Isolation and Identification of Exo-xylanase Producing Microorganism (Exo-xylanase 생산균의 분리 및 동정)

  • The xylanase producing microorganisms occurring on rotten woods were selectively isolated on the modified Czapek-Dox medium supplemented with 0.5% xylan as a sole carbon source. Among more than three-hundred isolates of xylanase producing microorganisms, only two bacterial isolates were turned out to be more potent xylanase producer than the reference strain of xylanase producer, Aureobaszdium pullulans NRRL Y-2311. The exo-xylanase producer, bacterial isolate No. 33 was identified as a strain of Pseudomonas sp. on the basis of morphological and biochemical characterizations as well as cellular fatty acid composition. Optima of pH and of temperature for enzyme reactions of xylanase were 5.5 and $50^{\circ}C$ respectively. The enzyme was stable in a range of pH 5.0~7.0 and below $45^{\circ}C$. Among the number of carbohydrate substrates, xylose was turned out to be a potent inducer of Pseudomonas sp. No.33 exo-xylanase. Among the raw materials tested, rice straw was the best material for xylanase production by Pseudomonas sp. strain No. 33.

  • Recently UAV(unmanned aerial vehicle) is frequently used not only for military purpose but also for civil purpose. UAV automatically navigates following the coordinates input in advance using GPS information. However it is impossible when GPS cannot be received because of jamming or external interference. In order to solve this problem, we propose a real-time segmentation and classification algorithm for the specific regions from UAV image in this paper. We use the super-pixels algorithm using graph-based image segmentation as a pre-processing stage for the feature extraction. We choose the most ideal model by analyzing various color models and mixture color models. Also, we use support vector machine for classification, which is one of the machine learning algorithms and can use small quantity of training data. 18 color and texture feature vectors are extracted from the UAV image, then 3 classes of regions; river, vinyl house, rice filed are classified in real-time through training and prediction processes.

  • The fracture mechanics analysis of a crack in a weld must consider residual stress generated during welding. The standard definition of the J-integral requires a path dependent value in the presence of a residual stress field. Therefore, it is necessary to develop a path independent J-integral definition for a crack in a residual stress field. This paper addresses the modification of the Rice-J-integral to produce a path- independent J-integral when residual stresses and external forces are present. The residual stress problem is treated as an initial strain problem and the J-integral proposed for this type of problems is used. A program which can evaluate the J-integral for a crack in a weld is developed using the proposed J-integral definition. The situation when only residual stress is present is examined as is the case when mechanical stresses are applied in conjunction with a residual stress.

  • The work has been carried out for the development of antifungal antibiotics possessing curative effect in the control of sheath blight disease of rice plant. Soil samples were collected from over 1600 spots throughout the country. More than 1300 specimens which seem to be the genus Streptomyces were isolated from the soil samples. Screening procedures consist of respective processes by four steps. Those are growth inhibition test in liquid culture, paper disk method, dendroid test and green house test. 102 isolates appeared to be active against Pellicularia sasakii when all specimens isolated were examined by the first growth inhibition test. Finally a strain of Streptomyces forming strong antifungal substances against P. sasakii was selected from a soil sample of Mt. Soyo, Kyeongi Province. Antifungal substances formed by the strain were isolated and purified from the culture broth and examined for antimicrobial activities as to be specific against fungi but not active on bacterial growth.

Chitinolytic and Chitosanolytic Activities from Crude Cellulase Extract Produced by A. niger Grown on Apple Pomace Through Koji Fermentation

  • Enzyme extracts of cellulase [filter paper cellulase (FPase) and carboxymethyl cellulase (CMCase)], chitinase, and chitosanase produced by Aspergillus niger NRRL-567 were evaluated. The interactive effects of initial moisture and different inducers for FP cellulase and CMCase production were optimized using response surface methodology. Higher enzyme activities [FPase $79.24{\pm}4.22$ IU/gram fermented substrate (gfs) and CMCase $124.04{\pm}7.78$ IU/gfs] were achieved after 48 h fermentation in solid-state medium containing apple pomace supplemented with rice husk [1% (w/w)] under optimized conditions [pH 4.5, moisture 55% (v/w), and inducers veratryl alcohol (2 mM/kg), copper sulfate (1.5 mM/kg), and lactose 2% (w/w)] (p<0.05). Koji fermentation in trays was carried out and higher enzyme activities (FPase $96.67{\pm}4.18$ IU/gfs and CMCase $146.50{\pm}11.92$ IU/gfs) were achieved. The nonspecific chitinase and chitosanase activities of cellulase enzyme extract were analyzed using chitin and chitosan substrates with different physicochemical characteristics, such as degree of deacetylation, molecular weight, and viscosity. Higher chitinase and chitosanase activities of $70.28{\pm}3.34$ IU/gfs and $60.18{\pm}3.82$ to $64.20{\pm}4.12$ IU/gfs, respectively, were achieved. Moreover, the enzyme was stable and retained 92-94% activity even after one month. Cellulase enzyme extract obtained from A. niger with chitinolytic and chitosanolytic activities could be potentially used for making low-molecular-weight chitin and chitosan oligomers, having promising applications in biomedicine, pharmaceuticals, food, and agricultural industries, and in biocontrol formulations.

Assessing the children's views on foods and consumption of selected food groups: outcome from focus group approach

  • The food choices in childhood have high a probability of being carried through into their adulthood life, which then contributes to the risk of many non-communicable diseases. Therefore, there is a need to gather some information about children's views on foods which may influence their food choices for planning a related dietary intervention or programme. This paper aimed to explore the views of children on foods and the types of foods which are usually consumed by children under four food groups (snacks, fast foods, cereals and cereal products; and milk and dairy products) by using focus group discussions. A total of 33 school children aged 7-9 years old from Selangor and Kuala Lumpur participated in the focus groups. Focus groups were audio-taped, transcribed and analyzed according to the listed themes. The outcomes show that the children usually consumed snacks such as white bread with spread or as a sandwich, local cakes, fruits such as papaya, mango and watermelon, biscuits or cookies, tea, chocolate drink and instant noodles. Their choices of fast foods included pizza, burgers, French fries and fried chicken. For cereal products, they usually consumed rice, bread and ready-to-eat cereals. Finally, their choices of dairy products included milk, cheese and yogurt. The reasons for the food liking were taste, nutritional value and the characteristics of food. The outcome of this study may provide additional information on the food choices among Malaysian children, especially in urban areas with regard to the food groups which have shown to have a relationship with the risk of childhood obesity.

Selected Dietary Intake among Californians of Korean Descent : Preliminary Findings

  • Research on diet, acculturation, and ethnicity has suggested that immigrants dietary patterns differ from people who do not have immigration experience. The purpose of this paper is to present preliminary descriptive findings concerning consumption of foods that are common in Korean and American diets, by people of Korean descent residing in California. Adults (18 years of age or older, N=1334) of Korean descent who could be reached by listed residential telephone number in California were randomly selected and telephone-interviewed by closely supervised bilingual (Korean-English) interviewers who were specially trained for this task. This study found that dietary patterns of Koreans living in California differed according to age and gender; also, the frequency of consumption of foods differed according to number of years of residence and acculturation. Koreans who reported higher levels of acculturation to American society ate less of traditional Korean foods such as Kimchi and ate significantly more higher-fat foods common in the American diet. Increased acculturation was also related to an increased frequency of alcohol drinking. Older Koreans tended to eat slightly more of traditional foods such as rice, Kimchi, fruits, and fish, compared to younger Koreans. Women tended to eat healthier foods than men, consuming green vegetables, Kimchi, milk, cheese/yogurt, fruits, and bread more frequently. Koreans reporting longer residences in the United States reported a greater consumption of pork, bread, and soda.

  • This paper discusses scientific books and journal articles in the fields of Entomology, Plant Pathology, Weed Science and Sericulture of North Korea. Most research in the fields of plant protection and sericulture are carried out in the respective research institute of their Academy of Agricultural Sciences. But scientific papers are published in a general agricultural journal. "Bulletin of Korean Academy of Agricultural Sciences", in the case of those in the field of plant protection and in a more specific journal. "Sericulture", in the case o the other. The main interests of entomologists, plant pathologists and weed scientists seem to be how to control pests on rice, com and apple. They do not appear to have many kinds of agricultural chemicals and concentrate their control strategy on the basis of non-chemical mans. Research papers in sericulture are evenly divided between on the silkworm and the mulberry tree, including their pest control.

Analysis of metabolites in wheat roots in response to salinity stress

  • Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.

Current Status on Molecular Genetic Study and Comparative Genomic Analysis of Virulence Related Genes in Xanthomonas oryzae pv. oryzae (벼 흰잎마름병균(Xanthomonas oryzae pv. oryzae)의 병원성 유전자의 분자유전학적 연구현황 및 비교유전체 분석)

  • Xanthomonas oryzae pv. oryzae (Xoo) is the causal agent of bacterial blight on rice. In this paper, current status on molecular genetic study of major virulence genes, hypersensitive response and pathogenicity (hrp), productions of extracellular polysaccharide (EPS), extracellular enzymes and lipopolysaccharides (LPS), avr genes were reviewed. The IS elements with 611 copies including 133 ORF IS were inserted in various regions of the Xoo genome and in expecially regions franking virulence genes. Whole genome sequence of X. oryzae pv. oryzae KACC10331 were used for defining genetic organization of the virulence genes. Futhermore, the virulence genes in Xoo genome were compared to those of other Xanthomonas species in Blast GenBank data base.
