Magnaporthe grisea, a causal agent of blast, forms a specialized infection structure, an appressorium, to infect host. Hydrophobicity of contact surface and cAMP have been suggested as a primary environmental signal and a second messenger to trigger and mediate appressorium formation in this fungus, respectively. To generalize these factors in field isolates of M. girsea, twenty isolates originated from rice and other gramineous hosts were tested. Seventeen including rice and non-rice isolates formed appressoria on hydrophobic surface, but none of isolates formed appressoria on hydrophilic surface. Eighteen isolates formed appressoria on hydrophilic surface in the presence of IBMX, an inhibitor of phosphodiesterase, except two rice isolates. These results strongly support the hypothesis that appressorium formation by M. grisea is induced by hydrophobic hard surface and regulated by the endogenous level of cAMP in the cells. Understanding fungal development is not only of biological interest but provides new targets for novel disease control strategies.
The genome of the rice blast fungus Magnaporthe oryzae contains several types of transposable elements (TEs), and some TEs cause genetic variation that allows M. oryzae to evade host detection. We studied how five abundant TEs in rice pathogens, Pot3, Pot2, MAGGY, Line-like element (MGL) and Mg-SINE, are expressed under diverse conditions related to growth, development, and stress. Expression of Pot3 and Pot2 was activated in germinated conidia and mycelia treated with tricyclazole. Retrotransposon MAGGY was highly expressed in appressoria and tricyclazole-treated mycelia. MAGGY and Pot2 were also activated during the early and late stages of perithecia development. MGL was up-regulated in conidia and during conidial germination but not during appressorium formation. No noticeable expression of Mg-SINE was observed under most conditions. Our results should help investigate if and how condition-specific expressions of some TEs contribute to the biology and evolution of M. oryzae.
Fungicide treatment is the most important method for the control of plant diseases caused by phytopathogenic fungi. But fungicide resistant strains have appeared in many phytopathogenic fungi. Until now, molecular mechanisms of fungicide resistance such as mutation of target protein, overproduction of target enzyme and detoxification of fungicide have been designated. Recently, it was demonstrated that active efflux of fungicides mediated by ATP-binding cassette (ABC) transporters also contributes to fungicide resistance in several filamentous fungi, such as Aspergillus nidulans, Penicillium digitatum and Botrytis cinerea.(중략)

Previously, we have reported a plant extract isolated from Lysimachia foenum gracum Herba as a new environment friendly biopesticide that has the mycelial growth inhibition effect on Magnaporthe oryzae, the pathogenic fungus of the rice blast disease. For the finding of additional biopesticide candidate, we tested the mycelial growth inhibitory effects about 700 species of plant extracts on PDA media. Among them, the extract of Anemarrhena asphodeloides showed prominent inhibitory effect of which $IC_{50}$ was $139.7{\mu}g/ml$. Mycelial radii of M. oryzae were measured on PDA medium containing the four organic solvent fractions isolated from total extract from A. asphodeloides. Ethyl acetate fraction showed the impressive inhibitory effect of $IC_{50}$, $54.12{\mu}g/ml$. In the subsequent rice field test for the total extract of A. asphodeloides, we obtained encouraging 62.0% control rate of rice blast disease without any phytotoxicity. It is almost equivalent to that of chemical pesticides implying the applicability of the extract as a new biopesticide. In further study, the analysis of active ingredients of the extract would be necessary for the development of a new biopesticide and for the verification of cellular mechanism by which the mycelial growth of M. oryzae inhibited.
Plant disease resistance occurs as a hypersensitive response (HR) at the site of attempted pathogen invasion. This specific event is initiated in response to recognition of pathogen-associated molecular pattern (PAMP) and subsequent PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI). Both PTI and ETI mechanisms are tightly connected with reactive oxygen species (ROS) production and disease resistance that involves distinct biphasic ROS production as one of its pivotal plant immune responses. This unique oxidative burst is strongly dependent on the resistant cultivars because a monophasic ROS burst is a hallmark of the susceptible cultivars. However, the cause of the differential ROS burst remains unknown. In the study here, we revealed the plausible underlying mechanism of the differential ROS burst through functional understanding of the Magnaporthe oryzae (M. oryzae) AVR effector, AVR-Pii. We performed yeast two-hybrid (Y2H) screening using AVR-Pii as bait and isolated rice NADP-malic enzyme2 (Os-NADP-ME2) as the rice target protein. To our surprise, deletion of the rice Os-NADP-ME2 gene in a resistant rice cultivar disrupted innate immunity against the rice blast fungus. Malic enzyme activity and inhibition studies demonstrated that AVR-Pii proteins specifically inhibit in vitro NADP-ME activity. Overall, we demonstrate that rice blast fungus, M. oryzae attenuates the host ROS burst via AVR-Pii-mediated inhibition of Os-NADP-ME2, which is indispensable in ROS metabolism for the innate immunity of rice. This characterization of the regulation of the host oxidative burst will help to elucidate how the products of AVR genes function associated with virulence of the pathogen.
한국(韓國)에서 농업관계연구(農業關係硏究) 기관(機關)에 의해 벼도열병(病) 방제(防除)에 관(關)한 실질적(實質的)인 연구(硏究)가 시작(始作)된 것은 전북일부지방(全北一部地方)에 목도열병(病)이 격발(激發)한 다음해인 1927년(年)부터였다. 과거 55년간(年間)에 걸쳐 많은 수도관계학자(水稻關係學者), 특(特)히 식물병리학자(植物病理學者)들의 노력(努力)에도 불구하고 도열병(病)은 아직도 가장 피해(被害)가 심(甚)하고 광범위(廣範圍)하게 발생(發生)하고 있는 병(病)중의 하나이다. 벼도열병(病)은 병원균(病原菌)의 특이(特異)한 병원성(病原性)의 변이(變異)때문에 방제(防除)가 매우 어렵다. 지난 70 년 동안의 병발생(病發生) 추이조사(推移調査)에서 도열병(病)의 발생(發生) 양상(樣相)에는 잎도열병형(病型), 목도열병형(病型), 지속적(持續的)인 발생형(發生型)의 3가지 각기 다른 발생(發生) 양상(樣相)이 존재(存在)함을 알았다. 일반적(一般的)으로 도열병(病) 발생(發生)은 잎도열병(病) 발생(發生)은 적어도 목도열병(病)은 심(甚)한 소위 잎도열병(病)과 목도열병(病)의 불연속현상(不連續現象)이 존재(存在)하는 특징(特徵)이 있다고 한다. 따라서 수량(收量)에 직접 영향(影響)을 미치는 목도열병(病)의 방제(防除)에 주의를 기울여야 한다. 과거 70 년 동안의 도열병(病)에 관(關)한 광범(廣範)한 조사(調査)와 연구(硏究)에서 도열병(病) 발생(發生)을 야기(惹起)시키는 주요인(主要因)은 병진항성(病振抗性)의 역전(逆轉), 질소질비료(窒素質肥料)의 과용(過用)과 만식(晩植), 그리고 지속적(持續的)인 강우(降雨)등으로 밝혀졌다. 그동안 한국(韓國)에서는 주(主)로 그룹 A에 속하는 곡양도(穀良都), 다마금(多摩錦), 은방주(銀坊主), 풍옥(豊玉) 둥 일본형(日本型) 수도품종(水稻品種)이 30 년이상(年以上) 재배(栽培)되어 왔다는 사실이 학자(學者)들에 의해 주목(注目)되어 왔다. 도열병(病)의 경우 동일(同一)그룹내(內) 품종간(品種間)의 포장지항성 차이(差異)는 매우 적으나 때로는 역학적(疫學的)으로 상당한 차이가 발견(發見)되기도 해서 도열병(病)의 성공적(成功的)인 방제(防除)를 위해서는 포장지항성 연구(硏究)에 보다 많은 관심을 기울여야 하겠다. IR 계통(系統) 품종(品種)들에 대(對)해 질소비료과용(窒素肥料過用)은 잎도열병(病)보다 목도열병(病) 발생(發生)에 더 조장(助長)하며 질소비료(窒素肥料) 소량시비시(少量施肥時)에도 IR 계통(系統) 품종(品種)들의 목도열병(病) 발생(發生) 비교적 심(甚)했고 도열병(病) 발생(發生)에 대(對)한 저온(低溫)의 영향(影響)은 매우커서 잎도열병반(病斑)의 전염능력(傳染能力)과 수잉기(穗孕期)의 엽초내 목도열병(病)의 감염(感染)을 현저히 증가(增加)시켰다. 출수전(出穗前) 3회(回), 출수후(出穗後) 2회(回)의 약제살포(藥劑撒布)가 도열병방제(病防除)에 가장 효과적(效果的)인 것으로 추정(推定)되었으며 도열병(病)에 대(對)해 완벽(完壁)한 단독(單獨) 방제방법(防除方法)은 없고 1950년경에 작성(作成)된 혼합방제법(混合防除法)이 수도(水稻)가 재배(栽培)되는 한 가장 효과적(效果的)일 것이다.
In order to study disease resistance mechanisms in rice against the rice blast fungus Magnaporthe grisea, we screened fungal elicitor-responsive genes from rice suspension-cultured cells treated with fungal elicitors employing differential hybridization (DH). By DH screening, 31 distinct rice clones were isolated and a majority of them were full-length cDNAs encoding pathogenesisrelated (PR) genes. Sixteen of the 31 genes were upregulated at 4, 8, and 12 h following fungal elicitor treatment. To elucidate the effect of signal molecules and biotic elicitors on the regulation of rice defense genes, we further characterized the transcriptional expression patterns of representative isolated PR genes; OsGlu1, OsGlu2, OsTLP, OsRLK, and OsPR-10, following treatment with fungal elicitor, phytohormones, cycloheximide, and inhibitors of protein phosphorylation. Jasmonic acid (JA) induced transcriptional expression of OsGlu1, OsTLP, and OsRLK, but not of OsGlu2 and OsPR-10 at any of the tested time points. Salicylic acid (SA) and abscisic acid weakly induced the expression of OsTLP and OsRLK. SA showed an antagonistic effect with fungal elicitor and JA. Cycloheximide suppressed all these genes upon elicitor treatment, except for OsGlu2. Staurosporine only induced the expression of OsRLK. Application of calyculin A strongly induced OsRLK expression, but suppressed the expression of OsGlu2. Our study yielded a number of PR genes that play a role in defense mechanisms against the rice blast fungus, as well as contribute towards the elucidation of crosstalk between phytohormones and other modifications during defense signaling.
