. The specific activity of CAT was decreased with ages of the plants. The activities of wildness and cultivate paddy fields was similar, but that in cultivate dry fields was lower than others. The tissue distribution in the parts of the roots was upper>lower>middle. The activity of CAT middle part of rasied in the Sebuck area was 5.359 unit/mg protein. The activities antioxidative in the cells cultured in MSID(Murashige and Skoog +2.4-D 1mg/$\iota$) was followings: 1564 for CAT. 30 for POD and 22200 unit/mg protein for SOD. These figures were lower than that in in vivo.

Objective : Brachial plexus injury can produce a intractable chronic neuropathic pain. This study was undertaken to assess the long term outcome of microsurgical dorsal root entry zonotomy[MDT]. Methods : Between October 1997 and December 2002, 21 patients received MDT because of a intractable pain resulting from brachial plexus injury. Of these, 19 patients were followed for more than 2 years. Fourteen of 19 patients were male and patient ages ranged from 22 to 69 years. Mean pain duration was 36.8 months and all patients had severe pain of $9{\sim}10$ visual analogue scale. To achieve complete destruction of abnormal dorsal horns, thermocoagulation of the posterolateral sulcus were performed and careful gluing was done to prevent postoperative adhesion and pain recurrence. Results : Of the 19 patients, 15 patients had excellent [>75% reduction in pain] and good [$51{\sim}75%$ pain relief] results in a average postoperative period of 4.1 years. One patient had a poor [less than 25% pain relief] result. Three patients were considered to have a fair result [$26{\sim}50%$ pain relief]. Postoperative complications were 2 transient ipsilateral ataxia and 1 CSF fistula that resolved without surgical revision. Conclusion : These results indicate that MDT provides excellent long-term pain relief in medically intractable chronic neuropathic pain following brachial plexus injury without significant complications.
Cooking basic information for indexing of fresh ginseng roots (Panax ginseng C. A. Meyer) was determined. Ginsenoside contents of various age fresh ginseng roots cultivated in Northeast Asia were quantitatively analyzed by HPLC. Average contents of total saponin and each ginsenoside of 4-year-old fresh ginseng root cultivated in Korea were higher than those cultivated fur longer periods (5- and 6-year-old). One-way analysis variance showed average contents of total saponin and each ginsenoside of 4-, 5-, and 6-year-old fresh ginseng roots were not statistically significant. Four-year-old fresh ginseng root cultivated at Geumsan, Korea contained the highest ginsenoside content among samples studied.
