Acid phosphatases (APases) play a role in the release of phosphate in organic complexes in soil. We investigated tissue- and isoform-specific responses of APases to phosphorus (P) deficiency in three rice genotypes; Dasan-byeo, Sobi-byeo, and Palawan. The levels of shoot APase activity per protein were similar in the three genotypes. They significantly decreased with P deprivation that was longer than seven days. Root APase activity per protein was two- to three-fold higher in Dasan than in Sobi and Palawan. In all genotypes the APase activity increased in P-deficient plants, but the increase was higher in Sobi and Palawan. After 21 days of P deprivation, secreted APase activity increased more than eight-fold in Dasan and two-fold in Sobi and Palawan. Isoform profiles of shoot and root APases were most diverse in Dasan. The activities of the major isoforms in P-deficient shoots decreased in all three genotypes. Depending on the genotypes, further increases in constitutive isoforms and new induction of one to four isoforms occurred in P-deficient roots. The results indicate that tissue and genotype differences in the response of APase to P deficiency are primarily facilitated by the different responses of the isoforms.
The mechanisms of resistance to oxyfluorfen (OF) and paraquat (PQ) were investigated in rice plants. Examination of the concentration dependence of oxyfluorfen- or paraquat-induced increase in conductivity showed that conductivities in the OF- and PQ-treated leaf squares were increased with 0.1 ${\mu}M$ oxyfluorfen and 0.01 ${\mu}M$ paraquat and further increased with higher concentrations. The levels of conductivity were approximately 10-times higher in the PQ-treated plants than in the OF-treated plants, indicating that the PQ-treated plants suffered more severe photodynamic damage than the OF-treated plants. The photooxidative stress caused by foliar application of either 50 ${\mu}M$ oxyfluorfen or 100 ${\mu}M$ paraquat increased the enzyme activities of ascorbate peroxidase and peroxidase 1 day after the herbicide treatments and then further increased their enzyme activities 2 days after the treatments. The activities of catalase began to increase 2 days after the oxyfluorfen and paraquat treatments. These antioxidant enzymes appear to play an essential part of defense mechanisms against oxyfluorfen and paraquat. Our results demonstrate that paraquat caused more severe oxidative stress, as indicated by a greater change in conductivity, thereby resulting in greater increases in antioxidant responses in plants, compared with those of oxyfluorfen.
To demonstrate the importance of transformation efficiency in independent event, molecular and cytogenetic analysis were conducted with genomic DNA and chromosome of transgenic plants produced by Agrobacterium tumefeciens LBA4404 (pSBM-PPGN: gusA and bar). Selection ratios of putative transgenic calli were similar in independent experiments, however, transformation efficiencies were critically influenced by the type of regeneration media. MSRK5SS-Pr regeneration mediun, which contains 5 mgL$^{-1}$ kinetin, 2% (w/v) sucrose in combination with 3% (w/v) sorbitol, and 500 mgL$^{-1}$ proline, was efficient to produce transgenic plant of rice from putative transgenic callus in the presence of L-phosphinotricin (PPT). With MSRK5SS-Pr medium, transformation efficincies of Nagdongbyeo were significantly enhanced from 3.7% to 6.3% in independent callus lines arid from 7.3% to 19.7% in plants produced, respectively. Stable integration and expression of bar gene were confirmed by basta herbicide assay, PCR amplification and Southern blotting of bar gene, and fluorescence in situ hybridization (FISH) analysis using pSBM-PPGN as a probe. In Southern blot analysis, diverse band patterns were observed in total 44 transgenic plants regenerated from 20 independent PPT resistant calli showing from one to five copies of T-DNA segments, however, the transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.
To establish the method of integrated weed management in rice nurserybed, phytotoxic factors of herbicide and antidote efficacy were evaluated at the Yeongnam Crop Experiment Station in 1982 and 1984. Seven hundred eighty four research items were carried out as weed control research since 1961. While 65% of these were belonged to rice research, only 6% was attributed to nurserybed among rice research. More herbicidal phytotoxicity exhibited when seedbed was pressed just after seeding than sand covered or uncovered seedbed and also this phytotoxic symptom enhanced by using intact seed compared to pregerminated seed. Rinsing practice of seedbed reduced the phytotoxic effect of butachlor and this effect was more pronounced with the number of rinsing operation increase and at the pressed plot. However, herbicidal efficacy was not significantly decreased by rinsing operation. Growth of rice seedling hardly affected where the herbicide was absorbed through root only compared to absorption from both of root and shoot for pyrazolate, butachlor and thiobencarb. Herbicide antidote `CGA 123'407' completely protected from the phytotoxic effect of pretilachlor without arising any adversal effect in weed control. However, without antidote, pretilachlor showed the most severe phytotoxic symptom among used herbicides.
Rice plants were grown for 42 days in the specially made micro-ecosystem(pot) containing two different soils treated with fresh and 60-day-aged residues of [$^{14}C$]quinclorac, respectively, to elucidate the behaviour of the herbicide quinclorac residues in the soils. Amounts of $^{14}CO_{2}$ evolved from two soils treated with different residues with and without vegetation were all less than 2.2% of the total $^{14}C$, indicating that there was little microbial degradation of quinclorac in soil. $^{14}C$-Radioactivity absorbed and translocated into rice plants from soil A and B containing fresh quinclorac residues was 8.4 and 24.2%, respectively, of the originally applied $^{14}C$, while 5.5 and 17.7%, in aged residue soils. These results indicate that larger amounts of $^{14}C$ were absorbed by rice plants from soil B with less organic matter and clay than soil A, and the uptake of [$^{14}C$]quinclorac and its degradation products decreased with aging in soil. After 42 days of rice growing, 84.5 and 61.8% of the $^{14}C$ applied freshly to soil A and B, respectively, remained in soil, whereas, in the case of aged soils, 86.3 and 67.7% of the $^{14}C$ applied did. Meanwhile, without vegetation, more than 98.3% of the $^{14}C$ applied, in both fresh and aged residues, remained in soil, suggesting that quinclorac was relatively persistent chemically and microbiologically. Most of the non-extractable soil-bound residues of [$^{14}C$]quinclorac were incorporated into the organic matter and largely distributed in the fulvic acid portion.
The research was carried out in tray ($0.12m^2$) in greenhouse ($30{\pm}2^{\circ}C$${\sim}$$25{\pm}2^{\circ}C$) equipped with Auto Foggy System (SAE KI RIN Co.) and results are summarized as follows: 1. Bifenox and bensulfuron retarded the growth of rice under foggy condition and the weeding efficacy among the herbicides tested was not different. 2. Weeds under foggy condition emerged at smaller amounts than under the non-foggy condition. bensulfuron, molinate and bifenox showed the high weeding efficacy. There was a tendency that molinate reduced the number of tillers at 20 and 60 days after transplanting. 3. Among the herbicides tested, only the molinate reduced the dry weight and yield of rice significantly.

Technology development for sufficient dry matter production of Chinese milk vetch (CMV) is most important in CMV-rice cultivation system in order to provide sufficient nutrients to rice plants. However, when the CMV plants are dominated by the weed, especially Alopecurus aequalis, the CMV growth could be reduced due to light and nutrient competition. In addition, A. aequalis is potential host of the rice dwarf virus disease. Therefore, control of A. aequalis is necessary to enhance the biomass production of CMV plants when CMV stands are insufficient. The use of chemical like sethoxydim (20%, ai) showed the highest control rate of 84% at early stage and was reduced as application was delayed. A. aequalis control did not change the CMV seedling stand before and after herbicide treatment and the reseeding stand in fall was rather increased 2.2 to 2.6 times. On the other hand, in untreated control, the CMV stand at May 15 and reseeding stand in fall was significantly reduced as compared with the before herbicide treatment. Control of A. aequalis increased the CMV dry matter production by 164% for 50% CMV coverage rate and 63% for 25% CMV coverage rate. This is equivalent to $12.3{\sim}16.4\;kgN$/10a which is greater than the recommended nitrogen rate of 9kg/10a. The result indicates that the control of A. aequalis is an efficient way to enhance dry matter production in CMV-rice cultivation system especially when CMV stand is poor.
The response of rice (Oryza sativa L.) cultivars to mixtures of 2, 4-D(2, 4-dichlorophenoxyacetic acid) and other herbicides was investigated to determine if there was an interaction between them. When 2, 4-D was applied, shoot growth of Taipei 309 was more affected than that of IR28 at all concentrations used. In contrast, when thiobencarb (S-[(4-chlorophenyl)methyl]diethylcarbamothioate), butachlor [N-(buthoxymethyl) -2-chloro-N-(2, 6-diethylphenyl) acetamide], and glyphosate [N-(phosphonomethyl)glycine] were applied, the shoot growth of Taipei 309 was less affected than that of IR28 at all herbicide concentrations. Combination of 2, 4-D and the lowest thiobencarb concentration was antagonistic for shoot length for both cultivars, but at higher concentrations, it was synergistic. Synergism for shoot fresh weight between 2, 4-D and thiobencarb was observed with IR28 at all concentrations but, for Taipei 309, synergism was observed only at lower 2, 4-D concentrations. Mixing 2. 4-D with butachlor resulted in greater inhibition in shoot length and fresh weight of IR28 than Taipei 309 at all concentrations indicating a synergistic interaction. With combinations of 2, 4-D and glyphosate, an antagonistic interaction for shoot length was observed for both cultivars. A synergistic interaction for shoot fresh weight was observed with IR28 when combinations of the highest concentration of glyphosate and 2, 4-D were applied but there was an antagonistic interaction with Taipei 309.
Various herbicides were applied to the young rice seedling from which endosperm were removed and their effects were determined at 40 days after transplanting. Generally, growth was retarded, but the number of tillers was not effected. Application of bensulfuron and pretilachlor showed slight growth inhibition compared to dimepiperate and pyrazolate. Injury due to herbicide application was dependent on the transplanting depth. Injury was severe when pretilachlor and pyrazolate were applied to rice transplanted 0 and 4cm deep, respectively. However, bensulfuron showed much injury regardless of transplanting depths with the tendency of general growth retardation. Dimepiperate was fairly safe at all transplanting depths. The application of herbicides at 3 and 6 DAT showed higher injury than that at g and 12 DAT. The degree of injury was severe when pretilachlor and bensulfuron were applied.
This study was conducted to evaluate the susceptibility of rice(Oryza sativa L.) cultivars to herbicide thiobencarb through the determination of protein patterns using SDS-PAGE. In both greenhouse and laboratory screening tests, IR 10198-66-2 and IR 9660-50-3-1 showed relatively tolerant response to thiobencarb, while IR 22, IR 31802-48-2-2-2 and IR 20656-R-R-R-6-1 were susceptible to it. The total protein content of susceptible cultivars markedly decreased as the thiobencarb concentrations increased, but tolerant cultivars such as IR 10198-66-2 and IR 9660-50-3-1 showed very slight changes suggesting that protein synthesis of susceptible cultivars may be inhibited by thiobencarb. The protein profiles of the tolerant cultivars were not much affected by thiobencarb treatment. However, the protein spots with molecular weights of 14.4 kD and 55 kD in the susceptible cultivar of IR 22 disappeared with the treatment of 3 ppm thiobencarb, indicating that differential susceptibilities of rice cultivars against thiobencarb can be attributed to their difference in protein metabolism affected by thiobencarb.
